1.The Screening of Bifidobacterium from Macrobian

Using colon adenocarcinoma cell line HT-29 as testing material, adhesion test was carried out for 24 strains of Bifidobacteria isolated from dejecta of longevous people aged one hundred years. The results showed that all strains could adhere to colon adenocarcinoma cells HT-29, among which the strains of TTF, Z2, TZ5 and (J-1) showed higher adhesive capability. Thus these four strains of Bifidobacteria were further tested for their endurance to stomach acid and bile acid. The production of vitamin B by these bacterial were also tested. All these tests indicated that the strain of TTF has high ability to produce Vitamin B_(1), B_(2), B_(6), B_(12), The survival rate after the treatment under pH 3.0 for 120 min was 93.11%, at the same time, the strain TTF can survive after being treated with 2% bile acid for 24h.
2.Discriptive systematic review for the application of problem-based learning in education of health service management

Objective To systematically evaluate the application of problem-based learning (PBL)in the teaching process of health management major in China.Methods Databases including CNKI (1979 to December the 2015),VIP (1989 to December the 2015),Wanfang (1982 to December the 2015) and PubMed were systematically retrieved.Any literature about PBL of health management major was included.Seffdeveloped data extraction form was used for collecting the information.Data were input and analyzed using Excel 2007.Results 15 papers were included in the analysis.The first author mainly came from the northeast region of China (7).Most (10) papers did not get the funding support.No paper was published in the journals contained in the Guide to the Core Journals of China.6 papers of experimental studies compared the performance difference of students of the PBL and lecture-based learning (LBL),which was statistically significant.Conclusion PBL is superior to the LBL.However,due to the large difference in the quality of literature,more studies were needed to determine the effect of PBL.Moreover,we should pay attention to the combination of PBL and LBL.
3.Effect of Hexue mingmu tablets in the treatment of hyphema

To evaluate the effect of Hexue mingmu tablets on traumatic hyphema caused by blunt ocular trauma.
●METHODS: Totally 150 patients of traumatic hyphema were divided into seven types by using ultrasound biomicroscopy combining with anterior segment abnormalities, each type was randomly classified as trial group and control group. The trial group was administered Hexue mingmu tablets, control group was treated by hemocoagulase.
●RESULTS: The absorbing time of trial group was shorter than that of the control group. And there was statistical significance between the two groups (P<0. 05).
● CONCLUSlON: Hexue mingmu tablets is an effective medicine to treat traumatic hyphema. Ultrasound biomicroscopy can be used as a routine examination method in traumatic hyphema.
4.A clinical analysis of sleep-related breathing disorders in patients with idiopathic pulmonary fibrosis

Objective To observe the incidence and clinical feature of sleep-related breathing disorder in patients with idiopathic pulmonary fibrosis (IPF). Methods Thirty-four IPF patients who were measured by polysomnography (PSG) were collected in the Department of Respiration of Tianjin Medical University General Hospital. According to the results of apnea hypoventilation index (AHI), patients were divided into pure IPF group (AHI<5 events/h, n=7) and IPF combined with obstructive sleep apnea hypopnea syndrome (IPF+OSAHS) group (AHI≥5 events/h, n=27). The PSG reports of two groups were analyzed, and the correlation between AHI and pulmonary function and oxygen saturation in sleep and at wake were analyzed. Results (1)Thirty-four IPF patients were all demonstrated sleep disorders, low sleep efficiency, increased proportion of stageⅠand stageⅡand decreased proportion of stageⅢand rapid eye movement (REM). The arousal index and the proportion of stageⅠand stageⅡwere higher in IPF+OSAHS group than those of pure IPF group (P<0.01), while the proportion of stageⅢwas lower in IPF+OSAHS group than that of pure IPF group (P<0.01). There was no significant difference in stage REM between two groups. (2)Twenty-seven patients (79%) combined with OSAHS, among which five subjects (15%) were mild OSAHS with 5 events/h≤AHI<15 events/h, and 22 subjects (65%) were moderate-severe with AHI≥15 events/h. The main type of sleep-disorder breathing was hypoventilation, which mainly happened in stage REM. (3) Thirty-four IPF patients showed sleep hypoxemia, and the oxygen desaturation index (ODI) was higher in IPF-OSAHS group than those of pure IPF group (P<0.05). (4)The AHI was positively correlated with body mass index (r=0.791, P<0.05), and was negatively correlated with forced vital capacity (FVC%pred) (r=-0.574, P<0.05) and forced expiratory volume in 1 second (FEV1%pred) in IPF patients (r=-0.664, P<0.05). The lowest oxygen saturation (LSO2) and mean oxygen saturation (MSO2) in sleep were positively related with oxygen saturation at wake (r=0.421 and r=0.464, P<0.05 respectively). Conclusion The IPF patients show severe sleep disorder and hypoxemia, which can be worsen by OSAHS and produce negative effect on daily life. We should initiate active treatment in patients with sleep-related breathing disorders.

