2.Breeding Actinobacillus succinogenes with Acid-tolerance by Genome Shuffling

A strain Actinobacillus succinogenes CGMCC 1593 was selected as the parent strain.After UV-EMS and UV-DES treatments respectively,seven mutated strains with subtle improvements in acid tol-erance were obtained,and were subjected for recursive protoplast fusion.Through three rounds of genome shuffling,four shuffled strains with both higher yield and acid tolerance were obtained.The shuffled strain namely F3-21 could even survive at pH 5.2.The comparison of the shuffled strains and the parent strain for succinic acid production was also studied here.After 48 h of shake-flask fermentation,the succinic acid concentration of F3-21 was 48% higher than that of the parent strain.When F3-21 was carried out in a 5 liter stirred bioreactor with pH controlled 5.6~6.0,the accumulation of succinic acid in 48 h fermentation attained 38.1 g/L,which was increased by 45% compared with that of the parent strain(26.2 g/L).While pH was controlled at 6.5~7.0,the production of succinic acid in 32 h fermentation attained 40.7 g/L.When F3-21 was carried out in fed-batch fermentation,succinic acid concentration of 67.4 g/L was reached in 72 h fer-mentation.These results indicated that the genome shuffling could improve the acid tolerance and the suc-cinic acid production of A.succinogenes CGMCC 1593.
3.The experimental study on effect of the spinal neuron flow with the nerve repair time

Objective To measure Ihe effect on rats spinal neuron flow according nerve roots repair time.Methods We adopted the experimental rats on the root avulsion and extravertebral foramen nerve root divison of C_(5~7).We divided them into four groupsin each which there were 16 ratsaccording the type of nerves root injury and repair timeGroup AC:the avulsed roots were reimplanted into the spinal cord and the transeeted roots were sutured to the proximal stump immediately.Group B,Dthe avulsed roots and the transected roots were reimplanted into the spinal cord or were sutured to the proximal stump in delayed 3 weeks each with 16 rats.At the different time point(3 weeks3 months6 months)through pathological examina- tion and immunohistological lechniques and nerve tracing techniqueswe examined the spinal cord and distal nerve trunk in order to observe the pathologic changes and axonal regeneration.Results Group A、C were much better than group B、D in the numberthe conformation and the degree of abatement of spinal motoneu- rons and nissl body.It is the same on the number and the development level of regenerating nerve fiber. Conclusion It had the advantage of neuronal protection and nerve regeneration that reparing the injured nerve roots earlv after nerve roots injury.
4.Arthroscopic rotator cuff repair using a suture-bridge technique in late middle-aged adults.

OBJECTIVETo evaluate the clinical results after arthroscopic rotator cuff repair using a suture-bridge technique in late middle-aged adults.
METHODSA retrospective analysis of 30 patients with rotator cuff tears, who were treated consecutively between December 2012 and December 2013 at our clinic, was carried out to evaluate the outcome of arthroscopic suture-bridge technique. The visual analogue scale (VAS), the range of motion (ROM), the rating scale of the University of California at Los Angeles (UCLA), and the shoulder index of the American Shoulder and Elbow Surgeons (ASES) were determined as the evaluation criteria.
RESULTSThe mean follow-up period was 13 months. The follow-up rate was 100%. The study included 0 small, 16 medium, 12 large and 2 massive rotator cuff tears. At the latest follow-up, the VAS score decreased significantly from preoperative 6.3±3.2 to 0.8±1.4 at rest,and from preoperative 7.7±2.1 to 1.7±1.6 during motion. The UCLA score improved significantly from 15.1±4.6 preoperatively to 31.2±5.0 at follow-up. The ASES shoulder index improved significantly from 30.2±16.0 preoperatively to 80.4±13.0 at follow-up. The ROM improved significantly. The satisfactory rate was 94%. There were no complications.
CONCLUSIONThe results of this study indicate that the arthroscopic suture-bridge technique is an effective and reliable treatment for patients with rotator cuff tear in late middle-aged adults.

5.Literature analysis of electroacupuncture stimulation index on the treatment of sciatica.

