6.High-sensitive Detection Method for Siderophores from Pseudomonas

CAS (Chrome azurol S) assay was a universal method for detecting the bacterial siderophores, and Sugar-Asn liquid medium has been applied to the studies on siderophores from Pieudomonas. In this paper, Asp has been substituted for Asn, and MSA-CAS agar plate was developed by integrating the MSA (sugar-Asp) medium and CAS bright blue dye, which has been used in the universal CAS assay. On the aspect of siderophores detection , 8 strains of 7 species from Pseudomonas had been screened on MSA-CAS agar plates and universal CAS assay respectively. The results showed that MSA-CAS agar was higher-sensitive and lower basic fluorescent than universal CAS assay.
7.Improvement and Analysis in Over- siderophores Production Bacteria Filtrating and Detecting

A novel stable blue agar plate which is convenient to preparing and more effective than the universal chrome azurol sulfonate (CAS) assay established by Schwyn and Neilands was designed by replacing MM9 growth medium and pipes with certain concentrate of phosphorus buffer solution which pH could be stabled at 6.8. It is more suitable for screening over- siderophores production bacteria. Since OD_ 630 of the sample is usually out of the range of spectrophotometer with CAS assay solution when quantifying the siderophores and the outcome is not steady,the measuring wavelength had been changed to 680 nm corresponding to the middle of max absorbance and the correlation between siderophores concentrations and OD was unchanged. But the detecting sensitivity is elevated by enlarged the absorbance differences among samples with different productivity of siderophores at 680 nm .

Objective To investigate the difference of tumor cell proliferation between the center and the edge tissues of human colon cancer and its relationship with the expression and methylation of p16 and p14 gene.Methods The expression of Ki67,P14 and P16 protein was assessed by immunohistochemistry,methylation status and mRNA expression of p16 and p14 gene in different area of colon cancer tissue were analyzed by methylation-specific PCR.RT-PCR and microdissection technique. 9.Comparisons of endoscopic and pathological characteristics between long and short segment Barrett's esophagus

Objective To investigate the similarities and differences of endoscopic and pathological char- acteristics between long and short segment Barrett's esophagus.Methods One hundred and twenty-eight cases of Barrett's esophagus identified both by endoscopy and pathology were enrolled in this retrospective study. Among them,40 cases were long segment Barrett's esophagus (LSBE) and 88 were short segment Barrett's esophagus (SSBE).The age distribution,sex distinction,endoscopic manifestations and pathological changes were assessed.Data were statistically analyzed by t-test or u-test.Results There were no differences in age distribution and sex distinction between LSBE and SSBE groups (P>0.05).The ring pattern was the most prominent type accounting for 62.5% in LSBE group.The island pattern was the most prominent type accounting for 85.2% in SSBE group.There were significant differences in the rates of specialized intestinal metaplasia between LSBE and SSBE groups(47.5% vs 14.8%,P<0.01).Moreover,among the special- ized intestinal metaplasia,low grade (15.0% vs 4.5%),medium grade (12.5% vs 3.4%) and high grade dysplasia (5.0% vs 0.0%) between LSBE and SSBE groups also had statistical differences (all P<0.05).Conclusions LSBE may have more tendency in dysplasia than that of SSBE.We should pay attention to the importance of endoscopic manifestations and pathological diagnosis.
10.Ursolic acid induces apoptosis in colon cancer HT-29 cells.

OBJECTIVETo study the antitumor effects of ursolic acid and mechanisms of its action.
METHODSThe cells of human colorectal carcinoma cell line HT-29 were treated with ursolic acid at different concentration. The proliferation inhibition was examined by MTT assay. Morphological examination, TUNEL method and flow cytometry were used to detect apoptosis. Immunohistochemical method was used to detect the expression of apoptosis related genes caspase-9 and bcl-2. The semi-quantification of protein expression was analyzed by pathological image-analysis.
RESULTSUA inhibited the proliferation of HT-29 cells moderately. Apoptosis of HT-29 cells was induced by ursolic acid treatment. The morphology of HT-29 showed changes such as nuclear chromatin condensation and fragmentation. Sub-G(1) peak was found by flow cytometry. The maximal apoptosis rate was 11.63%. The expression of caspase-9 gene was enhanced. The expression of bcl-2 gene was decreased. All these effects were in a dose-dependent and time-depend manner.
CONCLUSIONApoptosis in colon cancer HT-29 cells is one of the key mechanisms of ursolic acid action and its antitumor activity may be applicable for the treatment of cancers.
