1.In Vivo and in Vitro Antibacterial Activities of Cefodizime Sodium

OBJECTIVE: To evaluate the in vitro and in vivo antibacterial activities of cefodizime from both home and abroad. METHODS: The minimal inhibitory concentrations (MICs) of domestic and imported cefodizime, cefotaxime sodium against 300 clinical isolates of bacteria were tested by standard agar dilution method and the test tube dilution method. The in vivo antibacterial activities of cefodizime were determined in mice with experimental systemic infection caused by E.coli, K. pneumoniae and S.aureus. The values of ED50 were calculated by Bliss method. RESULTS: Both domestic and imported cefodizime had remarkable potency against majority gram- negative bacteria and gram- positive bacteria, including pneumococcus and beta streptococcus, and the MIC90 of cefodizime stood at 0.012~1.0?g?mL-1; and they showed medium grade antibacterial activity against bacillus cloacae, bacillus aerogenes and serratia but mild effects on MSSA and S.epidermidis, and the MIC50 stood at 4?g?mL-1. In this trial, bacillus aeruginosus, acinetobacter, enterococcus and MRSA showed resistance against the domestic and imported cefodizime. The in vivo antibacterial activities of domestic and imported cefodizime were similar and remarkable in mice infected with E.coli, K.pneumoniae and S.aureus. CONCLUSION: Domestic and imported cefodizime had excellent and similar antibacterial activity.
4. Determination of Plumbago content in Herba Plumbaginis Zeylanicae under different growing conditions

OBJECTIVE: To determine Plumbago content in Herba Plumbaginis Zeylanicae under different growing conditions and to provide experimental data for its domestication. METHODS: High performance liquid chromatography was used to determine Plumbago contents in the roots, stems and leaves of home-grown Herba Plumbaginis Zeylanicae. RESULTS: Plumbago content in Herba Plumbaginis Zeylanicae grown in mellow and clay soil was higher than in those grown in sand and crude soil. Potasium-fertilized and nitrogen-fertilized Herba Plumbaginis Zeylanicae contained higher Plumbago than those fertilized by phosphorus. Plumbago content was the highest in Herba Plumbaginis Zeylanicae grown in full light. Plumbago contents in roots, stems, and leaves of Herba Plumbaginis Zeylanicae grown under different bred conditions did not vary much. CONCLUSION: The research group's preliminary studies indicated that Plumbago was the main effective ingredient having anti-cancer and anti-inflammation bioactivity. Clay soil, cuttings, potassium fertilizer and nitrogen fertilizer, and full light were thought as ideal conditions for increasing Plumbago content. High quality Herba Plumbaginis Zeylanicae can be grown under these conditions. Copyright 2012 by the Chinese Pharmaceutical Association.

6.SSTR2 mRNA expression and correlation with proliferating cell nuclear antigen in human colorectal cancer

Objective To investigate somatostatin receptor subtype 2 (SSTR2) mRNA expression and its correlation with proliferating cell nuclear antigen (PCNA) in human colorectal cancer. Methods A total of 60 colorectal cancer samples were analyzed. The SSTR2 mRNA expression was examined by in situ hybridization (ISH) using multiphase oligoprobes. The PCNA were detected by immunohistochemical staining. A computerized image analysis system was utilized to estimate the relative contents of SSTR2 mRNA. Results The positive rate of expression of SSTR2 mRNA and relative contents in early and well-differentiated colorectal cancer were higher than those in advanced and poorly differentiated lesion. A negative correlation between SSTR2 mRNA expression and PCNA was found in colorectal cancer. Conclusion In the samples of colorectal cancer, the expression of SSTR2 mRNA is correlated with the degree of differentiation and clinical stage, and it plays an important role in reflecting the degree of malignancy and prognosis.
7.Computed tomography diagnosis of cardiovascular involvement in Behcet syndrome

Objective To evaluate the computed tomography (CT) characteristics of cardiovascular involvement in Beheet syndrome. Methods Eleven patients with clinically diagnosed Behcet syndrome were studied retrospectively from July 1995 to December 2007. Electron beam CT or 64-slice helical CT scanner was used and CT characteristics were reviewed. Results Eleven patients were diagnosed according to the criteria reported by the international study group for Behcet syndrome. Of them, 4 patients presented with aortic valve prolapse (2 patients with mitral valve prolapse), false aneurysm of right coronary artery was demonstrated in 2 patients, false aneurysm of left subclavian artery, aortic aneurysm and penetrating ulcers, aortic arch false aneurysm, aortic dissection, pulmonary embolism and interatrial scptum aneurysm in 1 case, respectively. Conclusion CT is a very useful method for the diagnosis and foUow-up of Behcet syndrome.

9.Long-term effects of oxypurinol on oxidative modification of actin and myocardial contractility in mice with heart failure

Objective To investigate the long-term effects of xanthine oxidase inhibitor,oxypurinol on myocardial contractility of post-ischemic heart failure in mice,and explore the underlying mechanism. Methods One hundred and twenty SV120 mice were randomly assigned into myocardial infarction control group,sham operation group and Oxy treatment group.Post-ischemic heart failure were induced by left anterior descending coronary artery ligation in myocardial infarction control group and Oxy treatment group,and mice in Oxy treatment group and sham operation group were orally administered with 0.5 mmol/L Oxy each day.Nine to eleven months after treatment,echocardiography was performed in all groups.Trabeculae from the right ventricle of mice were dissected for assessment of changes in excitation-contraction coupling.Sarcomere length was measured by laser diffraction.Intracellular free Ca~(2+) concentration([Ca~(2+)]_I)was detected with fluorescent dye Fura-2,which was microinjected iontophoretically into cells. Steady-state force-[Ca~(2+)]_I was achieved by addition of ryanodine and increasing the stimulus frequency to induce tetanization,and the relationship between myocardial contractility and intracellular Ca~(2+) transients was analysed.Besides,Western blotting was performed to determine the oxidation of myofilament proteins. Results Long-term oral administration of oxypurinol significantly improved myocardial contraction function and reduced ventricular wall thickness.Programming of excitation-contraction coupling was significantly improved,and maximal Ca~(2+) activated force(F_(max))in steady-state wag also significantly increased.Western blotting revealed the oxidative modification of actin in mice of Oxy treatment group was significantly inhibited compared with that of myocardial infarction control group. Conclusion Long-term treatment with Oxy improves the cardiac contraction function and boosts the cardiac force dramatically in post-ischemia heart failure.The increase in contraction is the result of increased myofilament Ca~(2+) responsiveness.Thus,antioxidant oxypurinol,by preventing oxidative damage to contractile proteins,can augment contraction with little changes in[Ca~(2+)]_I,represents new class of inotropic agents with advantages of reducing Ca~(2+) overload,and offers new promises in management of heart failure in the future.
