1.Clinical effect of tolterodine on detrusor hyperreflexia of patients with spinal cord injury

ObjectiveTo evaluate th effect and safety of tolterodine on detrusor hyperreflexia of patients with spinal cord injury.Methods20 cases of spinal cord injury patients with detrusor hyperreflexia were treated with tolterodine (2mg, twice daily), and the data of urodynamics and voiding diary before and after treatment were evaluated.ResultsAfter 12 weeks of treatment, it showed a significant increase from baseline in mean voiding volume (P<0.01), functional bladder volume (P<0.05) and interval of voiding (P<0.01). The bladder volume at first contraction significantly increased from (62.62±36.37)ml to (126.75±34.64)ml (P<0.01), the maximal pressure of detrusor contraction significantly decreased from (74.81±28.60) cm H2O to (61.90±16.22) cm H2O (P<0.05), the maximal amplitude of wave significantly decreased from (47.24±30.42) cm H2O to (39.36±25.28) cm H2O.ConclusionTolterodine is effective and safety to detrusor hyperreflexia of patients with spinal cord injury, and has a better therapeutic compliance and less adverse reactions.
2.Expression of integrin β1 in detached and reattached retina of rabbits

ObjectiveTo investigate the expression of integrin β1 during retinal detachment and reattachment of rabbits.Methods24 rabbits were used to make retinal detachment and reattachment model by using hyaluronidase and micropipette. The expression of integrin β1 were observed with hybridization in situ.ResultsThe expression of integrin β1 in reattached retina was lower than that in detached retina.ConclusionRetinal reattachment may inhibit the development of proliferative vireoretinopathy.
3.Level of proliferating cell nuclear antigen and interleukin-1β in rats' experimental retinal detachment

ObjectiveTo investigate the possible effect of interleukin-1β(IL-1β)in experimental retinal detachment,and the role in proliferation.MethodsThe experimental retinal detachment and reattachment in different time were made using Sprague-Dawley rats. The expression of proliferating cell nuclear antigen (PCNA) in neuro-retina was tested with flow cytometry at different time. IL-1β in neuro-retina were analyzed by labeling with polyclonal IL-1β antibody. The level of IL-1β in neuro-retina were tested with radio-immune method. IL-1β antibody 1000 ng,IL-1Ra 20 ng were injected in the subretinal space of some rats before PCNA reached to its high point respectively,and the expression of PCNA of them were compared with that of control which were injected with 0.01 M PBS.ResultsIL-1β expressed in Muller cell,astrocyte,vascular endothelial cell in neuro-retina and reached to its peak at the 7th day after detachment,then declined and continued at a low level as long as the retina detached.The expression of PCNA began at the second day after detachment, and reached a maximum at about 10 days after,then declined and continued at a low level as long as the retina detached. IL-1βantibody and IL-1Ra could restrain the expression of PCNA.ConclusionProinflammatory cytokine IL-1β is the key factor in proliferation of experimental retinal detachment.

8.Change of IL-6 in detached and reattached retina

ObjectiveTo determine the source of IL-6 in detached retina and the change of IL-6 level in detached and reattached retina.MethodsRetinas of SD rat were examined after subretinal injection of 1.4% Healon GV at different period of time, and the level of IL-6 in detached and reattached retina were detected by radio-immune histochemistry method. Wax-embeded sections were labeled with IL-6 antibody to determine the location of IL-6.ResultsDetached retina with normal vitreous and inner limiting membrane could only induce the subretinal fibrosis. This kind of fibrosis reached to its peak at 10th day and then remolded with time. Retinal pigment epithelial (RPE) cell, Muller cell, endothelial cell, glial cell were labled with IL-6, and the level of IL-6 in neuro-retina reached to its peak at 3rd-4th day and then downed to normal within a few days. The level of IL-6 in reattached retina was lower than in detached retina. The expression of IL-6 in RPE of detached area was stronger than in attached area.ConclusionIL-6 takes active part in wound healing process induced by the separation of RPE and neuro-retina. Reattachment can lower the expression of IL-6 in retina.
9.Differentiation of Dendritic Cells from Embryonic Stem Cells

