1.Diagnostic value of BNP and NT-proBNP for acute respiratory distress syndrome

Objective:To explore significance of brain natriuretic peptide (BNP)and N terminal pro brain natriuretic peptide (NT-proBNP)for diagnosis of acute respiratory distress syndrome and its guidance of treatment.Methods:A total of 124 cases with definite organic heart disease and sudden respiratory distress syndrome (measurement group) received measurement of BNP and NT-proBNP to judge whether they suffered from heart failure or not.Another 110 patients with acute respiratory distress syndrome but no receiving BNP and NT-proBNP measurement were en-rolled as no measurement control group.Relevant data,including diagnosis time,length of hospital stay,hospitali-zation cost and mortality rate etc,were collected in all patients.Results:Compared with no measurement control group,there were significant reductions in diagnosis time [(24.2±6.4)min vs.(16.3±5.2)min],length of hospi-tal stay [(12.5±3.5)d vs.(8.5±4.5)d]and mortality rate (8.18% vs.4.84%)in measurement group,P<0.05 all;there was no significant difference in mean hospitalization cost between two groups (P>0.05).Conclusion:Measurement of brain natriuretic peptide and N terminal pro brain natriuretic peptide possesses important value for early diagnosis,elevating therapeutic effect and improving prognosis in patients with acute respiratory distress syn-drome.
2. Role of PTEN gene in multidrug resistance reversal in leukemia cells

Objective: To investigate the mechanism by which wild-type PTEN gene reversing multi-drug resistance (MDR) in human leukemia K562/ADM cells resistant to adriamycin (ADM). Methods: The recombinant adenovirus containing green fluorescent protein and PTEN (Ad-PTEN-GFP) or empty vector (Ad-GFP) was transducted into K562/ADM cells resistant to ADM. The transduction efficiency was assessed by flow cytometry (FCM). Then the cells were treated with different concentrations of ADM, cytarabine (Ara-C) or arsenic trioxide(As203) 3 days after transduction. The proliferation of K562/ADM cells was examined by MTT assay, the apoptosis rate was assessed by FCM, and the IC50 of different drugs was used to calculate the drug resistance reversal fold (RF), so as to observe the effect of PTEN on reversing MDR of the 3 drugs. PTEN, NF-κB, MDR1, MDR-associated protein (MRP) and apoptosis related genes (Bcl-2, Bcl-xL, Bax) were detected by fluorescence quantitative PCR. PTEN, Akt, p-Akt and NF-κB protein levels were detected by Western blotting analysis. Results: The proliferation inhibition rate and apoptosis rate of cells in Ad-PTEN-GFP plus chemotherapeutic groups were significantly higher than those Ad-GFP plus chemotherapeutic groups at 3 days after infection (MOI: 200) (P<0. 05). PTEN transduction promoted the sensitivity of K562/ADM cells to ADM, Ara-C and As203, with the RF being 3. 80, 2. 65 and 2. 64 folds, respectively. K562/ADM cells in Ad-PTEN-GFP group had lower p-Akt and NF-κB (P65) protein levels and lower NF-κB, MDRl, Bcl-2 and Bcl-xL mRNA levels, and up-regulated Bax mRNA level compared with those in Ad-GFP group. Conclusion: Wild-type PTEN gene may reverse drug resistance via inhibiting Akt pathway and regulating its downstream signaling molecules, such as NF-κB, MDR1, Bcl-2 and Bax.

4.Study on the Effect of Biological Reductive Dechlorinationof 2,4-Dichlorophenol by Zero-valent Iron

A batch laboratory test was conducted to examine the effect of biological reductive dechlorination of 2,4-Dichlorophenol(2,4-DCP) by the addition of zero-valent iron(Fe0) in the anaerobic system, through inoculating the anaerobic mixed microorganism acclimated for two months. Meanwhile, several factors that affected "Fe0+ microbe" system were also being discussed. The results showed that, "Fe0+ microbe" system accelerated the biological dechlorination of 2,4-DCP effectively compared to the individual use. The opti-mum quantity of added Fe0 and inoculation was 0.5 g/L and 376.2 mgVSS/L in the combined system respec-tively. It showed the most effective transformation efficiency for 2,4-DCP when initial pH=8.0, whereas it become weaker when initial pH are keeping in acid condition. There existed a proportion between quantity of added Fe0 and inoculation. It enhanced degradable effect of 2,4-dichlorophenol when increased the quan-tity of inoculation at suitable ranges, which generated more enzyme or enzymatic series degraded pollutant.
5.Clinical observation of elderly patients with coronary heart disease undergoing coronary artery bypass grafting through descending thoracic aorta

Objective To investigate the clinical effectiveness of coronary artery bypass grafting through descending thoracic aorta in elderly patients with coronary heast disease and to decrease the post-operative complication.Methods Thirteen elderly patients underwent coronary bypass surgery with minimally invasive direct coronary artery bypass (MIDCAB).Age range from 70 to 82 years with a mean of(72.1?6.0)years.Patients suffered from multi vessel disease.Many minimally invasive techniques of“Y”blood vessel graft anastomosis,anastomosis of blood vessel graft to descending aorta,minimally invasive direct,thoracoscope assist were used.Results All patients were survived.The mean duration of intubation was (6.9?0.9) hours.The average ICU stay was (2.5?0.5)days.No patients received blood transfusion.During the short-term follow-up(3 to 14 months) patients had no complaint of angina,Conclusions The technique of“Y”blood vessel graft anastomosis,descending aorta blood vessel graft,minimally invasive direct and thoracoscope assist in combination with coronary artery bypass grafting is a safe and cost-effective new procedure for elderly patients with multi-coronary artery disease.
6.The clinical study on the surgical treatment of thoracic aortic aneurysm associated with coronary artery disease.

