Aim To investigate the effect of procyanidins on apoptosis in brain of experimental rats with intracerebral hemorrhage(ICH) and its possible mechanisms.Method Rat intracerebral hemorrhage model was established by injecting collagenase into the brain caudate nucleus.Fifty-four health Sprague-Dawley rats were divided into six groups randomly: sham operation group,model group of intracerebral hemorrhage,treatment groups with procyanidins at doses of 50 mg?kg~-1,100 mg?kg~-1 and 200 mg?kg~-1,and positive control group with nimodipine at dose of 10 mg?kg~-1.Twentyfour hours after operation,the content of glutathione(GSH) and activity of catalase(CAT) were measured.The levels of bax and bcl-2 were detected by method of immunohistochemistry,and the apoptosis in brain tissues was detected by method of TUNEL.Result In procyanidins groups(50,100 and 200 mg?kg~-1),the content of GSH and the activity of CAT were significantly higher than those in ICH group(P

Aim To study the changes of the biological behavior of dermal fibroblasts in Ⅲ skin burns wound in rats using chitosan.Methods Wistar rats were randomly divided into 5 groups as follows:1% chitosan(W/V)group,2% chitosan(W/V)group,4% chitosan(W/V)group,bFGF(basic fibroblast growth factor) group and the control group.Rats were made for Ⅲ skin burns.The wound healing time was recorded,and the wound healing rate was calculated.Then the cell cycle and apoptotic dermal fibroblasts were determined and the amount of Hydroxyproline(HOP) in the skin tissue was analyzed.Results The wound healing rate of 4% chitosan(W/V) group was higher and the wound healing time of 4% chitosan(W/V) group was shorter than that of the control group.On the 7th,14th day post-injury,the content of protein of 4% chitosan(W/V) group was higher than that of the control group.The content of HOP of 2% and 4% chitosan(W/V) group was highest on the 7th day post-injury. Compared with that in control group,the percentage of cells of S stage in 4% chitosan(W/V) group was aboundant,and was reduced in apoptotic dermal fibroblasts.Conclusion The changes of cellular biological behaviors might be one of the mechanisms of that Chitosan could promote the wound healing of Ⅲ skin burns in rats significantly.

目的探讨不同年龄组脑卒中患者康复结局的预测模型。方法对462例首次发病的脑卒中患者的临床资料进行回顾性分析。采用自行设计的日常生活活动能力(ADL)评定量表对其进行评定。结果总回归方程为:ADL出院=55.09+0.593ADL入院-0.17年龄-0.02OAI,复相关系数R=0.75,决定系数R2=0.56,校正的决定系数R2adjus=0.56;60岁以下年龄组回归方程为:ADL出院=52.27+0.51ADL入院-0.03OAI,R=0.69,R2=0.48,R2adjust=0.48;60岁以上年龄组回归方程为:ADL出院=34.46+0.73ADL入院,R=0.8,R2=0.64,R2adjus=0.64。60岁以上年龄组的预测模型准确性较高。结论按年龄分组可提高对脑卒中患者康复结局预测的准确性。

Objective：This study was designed to express chimeric anti-VEGF (vascular endothelial growth factor) antibody in dihydrofolate reductase-deficient Chinese hamster ovary (CHO-dhfr-)cells at high-level, and explore an optimum method to obtain high-level expression cells clone. Methods：The light chain and heavy chain genes of chimeric anti-VEGF antibody were induced into CHO-dhfr-cells using a novel eukaryotic high-level expression vectors system for genetic engineering antibodies. High-level expression was achieved after subcloning and several rounds of co-amplification of methotrexate (MTX). Biological features and productive amount of chimeric antibody was charactered by ELISA. Result：The cells strain that secret anti-VEGF chimeric antibody at the highest level of 28 μ g/ml was established. The cells were subcloned following each round of co-amplification of MTX, while greatly different results were obtained using three methods. The chimeric antibody contained constant regions of human immunoglobin and had the specificity against VEGF by ELISA. Conclusion：The anti-VEGF mouse-human chimeric antibody was expressed at high-level successfully in CHO cells. This may be an optimum method to obtain high-level expression cells clone for the eukaryotic high-level expression vectors system.

Objective To investiagte the serum adiponectin concentration in preeclampsia and its relationship with serum leptin and soluble leptin receptor levels.Method The level of adiponectin,leptin and soluble leptin receptor in serum were measured by enzyme-linked immunosorbent assay(ELISA)in 38 patients with preeclampsia and 42 patients as control.The relationship of free leptin index(leptin/soluble leptin receptor) to preeclampsia was analyzed.Results There were no significant differences in maternal age,gestational age and body mass index(BMI)between two groups.But the gestational age and birth weight were significantly lower in preeclampsia than in control.The patients with preeclampsia had significantly higher levels of serum adiponectin, leptin and free leptin index(1691.7?g/ml,37.5 ng/ml and 0.95 respectively)than the control(689.4?g/ml, 19.3 ag/ml and 0.49,respectively).But there was no significant difference in serum level of soluble leptin receptor between the groups(35.0 ng/ml vs 42.2 ng/ml).Serum adiponectin was not significantly correlated with the level of leptin,soluble leptin receptor and free leptin index.Area of serum adiponectin,leptin and free leptin index in preeclampsia under the ROC curve were less than 0.5.Conclusions The patients with preeclampsia have paradoxical higher serum levels of adiponecfin and more bioavailability of leptin,suggesting these may be important factom of this complication.

