1.Regulation of endometrial stromal cells through the Wnt/beta-catenin signaling pathway

BACKGROUND:Increasing number of genes and signaling pathways are involved in regulating stem cels periodicaly, and wherein Wnt/β-catenin signaling pathway is an important pathway of stem cels.
OBJECTIVE:To investigate the effects of Wnt/β-catenin signal pathway on mouse endometrial stromal cels.
METHODS:After injection of Wnt/β-catenin pathway inhibitor or activator, endometrial tissues from Balb/c mice were obtained, some of which were used for detection of invasion of endometrial stromal cels and western blot detection, and the rest of which were used for preparing animal models of endometriosis folowed by immunohistochemical detection.
RESULTS AND CONCLUSION:The expression of β-catenin and GSK3β proteins was significantly higher in the activator group than the inhibitor group (P < 0.05). The number of transmembrane cels was significantly higher in the activator group than the inhibitor and control groups (P < 0.05). Immunohistochemical findings showed positive expression of E-cadherin in ectopic endometrial tissues of the inhibitory group, and strongly positive expression of vascular endothelial growth factor in ectopic endometrial tissues of the activator group. These findings indicate that Wnt/β-catenin signaling pathway may cause endometriosis by strengthening ectopic endometrial implantation, invasion and metastasis.
2.MiR-18 a and estrogen receptor alpha expression in single and multiple uterine fibroids

Objective To detect the expression of miR-18a and estrogen receptor alpha (ER alpha) in single and multiple uterine flesh tumor tissues, discuss the relationship between miR-18a and ER alpha, and their effect in single and multiple uterine fibroids.Methods The expression of miR-18a and ER alpha in single and multiple uterine fibroids tissue paraffin section were detected by in situ hybridization method and immunohistochemical method, respectively.And the correlation between the miR-18a and ER alpha were evaluated.Results The expression of ER alpha in multiple uterine fibroids group was significantly higher than that of single uterine fibroids tissues (P<0.05);while miR-18a was weaker than that of single uterine fibroids tissues(P <0.05).The correlation results showed that miR-18a expression was correlated negatively with ER alpha expression either in single(r =-0.4421) and multiple uterine fibroids(r =-0.4181).Conclusion The expression of miR-18a is low in multiple uterine fibroids, while ER alpha had high expression.miR-18a could bea new target for the treatment of multiple uterine fibroids.
3.Nutrition Status of Premature Neonates in Early Postnatal Stage in Neonatal Intensive Care Unit

Objective To study the nutrition status of premature infants in the early postnatal stage.Methods According to the physical development standard,the premature neonates for treatment in our NICU from Jun.2007 to Jun.2009 were enrolled and divided into normal growth group and extrauterine growth restriction(EUGR) group.The nutrition status of these premature neonates were evaluated respectively.SPSS 13.0 software was used to analyze the data.Results Compared with EUGR group,the durations of regaining birth weight were shorter in normal growth group(t=-13.83,P0.05).Conclusions Premature neonates are prone to encounter extrauterine growth retardation.Extrauterine growth retardation had great impact on premature neonates.Premature neonates should be strengthened with nutrition supplement in order to reduce the incidence of extrauterine growth retardation.
4. Edaravone inhibits pain sensitivity through decreasing pJNK expression in dorsal root ganglia and spinal cord in rats with spinal nerve ligated

Objective: To investigate the effect of edaravone on the pain sensitivity in rats with spinal nerve ligated and to probe into the related mechanism. Methods: Male adult SD rats were randomly divided into 3 groups: a sham (Sham) group, a spinal nerve ligation (SNL) group and edaravone(Eda) group. The paw withdrawal mechanical threshold(PWMT) was measured before and after ligation (once daily for 7 days). Rats were sacrificed at specified time points and the left(operation side) L4 and L5 dorsal root ganglia(DRG) and the right (control side) L5 DRG were obtained and immunostained to observe the changes of pJNK in DRG neurons and spinal cords, so as to observe the effect of edaravone on pJNK. Results: Edaravone can reduce the mechanical hyperalgesia induced by spinal nerve ligation. Immunostaining showed that the SNL group had an increased pJNK in the ipsilateral DRG neurons (L5) 24 hours after ligation; double immunofluorescence indicated that the expression of pJNK in the ipsilateral spinal astrocytes was increased 3 days after ligation. Edaravone can reduce pJNK expression in DRG neurons and spinal cords at corresponding time points. Conclusion: Edaravone can relieve the neuropathic pain induced by spinal nerve ligation, and the mechanism might be related to the inhibition of pJNK expression in DRG neurons and spinal cords.
5. Role of transient receptor potential vanilloid 1 channel in pancreatitis

Transient receptor potential vanilloid 1 (TRPV1) is a non-selective cation channel, which can be activated by multiple pathways during the course of the diseases. Recent studies indicate that primary sensory neurons of the pancreas express TRPV1 receptor and the activation of TRPV1 receptor promotes pancreatic inflammation. Moreover, blockade of these transient receptor potential channels can greatly ameliorate the pain response in experimental pancreatitis.
6.Effect of Temperature on Lycopene Prodmction by Recombinant E.coli and Control Strategy

Batch fermentation of lycopene by recombinant E. c ol i ZYL-2 at various temperatures ranging between 23℃~37℃ were studied in 7L fermentor. At 33℃, cell specific growth rate in earlier stage of culture was higher, and the time obtained maximum cell dry weight was shorter at other tempe rature. while after 9 h, lycopene specific production rate was higher at 28℃. Based on these results, a two-stage temperature control strategy was developed in which 33℃ was used for fermentation for the first 9 h and the temperaturew as switched to 28℃ after 9h. Using this temperature-shift strategy, the maxim al lycopene content and productivity reached 605.25?g L-1 and 28.82?g L-1h-1. The lycopene fermentation level obtained by the strategy was higher than those in single temperature-control experiments.
7.Factors affecting the recovery of spinal functions in patients with spinal metastatic carcinoma