OBJECTIVETo analyze the factors which influence the safety and prognosis of aorta replacement combined with coronary artery bypass grafting (CABG) for thoracic aortic aneurysm associated with coronary artery disease.
METHODSFrom May 1982 to October 2002, 67 patients with thoracic aortic aneurysm were admitted, and 24 of them combined with CABG. Of the 24 patients, 9 received descending aorta replacement combined with CABG, and the other 15 received the ascending aorta replacement combined with CABG. The treatment results were compared with the other 43 patients only undergoing the thoracic aortic replacement.
RESULTSThe mortality rate of the patients with aorta replacement combined with CABG was 13% (3/24). Though the descending aorta replacement combined with CABG could make the cardiopulmonary bypass time and selective cerebral perfusion time longer, (278 +/- 54) min and (188 +/- 59) min respectively, no significant difference was observed in postoperative complications, 3-year survival rate, 3-year-cardiac-event-free rate compared with the patients only undergoing the thoracic aortic replacement (P > 0.05).
CONCLUSIONSThe aorta replacement combined with CABG can be performed safely, and the revascularization for coronary artery disease is useful for preventing occurrence of cardiac events.

7.Effect of jiangu erxian pill on proliferation and cell cycle of human osteoblast.

OBJECTIVETo further explore the effect of Jiangu Erxian Pill (JGEXP) on proliferation and cell cycle of human osteoblast on the basis of previous clinical and experimental studies.
METHODSHuman primary osteoblast were isolated and cultured. The cell proliferation was tested by 3H-thymine incorporation and methyl thiazolyl tetrazolium (MMT) method and the cell cycle was determined by flow cytometry technique.
RESULTSIn the medium and high dosage JGEXP groups, the cell proliferation rate and index, and percentage of diploid synthesis phase (S phase) cells were significantly higher than those in the blank control group (P < 0.01 or P < 0.05), and similar to those in the estrogen group; and the cell apoptosis rate and percentage of G0-G1 stage cells were lower than those in the blank control group (P < 0.01 and P < 0.05).
CONCLUSIONJGEXP could effectively promote the cell proliferation and differentiation, and prevent the cell apoptosis of osteoblast in vitro.

8.Expression of gastrin and gastrin releasing peptide in patient's with gastric cancer by using tissue chip technique.

OBJECTIVETo study the expression of gastrin(GAS) and gastrin releasing peptide(GRP) in patients with gastric cancer and investigate the clinical significance.
METHODSThe expression of GAS and GRP in sixty patients with gastric cancer was detected by using tissue chip technique and immunohistochemical methods.
RESULTSThe positive rates of GAS and GRP were 30.0% and 11.7% respectively in 60 cases with gastric cancer. The positive rates of GAS and GRP were higher in moderately and poorly differentiated cancers than those in well differentiated cancer (P< 0.05). The positive rates of GAS and GRP were significantly higher in mucinous adenocarcinoma and signet-ring cell carcinoma than those in other types of gastric cancer (P< 0.05). The positive expression of GAS and GRP in gastric cancer was correlated with lymph node metastasis (P< 0.05).
CONCLUSIONTissue chip technique is a feasible,rapid,economic and accurate approach for screening clinical tissue specimens on a large scale.

9.Endoplasmic reticulum stress promotes the apoptosis of testicular germ cells in hyperlipidemic rats.

OBJECTIVETo investigate the role of endoplasmic reticulum stress in the apoptosis of testicular germ cells in hyperlipidemic rats.
METHODSWe randomly assigned 42 four-week-old male Wistar rats into a normal control group (n = 12) and a high-fat group (n = 30) to be fed on a normal diet and a high-fat diet, respectively, for 10 weeks. Then we measured the concentrations of triglyceride (TG) and total cholesterol (TC) in the serum using an automatic biochemistry analyzer, detected the apoptosis of testicular germ cells by TUNEL staining, and determined the protein and mRNA expressions of GRP78 and. caspase-12 in the testis tissue by immunohistochemistry and RT-PCR, respectively.
RESULTSThe concentrations of TG and TC were significantly increased in the animals of the high-fat group ([3.00 ± 0.92] and [3.04 ± 0.39] mmol/L) as compared with the control rats ([1.43 ± 0.41] and [1.55 ± 0.23] mmol/L) (P < 0.01), and so was the apoptosis index of the testicular germ cells ([37.17 ± 2.74]% vs [5.16 ± 0.81]%, P < 0.01). The high-fat group, in comparison with the control, also showed remarkably upregulated protein and mRNA expressions of GRP78 (0.32 ± 0.03 and 0.86 ± 0.05 vs 0.19 ± 0.01 and 0.37 ± 0.03, P < 0.01) and caspase-12 (0.34 ± 0.02 and 0.87 ± 0.01 vs 0.12 ± 0.01 and 0.34 ± 0.03, P < 0.01) in the testis tissue.
CONCLUSIONThe apoptosis of testicular germ cells is increased in hyperlipidemic rats, which may be attributed to endoplasmic reticulum stress.
