Objective To investigate the effect of local delivery of paclitaxel on bile duct scar formation and biliary stricture in guinea pig. Methods Models of end-to-end anastomosis of the common bile duct were established in 42 guinea pigs. The animals were randomly divided into paclitaxel and control groups. In the paclitaxel group, the drug solution(1000 ?mol/L,0.05 ml) was smeared onto the anastomotic wound before closing the abdomen. The animals were killed in 3 days,2 weeks, and 1 month after the operation(7 in each group at each time point). The sepcimens of the common bile duct were obtained for histological examination.Transmission electron microscopy was employed to observe the ultrastructure of paclitaxel-induced apoptotic cells. Results Histological examination showed inflammatory reaction and hyperplasia of the bile duct wall. The bile duct wall of the paclitaxel group was significantly thiner than that of the control group in 3 days and 1 month after the operation [3 days:(574.41?24.53)?m vs(802.98?24.42) ?m,t=-6.604,P=0.000;1 month:(1383.36?36.64) ?m vs(1518.56?34.89)?m,t=-2.672,P=0.020].Paclitaxel-induced cell apoptosis and injured cell organs were found by transmission electron microscopy. Conclusion Single local delivery of paclitaxel can inhibit biliary stricture in one month.

It is a common phenomenon that medical research is out of line with clinical work at present. In fact, there is intrinsic interrelation between clinical work and research. Thinking and exploration at clinics are part of the research, and any research based on chnical work not only has great value, but also receives strong support from the state. However, to translate clinical research into clinical practice is difficult, which requires evidence-based methodology, a knowledgable team with persistence andtechnologies, et al.

Objective To investigate the effects of total flavonoids of Herba Epimedii(HEF)on the metabolism of typeⅠcollagen and the expression of cathepsin K in the bone of ovariectomized(OVX)rats. Methods Fifty-four female SD rats were allocated into 6 groups;OVX group, sham operation group,OVX rats followed by three doses of HEF(40,80 and 160 mg?kg~(-1)?d~(-1))and nilestriol(0.1 mg?kg~(-1)?d~(-1))for 12 weeks respectively.Bone mineral density(BMD)of whole body was determined by dual-energy X-ray absoptiometry. The level of cross-linked N-telopeptide of typeⅠcollagen(NTx)in the urine were determined by ELISA.The amounts of typeⅠcollagen protein and cathepsin K protein in bone tissue were detected by immunohistochemical method and Western blotting. Results Compared with OVX group, the total BMD values in the HEF treated groups were increased(all P<0.05),and the expression levels of typeⅠcollagen in three HEF treated groups rose significantly in a dose-dependent manner after 12 week, and simultaneously, both the expression of cathepsin K in bone and the level of NTx/Cr were reduced markedly(P<0.05),being most significant(P<0.01)in the group treated with the highest dose of HEF(160 mg?kg~(-1)?d~(-1)).Conclusion HEF seems to be able to elevate BMD and improve bone quality of rats via promoting synthesis and inhibiting proteolysis and absorption of typeⅠcollagen in the bone.

Recently, study on cencer stem cells has been a focus of study.Cancer stem cell is a small population of cencer cells possessing the properties of stem cells:self-renewal,differentiation and proliferation. To date, the existence of cancer stem cells has been proven in acute and chronic myeloid leukemia, breast cancer, brain tumors, liver cancer and colon cancer,etc..In this article we reviews the current progress on cancer stem cells, including the defination, existing evidence, research methods, and challenges in clinical application.

By the breeding model of physics mutagenests (60Co,UV light) and re-strong in the body of insects, We obtained a Bt strain Bt99985 with high insecticidal activity. The shake flask and 40t tank fermentation tests showed that, this strain showed special activity against Spodoptera exjgua, Comparing with original Bt strain: 93005, CK strain: HD-1-580 and GC-91, the value of potency increased respectively by 429%, 655% and 114% in shake flask test. Comparing with GC-91, the value of potency increased by 180% in 40t tank fermentation test.

Background The main mechanism of diabetic retinopathy (DR) is the formation of retinal neovascularization. Vascular endothelial growth factors (VEGF) is primarily factor for DR. Objective The goal of this study was to investigate the effects of angelicae sinensis decoction for supplementing blood ( ASD), a kind of traditional prescription of Chinese herbal medicine, on vascular endothelial growth factor (VEGF) level in vitreous and serum of STZ-induced type 2 diabetes mellitus. Methods Thirty-four SPF male SD rats were fed using high fat diet for 8 weeks and then the streptozocin of 25 mg/kg was intraperitoneally injected to create the type 2 diabetic models. ASD (3.6 g/kg) and 10 mg/kg Gli of 5 mi were administered intragastrically from the 4th day of high fat die though 8 weeks respectively. Other 10 matched rats were used as normal control group. The body weight, blood glucose level ,triglycerides (TG) and high density lipoprotein (HDL) were detected in 4 and 8 weeks after usage of drug. The vitreous and serum samples were obtained in the 8 th weeks for the VEGF detection by enzyme-linked immunosorbent assay (ELISA). This experiment comlied with the Regulations for the Administration of Affair Concerning Experimental Animals by State Science and Technology Commission. Results The body weight and HDL in DM group were significantly lower than those of normal control group in 4 or 8 weeks ( P<0. 01 ), but in ASD and Gli groups ,the body weight and HDL were higher than those of the DM group (P<0. 05 ). the blood glucose level and TG were elevated in DM group compared with normal control group (P<0. 01 ), but those in ASD group and Gli group were decreased in comparison with DM group ( P<0.05 ). EIISA revealed that no significant differences were found in VEGF levels between normal control group and ASD group or Gli group (P>0. 05 ), but in DM group, the serum VEGF level (147.65±4. 55 ng/L) was reduced and vitreous VEGF level (673.45± 100. 85 ng/L) was raised, showing the considerable differences (P< 0.05 ). No any correlation was found between serum VEGF level and vitreous VEGF level (r=0.314,P>0. 05). Conclusion ASD can decrease the VEGF level in vitreous, implying that ASD postpone the occurrence of diabetic retinopathy in DM rat.
