2.The Effect of Quercetin Targeted p53 Gene and Protein Expression in NB4 Cell Lines

Objective In order to study the value of drug treatment for leukemia, quercetin was delivered to the cultured NB4 cell line. MethodsUsing RT- PCR and western blot, we studied the expression of mutant p53 gene and protein after inducing 24, 48, 72, 96 and 120h by the quercetin with different concentrations (30, 60, 90μmol/L). Results We verified the results that quercetin with concentration of 30 ~90μmol/L could successfully inhibit the expression of mutant p53 protein, but the mutant p53 gene did not have. ConclusionThat data suggested that quercetin would be an effective method for the therapy of leukemia.
3.Effect of coronary artery disease on QT dispersion

Objective To observe the effect of coronary artery disease on QT dispersion (QTd). Methods The QTd and QTcd in 245 patients underwent percutaneous coronary angiography (CAG) were analyzed. It was evaluated The relations between the QTd and the degree of coronary stenosis and the number of disease vessels. Results The QTd and QTcd were significantly prolonged in patients with coronary artery stenosis than those in patients with normal coronary artery(P0.05). Conclusion There is no relation between the number of disease coronary artery and the QTd.

5.Effect of Hexue mingmu tablets in the treatment of hyphema

To evaluate the effect of Hexue mingmu tablets on traumatic hyphema caused by blunt ocular trauma.
●METHODS: Totally 150 patients of traumatic hyphema were divided into seven types by using ultrasound biomicroscopy combining with anterior segment abnormalities, each type was randomly classified as trial group and control group. The trial group was administered Hexue mingmu tablets, control group was treated by hemocoagulase.
●RESULTS: The absorbing time of trial group was shorter than that of the control group. And there was statistical significance between the two groups (P<0. 05).
● CONCLUSlON: Hexue mingmu tablets is an effective medicine to treat traumatic hyphema. Ultrasound biomicroscopy can be used as a routine examination method in traumatic hyphema.
6.The circadian ryhthm variation of melatonin receptor in peripheral leukoc ytes in patients with acute cerebral infarction

Objective: To investigate the circadian ryhthm v ariation of 2-125I-iodomelatonin (125I-Mel) binding sites in the human peripheral leukocytes in patients with acute cerebral infarction. Methods: Melatonin binding sites in the human peripheral leukocytes were studied using 125 I-Mel as a radioligand (radioligand binding assays).A ll patients [age: (70.18±11.70) years] were diagnosed by CT according to the standard [Chin J Nerv, 1996,29(6):379]. We also studied 15 age-match ed healthy old people as the control [age: (68.33±7.76) years]. Resul ts: The circadian ryhthm variation of 125 I-Mel specific binding in control remained significant (P<0.01) with a higher value at midlight [ (0.16±0.049) fmol/106] than at middark [(0.078±0.035) fmol/106] with one point analysis. There was no significant variation in the patient grou p (P>0.05). The specific binding in the peripheral leukocytes at midlight in the patients with acute cerebral infarction were lower than that of the control . Conclusion: The expression of Mel receptor decreases in the pa tients wit h acute cerebral infarction and the circadian ryhthm variation of 2-125I -Mel appears abnormal.

The chemical investigation on Ganoderma philippii led to the isolation of sixteen compounds by silica gel and Sephadex LH-20 column chromatography. On the basis of spectroscopic data analyses, their structures were elucidated as 2, 5-dihydroxyacetophenone (1), methyl gentisate (2), (S) -dimethyl malate (3), muurola-4, 10 (14) -dien-11beta-ol (4), dihydroepicubenol (5), 5-hydroxymethylfuran carboxaldehyde (6), ergosta-7, 22E-dien-3beta-ol (7), ergosta-7, 22E-dien-3-one (8), ergosta-7, 22E-diene-2beta, 3alpha, 9alpha-triol (9), 6/beta-methoxyergo-sta-7, 22E-dien-3beta, 5alpha-diol (10), ergosta-4, 6, 8(14), 22E-tetraen-3-one (11), ergosta4, 6, 8-(14), 22E-etetraen-3beta-ol (12), 5alpha, 8alpha-epidioxy-ergosta-6, 22E-dien-3beta-ol (13), 7alpha-methoxy-5alpha, 6alpha-epoxyergosta-8-(14), 22E-dien-3beta-ol (14), ergosta-8, 22E-diene-3beta, 5alpha, 6beta, 7alpha-tetraol (15), and ergosta-5, 23-dien-3beta-ol, acetate (16). All the compounds were obtained from this fungus for the first time, and compounds 4 and 5 were isolated from the Ganoderma genus for the first time.

8.Ultrasound-induced uterine smooth muscle contraction

Objective To study the influence and dose effect of ultrasound on the contraction of uterine smooth muscle in rats.Methods Estradiol benzoate was injected into rats three days before conducting an in-vitro experiment.Their uteri were resected and irradiated with ultrasound(0.8 MHz,3 W/cm~2,0-40 rain).The contrac- tion frequency and amplitude were recorded using an MS-302 biological experiment system.Results It could be seen that the contraction frequency and amplitude,and general contractile activity were significantly increased during ultrasonic irradiation(P<0.01).The increased contraction frequency and amplitude lasted for ten minutes,and then the normal contraction pattern resumed.The contraction frequency as well as the percentage change in eontraction fre- quency were highest during the first 15 minutes of ultrasonic irradiation;the contraction amplitude as well as the per- centage change in amplitude were highest during 40 minutes of ultrasonic irradiation.Contraction activity was at its highest for 30 minutes,but the percentage change in activity was highest for 20 minutes.Conclusions Ultrasound can induce uterine smooth muscle contraction in rats.This biological effect is related to the irradiation time.
9.Mutation analysis on DACT1 gene in children with neural tube defects in northern Chinese Han population

Objective To investigate the correlation between neural tube defects (NTDs) and DACT1 gene, and provide the basic data for disease diagnosis and genetic counseling. Methods Blood samples were obtained from 163 NTDs patients and 480 unrelated healthy individuals. Mutation detection of DACT1 gene and DNA direct sequencing was carried out by PCR amplification. Bioinformatics analysis of these mutated loci was performed. Results Six mutations were found in NTDs patients, including 4 missense mutations (p.R45W, p.D142G, p.N356K and p.V702G). But these mutations were not found in 480 healthy individuals. Three mutated amino acid residues (p.45R, p.142D and p.356N) were highly conservative in evolution, and the mutated carriers were female patients, and suffered from anencephaly. Conclusion DACT1 gene mutation may be a risk factor of NTDs in Han population of northern China.
