China Journal of Chinese Materia Medica 2015;40(22):4384-4388
Licorice seedlings were taken as experimental materials, an experiment was conducted to study the effects of exogenous sucrose on growth and active ingredient content of licorice seedlings under NaCl stress conditions. The results of this study showed that under salt stress conditions, after adding a certain concentration of exogenous sucrose, the licorice seedlings day of relative growth rate was increasing, and this stress can be a significant weakening effect, indicating that exogenous sucrose salt stress-relieving effect. The total flavonoids and phenylalanine ammonia lyase (PAL) activity were significantly increased, the exogenous sucrose can mitigated the seedling roots under salt stress, the licorice flavonoid content in the enhanced growth was largely due to the activity of PAL an increased, when the concentration of exogenous sucrose wae 10 mmol x L(-1), PAL activity reaching a maximum, when the concentration of exogenous sucrose was 15 mmol x L(-1), PAL activity turned into a downward trend, the results indicating that this mitigation has concentration effect. After applying different concentrations of exogenous sugar, the contents of liquiritin changes with the change of flavonoids content was similar. After applying different concentrations of exogenous sucrose, the content of licorice acid under salt stress was higher than the levels were not reached during salt stress, the impact of exogenous sucrose concentration gradient of licorice acid accumulation was not obvious.

3. Expression of hypoxia inducible factor-1α in HepG2 cells cultured under hypoxia and its relationship with vascularization and apoptosis-related protein
Academic Journal of Second Military Medical University 2010;27(12):1328-1332
Objective: To investigate the expression of hypoxia inducible factor-1α(HIF-1α) in HepG2 cells cultured under hypoxia for different time periods, and to assess its relationship with vascularization and cell apoptosis. Methods: HepG2 cells were cultured in a medium containing cobalt chloride (125 μmol/L) for different time periods (0, 1, 2, 4, 6, and 8 hours). RT-PCR was used to measure the expression of HIF-1α mRNA, VEGF mRNA, and bcl-2 mRNA, and bax mRNA at the above time points; Western blot was used to determine the expression of Caspase-3 protein and the activity of Caspase 3 enzyme. Results: After 1-2 hour culture under hypoxia, the expression of HIF-1α mRNA, VEGF mRNA, and bcl-2 mRNA began to increase gradually, peaked at 4 h, and then decreased, but were still higher than that before culture. There was no obvious change in the expression of bax mRNA and the ratio of bcl-2 to bax mRNA expression had a similar change with that of bcl-2 mRNA expression. The changes of Caspase-3 protein/mRNA and Caspase-3 enzyme activity were contrary to that of HIF-1α. Conclusion: HIF-1α may inhibit the apoptosis of hepatic cancer cells through regulating the expression of VEGF, bcl-2, bax, and Caspase-3, and the inhibition is related to the severity of hypoxia.

6. Taste comparison of unprocessed and processed Siegesbeckiae puescens based on electronic tongue
Chinese Traditional and Herbal Drugs 2017;48(4):673-680
Objective: To study the taste of unprocessed and processed Siegesbeckiae puescens (SP) and establish a taste discrimination model. Methods: The sour, bitter, astringent, salty, and sweet values of 12 batches of unprocessed and processed SP were measured by electronic tongue using a paired t-test, principal component analysis (PCA), and the linear discriminant factor analysis (LDA) to study the changes of the taste of unprocessed and processed SP. Results: The paired t-test showed that the bitterness and saltiness of processed SP decreased; Astringency, sweetness, and sourness did not significantly change. The PCA can distinguish unprocessed and processed SP. The model of the taste of unprocessed and processed SP was established through the LDA, and conduct cross validation; Its correct rate reached 100%. The fingerprint of taste of unprocessed and processed SP was established through radar-graph. Conclusion: Electronic tongue can identify sour, bitter, astringent, salty, and sweet values of traditional Chinese medicine accurately. It can reflect the numerical change of taste of unprocessed and processed SP combining with statistical method. Based on it, it can be inferred that the relationship between the change of taste and clinical efficacy before and after processing of SP. Besides, the discriminant model can be used to distinguish unprocessed and processed SP.