OBJECTIVETo summarize the main electroacupuncture stimulation indexes used in clinical treatment and experimental study on sciatica, and to investigate the optimal stimulation index for sciatica treatment with electroacupuncture.
METHODSThe data was obtained on line from the computerized general and specialized databases such as CBM, VIP, CNKI, and MEDLINE. Data analysis was focused on the different effects of electroacupuncture with different stimulation indexes.
RESULTS(Clinical research suggested that the commonly used electroacupuncture stimulation indexes included frequency of 1-20 Hz, electric current of 0.2-0.8 mA, continuous and dilatational waveform. The intensity of stimulation was endurable for patients. (2) The frequency of electroacupuncture used for animal experimental research was mainly concentrated among 2-20 Hz, which 2 Hz and 5 Hz had better effects for sciatica treatment with electroacupuncture. The scale of electric current was 0. 4-10 mA with intermittent waveform. The intensity of stimulation was decided by a visible muscle contraction occurred during the acupuncture.
CONCLUSIONDifferent kinds of electroacupuncture stimulation indexes have different effects on the treatment of sciatica. Therefore, it is still needed the more evidence from higher quality clinical trials to investigate the optimal electroacupuncture stimulation indexes on the treatment of sciatica.

6.Effects of Fructose,Glucose and Exogenous Nitric Oxide Donor Sodium Nitroprusside on Germination of Psychotria rubra(Lour.)Poir. Seeds Under Drought Stress

Objective To observe the effect of fructose(Fru),glucose (Glc)and exogenous nitric oxide donor sodium nitroprusside(SNP)on the germination of Psychotria rubra(Lour.)Poir. seeds under drought stress,and to explore the optimal concentration for clearing the drought stress. Methods We established the drought stress model of Psychotria rubra(Lour.)Poir. seeds with 15%(volume fraction)PEG6000 and 0.4 mol/L D-mannitol, measured the germination rate and germination index of Psychotria rubra (Lour.)Poir. seeds under drought stress combined treatment with different concentrations of Fru (0,300,500 μmol/L),Glc (0,300,500 μmol/L) and SNP(0 , 100 , 200 μmol/L). Results The germination process of Psychotria rubra(Lour .) Poir . seeds was significantly suppressed by drought stress. Fru,Glc and SNP could significantly promote the germination of Psychotria rubra (Lour.) Poir. seeds under drought stress. The optimal condition for clearing drought stress induced by 15% PEG6000 was Fru 300 μmol/L, Glc 500 μmol/Land SNP 200 μmol/L, and the germination rate was 95% under the optimal condition . The optimal condition for clearing drought stress induced by D-Mannitol was Fru 500 μmol/L, Glc 500 μmol/L and SNP 100 μmol/L, and the germination rate arrived to100%. Conclusion Fru,Glc and SNP can significantly alleviate the negative impact induced by drought stress for the Psychotria rubra(Lour.)Poir. seed germination,improve the drought-resistant ability of the seeds.
7.The immunological effects of three doses of a live attenuated hepatitis A vaccine (H2 strain) in 8 years.

OBJECTIVETo observe the immunological effects of three doses of H2 strain live attenuated hepatitis A vaccine 8 years after the administration and to compare with that of one dose of the vaccine.
METHODSIn a country area, 110 children of 1 to 7 years old susceptible to HAV were screened and administered with one dose of the vaccine, as group B; Group A were 42 children from one of the villages and administered with 3 doses of the vaccine according to 0, 2, 6 month schedule. Blood samples were taken for the children 1, 2, 6, 7, 8, 12, 24, 36 and 96 months after the administrations respectively and detected for anti-HAV antibody.
RESULTSFor group B, the sero conversion rate of anti-HAV and GMC reached peak at 92.2% and 126.2 mIU/ml respectively, and then, began to drop with time; For group A, after 2 dose of the vaccine, the sero-conversion rate reached 100%, and the GMC reached peak of 2 739 mIU/ml one month after the third dose at 7 months. So that, group A has a better short-term immunological effects than that of group B. During 36 through 96 months, the anti-HAV positive rate in group B was 75%-71% and 80-89 mIU/ml respectively, and comparatively in group A were 100% and 918.2-480.6 mIU/ml respectively. The differences between group A and B were significantly important.
CONCLUSIONA 3-dose schedule administration of H2 strain live attenuated hepatitis A vaccine has better immunological effects than 1-dose schedule in 8years and further observations are needed.

