1.Expression of Bax and apoptosis in enterocytes of neonatal rats with necrotizing enterocolitis

Objective To investigate the correlation of Bax and apoptosis in enterocytes of neonatal rats with necrotizing enterocolitis (NEC).Methods Forty-eight neonatal rats (1 day) were randomly divided into control group (n =24) and NEC model group (n =24) by use of odd and even.The rats in control group were maternal breast-fed.The rats in NEC model group were separated from their mothers.To be given formula feeding,cold exposure after hypoxic-reoxygenation treatment.The intestinal tissue located at the boundary of ileum and caecum of two groups were gained on the 24 h,48 h and 72 h with which that all rats were sacrificed by cutting neck.Section of intestinal tissue were stained with immunohistochemistry to detect the expression of Bax and were stained with TdT mediated dUTP nick end labeling(TUNEL) to evaluate the apoptosis in each group.Results The integrate optical density (IOD) value of expression of Bax in the NEC model group began to increase on the 24 h [(1 005.06-± 11.96) IOD] and reached the summit on the 72 h [(3 340.66 ±68.72)] compared with the control group[(666.55 ± 15.77)IOD].A few of TUNEL positive cells began to increase with time dependence.A lot of TUNEL positive cells could be found in NEC model group on the 24 h [(15.04 ± 0.24) %],and the apoptotic index reached the peak on the 72 h [(35.65 ±0.61) %] compared with the control group[(4.73 ±0.04) %,P <0.01]-There was a significantly positive correlation between the cell apoptosis and the ratio change of Bax in NEC model group (r =0.94,P < 0.01).Conclusion There is a significantly positive correlation between the cell apoptosis and the ratio change of Bax in enterocytes of neonatal rats with NEC.The cell apoptosis in enterocytes of neonatal rats with NEC maybe be induced by Bax.
2.Experimental study of the bioresorbable collagen membrane used for guided bone regeneration around dental implants.

OBJECTIVETo investigate the effects of using guided bone regeneration (GBR) technique with an bioresorbable collagen membrane for repairing bone defects around dental implants.
METHODSIn 12 mongrel dogs, pure titanium implants were immediately implanted in mandibles after extraction of the mandibular third and fourth premolars of both sides, in which standard bone defects of 3 mm x 3 mm x 5 mm were created at mesial side of the implants. The defects on right side were covered with Co membrane, the left side was uncovered as control. The dogs were sacrificed 1, 2, 4, 6 months postoperatively. The specimens were removed and studied by gross observation, X-ray radiograph, histological examination, scanning electron microscope (SEM) and pull-out test.
RESULTSThe quantity and quality of new bone formation in experimental side which bone defects covered with Co membrane were much better than that in the control side.
CONCLUSIONWith the biodegradable property and excellent biocompatibility, Co membrane can be used for guided bone regeneration to promote the bone repair progress, and the promotion acts mainly at early stage of bone healing.

3.Identification of the fumatory Radix Bupleuri with sulfur by static headspace GC-MS.

OBJECTIVETo identify whether Radix Bupleuri (Bupleurum chinense) was fumed with sulfur.
METHODA static headspace GC-MS method was used to detect sulfur in the fumatory Radix Bupleuri, the authentic samples free of sulfur was detected as reference.
RESULTSulfur was detected in six samples from nine samples collected in different locations.
CONCLUSIONThe method can be used to detect sulfur rapidly in the fumatory Radix Bupleuri with sulfur.

5.Prospective study of gracilis muscle repair of complex rectovaginal fistula and rectourethral fistula.

OBJECTIVETo assess the efficacy and experience of gracilis muscle transposition for complex rectovaginal fistula (RVF) and rectourethral fistula (RUF).
METHODSNineteen patients underwent gracilis muscle transposition for complex RVF and RUF from May 2009 to November 2011 in the Beijing Shijitan Hospital and the clinical data were prospectively collected. The success rate and complications were recorded. SF-36 quality of life score, Wexner fecal incontinence score, and female sexual function score before surgery and 6 months after surgery were recorded.
RESULTSIn 19 patients, there were 8 males (RUF) and 11 females (RUF). The times of failed attempt repair preoperatively ranged from 0-3 (mean, 1.0). The diameter of the fistula ranged from 0.5-3.0 cm (mean, 1.6), and all fistulas located above the sphincter. The operative time ranged from 145-400 minutes (median, 240). The postoperative hospital stay ranged from 10-39 days (median 21). Early postoperative complications included thigh pain and numbness in 2 cases, leg numbness in 2 cases. No long-term complications were noticed. The follow-up period ranged from 6-35 months (median, 18). The gracilis muscle transposition had a healing rate of 94.7% (18/19). As compared with the preoperative level, Wexner score decreased from 10.0±8.8 to 2.9±5.8, and the continence function improved significantly (P=0.002). Sexual function score of 11 female patients increased from 1.0±1.8 to 4.0±4.0, and the sexual function had a significant improvement after surgery (P=0.022). SF-36 quality of life scores improved significantly (P<0.001).
CONCLUSIONSGracilis muscle transposition for complex rectovaginal fistula and rectourethral fistula has high success rate with mild and rare complications.

6.The clinical staging and tissue bacterial quantification in the diagnosis of burn wound sepsis.

