1.Initial study of contrast-enhanced ultrasound 5-point scoring system in the diagnosis of breast tumor

Objective To explore the application of contrast-enhanced scoring system in the diagnosis of breast tumor.Methods 249 patients with 260 lesions were examined by elastography and contrastenhanced ultrasound before surgery.All the lesions were scored according to the two different methods.Pathological results were made the golden criteria.The diagnosis accuracy of the two methods were compared.Results There were 145 benign and 115 malignant tumors.The diagnosis accuracy,specificity and sensibility of elastography were 85.0%,88.3%,80.1 % respectively.The critical value of contrastenhanced ultrasound was between 3 and 4.The diagnosis accuracy,specificity and sensibility of contrastenhanced ultrasound were 88.9%,87.6%,90.4% respectively.Area under ROC curve of the two methods were 0.901 and 0.910,the difference was not statistically significant.Conclusions Contrast-enhanced ultrasound 5-point scoring system was an initial quantitative criterion for breast tumor diagnosis.The diagnosis accuracy was parallel to that of elastography.
2.High-throughput pyrosequencing of the complete chloroplast genome of Magnolia officinalis and its application in species identification.

Chloroplast genome sequences have comprehensive application prospects in DNA barcoding and chloroplast engineering in traditional Chinese medicine. The complete chloroplast genome of Magnolia officinalis sequenced by high-throughput pyrosequencing and a sequencing procedure was established. Fourteen contigs were obtained after de nove assembly. The sequencing percent of coverage was 99.99%. The chloroplast genome is 160 183 bp in size, and has a typical quadripartite structure with the large (LSC, 88 210 bp) and small copy (SSC, 18 843 bp) regions separated by two copies of an inverted repeat (IRs, 26 565 bp each). chloroplast genes were successfully annotated, of which 17 genes located in each IR region. The chloroplast genome features in Magnolia officinalis are nearly identical to those from other Magnoliid chloroplast genomes. Phylogenetic analyses were performed based on 81 shared coding-genes for a total of 9 Magnolia samples of 5 closely related species. Results showed that distinguishing among species was generally straightforward at the species and population level. This study confirmed the effectiveness of our chloroplast genome sequencing procedure. The chloroplast genome can provide distinguishing differences to help identify Magnolia officinalis and its closely related plants.

3.Clinical studies on different coronary artery interventional therapies through femoral artery or radial artery approaches

Objective To compare the clinical efficacies regarding femoral artery or radial artery approaches on coronary artery interventional therapies.Methods 360 patients were randomly divided into intervention group via femoral artery (TFI) or transradial coronary intervention (TRI) group.Postoperative observation on the two said groups of patients with vascular lesion characteristics,feasibility factors ( success rate of interventional therapy puncture,time of operation and hospitalization)and complications,were made.Results There were no significant differences between the two groups on the characteristics of vascular lesion (P>0.05).Success rates of the two groups were 97.78% and 96.67% respectively.The differences on success rate,time of operation were not statistically significant (P>0.05) while the average time of puncture,the mean duration of hospitalization and the rates of complications were significantly different ( P< 0.05 ).Conclusion The two methods under comparison had similar success rate and feasibility.However,the complications related to radial artery interventional therapy were much less than the femoral artery interventional therapy.As having better safety,radial artery interventional therapv seemed to have applicable value on clinical oractice.
4.Expression of a testis-specific gene 1700001022RIK in mice and its bioinformatic analysis.

OBJECTIVETo identify the expression characteristics of the 1700001022RIK (RIKEN cDNA 1700001022) gene in mice and explore its function by bioinformatic analysis.
METHODSUsing the expression profile of gene microarray, we detected the expression of a new testis-specific gene, 1700001022RIK, in mice. We analyzed its expression characteristics in the testis tissue and their changes in different developmental stages of the testis by RT-PCR, real-time RT-PCR, Western blot, and immunohistochemistry. We performed bioinformatic analysis using a bioinformatic software.
RESULTSThe 1700001022RIK gene was specifically expressed in the mouse testis in an age-dependent manner, most highly in the adult mice. The 1700001022RIK protein was mainly expressed in the spermatogonia, spermatocytes, and round spermatids of the adult mice. Bioinformatic analysis showed that the 1700001022RIK protein amino acid sequence had a high similarity in human and mice, which indicated that this gene was highly conserved in mammals.
CONCLUSION1700001022RIK is a testis-specific gene mainly expressed in the spermatogonia, spermatocytes, and round spermatids of seminiferous tubules, which might be involved in the regulation of spermatogenesis.

