2.Effects of ginkgo biloba extract 761 on contraction of gastrocnemius of fixed hindlimb in rats

Objective To study the preventive and therapeutic effects of ginkgo biloba extract 761 on disused skeletal muscle atrophy by observing its effects on the wet weight, protein content and contraction of the gastrocnemius of fixed hindlimb in rats. Methods 24 SD rats were divided into blank group, contrast group and experimental group. Their right hind limbs were fixed for 4 weeks with plastic plaster from metatarsus to 1 cm up the knee. The knee joint was fixed in 100? flexion and the ankle joint in 60? plantar flexion. Rats in the contrast group were perfused with 3mL distilled water, and rats in the experimental group were perfused with ginkgo biloba extract 761 (120 mg/kg, 14 mg/mL). The whole gastrocnemius muscle of the right hindlimb was dissected 4 weeks later to measure its contract tension, wet weight and protein content. Objective:To investigate the influence of p33~ING1b and wt-p53 on the biological activities of pancreatic cancer cells. Methods: Sense and anti-sense cDNAs of p33~ING1b were cloned into pcDNA3 eukaryotic expression vector separately, and the recombinant plasmids were solely or co-transfected with wt-p53 into pancreatic cell line SW1990. Flow cytometry was used to analyze the changes of cell cycle, Western blotting was used to evaluate the expression of p33~ING1b and p53 protein, and MG-P-MY staining was applied to observe cell apoptosis. 4.Growth activity of fetal liver stem cells in polyanhydride-three-dimensional vector-glucan

OBJECTIVE: To observe effects of polyanhydride-three-dimensional vector-glucan material on the fetal liver stem cell adhesion and proliferation.METHODS: The two-step collagenase perfusion digestion and bliquid percoll discontinuous density gradient centrifugation was used to isolate fetal liver stem cells. Fetal liver stem cells at the third passage were incubated on the polyanhydride-three-dimensional vector-glucan material. Inverted microscope was utilized to observe cell adhesion and growth status. Cell adherent rate, proliferation activity were calculated, and cell number was counted. Cell-vector was obtained for tissue section. Using hematoxylin-eosin staining, cell growth in the vector was observed under the optical microscope. At 7 days,immunofluorescence staining and flow cytometry were used to determine marker expression.RESULTS: Polyanhydride-three-dimensional vector-glucan promoted grow and adhesion of liver stem cells. There was the active function of the liver stem cells within carrier materials. In the three-dimensional surface and the internal culture, liver stem cell proliferation was sustained. After 10 days, the polyanhydride common culture-three-dimensional vector-glucan on stem cells was non-toxic, and human fetal liver stem cells could be attached to the polyanhydride-three-dimensional vector-glucan stent. The cell proliferation was better and dynamic sustained expression of markers. 7-days training received 19.7 percent increase in the number of cells.CONCLUSION: Polyanhydride-three-dimensional vector-glucan promotes the proliferation of liver stem cells, and liver stem cells can be used as the vector in liver tissue engineering.
6.Ecological Significance and Processes in Research of the Viable but Nonculturable State in Bacteria

The viable but noncuturable (VBNC) state is a survival strategy when bacteria are exposed to en- vironment stress. The VBNC state is part of the life cycle of non-differentiating bacteria, and it has a far-reaching impact on traditional bacteriology. Cells in the VBNC state fail to grow on the routine bacterio- logical media, here its significance in human health and environment science are detailed. Cells entering the VBNC state exhibit dwarfing and a number of metabolic changes in respiration rates and macromolecular synthesis. This paper summarized the variations in DNA and protein comparing to the culturable cells. We also discussed the ability of cells to resuscitate from the VBNC state and return to an actively metabolizing and culturable form. Some new methods for monitoring the VBNC state were listed. Finally the future was suggested.
7.Survey on the knowledge, attitudes and behavior for foodborne disease in medical staff

