1.Nitric oxide signal transmission pathway and its regulatory effect on skeletal muscle glucose uptake during exercise
Chinese Journal of Tissue Engineering Research 2007;0(15):-
BACKGROUND: Nitric oxide (NO) can influence glucose transportation. Although its signal transmission pathway is not certain, it has been confirmed that the pathway is different from insulin. OBJECTIVE: To explore NO signal transmission pathway and the regulatory mechanism for the glucose uptake of skeletal muscle during exercise. RETRIEVAL STRATEGY: Using the keywords of "nitric oxide, signal, glucose", we searched the Pubmed database for the relevant articles about the transmission pathway and its regulation of glucose uptake in skeletal muscle published from January 1996 to November 2007. Meanwhile, the related foreign language books were retrieved manually from national library. Articles about the regulation of NO to muscular blood flow, signal transmission and glucose under exercise stress in human and gnawer were selected. Pathologically related basic studies were excluded. LITERATURE EVALUATION: Sixty related literatures (books) were collected, and 30 were accorded with the inclusive criteria, of which 5 were review articles, 23 were basic researches and 2 were related books. Meanwhile, 20 articles were related to the effects of NO on blood flow regulation and vasodilation, 4 articles and 2 books were related to signal transduction and 4 were about its signaling effect during exercise. DATA SYNTHESIS: NO is a signal molecule. It can mediate various biological phenomenon and displays strong vasodilation effect. The production of NO is increased in vivo during exercise, and NO can stimulate glucose uptake in skeletal muscle through its transduction. CONCLUSION: The generation of NO during exercise has positive effect on glucose uptake in skeletal muscle. The contraction and glucose uptake are closely correlated to the generation and transmission of NO, but the mechanism is still unclear.

Chinese Journal of Rehabilitation Theory and Practice 2006;12(4):293-294
ObjectiveTo investigate the features of pulmonary function and influencing factors in patients with cervical spinal cord injury (SCI).MethodsThe pulmonary function data of 48 patients with cervical SCI were analyzed. The correlations between the percent predicted values of vital capacity (VC), maximal voluntary ventilation (MVV), forced expiratory volume of 1st second (FEV1) and V75 (expressed as VC%, MVV%, FEV1%, V75%) and ASIA motor/sensory scores were analyzed. Then, four multiple linear regressions were run to use VC, MVV, FEV1, V75 as an independent variable respectively.ResultsThe percent predicted values of the pulmonary function (VC%, MVV%, FEV1%, V75%) in patients with cervical SCI obviously decreased, which suggested that these patients had a restrictive ventilation disorder. There were significant positive correlations between VC%、FEV1%、V75% and ASIA motor scores (by bivariate correlation tests), no significant correlations between any of pulmonary function parameters and the ASIA sensory scores. Of four linear regression models, the height showed as a protecting factor in 3 linear regression models and weight did in the other one respectively. Of parameters associated with SCI, just ASIA motor scores showed as a protecting factor in 3 linear regression models, no sensory scores or duration appeared in any linear regression models.ConclusionBesides the height and weight, the ASIA motor score is a main factor influencing the pulmonary function.

Objecfive To establish the quality standard of Zaodi Anshen Pills. Methods TLC method was used to identify Spinosae and Radix polygoni Multiflori. HPLC method was used to determine the schisandrin, deoxyschizandrin and schisandrin B. Eclipse XDB-C18 column (250 mm×4.6 mm, 5 μm) was adopted by gradient elution using MeOH-water solution as the mobile phase. The detection wavelength was at 254 nm, column temperature was at 40 ℃ and the flow rate was 1.0 mL/min. Results The TLC spots were clear without interference of the negative control. The linear ranges of schisandrin, deoxyschizandrin and schisandrin B were within 338-1690 ng (r 2=0.999 1), 128-640 ng (r 2=0.999 3) and 236-1180 ng (r 2=0.999 4), and the average recoveries were 99.2%, 99.0% and 98.9% respectively. Conclusion This method is accurate, reliable, specific and reproducible, and can be used for quality control of Zaodi Anshen Pills.

Objective To establish a normal lumbar spine 3-dimensional(3D) finite element model(FEM) and discuss the biomechanics of the lumbar motions. Methods A geometrical model was constructed on the basis of a healthy adult male's lumbar CT image. The lumbar spine FEM was established by finite element analysis software. After setting the boundary and loading the burden, the lumbar spine's range of motion and stress distribution in different institutions such as anterior flexion, posterior extension, left or right bending and axial rotating were recorded. Results The lumbar spine FEM included 114 953 tetrahedron units. The results of the FEM matched the results of the in vitro experiment of biomechanics. Conclusion The lumbar spine FEM can imitate the motions of the lumbar spine.

BackgroundOsteopontin is highly expressed in many metastatic tumors, but its expression in uveal melanoma and the relationship of osteopontin with metastasis is still under investigation. Objective This study was to investigate the relationship between osteopontin expression and uveal melanoma metastatic potential. Methods Fifty samples of uveal melanoma were collected in Beijing Tongren Hospital. The expressions of osteopontin in the samples were detected using immunohistochemistry, and expressions of osteopontin mRNA in different metastatic potential uveal melanoma cell lines (MU M-2B,C918,OCM-1A) were analyzed using quantitative polymerase chain reaction (Q-PCR).ResultsOsteopontin was positively expressed in 10 samples of 13 patients with metastasis and 14 samples of 37 patients without metastasis, showing a significant difference between them (x2=5.888,P=0.000).There were 11 samples and 13 samples exhibiting the positive expression of osteopontin in 15 epithelial type and 35 non-epithelial type of uveal melanoma respectively with the significant difference (x2=5.510,P =0.000).The positive expressing numbers of osteopontin were 20 tumors of 30 affected tumors and 4 tumors of 20unaffected ciliary tumors with the significant difference (x2 =10.470,P =0.000 ).No significant differences in the expression of osteopontige were found between different gender, age, eyes, largest basal diameters and invasion of scleral canal( P =0.536,0.256,0.802,0.848,0.555 ).The relative expression value of osteopontin mRNA was in turn 1.00±0.04,0.91 ±0.03,0.08±0.01 in MUM-2B,C918,OCM-1A with the significant difference among three cell lines( F=33.135,P<0.05).The osteopontige mRNA expression was significant enhanced in high metastatic potentialcell lines( MUM-2B,C918 ) compared with low metastatic potential cell lines( OCM-1 A) (P<0.05 ),but no significant difference was seen between two high metastatic potential cell lines(MUM-2B,C918) (P=0.804).Conclusions Osteopontige expression is associated with the uveal melanoma cell line invasion and metastasis potential. These results imply that osteopontige expression maybe an indicator of uveal melanoma invasion and metastasis ability.
