1.The expressions and clinical sig nificances of p27 and proliferating cell nuclear antigen and nuclear-associated antigen in synovial sarcoma

Objective To investigate the association of the expressions of p27 and proliferating cell nuclear antigen(PCNA)and nuclear-associated antigen(ki-67)proteins in synovial sarcoma,and the relationship with its prognosis.Methods The expression of p27,PCNA and ki-67 in 36 synovial sarcoma and 10 normal synovial tissue were immunohistochemically examined.The relationship between the expressions of p27,PCNA and ki-67 proteins in synovial sarcoma with its prognosis by clinical stage,tissue grades and prognosis was investigated.Results (1)In normal synovial tissue,the expressions of PCNA(0) and ki-67(0) were positive,p27 were positive100%(10/10).but the expressions of PCNA[86.1%(31/36)]and ki-67[69.4%(25/36)]in most synovial sarcoma were positive,p27 were negative[11.1%(4/36)],significant differences were found among these expressions(χ2=25.16,χ2=9.18,P<0.05).(2) p27 positive-expression correlated significantly wiht thehistological grade of synovial sarcoma(χ2=9.19,P<0.05)and prognosis(χ2=5.98,P<0.05).The positive expressions of PCNA and ki-67 were positively related with the clinical stage(χ2=7.40,χ2=8.12,P<0.05)and tissue grades(χ2=7.17,χ2=9.18,P<0.05)and prognosis(χ2=13.03,χ2=10.66,P<0.05)of synovial sarcina(3) There were negative correlations between p27 and PCNA(r=-0.63,P<0.05) and between p27 and ki-67(r=-0.53,P<0.05).But positive correlation between PCNA and ki-67(r=0.63,P>0.05)was found.Conclusions Expressions of PCNA and ki-67 in most synovial sarcomas are positive,and that of p27 is negative,each plays an important role in the proliferation of synovial sarcoma.Combination of measuring of the p27,PCNA and ki-67 is valuble in predicting the prognosis.
2.HPLC enantioseparation, absolute configuration determination and anti-HIV-1 activity of (±)-F18 enantiomers.

Racemic (±)-F18 (10-chloromethyl-11-demethyl-12-oxo-calanolide A), an analog of nature product (+)-calanolide A, is a new anti-HIV-1 nonnucleoside reverse transcript inhibitor (NNRTI). A successful enantioseparation of (±)-F18 offering (R)-F18 and (S)-F18 was achieved by a chiral stationary phase prepared HPLC. Their absolute configurations were determined by measurement of their electronic circular dichroisms combined with modem quantum-chemical calculations. Further investigation revealed that (R)-F18 and (S)-F18 shared a similar anti-HIV activities, however, (R)-F18 was more potent than (S)-F18 against wild-type virus, K101E mutation and P225H mutation pseudoviruses.

4.A feasibility study of recombinant adeno-associated virus(rAAV)as a vector for transferring a target gene to retina

AIM:To study the feasibility of recombinant adeno-associated virus(rAAV)as a vector to transfer the green fluorescent protein(GFP)gene as a target gene into rabbit retina.METHODS:Intravitreal injection of rAAV-gfp was performed in either eye for each rabbit with the other eye taken as control.At the 3rd,7th,and 14th day after injection,the eyeballs were removed,and the retinas were flat-mounted on glass slides to inspect the retinal fluorescence,respectively.RESULTS:After intravitreal injection of rAAV-gfp,the presence of fluorescent spots in the cytoplasm of retinal cells indicated that GFP gene was efficiently transferred and expressed in the rabbit retina.CONCLUSION:Recombinant adeno-associated virus is a reliable and simple vector for transferring target gene,e.g.,GFP gene,to the retina.
5.Recent progress in interferon induced protein GBP1 research.

Guanylate-binding protein 1 (GBP1) is an interferon induced protein, that belongs to the guany late-binding protein family. GBP1 is widely involved in anti-infection immune responses, anti-tumor activity and various biological reactions. Recent studies have proved that IFN-alpha, IFN-beta, IFN-gamma, IL1alpha, IL1beta, TNF-alpha and LPS can induce GBP1 expression; hence, the diverse biological functions of GBP1 have been gradually deduced and exploited. Many studies have been performed over recent years to understand the exact mechanisms that underlie the anti-infection and anti-tumor properties of GBP1. This review describes the molecular structure, biological activity, anti-infective properties and other functions of GBP1, in order to provide insights into the divergent roles of GBP1 in the regulation of various biological processes.

6.Changes of heme oxygenase-1 and inflammatory factors in patients with end-stage renal failure following hemodialysis.

OBJECTIVETo evaluate the changes in heme oxygenase-1 (HO-1), C-reactive protein (CRP), interleukin-6 (IL-6), and malondialdehyde (MDA) levels in patients with end-stage renal failure (ESRF) following hemodialysis.
METHODSThe urea nitrogen and the blood creatinine levels were determined with an automatic biochemistry analyzer, HO-1 and IL-6 levels by enzyme linked immunosorbent assay (ELISA), CRP by chemoluminescence technique, and MDA by thiobarbituric acid assay.
RESULTSHO-1, CRP, IL-6, and MDA levels were significantly elevated in patients with ESRF in comparison with the control group. After hemodialysis, HO-1 and CRP levels increased and IL-6 and MDA levels decreased significantly.
CONCLUSIONIn ESRF patients who have elevated inflammatory factors and HO-1 levels, hemodialysis can increase HO-1 and CRP levels and lower IL-6 and MDA levels.