Objective To investigate the knowledge,attitude and behavior of medical staffs on foodborne diseases.Methods To investigate the knowledge,attitude and behavior of medical staffs on foodborne diseases by random sampling following the questionnaire.Results There were 332 medical workers attending the survey.The medical staffs were lack of knowledge for foodborne disease,percent of pass was 34.33%.The pass rate of attitude and behavior were 88.88% and 98.19%.The knowledge score of medical staff of top three hospital (6.49± 2.18)were lower than the community health service center(7.04±2.26) (P<0.05),but attitude and behavior score of this two staffs had not statistical differences(P>0.05).Conclusion It is necessary to help medical staff to improve the attitude of foodborne diseases,and improve the monitoring of underreporting case.
8.Correlation of video fluoroscopic swallowing study and clinical swallowing function of patients with post-stroke dysphagia

Objective To investigate the correlation between clinical presentations and the findings of video fluoroscopic swallowing study (VFSS)in patients with post-stroke dysphagia. Methods A total of 56 consecutive patients with post-stroke dysphagia admitted to Wenzhou Hospital of Traditional Chinese Medicine Affiliated to Zhejiang Chinese Medical University from June 2012 to May 2014 were enrolled. Four different kinds of food were selectively used to complete clinical assessment of swallowing function and VFSS respectively. The SPSS 20. 0 statistical software was used to perform univariate and multivariate Logistic regression analyses for all observed indexes of the 2 methods. Conclusion Some clinical manifestations are closely correlated with the swallowing dysfunction revealed by VFSS. With the help of this law,it may more simply and safely determine the degree and type of dysphagia in patients,and provide guidance for patients with dysphagia after stroke who can not accept VFSS in the rehabilitation treatment.
9.Changes of the biological behavior of dermal fibroblasts in Ⅲ skin burns wound in rats using chitosan

Aim To study the changes of the biological behavior of dermal fibroblasts in Ⅲ skin burns wound in rats using chitosan.Methods Wistar rats were randomly divided into 5 groups as follows:1% chitosan(W/V)group,2% chitosan(W/V)group,4% chitosan(W/V)group,bFGF(basic fibroblast growth factor) group and the control group.Rats were made for Ⅲ skin burns.The wound healing time was recorded,and the wound healing rate was calculated.Then the cell cycle and apoptotic dermal fibroblasts were determined and the amount of Hydroxyproline(HOP) in the skin tissue was analyzed.Results The wound healing rate of 4% chitosan(W/V) group was higher and the wound healing time of 4% chitosan(W/V) group was shorter than that of the control group.On the 7th,14th day post-injury,the content of protein of 4% chitosan(W/V) group was higher than that of the control group.The content of HOP of 2% and 4% chitosan(W/V) group was highest on the 7th day post-injury. Compared with that in control group,the percentage of cells of S stage in 4% chitosan(W/V) group was aboundant,and was reduced in apoptotic dermal fibroblasts.Conclusion The changes of cellular biological behaviors might be one of the mechanisms of that Chitosan could promote the wound healing of Ⅲ skin burns in rats significantly.
10.Analyzing Role of △~5 Desaturase in Biosynthesis of Arachidonic Acid Using Real Time PCR

Arachidonic acid is an essential fatty acid in human nutrition and a biogenetic precursor of the biologically active prostaglandins and leukotrienes. ?~5 desaturase catalyzes the ?~5 dehydrogenation of di-homo-?-linolenic acid to form arachidonic acid in biosynthetic pathway of arachidonic acid. Using real time PCR technology,the transcriptional expression levels of gene encoding ?5 desaturase in three Mortierella alpina strains M10,M6 and M23,and in different growth phase of high arachidonic acid yielding strain M6,were determined. Results showed that there was a distinct corelationship between mRNA transcript level of ?~5 desaturase gene and biosynthesis of arachidonic acid. Results indicated that ?~5 desaturase plays an important role in arachidonic acid biosynthesis.