5.A clinical efficacy and safety study on coronary heart disease and angina treatment with Puerarin Injection

Objective To observe the clinical efficacy and safety of Puerarin Injection treatment on angina pectoris.Methods 388 patients with angina pectoris,enrolled to Tianjin Fourth Central Hospital during January 2009 and February 2011 were selected and randomly divided into treatment or control groups with 194 patients of each.Based on the conventional therapy program,one group was given Puerarin Injection as treatment group while,the other was given with Danshen Injection as control group.Clinical efficacy,the attack rate of angina pectoris,oxygen consumption,iadices on electrocardiogram,haemorheology and other adverse reactions among the two groups were compared.Results The total efficacy of the treatment group (88.14%,171/194)was significantly higher than the control group (61.86%,120/194) and the difference was statistically significant (P<0.05).During the treatment,no significant adverse events were noticed in both of the two groups of patients.Conclusion The Puerarin Injection treatment program on angina pectoris seemed effective and safe.
6.Flavonol glycosides from Lysimachia clethroides.

Eleven flavonol glycosides were isolated from the ethanol extract of Lysimachia clethroides by a combination of various chromatographic techniques including column chromatography over silica gel, Sephadex LH-20, and reversed-phase HPLC. Their structures were identified as astragalin (1), isoquercitrin (2), isorhamnetin-3-O-β-D-glucopyranoside (3), quercetin-3-O-β-D-6"-acetylglucopyranoside (4), quercetin-7-O-β-D-glucopyranoside (5), prunin (6), 2-hydroxynaringin-5-O-β-D-glucopyranoside (7), kaempferol-3-O-rutinonoside (8), kaempferol-3-O-robinobioside (9), rutin (10) and kaempferol-3,7-di-O-β-D-glucopyranoside (11). Among them, compounds 4, 7 and 11 were obtained from the Lysimachia genus for the first time, while compounds 3, 5 and 9 were firstly reported from this plant. In the preliminary assays, compounds 2, 6 and 8 possessed significant inhibition against aldose reduc- tase, with IC50 values of 2.69, 1.00, 1.80 μmol · L(-1), respectively; none of compounds 1-11 exhibited obvious cytotoxic activity (IC50 > 10 μmol · L(-1)).

7.Preparation optimization for Bishutongqiao Mixture

AIM To optimize the preparation of Bishutongqiao Mixture.METHODS With solid total mass,contents of astragaloside A and calycosin-7-glucoside as evaluation indices,water addition,extraction time and extraction times as influencing factors,the aqueous extraction process was optimized by orthogonal test.With the contents and retention rates of astragaloside A and calycosin-7-glucoside as evaluation indices,the effects of normal pressure and decompression on the concentration process were investigated.And the effects of solubilizer (polysorbate 80),taste-masking agent (sucrose) and bacteriostatic agent (sodium benzoate) amounts on the forming process were studied as well.RESULTS The optimal aqueous extraction conditions were determined to be ten times for water addition,1 h for extraction time,and extracting twice,the comprehensive score was 91.67.The contents and retention rates of astragaloside A and calycosin-7-glucoside at normal pressure concentration were higher than those at decompression concentration.The optimal forming process was obtained at polysorbate 80,sucrose and sodium benzoate amounts of 1.43%,15% and 0.3%,respectively.CONCLUSION This stable and feasible method can be used for the preparation of Bishutongqiao Mixture.
8.Tea Polyphenols Down Regulated Expression of Cyclin D1 in Nasopharyngeal Carcinoma Cells