Objective To study the significance of Titin-ab and Ryanodine receptor-ab (RyR-ab) in the myasthenia gravis (MG) patients and the expression of Titin and RyR epitopes in thymoma of myasthenia gravis.Methods Using ELISA methods,the titer of the Titin-ab,RyR-ab in the sera of 62 patients with MG,as well as 45 cases non-MG with other neurologic disorders and 50 case of normal controls were determined,and Titin and RyR were studied with immunohistochemistry stain in the 19 samples of thymic tissues from 9 cases of MG with thymoma (MGT),6 cases of MG with thymic hyperplasia (MGH), 2 cases of MG with thymic atrophy (MGA),and 2 cases of non-MG with thymic carcinoma (NMGTC). Results The positive rate of Titin-ab in MG was 35.5% (22/62),with the highest being 82.3% (14/17) in MGT group.The positive rate of RyR-ab was 24.2% (15/62),the highest being 76.5% (13/17) in MGT group.Titin receptor epitopes were expressed in the transmembrane and cytoplasm region of thymoma epithelial cells of 7 cases of MGT patients,and RyR epitopes in the transmembrane region of thymoma epithelial cells of 6 cases of MGT;but no Titin nor RyR epitopes was identified in controls and such thymic pathological patterns,as MGH,MGA,NMGTC.Conclusions Titin-ab and RyR-ab are mostly found in MGT patients;Titin and RyR epitopes are expressed in neoplasm epithelial cells of thymoma with myasthenia gravis;it's a result of autoimmunization of Titin and RyR epitopes irritated by Titin and RyR specific T cells activated by the change of pathogenic microenvionment inside the thymoma.

One hundred and fifty two actinomycetes were isolated from forty two radiation-polluted soil samples,using six different isolation media. Sixty cultures were chosen for 16S rRNA gene sequence and systematic analysis,which based on their morphology and ARDRA. Results of 16S rRNA gene sequences blasting showed that the strains were assigned to 12 recognized genera of actinomycetes,most of them fall within Streptomyces genus and a great deal of strains belonged to rare actinomycetes,which indicated a rich diversity of actinomycetes in the radiation-polluted soil.

Objective To investigate the expression of complement split product C4d and its significance in renal allograf tissue of chronic rejection in rats.Methods The healthy closed population Wister rats and SD rats were used as donator and acceptor in renal transplantation.The chronic rejection model of renal transplant in rats was established and the rats were divided into 2 groups.The rats in experimental group were given Mycophenolate mofetil(MMF)(10 mg/kg) and those of the control group were given nothing except CsA(5 mg?kg~(-1)?d~(-1)) for 10 days.At the 12th week of renal transplantation,the allograft was tested by light microscope,and the pathological changes of renal grafts and the expression of C4d in peritubular capillaries were observed.Results On the 12th week of renal transplantation,the morphology changes of chronic rejection was observed in the experimental group and obvious C4d deposition was detected in peritubular capillaries of renal allograft tissue,with significant difference compared with those of the control group(all P

OBJECTIVETo compare clinical effects of different fixations in order to provide the best therapeutic choice for comminuted patellar fractures.

METHODSFrom February 2003 to June 2009, 56 patients with comminuted patellar fractures were treated with three methods of fixation. Of them, 18 patients (group A) were treated with Kirschner-wire with steel wire fixation, there were 9 males and 9 females, ranging in age from 32 to 52 years with an average of (47.62 +/- 4.82) years; 13 patients (group B) with circular silk thread fixation and plaster immobilization, there were 3 males and 10 females,ranging in age from 38 to 65 years with an average of (48.58 +/- 8.28) years; 25 patients (group C) with memory alloy patella holder fixation, there were 9 males and 16 females, ranging in age from 32 to 68 years with an average of (48.36 +/- 9.59) years. According to criteria of Böstman, eight indexes were compared, including the range of motion of the knee, pain, walking and so on.

RESULTSAll patients were followed up from 8 to 26 months with an average of 15.6 months. All the fractures healed. The Böstman scoring of group A, B, C were respectively 26.00 +/- 4.6, 22.08 +/- 5.31, 26.44 +/- 3.77. The clinical effects of group A and C were better than that of group B (P < 0.05).

CONCLUSIONThe method of Kirschner-wire with steel wire fixation or memory alloy patella holder fixation can obtain satisfactory effects in treating comminuted patellar fractures because of steady fixation and early function recovery.