OBJECTIVE:To investigate the effects of Xuebijing injection on inflammatory factors, oxidation stress level and thrombosis risk in patients with acute non-ST elevation myocardial infarction(NSTEMI). METHODS:A total of 130 NSTEMI pa-tients selected from our hospital during May 2015-Aug. 2016 were divided into control group and observation group according to random number table, with 65 cases in each group. Control group was given routine treatment as lowering blood pressure,lower-ing lipid, anticoagulation and antiplatelet. Observation group was additionally given Xuebijing injection 50 mL,ivgtt,q12 h,on the basis of control group. Both groups were treated for a week. The levels of inflammatory cytokines(TNF-α ,IL-6,IL-10, hs-CRP),SOD,MDA,MMP-9,platelet aggregation rate and fibrinogen were observed in 2 groups before and after treatment. The treatment course, the occurrence of MACE and bleeding event were also recorded. RESULTS:Before treatment, there was no statistical significance in the levels of inflammatory factors,SOD,MDA,MMP-9,platelet aggregation rate or fibrinogen between 2 groups(P>0.05). After treatment, the levels of TNF-α,IL-6,hs-CRP,platelet aggregation rate or fibrinogen in 2 groups were decreased significantly, while the level of IL-10 was increased significantly;the improvement of above indexes in observation group was significantly better than control group. The levels of SOD and MMP-9 in 2 groups were increased significantly, while the level of MDA was decreased significantly. The level of SOD in observation group was significantly higher than control group, while the level of MDA was significantly lower than control group, but the level of MMP-9 was significantly lower than control group, with statistical significance (P<0.05). There was no statistical significance in the incidence of MACE (16.92% vs. 20.00%)or bleeding event(4.62% vs. 7.69%)between 2 groups(P>0.05). CONCLUSIONS:Xuebijing injection shows signifi-cant therapeutic efficacy for NSTEMI,effectively inhibits inflammation, and oxidative stress and reduces the risk of thrombosis with good safety.

9.Construction of granulocyte-macrophage colony stimulating factor gene eukaryotic expressing plasmid and identification of its biological activity
Journal of Applied Clinical Pediatrics 2004;0(07):-
Objective To construct mouse granulocyte-macrophage colony stimulating factor (mGM-CSF) gene eukaryotic expressing plasmid pcDNA3-GM-CSF, to transfect the recombinant into erythroleukemia cell line FBL-3, and identify their biological activity. Methods GM-CSF gene eukaryotic expressing plasmid was constructed by subclone and recombinant was transfected into FBL-3 cells by electroporation. After screening by G418 and cloning by limiting dilution,we obtained positive cell clones(FBL-3-GM-CSF). PCR and RT-PCR were used to identify the integration and stable expression of GM-SF gene in FBL-3-GM-CSF cells. The biological activity was confirmed by the hematopoietic progenitor cell proliferative assay and hematopoietic progenitor cell colony formation assay. Results Mouse GM-CSF cDNA was amplified from the prokaryotic expressing plasmid PET-30a(+)-GM-CSF by PCR firstly and BamH Ⅰ and EcoRⅠrestriction sites were introduced. The inserted fragment was cut by BamH Ⅰ and EcoR Ⅰ digestion and ligated into pcDNA3 vector. The pcDNA3-GM-CSF eukaryotic expressing plasmid was constructed. The recombinant was cleared with appropriate endoneucleases and sequenced. The findings showed that the orientation of the insert was correct, while no rearrangement or mutation was found. PCR and RT-PCR assay showed that GM-CSF gene had integrated into FBL-3-GM-CSF cells and stably expressed. The hematopoietic progenitor cell proliferative assay and hematopoietic progenitor cell colony formation assay demonstrated that the cultured supernatant of FBL-3-GM-CSF cells of expressing GM-CSF should obviously stimulate proliferation of murine marrow mononuclear cells, and could stimulate hematopoietic progenitor cell colony formation. The number of colony formation was 54.67?4.83. The rate of colony formation was 0.547 %.Conclusions GM-CSF gene eukaryotic expressing plasmid is constructed successfully. A cell clone, which can express stably GM-CSF gene and possess biological activity,is obtained. Our studies have founded the base for the preparation of GM-CSF gene-modified vaccine of tumor cell and the study of feasibility of immune therapy of leukemia.