6.Studies on Shedding Virus and the Formation of Aerosol of H_9N_2 Subtype AIV

In order to study the regularity of shedding virus from infected SPF chickens and the formation of aerosol of H9N2 subtype AIV, SPF chickens were bred in a positive and negative pressure isolator. Aerosol samples were collected by AGI-30 (All Glass Impinger-30) extractor, and simultaneously trachea and cloaca samples were collected by tracheal swabs and cloacal swabs in different periods after challenged with vi- ruses. The above-mentioned samples were detected by HI, Dot-ELISA and RT-PCR methods. The results in- dicated that aerosols were isolated from the 4 days to the 43 days after inoculation. It was proved that H9N2 subtype AIV could copy themselves in respiratory passage and cloaca, and then could formation of aerosols. AIV H9N2 subtype could be isolated from cloacal and tracheal swabs 3 days after inoculation and lasted for 45 days, viruses were detected from all infected SPF chickens on 7 days.
7.Effects of Genistein on the proliferation and expression of survivin in salivary adenoid cystic carcinoma cell line SACC-83.

OBJECTIVETo investigate the anti-proliferation effect of tyrosine protein kinase inhibitor, Genistein, on human salivary adenoid cystic carcinoma cell line SACC-83, and its effect on Survivin expression.
METHODSSACC-83 cells were treated with different concentration Genistein for different time, cell survival rate was calculated with MTT assay, apoptosis was detected with flow cytometry, the expression of Survivin was quantitatively analyzed by Western blotting and FluorChem V2.0 software.
RESULTSWhen treated with Genistein of certain concentration for certain time, SACC-83 cell growth was significantly inhibited. With the increase of concentration and elongation of acting time, the inhibitory effects increase. Treated with 220 micromol/L Genistein for 72 hours, SACC-83 cell growth was significantly inhibited, cell apoptosis was induced (P < 0.01), and the expression of Survivin decreased.
CONCLUSIONGenistein inhibits the growth of human salivary adenoid cystic carcinoma cell line SACC-83, and induces cell apoptosis; the decrease of Survivin expression may be one of the mechanisms of Genistein inducing apoptosis.

8.Changes in subcortical white matter and corpus callosum volumes in patients with type 2 diabetes mellitus.

