Human immunodeficiency virus(HIV) infection may result in serious impairment of the immune system in HIV-posi-tive patients. Both HIV productions and highly active antiretrovi-ral therapy (HAART) drugs can cause neuropathy, including the HIV associated neurocognitive disorders (HAND) and the peripheral neuropathy. Several animal models have been devel-oped in an attempt to study the mechanisms of HIV relative neu-ropathies. This review focuses on researching HIV-1 associated neuropathies by application of HIV-1 animal models.
2.Application of a method of improving the quality of sampling in review to determine the light areas of endemic fluorosis in quality control

ObjectiveTo study an application of a method of improving the quality of sampling in review to determine the light areas of endemic fluorosis(referred to as endemic fluorosis) in quality control. Methods Of 15 light endemic fluorosis township(town), six were randomly sampled, and the prevalence of dental fluorosis in 22 village primary school children aged 8 to 12 were reviewed to determine the improved quality of sampling in Xuyong county Sichuan province. ResultsSix townships(towns) were selected by simple random sampling from 15 endemic fluorosis townships(towns) for review inspection in Xuyong country. A total of 22 villages were verified, accounting for 22.7% of the total 97 villages verified. Of the 416 children for review inspection of dental fluorosis, 383 children were positive. The consistent rate of children' s dental fluorosis was 92.07%, and the verification to be slight villages was up to 21 endemic villages, accounting for 95.45%. ConclusionsThe application of a method of improving the quality of sampling can improve the efficiency of quality control, and improve the accuracy. It is a novel quality control method.
3.Advance on inducing ferroptosis of tumor cells based on nanodelivery technology

At present, cancer is still one of the most serious threats to human health. Despite the wide application of multiple cancer therapies in clinical practice, the therapeutic effects of most cancers are still far from satisfactory. In recent years, the discovery of regulated cell death may be a good first step on the road to treat cancer. Ferroptosis is triggered by lipid peroxidation of unsaturated fatty acids in cell membrane catalyzed by iron ion. It has been widely concerned as an emerging target for cancer therapy. With the booming of biomedical nanotechnology, ferroptosis as an emerging therapeutic target has attracted extensive attention. Here, we review the advance on the intersection of ferroptosis and biomedical nanotechnology. First, the research background of ferroptosis and nano-preparation as well as the feasibility of ferroptosis-based nano-drug delivery systems (nano-DDS) for cancer treatment are presented and analyzed. Then, the strategies for inducing ferroptosis based on nano-DDS are summarized, mainly including: the promotion of Fenton reaction, the inhibition of glutathione peroxidase 4 (GPX-4) and the restriction of the cysteine-glutamate exchange transporter (system Xc-). Furthermore, the combination therapy strategies based on biomedical nanotechnology induced ferroptosis are also discussed. Finally, we shine the spotlight on the prospects and challenges of ferroptosis-based nanotherapeutics in clinical application.

5.Low-dose urokinase combined with low-molecular weight heparin nadroparin calcium and ozagrel sodium in patients with acute cerebral infarction

[Objective]To explore an individualized treatment measure enjoying more practical,effective and safe characteristics through evaluating the efficacy and safety of combined medications of low-dose urokinase,low-molecular weight heparin nadroparin calcium and ozagrel sodium in treating patients with acute cerebral infarction.[Methods]One-hunderd patients with acute cerebral infarction patients were recruited in this trail,and grouped according to different treatment times:Group A (n=40,from January 2005 to February 2008,being selected into the group in accordance with standards of China Guideline for Cerebrovascular Disease Prevention and Treatment) and Group B (n=60,from March 2008 to June 2011,being selected into the group in accordance with indications for onset time within 24 h and allowing age more than 75 years).Standard thrombolytic therapy (high dose urokinase) was performed on Group A and combined medications of low-dose urokinase,low-molecular weight heparin nadroparin calcium and ozagrel sodium (triple antithrombotic therapy) were performed on group B.National Institute of Health Neurological Deficit Scale (NIHSS) and Evaluation Standard of Clinical Efficacy were used to evaluate the recovery of neurological function before treatment and 24 h,7 and 14 d after treatment.[Results] NIHSS scores after therapy rapidly decreased in both groups as compared with those before treatment (P<0.05).The NIHSS scores of Group B at 24 h,and 7 and 14 d after treatment were significantly decreased as compared with those of Group A (P<0.05).The efficacy rate of Group B was significantly higher than that of Group A (P<0.05).Intraparenchymal hemorrhage rate was 10.0% (4/40) in Group A and 3.3% (2/60) in Group B.[Conclusion] Triple antithrombotic therapy is more effective and relatively safer than standard thrombolytic therapy in treatment of patients with acute cerebral infarction.
6.Ribosome display screening of a novel human anti-IgE scFv fragment.