OBJECTIVETo investigate and re-evaluate the relationship between burn wound sepsis and tissue bacterial quantity.
METHODSThirty-two patients admitted during past 5 years were enrolled in the study. Bacterial isolation and quantity in burn wound tissue were carried out. Meanwhile clinical signs were evaluated for the staging of burn wound sepsis.
RESULTS1) Bacterial invasion could be identified in 123 pieces of tissue samples from 32 patients. Samples with tissue bacterial quantity > or = 10(5)/g were found in 82 subeschar tissue samples, and 41 samples with bacteria <10(5)/g. Subeschar tissue samples with bacterial quantity > or = 10(5)/g could be determined in 68 samples from 18 patients, and < 10(5)/g in 20 samples from 5 cases. In addition, samples of subeschar tissue with bacterial quantity > or = 10(5)/g could only be found in some of the samples form 9 cases. 2) Burn wound sepsis could be classified into I-IV stages according to tissue bacterial quantification and clinical signs.
CONCLUSIONBurn wound sepsis could be established by identification of bacterial invasion into living tissue with clinical symptoms of toxemia.

7.Literature review report on safety evaluation about Kudiezi injection.

OBJECTIVETo explore safety evaluation information about Kudiezi injection used in the clinical practice.
METHODThree Chinese literature databases were searched and 96 articles were downloaded. Safety information about the study question was extracted by researchers in the Excel form. Finally, 22 articles including 24 case reports were analyzed in this article. Five aspects about safety information were summarized.
RESULTIt was found that all the adverse drug reaction reports in the literature were improper. There was no clear causality relation inference. It was inaccurate that those reports were identified as adverse drug reactions.
CONCLUSIONAlthough 24 case reports were identified as adverse drug reaction the evidence for each report was insufficient. There is no scientific causality judgment among these reports, which were implied little significance to future health decision about Kudiezi injection.

8.Clinical, genetic and cerebral ultrastructural features of late infantile neuronal ceroid lipofuscinosis:a ease report

Objective To study the clinical, histopathological and inheritance features of late infantile neuronal ceroid lipofuscinosis (LINCL). Methods The clinical manifestations and family history of a 4-year-old girl with an established diagnosis of LINCL were investigated and the findings in EEG, magnetic resonance imaging (MRI) and histological examination were analyzed. Results EEG of the patient showed diffuse background slowing with bursts of generalized spike-and-wave discharges or polyspike-and-wave activity. Brain MRI for her and her brother revealed brain atrophy, especially diffuse cerebellar atrophy. Histopathological examination also showed diffuse damages in the gray matter where numerous degenerated and atrophic neurons were found. Some immature neurons occurred in the disrupted cortical lamination. Electron microscopy revealed numerous osmiophilic granular lipofuscin inclusions in the cytoplasm of the neurons. Conclusion This patient presented with typical clinical and cerebellar ultrastructural features of LINCL, but the inheritance characteristics of the patient and the prominent lipofuscin pigments in the neurons suggest a case of new LINCL variant.
9.Influence of insertion torque on biomechanical characteristics at implant-bone interface

Objective To evaluate the influence of different insertion torque values on stress and strain distributions at implant-bone interface. Methods The three-dimensional finite element model of the whole mandible with dental implants for immediate loading was created by CT scanning and self-developed USIS (universal surgical integration system) software. The insertion torque values of dental implants were supposed to be 0、15、25 N•cm3, respectively. The values of Von Mises stress and strain at implant-bone interface were calculated with ANSYS software, when the dental implants were loaded with vertical and buccolingual force at a 45°oblique angle of 150 N. Results When the dental implants under the three insertion torques were loaded with the vertical force, the maximum Von Mises stress was 33.6, 56.4, 69.6 MPa and the maximum strain was 5 157, 8 645, 15 630 με, respectively, while loaded with the buccolingual force, the maximum Von Mises stress was 95.3, 100.6, 108.3 MPa and the maximum strain was 17 110, 18 690, 21 380 με, respectively. Conclusions With the increased torque value of dental implants, the stress and strain at the implant-bone interface were both increased, but the increase was much slower under buccolingual loading than that under vertical loading.
10.Pro-oxidation status of rotator cuff tissue and expression of Beclin-1 and mTOR in patients with rotator cuff injury.

OBJECTIVE:
To explore pro-oxidative state of rotator cuff tissue and expression levels of Beclin-1 and mam-malian target of rapamycin(mTOR) in patients with acute and chronic rotator cuff injury, and then analyzed relationship between rotator cuff injury and oxidative stress and autophagy.
METHODS:
Forty patients with rotator cuff injury were seleceted from July 2019 to December 2020, and divided into male chronic injury group, male acute injury group, female chronic injury group, and female acute injury group, 10 patients in each group. All patients were performed rotator cuff repair under arthroscopy. The sample of tendon at the rotator cuff injury site of the patient was taken during operation, and total reactive oxygen species (ROS) and superoxide dismutase(SOD) were detected by detection kit;expression of Beclin-1 and mTOR mRNA were detected by reverse transcription-polymerase chain reaction (RT-PCR), and Western-blot was applied to detect protein expression of Beclin-1 and p-mTOR/mTOR.
RESULTS:
There were no significant difference in expression of ROS, SOD, Beclin-1mRNA and mTOR mRNA between male and female chronic injury groups, and between male and female acute injury groups (P>0.05); ROS, SOD and Beclin-1mRNA in male chronic injury group were higher than those in male chronic injury group, while mTOR mRNAand protein decreased (P<0.05);ROS, SOD and Beclin-1 mRNA in female chronic injury group were up-regulated compared with female acute injury group, while mTOR mRNA was down-regulated (P<0.05).
CONCLUSION
Chronic rotator cuff injury is more likely to stimulate the pro-oxidation state of rotator cuff tissue than acute rotator cuff injury, which could up-regulating expression of autophagy factor Beclin-1 and down-regulating expression of mTOR. Therefore, patients with chronic rotator cuff injury may have higher levels of oxidative stress and autophagy.
