2.Application of body strait jacket design in mental patients

Objective To explore the application effect of self-developed strait jacket in mental patients.Methods A total of 200 mental patients meeting indications for protective restraint were selected from a tertiary first-class psychiatric hospital using convenience sampling,and were restrained using self-developed strait jackets,application effects and numbers of adverse events occurring during restraint were observed;patients' satisfaction with restraint tools was investigated as well.Results Cases of adverse events occurring during protective restraint were:3 patients developed dysaemia (1.50%),3 had self-injury (1.50%),4 were injured by others (2.00%),5 developed skin scratching (2.50%),7 threw off restraint by themselves (3.50%),13 presented violent and aggressive behaviors (6.50%),and no patient developed swelling of limbs.According to satisfaction survey,71.00% of the patients demonstrated acceptance of this restraint tool,76.50% felt comfortable,83.50% held that this strait jacket could meet the needs of free activities,76.00% felt safe,and satisfaction to body strait jacket came to 83.00%.Conclusion The manufacture of body strait jacket is simple and it is comfortable to wear,patient's compliance is high,and restraint effect is satisfactory;this body strait jacket reduces the incidence of adverse events,and improve patient's satisfaction to restraint tool.
3.Nitric Oxide Regulated Expression of ?-Aminobutyric Acid B Recepto r Subunits during Recurrent Febrile Seizures

Objective To explore the effect of nitric oxide (NO) on ?-aminobutyric acid B receptor (GABA_BR) subunits during recurrent febrile seizures (FS).Methods Twenty-four Sprague-Dawley rats aged 21 days were randomly divided into 4 groups: control group (37.0 ℃ water,n=8), FS group (45.2 ℃ water,n=8), FS + SNP group (45.2 ℃ water,n=8), FS+L-NMMA group (45.2 ℃ water,n=8). FS rats were induced 10 times in a warm-water bath, once every 2 days. The plasma level of NO was detected by the spectrophotometer. The expressions of GABA_BR subunit mRNA and c-fos gene were examined by in situ hybridization. The expressions of GABA_BR subunit and Fos protein were observed by immunohistochemistry. Results The plasma level of NO increased in FS + SNP group while decreased in FS+L-NMMA group compared with that in FS group. The expressions of GABA_BR_2 were down-regulated in FS+SNP group, while GABA_BR_1 hardly changed compared with those in FS group. In FS+L-NMMA group, both the expression of GABA_BR_2 and GABA_BR_1 up regulated compared with those in FS group. The expressions of c-fos gene and Fos protein were significantly enhanced after recurrent FS. SNP elevated the expressions of c-fos gene and Fos protein, while L-NMMA down regulated the expressions of them.Conclusion NO may play a regulatory role through modulating GABA_BR function in the pathogenesis of recurrent FS.

Objective To explore the effect of ? - aminobutyric acid B receptor(GABABR)on brain damage induced by recurrent febrile seizures (FS). Methods Rats were randomly divided into four groups: control group (37. 0 ℃ water, n = 8), FS group (45.2 ℃ water,n=8), FS + baclofen group (45.2 ℃ water,77 = 8), FS + phaclofen group (45. 2 ℃ water,n=8). FS in rats were induced for ten times in a bath of warm water, once every 2 days. The intensity, latency and duration of the seizure in rats were recorded. The expression of c - fos gene and Fos protein were examined by in situ hybridization and immunohistochemistry, respectively. Results Compared with those of FS group, the seizure latency gradually prolonged, and the seizure duration was shortened in FS + baclofen group. In FS+ phaclofen group, the seizure latency was shorter and the seizure duration was longer than those of FS group. The seizure intensity was lessened in FS + baclofen group while aggravated in FS + phaclofen group compared with that of FS group. The expression of c - fos gene and Fos protein increased significantly after recurrent FS. Baclofen down regulated the expression of c -fos gene and Fos protein, while phaclofen enhanced the expression of them. Conclusion The study by using the agonist and the inhibitor of GABABR showed that GABABR might play a crucial role in the development of FS- induced brain damage.

OBJECTIVEThe research on flavonoid in the recent years is extensive, such as Soybeans flavone, Baicalensis flavone, Epimedium flavone. Experiments show the effects of flavonoid is closely related to human health. There are a lot of reports about Ginkcobiobal flavone. In order to make further progress research on ginlcgobiobal flavone, we sum up the articles and reports on ginlcgobiobal flavone in the recent years.
METHODTo searche the articles about ginlcgobiobal flavone studies in the past five years.
RESULTGinlcgobiobal flavone is not only a vasodilator, but also has the effects of anti-inflammation, analgesia, lowering blood lipids, preventing senile and inhibiting tumor, treating leukaemia, regulating gene and biotransformation.
CONCLUSIONGinlcgobiobal flavone has the potential value for drug research and development.

8.Disruption of hom Gene Encoding for Homoserine Dehydrogenase of Corynebacterium glutamicum

The hom gene encoding for homoserine dehydrogenase was amplified from the genomic DNA of Corynebacterium glutamicum ATCC 13032.After the kanamycin-resistant gene(Km)cassette from plasmid pET28a was inserted into the center of hom,the hom::Km cassette was then electroporated into the competent cell of C.glutamicum ATCC 13032.And kanamycin-resistant clones were obtained.PCR was performed to confirm whether the Km gene was integrated into the hom gene of these clones and the recombinant strains of hom-disrupted were screened out.Fermentation results showed that the lysine yield of the hom-disrupted strain C.g-hom::Km-8 reached 4.7 g/L,which was 6.7 times that of C.glutamicum ATCC 13032.
9.Genetic Transformation of Candida glycerinogenes by REMI and Electroporation

In order to isolate genes related with the osmoadaptation and glycerol metabolism of Candida glycerinogenes, a transformation system based on the dominant selectable marker Zeocin and restriction enzyme-mediated integration (REMI) was established. Effects of seven restriction enzymes on transformation efficiency of C.glycerinogenes were tested. Transformation conditions were optimized in the presence of Hind III. Under the optimal conditions of OD_ 600 ≈1.3, voltage of 1.5 kV, 2.0?10~9 competent cells/mL, 100 units of Hind III added, the transformation efficiency was up to 129 trnaformants/?g DNA. 58% of transformants were stable on nonselective medium. These results suggest that REMI technique would be beneficial to the genetic transformation of C.glycerinogenes.
10.Determination of Dihydroxyacetone in Fermentation Broth by HPLC

A method to determine dihydroxyacetone (DHA) in fermentation broth was developed by high performance liquid chromatography (HPLC). DHA was separated on a Alltima C18(5?m,250?4.6mm). The mobile phase was 0.5% methanol solution (pH adjusted to 3.0 with H3PO4), the flow-rate was 1.0 ml/min and the detective wavelength was 200 nm. The detection limits of DHA was 0.1 g/L~10.0 g/L. 6.2 g/L DHA in the fermentation broth was detected by HPLC, which was in agreement with the result by spectrophotometric method.The method was applicable for DHA determination in the fermentation process.