Total mRNA was extracted from lymphocytes separated from the peripheral blood of allergic patients, and then variable region of heavy chain (VH) and variable region of light chain (VL) cDNA library were constructed by RT-PCR. Human scFv templates for rabbit reticulocyte lysate ribosome display were assembled by primers and linker peptide (Gly4Ser)3. mRNA bound in antibody-ribosome-mRNA complexes was recovered using in-situ single primer RT-PCR, and three rounds of anti-IgE scFv DNA were enriched. The target DNA fragments were double enzyme digested and ligated into plasmid pET22b (+), followed by transformation in E. coli Rosseta (DE3). Positive clones were screened using clone PCR, Dot blotting and antigen ELISA. The correct lengths of VH (400 bp) and VL (710 bp) PCR products were obtained. The expected 1,000 bp ribosome display templates were also observed in agarose gel electrophoresis. After three rounds of ribosome display target sequences were effectively enriched, leading to a library of 10(13) members. Antibodies with the highest ELISA value for IgE were generated in the strain pET-IgE-6. A human anti-IgE scFv library was successfully constructed as described herein. Ribosome display using single primer in-situ RT-PCR as the recovery procedure effectively enriched target sequences. Anti-IgE scFv with high affinity and specificity were identified. The prepared human anti-IgE scFv fragment might be self-developed to a lead drug for treating asthma. Our study provides an alternative method for rapid discovery of human antibodies of therapeutic importance.

7.Adult prostate sarcoma: a clinicopathologic study of 15 cases.

OBJECTIVETo clarify the clinical and morphological features of adult prostate sarcoma (APS) and to further improve the knowledge and diagnostic accuracy for APS.
METHODSFifteen cases of APS were observed and analyzed on the clinical symptom, pathological features, treatment and prognosis.
RESULTSAge of onset ranged from 22 to 77 years (mean 46.3 years). The majority of cases were presented with dysuresia. By digital rectal examination and imaging of the prostate, APS was often identified as a large tumor mass. There were 6 cases of leiomyosarcomas, 6 embryonal rhabdomyosarcomas, and 3 fibrosarcomas in this series. Follow-up data were available for 12 cases: 7 cases died of the disease between 9 days and 360 days after surgery. Among 5 survived patients, 3 cases had recurrence after 2 to 24 months follow-up.
CONCLUSIONSAPS is a rare tumor that typically has clinical features: earlier age of onset, fast-appeared urinary tract symptoms, significant mass effects, and poor outcome. Level of prostate specific antigen (PSA) is usually normal or lower. Final diagnosis relies on the features of histology and immunohistochemistry expression profile.

8.A case-control study of an influenza A (H1N1) outbreak in a hospital.