BACKGROUND: Tumor spinal metastasis is often complicated with pain,limitations in movement, paralysis and poor prognosis.OBJECTIVE: To investigate the factors affecting the recovery of the spinal functions in patients with spinal metastatic carcinoma and its therapeutic methods so as to improve the living quality of the patients with a terminal cancer.DESIGN: A case-controlled observation.SETTING: The Department of Tumor and Department of Radiotherapy of the Affiliated Hospital of Guangdong Medical College.PARTICIPANTS: We selected 162 patients with spinal metastatic carcinoma with different degrees of paralysis who received treatment at the Department of Oncology and the Department of Radiotherapy of the Affiliated Hospital of Guangdong Medical College between March 1992 and June 2001. They participated in this experiment voluntarily.METHODS:①Radiotherapy and chemotherapy; All the patients were given symptomatic treatment with simple radiotherapy or a combined therapy of simple radiotherapy and chemotherapy, in which the dose of chemotherapy was 2 Gy/time,5 times a week,3 weeks in total. A combined method was used for chemotherapy according to the nature of the different primary tumors. The chemotherapy was conducted for 3 therapeutic courses in average ranging from 2 to 6 courses of treatment with 21 days for each course. ② Physical rehabilitation therapy included exercise therapy, massage (twice/day), functional electrical stimulation (once a day),bladder training and toilet training (once or twice every day), 10 days as a therapeutic course, 3 successive therapeutic courses in total. After the treatment, analgesic pain, muscle power of the limbs and the recovery of defecation function were observed.MAIN OUTCOME MEASURES: Pain-alleviating, muscle power of the limbs and the recovery of defecation function after the operation. RESULTS: Totally 162 patients with spinal metastasis carcinoma were recruited, and all the patients entered the stage of result analysis at the end of therapy with no loss in the midway. ① Pain complete alleviating was in 97 cases (59.88%); no alleveclting in 0 case. ② The condition was remarkably improved in 127 cases(78.40%), and it was not improved in 35 cases (21.60%). The improved rate of muscle power of the patients with one focus and 2 adjacent focuses was 92.98%. The improved rate of the patients with multiple foci was 64.59%. The muscle power of the patients with a disease history of less than 2 weeks was improved remarkably, but there was no change in muscle power in patients with a disease history of more than 2 months. CT or MRI showed that spinal cord at the affected segment of the 21 patients suffered severely compression before the treatment, and only 2 cases were improved in muscle power, with no other changes.③The defecation function of 162 patients all became normal.CONCLUSION: Patients with spinal metastatic carcinoma complicated with paralysis have different degrees of relief and amelioration in pain,muscle power of the limbs and defecation function after radiotherapy,chemotherapy and synthetic rehabilitation therapy. Factors affecting the recovery of spinal function mainly included the length of disease course,compressed degree of the spinal cord, the quantity of metastatic focus, the health status of the patients and therapeutic time and the quality of primary rehabilitation and so on. Mental help to the patients should be paid attention to so as to improve the quality of life at the same time.

9.Analysis of early warning indicators of tuberculous peritonitis in patients with cirrhotic ascites

Objective To analyze the early warning indicators of tuberculous peritonitis (TBP)in patients with cirrhotic ascites.Methods A retrospective analysis was performed on the clinical data of 62 patients with cirrhotic ascites hospitalized in the Second Affiliated Hospital of Chongqing Medical University from January 2010 to December 2012.The chi-square test and Mann-Whitney U test were used for uni-variate analysis,while the logistic regression model was used for multivariate analysis.Results There were significant differences between TBP group and non-TBP group in sex,history of primary peritonitis,erythrocyte sedimentation rate (ESR),serum tuberculosis antibody, percentage of lymphocytes in ascites (≥50%),and prothrombin activity (PTA)(P<0.05 for all).The odds ratios (ORs)of history of primary peritonitis,PTA,ESR,and percentage of lymphocytes in ascites (≥50%)for development of TBP in patients with cirrhotic ascites were 1.933,3.205,2.716,and 11.701,respectively (P<0.05 for all),and the areas under the receiver operating characteristic curve were 0.831,0.815,0.775,and 0.935,respectively (P<0.05 for all).Conclusion History of primary peritonitis,PTA,ESR,and per-centage of lymphocytes in ascites (≥50%)are the early warning indicators of TBP in patients with cirrhotic ascites;these early warning in-dicators,especially percentage of lymphocytes in ascites (≥50%),have good predictive values.
10.Measurement of exopolysaccharides produced by Streptococcus mutans from caries-free and caries-active children

Objective: To compare the quantities of water-soluble, water-insoluble glucans synthesized by S. mutans in 3-4 years old children with high dmft and no caries. Methods: The quantities of water-soluble and water-insoluble glucans synthesized by S. mutans from dental plaques of children with high dmft (dmft≥5) and caries-free (dmft=0) were measured using the anthrone method. Results: S. mutans isolated from the children with high dmft had stronger ability in producing both water-soluble and water-insoluble glucans than those isolated from the caries-free children (0.0332 mg/ml v.s. 0.0192 mg/ml and 0.0357 mg/ml v.s. 0.0215 mg/ml respectively). Children with high dmft harbored more isolates with higher water-soluble or water-insoluble glucans producing ability than caries-free children. Water-insoluble glucans produced by all the clinical isolates is higher than water-soluble glucans (0.0301 mg/ml v.s. 0.0267 mg/ml) and the difference was statistically significant. Conclusion: There may be a correlation between the extracellular glucans producing ability of S. mutans and the caries susceptibility in deciduous teeth.