8.Study on relationship between length of homologous sequences and chromosomic recombination rate in Saccharopolyspora erythraea.

In order to study the relationship between lengths of homologous fragments and chromsomic recombination rate in Saccharopolyspora erythraea, three homologous sequences, with mutant loci and different flanking sequences, (26bp + 27bp), (500bp + 576bp) and (1908bp + 1749bp), were synthesized by chemical reaction or PCR amplification, and cloned into pWHM3 to construct homologous recombination plasmids, pWHM1113, pWHM1116 and pWHM1119. When the plasmids were transformed into protoplast of Saccharopolyspora erythraea A226 under PEG mediated, on an average 30, 69 and 170 transformants grew on each plate for the three plasmids respectively, but chromosomic integration frequency were 0, 2% and 19% among corresponding transformants. Both pWHM1116 and pWHM1119 could take double crossover recombination, and exchange the mutant loci in the chromosome. It was concluded that when the flanking sequences were equal or more than (500bp + 576bp), they could take effective single and double recombination with Saccharopolyspora erythraea chromosome.

9.Magnetic resonance imaging for the wrist joint of the coal miners in vibration department.

OBJECTIVETo study the magnetic resonance imaging (MRI) in the wrist joint of coal miners who work in excavation and vibration department.
METHODSForty-three coal miners with the hand-arm vibration disease served as the observation group while 20 workers who were not working in the vibration department acted as the control group. The patients in the observation group were divided into five subgroups according to the time when they received vibration. The regularity of the development of signs and symptoms of MRI was observed and analyzed.
RESULTSThe hydroarthrosis was most found in MRI. There were significant difference in hydroarthrosis (chi(2) = 8.80, P < 0.01), osteoporosis and osteomyelitis (chi(2) = 3.91, chi(2) = 5.01, P < 0.05 respectively) between the observation group and the control group. The edema of bone marrow and the avascular necrosis of ossa carpi were found only in the observation group and not found in the control group. The hydroarthrosis and the edema of bone marrow occurred most in the early stage of vibration. The signal in the edema of the bone marrow of the distal end of the radius was decreased in the GE sequence T(2)WI with the specificity.
CONCLUSION(1) Changes in the wrist joint occur in the early stage of the vibration work, and can be found in the MRI. (2) The edema of the bone marrow of the distal end of the radius is of great value in the diagnosis of the hand-arm vibration disease.

10.Effects of acupuncture on blood-lipid levels in the patient of cerebral infarction with hyperlipidemia.

OBJECTIVETo observe therapeutic effect of acupuncture method for consciousness-restoring resuscitation on cerebral infarction with hyperlipidemia.
METHODSSixty cases of apoplexy with hyperlipidemia were randomly divided into a consciousness-restoring resuscitation acupuncture group (treatment group) and a Chinese medicine control group (control group), 30 cases in each group. TCM symptomatic scores and indexes of blood lipids were detected before and after treatment in the two groups.
RESULTSThe total effective rate for improvement of blood lipids was 72.4% in the treatment group, which was not different from that (65.5%) in the control group (P> 0.05); the total effective rate for clinical symptoms was 89.7% in the treatment group, which was better than that (62.1%) in the control group (P < 0.05). The treatment group was superior to the control group in improvement of numbness and twinge in extremities, dizziness, distention and oppressed feeling in chest and hypochondrium, anorexia, sleepy and weakness (P < 0.05).
CONCLUSIONAcupuncture method for consciousness-restoring resuscitation can regulate metabolic disorder of lipids and improve main clinical symptoms in the patient of cerebral infarction with hyperlipidemia.