Objective: This stduy was designed to investigate the molecular mechanism of cell proliferation inhibited by tea polyphenols in nasopharyngeal carcinoma cell line CNE-LMP1. Method: Light microscope, MTT assay, flow cytometry, Western blot,and reporter gene analysis were applied to investigate the effects of tea polyphenols and(-)-epigallocatechin-3-gallate (EGCG) on the cell cycle and related molecular mechanisms. Result: After treatment of CNE-LMP1 cells with tea polyphenols(200 μ g/ml), the number of proliferating cells was obviously decreased as determined by light microscopy, and the survival rate of cells decreased from 100% to 38.1% as determined by MTT assay. With the increase of tea polyphenols concentrations, the number of cells in S-phase was obviously decreased from 22.20% to 13.16% (200 μ g/ml), and the number of cells in Phase G1 were elevated from 68.50% to 74.08% . It suggested that tea polyphenols could arrest the cell cycle at G1/S checkpoint. The transcription and the expression of cyclin D1 were obviously declined 4-5 fold. Furthermore, after treatment with tea polyphenols, transactivities of NFκ B and AP-1 was obviously down-regulated in CNE-LMP1 cells for 5-6 and 7-8 fold, respectively. Conclusion: Tea polyphenols could arrest cell cycle and inhibit cells proliferation of nasopharyngeal carcinoma, and down-regulation of the gene transcription and expression of cyclin D1 might also be involved in these events.
9.An experimental study of inhibiting effect of decoy oligodeoxynucleotides on the gene expression of collagen in NIH3T3 cell.

OBJECTIVETo investigate the effect of activator protein-1 (AP-1) decoy-oligodeoxynucleotides (Decoy-ODNs) on the expression of fibroblast alpha2 type I collagen, so as to explore the gene therapy of pathologic scar.
METHODSDecoy-ODNs targeting AP-1 were designed and synthesized. NIH3T3 cells were transfected by cationic liposomes. The distribution of Decoy-ODNs in the cells was investigated. The inhibiting effects of Decoy-ODNs on AP-1 were determined by electrophoretic mobility shift assay (EMSA). And the effects of Decoy-ODNs on the collagen synthesis in the cells were analyzed by RT-PCR.
RESULTSAP-1 Decoy-ODNs could competitively inhibit the AP-1 in vitro activity. Cationic liposomes could play roles by effectively transfecting Decoy-ODNs into the plasma and nucleus. The mRNA expression of fibroblast alpha2 type I collagen decreased evidently after 24 hours of Decoy-ODNs action.
CONCLUSIONDecoy-ODNs could inhibit the mRNA expression of fibroblast alpha2 type I collagen by antagonizing AP-1.

10.Perioperative cortisol circadian secretion and neuropsychological states in patients undergoing coronary artery bypass grafting surgery.

OBJECTIVETo investigate the relationship between the circadian rhythm of perioperative cortisol secretion and neuropsychological states in patients undergoing coronary artery bypass grafting surgery.
METHODSForty male patients scheduled for elective coronary artery bypass grafting (CABG) under hypothermic cardio-pulmonary bypass (CPB) or off-pump were enrolled in this study. They were allocated into CPB group or off-pump group with 20 patients in each group. Blood samples were withdrawn during surgery at specific time-points and every 3 h for 24 h in the immediate postoperative period. Plasma cortisol was measured by radioimmunoassay. All subjects were investigated preoperatively as well as 7 to 10 d and 3 months postoperatively with a comprehensive neuropsychologic assessment, while depression and anxiety were assessed by Self-Rating Depression Scale and the State-Trait Anxiety Inventory respectively.
RESULTSDuring postoperative 24 h, three patients in the CPB group and 7 patients in the off-pump group were demonstrating a circadian secretion pattern, while they were disturbed in the remaining patients in both groups. Postoperative depression scores of patients in both groups were significantly higher than preoperative values. Postoperative anxiety scores of patients in the CPB group were significantly higher than those in the off-pump group. The CABG with CPB patients showed a significant deficit in the Digit Span subtest of the WAIS-R and the Stroop colour word interference test. The disturbed cortisol circadian secretion in the CPB group correlated with depression and the Stroop colour word interference test, whereas in the off-pump group it correlated with depression, Digit Span subtest (forward), symbol digit modalities test and the Stroop colour word interference test. Degree of depression correlated with some items of cognitive dysfunctions.
CONCLUSIONPerioperative secretion rhythm of cortisol in patients undergoing CABG surgery with CPB or off-pump was disturbed. The disordered cortisol may correlate directly or indirectly through mood with neuropsychological deficits.