Objective To investigate the method of directed differentiation dendritic cells from embryonic stem cells(ESC) and to amplify high purity DCS in vitro for immunity therapy.Methods E14 ESC line were generated ESC-derived dendritic cells(ES-DC) in complete medium further supplemented with granulocyte-macrophage colony-stimulating factor(GM-CSF) and interleukin-3(IL-3).ES-DCs was used flow cytometry to determine CD11c,CD80,CD86,MHC-Ⅱ cell surface phenotype. Lipopolysaccharide (LPS) were added to induce the ES-DCs matured. The matured ES-DCs was harvested 24 hours later to be identified with morphology, transmission electron microscopy, analyzed by flow cytometry and compared with the immatured ES-DCs phenotype. The antigen presenting was evaluated by mixed lymphocyte responses.Results The ES-DC had obviously dendritic processes under scanning electron microscope . The immature DCs express low level of CD11c(4.33±0.23)%,CD80 (7.62±0.19) %, CD86 (4.77±1.22) % and MHC-Ⅱ (9.68±0.15) %, but the mature DCs express higher lerve of CD11c(47.36±2.68)%,CD80 (74.4±1.47) %, CD86 (29.77±2.00) % and MHC-Ⅱ (87.56±2.75) %. MLR showed that ES-DCs could effectively stimulate lymphocyte to proliferate.Conclusion These results provide evidence that DCs can be generated from E14 ESC with GM-CSF and IL-3, express high level of CD11c,CD80, CD86, MHC-Ⅱ and can effectively stimulate lymphocyte to proliferate. ES cells may become new origin for DCs which provided the immunotherapy.
10.A comparative study on accommodative microfluctuation between emmetropic and myopic school-aged children

Background The incidence of myopia is increase.Some researches documented that formation of myopia is closely related with weakness of the accommodative response,enhancement of accommodative lag and accommodative microfluctuations in short-distance use of eyes.However,there still is controversy.Objective This study sought to compare the accommodative microfluctuations among emmetropic and myopic school-aged children at reading,and to discuss its potential relationship with the onset and development of myopia.Methods A casecontrolled study was designed.Eighty-nine children aged 8-12 years old were recruited in this study,including 47emmetropic children and 42 myopic children.Refractive error were checked by subjective refraction in phoropters and binocular vision and stereopsis were examined in all the subjects.A Grand Seiko WAM5500 auto-refractor was used to measure the accommodative responses and accommodative microfluctuations with different stimulus in 40 cm and 25 cm.The differences in accommodative response and accommodative microfluctuations at 25 cm and 40 cm reading distance were compared between the emmetropic children and myopic children using independent sample t test,and change of accommodative microfluctuations in myopic children at 25 cm and 40 cm reading distance was evaluated by paired t test.Results When the reading distance was 25 cm and 40 cm,the accommodative responses of emmetropes were (2.67 ±0.31) D and (1.70 ±0.23) D,and they were higher than (2.31 ± 0.33) D and (1.49 ±0.24) D of myopes,showing significant differences (t =5.330,P =0.000; t =4.140,P =0.000).Accommodative microfluctuation of myopes was(0.35 ±0.16)D in 25 cm reading distance and that of emmtropes was(0.26±0.08)D,with significant difference between them (t =3.180,P =0.002).However,there was not significant difference in accommodative microfluctuation at 40 cm reading distance between the myopic children and emmtropie children [(0.27±0.10) D vs.(0.24±0.09) D] (P=0.220).In myopic children,the accommodative microfluctuation at 25 cm reading distance was(0.35±0.16) D,showing a much increase than(0.27±0.10) D at 40 cm reading distance(t=3.850,P =0.000),but an insignificant difference in the accommodative mierofluctuations was seen between the 25 cm and 40 cm reading distance in the emmetropic children (P =0.145).Conclusions With the increased accommodative stimulus,myopic children present lower accommodative responses and larger accommodative microfluctuations.