7.Rapid detection of Streptococcus mutans and streptococcus sobrinus in human saliva by nested polymerase chain reaction.

OBJECTIVETo establish a simple and rapid method to detect Streptococcus mutans and streptococcus sobrinus simultaneously in human saliva.
METHODSChromosomal DNA from the bacteria was obtained by the extraction method with phenol-chloroform. A nested PCR method with two sets of primers specific for portions of the glucosyltransferase genes (gtfB of S. mutans and gtfI of S. sobrinus), was optimized to detect S. mutans and S. sobrinus from standard strains, clinical strains and directly in human saliva.
RESULTSThe first process of nested PCR was capable of amplifying DNA fragments specific for these species from chromosomal DNA extracted from 10(5) CFU cells of standard and clinical strains, or from 1 ml clinical saliva samples containing 10(5) CFU cells of either species. a second process of nested PCR, using the first PCR product as a template with new internal primers to detect 10(3) CFU of either streptococcal species in 1ml saliva samples.
CONCLUSIONNested PCR could detect S. mutans and S. sobrinus rapidly and simply in human saliva. This finding would be important to studies of elucidation the role of these two streptococcal species in the etiology of dental caries.

8.Study on differential proteomic expression in human liver cells stimulated by trichloroethylene with proteomics.

OBJECTIVETo explore the differential proteomic expression in human liver cells L-02 induced by different dosages of trichloroethylene (TCE).
METHODSHuman liver cells L-02 were treated with different concentrations of TCE and the solvent control (dimethylsulfoxide). The total cellular proteins were separated using 2DE and visualized with silver staining after TCE treatment. The images were analyzed with Image Master 2D Platinum 5.0 analysis software. The differentially expressed protein spots were identified by matrix assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF-TOF-MS).
RESULTSFifteen protein spots with significant difference were found, and went upward or downward or disappeared after the stimulation of TCE with different dosages, which indicated that TCE induced the change of the proteomic expression in the liver cells. The mass spectrum identification and the IPI human database retrieval were used for identifying 9 proteins related to the L-02 Liver cells induced by TCE.
CONCLUSIONThe result provides an insight to TCE-related molecular mechanism and which might be useful for further study of the TCE-associated proteins and molecular markers.

9.Open reduction and mini-plate internal fixation for the treatment of fracture and dislocation of coccyx.

OBJECTIVETo explore the therapeutic methods of fracture and dislocation of coccyx and evaluate its curative effects.
METHODSFrom May 2002 to March 2010,56 patients with fracture and dislocation of coccyx were divided into surgical treatment group and non-surgical treatment group. There were 7 males and 20 females in surgical treatment group with an average age of (48.1 +/- 0.6) years (ranged, 29 to 62 years), treated with open reduction and mini-plate internal fixation. There were 8 males and 21 females in non-surgical treatment group with an average age of (47.5 +/- 0.9) years (ranged, 19 to 54 years),treated with manipulative reduction. All patients were underwent X-ray examination and were finally diagnosed before treatment. Clinical symptoms and Visual Analogue Scales (VAS) of all patients were statistically analyzed before and after treatment.
RESULTSThere was no significant difference between two groups in gender, age, BMI index and VAS evaluation. All patients were followed up from 12 to 25 months with an average of 17.2 months. In surgical treatment group,there were 26 cases with I/a incision and 1 case with II/a incision; the excellent rate of clinical symptom was respectively 92.6% and 100% at leaving hospital and final follow-up; the improvement rate of VAS was 97.6% and was excellent result;internal fixtures were removed at the 1 to 2 years after treatment and no unwell symptoms occurred; VAS of all patients in the group was 0 point. In non-surgical treatment group,the excellent rate of clinical symptom was respectively 72.4% and 82.8% at leaving hospital and final follow-up; the improvement rate of VAS was 72.1% and was good result. There was significant difference in clinical results between two groups (P<0.05).
CONCLUSIONThe results indicated that fracture and dislocation of coccyx should be treated in time. For the treatment of patients with manipulative reduction failures, instability reduction by X-ray examination and serious rectal irritation, open reduction and mini-plate internal fixation can obtain satisfactory results.

10.Survey of the prevalence of chronic prostatitis in men with premature ejaculation.

OBJECTIVETo investigate the prevalence of chronic prostatitis in men with premature ejaculation.
METHODSThe segmented urine specimens before and after prostatic massage and the expressed prostatic secretion specimens from 106 patients with premature ejaculation and 38 controls were evaluated by microscopic and/or bacteriological studies. The prevalence of premature ejaculation was also investigated in 120 patients with chronic prostatitis.
RESULTSProstatic inflammation was found in 46.2% and chronic bacterial prostatitis in 34.7% of the subjects with premature ejaculation, respectively. Compared with the controls, the findings were statistically significant (P < 0.05). The prevalence of premature ejaculation in the patients with chronic prostatitis was 47.5% (57/120).
CONCLUSIONSChronic prostatic inflammation may play a role in the pathogenesis of some cases of premature ejaculation and it is important to give a careful examination of the prostate before initiating any therapy for premature ejaculation.