OBJECTIVEThis study aimed to explore the epidemiological factors of an influenza A (H1N1) outbreak in a hospital.
METHODSGeneral data were collected via face-to-face interview and telephone survey. Total 132 individuals including medical and nursing staffs (37), in-patients (39) and patients' family members (56) who were exposed to the pediatric surgery ward during August 11 - 18, 2009, were investigated. The case group included 35 cases according to the diagnostic criteria for influenza A (H1N1). The other 97 persons were grouped as control. A case-control study was then conducted to explore the epidemic factors, and layering analysis was applied to determine the interactions among these factors.
RESULTSThe overall incidence in this study was 26.5% (35/132), which included 12 confirmed and 23 suspected cases, and there was no severe case. The first case was a child with the influenza-like symptoms before admission on August 11. The onsets of these cases were during August 7 - 17. The cases were distributed in 9 of 13 rooms, and there was no room aggregation in the cases distribution (χ(2) = 0.00, P > 0.05). Twelve of 25 oropharyngeal swabs were influenza A (H1N1) nucleic acid positive. The case-control study showed that exposure to the enema room accounted for 93.10% (27/29) in cases and 72.73% (48/66) in control; OR = 5.06, 95%CI = 1.01 - 34.23), long time exposure to ward was 71.43% (25/35) in cases and 44.33% (43/97) in control; OR = 3.14, 95%CI = 1.27 - 7.90), and short distance contact with the nurse LIU (76.46% (26/34) in cases and 50.52% (49/97) in control; OR = 3.18, 95%CI = 1.22 - 8.54) were the risk factors. However, keeping the window open (27.59% (8/29) in cases and 68.18% (45/66) in control; OR = 0.14, 95%CI = 0.05 - 0.39) and hand washing (25.71% (9/35) in cases and 76.29% (74/97) in control; OR = 0.11, 95%CI = 0.04 - 0.28) were the protective factors. The longer time exposure to ward had the higher risk (ratios of cases to control were 4:20 (0 - 1 day), 6:34 (2 - 4 days) and 25:43 (≥ 5 days); χ(2)(trend) = 5.737, P < 0.05). In contrast, hand washing with more frequencies (ratios of cases to control were 26:23 (0 - 1 time one day), 7:9 (2 - 3 times one day) and 2:65 (≥ 4 times one day); χ(2)(trend) = 37.136, P < 0.01) and the longer time window opening (ratios of cases to control were 21:21 (no), 4:13 (a few) and 4:32 (often); χ(2)(trend) = 13.830, P < 0.01) had the lower risk. Nevertheless, layering analysis excluded long time exposure to ward from the risk factors (for individuals with more frequent hand washing, 6.90% (2/29) exposed in cases, 7.14% (1/14) exposed in control, OR = 0.97, 95%CI = 0.06 - 29.51; for individuals keeping window open, 21.21% (7/33) exposed in cases, 8.33% (1/12) exposed in control, OR = 2.55, 95%CI = 0.26 - 60.87), indicating the main risk factors in this outbreak were exposure to the enema room and short distance contagion with the infected nurse.
CONCLUSIONThe influenza A (H1N1) outbreak in this hospital was induced by an inpatient infected with influenza A (H1N1) virus before admission. Infected medical staffs keeping on work and exposure to the same place, e.g.the enema room in this study might spread the influenza A (H1N1) virus, and frequent hand washing and keeping the window open are the most effective and economic methods to prevent influenza A (H1N1) infection.

10.Inflammatory pseudotumor-like follicular dendritic cell tumor of spleen.

OBJECTIVETo study the clinicopathologic features of inflammatory pseudotumor-like follicular dendritic cell tumor of spleen.
METHODSOne case of inflammatory pseudotumor-like follicular dendritic cell tumor of spleen was examined macroscopically and microscopically. Immunohistochemical study for CD21, CD23, CD35, clusterin, S-100 protein, vimentin, smooth muscle actin, CD1a, CD68, ALK protein, CD30, CD31, CD34, CD3 and CD20 was performed on formalin-fixed, paraffin-embedded sections by standard EnVision method. In-situ hybridization for Epstein-Barr virus (EBV)-encoded RNA was also carried out.
RESULTSMacroscopically, inflammatory pseudotumor-like follicular dendritic cell tumor was large in size, tan-colored, soft to rubbery in consistance and associated with central hemorrhage and necrosis. Histological examination showed scattered follicular dendritic cells admixed with abundant lymphocytes and plasma cells in the background, simulating inflammatory pseudotumor. On high-power magnification, the follicular dendritic cells possessed a moderate amount of pale to lightly eosinophilic cytoplasm, with indistinct cell borders. The nuclei were ovoid or spindly, with vesicular or stippled chromatin and small distinct, often centrally located, nucleoli. Some of the tumor cells showed nuclear pleomorphism and contained irregular foldings of nuclear membrane, coarse chromatin and prominent eosinophilic nucleoli. Mitotic figures were rarely identified. Immunohistochemical study showed that the tumor cells were positive for vimentin, clusterin, smooth muscle actin and CD68. They were weakly and focally positive for CD35 and S-100 protein, but negative for CD21, CD23, CD1a, ALK protein, CD30, CD31 and CD34. Most of the background lymphocytes were of T-lineage (CD3-positive) ,some were CD20 (B-cell marker)-positive. EBV RNA was demonstrated in the tumor cells by in-situ hybridization analysis.
CONCLUSIONSInflammatory pseudotumor-like follicular dendritic cell tumor is a rarely encountered low-grade malignancy with distinctive morphologic pattern. It is associated with EBV infection.