OBJECTIVETo evaluate the changes in subcortical white matter and corpus callosum volumes in patients with type 2 diabetes mellitus (T2DM).
METHODSHigh resolution three-dimensional T1-weighted fast spoiled gradient recalled echo magnetic resonance images were obtained from 16 T2DM patients and 16 normal controls, and 11 T2DM patients also received the same magnetic resonance imaging scans after insulin therapy for 1 year. Volumetric analysis was performed and analysis of covariance and paired t test were applied.
RESULTSA lower volume was demonstrated in T2DM patients than that in the normal controls in bilateral caudal middle frontal lobe, bilateral pars opercularis, right inferior parietal lobe, right inferior temporal lobe, right middle temporal lobe, and left transverse temporal lobe (all P<0.05). After insulin therapy for 1 year, the increased subcortical white matter of brain regions were demonstrated in left medial orbitofrontal lobe, right caudal middle frontal lobe, right inferior parietal lobe, right lingual lobe, right inferior temporal lobe, right middle temporal lobe, left transverse temporal lobe, left entorhinal lobe and left insular lobe at the follow-up level in T2DM patients compared with baseline level (P<0.05). The decreased volumes of subcortical white matter were observed in the bilateral paracentral lobe, right superior forntal lobe and right precuneus lobe in T2DM patients on insulin therapy for 1 year (P<0.05). A decreased trend of subcortical white matter was involved in corpus callosum (P>0.05) and a significant decreased volume was demonstrated in the middle posterior part of corpus callosum after insulin therapy for 1 year(P<0.05).
CONCLUSIONST2DM patients have decreased subcortical white matter of multiple brain regions, and the atrophy of subcortical white matter can be improved by the insulin therapy. The volume analysis of subcortical white matter is a simple and effective tool for the evaluation of white matter damage in T2DM patients.

9.Preliminary study of clinical significance of decreased D(L)CO in patients with left ventricular heart failure.

OBJECTIVEThis study aimed to investigate the feature of D(L)CO (Diffusion Lung Capacity for Carbon Monoxide) in CHF (left ventricular heart failure) patients, underlying pathophysiological mechanism and clinical significance.
METHODSWe retrospectively studied the D(L)CO, pulmonary ventilation function, cardiopulmonary exercise testing and related clinical information in severer HF patients.
RESULTSPeak VO2 severely decreased to 34 ± 7 percentage of predicted(%pred) and anaerobic threshold to 48 ± 11%pred in all patients. D(L)CO moderately decreased to 63 ± 12%pred and there were 25 patients lower than 80%pred. FVC, FEV1, FEV1/FVC and TLC were 75 ± 14%pred, 71 ± 17%pred, 97 ± 11%pred, and 79 ± 13%pred, which indicated borderline or mild restrictive ventilatory dysfunction. The decrease of D(L)CO was more severe than those of TLC, FEV1 and FVC.
CONCLUSIONFor patients with severe CHF, cardiopulmonary exercise function is extremely limited, D(L)CO generally moderately declines and ventilation function is merely mildly limited. D(L)CO is the parameter for cardiopulmonary coupling, reflecting limitation of the cardiovascular dysfunction while without ventilatory limit.

10.Prokaryotic expression of recombinant human bone morphogenetic protein-2 and preparation of monoclonal antibodies.

OBJECTIVETo express and purify the recombinant human bone morphogenetic protein-2 mature peptide (rhBMP-2m) in prokaryotic system and to develop highly-specific monoclonal antibodies.
METHODSAn engineered E. coli strain expressing rhBMP-2m was fermented. The bacterial cells were firstly lysed and then the rhBMP-2m inclusion bodies were isolated by centrifugation. After the inclusion bodies had been solubilized by high-concentration denaturing agents, denatured rhBMP-2m was purified by cation ion-exchange chromatography. Biologically active rhBMP-2m was obtained by refolding of purified denatured rhBMP-2m through direct dilution. The refolded rhBMP-2m was used to immunize Balb/c mice to develop anti-rhBMP-2m monoclonal antibodies using classic hybridoma technique.
RESULTSrhBMP-2m with a purity greater than 95% was obtained on reduced SDS-PAGE. The refolded rhBMP-2m was measured to be bioactive by the induction of alkaline phosphatase activity in MC3T3-E1 cells. Two hybridoma cell lines that stably secreted anti-rhBMP-2m antibody were developed from the immunized mice.
CONCLUSIONBioactive rhBMP-2m protein and its monoclonal antibodies were successfully prepared, which will provides a solid base for future studies on rhBMP-2.
