1.The effects of medical undergraduates' creativity tendency on participation in scientific research projects

Objective To analyze the effects of medical undergraduates' creativity tendency on participation in scientific research.Methods The self-administered questionnaire was used to survey the general characteristics,Williams creativity tendency,participation in scientific research projects of the junior students who majored in 10 medical specialties such as clinical medicine.The participation in scientific research includes the following statuses:those who do not participate in the scientific research project(NPSR,group A),those only participating in the teachers' scientific research project (PTSR,group B),those participating in college students' scientific research project as team members (TMSR,group C),and those participating in college students' scientific research project as project leaders(PLSR,group D).The differences of four dimensions scores and total scores of creativity tendency among four groups were tested using one-way analysis of variance.The effects of creativity tendency on participation in scientific research were tested with multinomial logistic regression analysis.Results The survey recycled 2 286 copies of effective questionnaires.About 39.1%(894/2 286) students participated in scientific research.The students were divided into A group(n=1 392),B group (n=108),C group(n=626),D group(n=160).The scores of adventure,curiosity,imagination,chal lenge and total scores had statistically significant differences among the four groups(P=0.000).The scores of adventure,curiosity,challenge and total scores of group C and D were higher than those of group A(P<0.05),respectively.With NPSR as a reference category,the scores of adventure(P=0.011),imagination (P=0.023),challenge (P=0.025) of PLSR had statistically significant partial regression coefficients,and the scores of adventure(P=0.009),curiosity(P=0.002) of TMSR had statistically significant partial regression coefficients.Conclusion High creativity tendency may be one of influencing factors that promote students to participate in college students' scientific research project.Various comprehensive measures should be taken to cultivate students' creativity and improve students' scientific research ability.
2.Antioxidative effect of schisanhenol on adipokine expression in 3T3-L1 adipocyte

Objective To explore the effect of schisanhenol on adipokine expression in 3T3-L1 adipocyte and its related mechanism. Methods 3T3-L1 adipocyte was cultured in vitro and induced to differentiation and maturity. Glucokinase was added to culture medium to make an oxidative model. The expression of adipocytokines were detected under the circumstance of different doses and at different time points of schisanhenol. Results The expression of adiponectin, leptin, resistin and visfatin were decreased with the increase of glucokinase concentration. Concentration-dependent inhibition effect was most obvious in leptin (25 mU/ml Glucokinase vs Blank group, t =7.29, P<0.01). With pretreatment of oxidative stress, the adipocytokines increased as the doses of schisanhenol increased (t=6.31,P<0.01 in adiponectin;t=5.92, P<0.01 in leptin; t=3.77, P<0.05 in resistin; t=3.63,P<0.05 in visfatin). With the extension of schisanhenol effect, the expression of four adipokines showed the process of first decrease-then increase'. The effects of schisanhenol on adipokines were parallel with the alteration of oxidative stress. Conclusions Schisanhenol increased adipocytokines expression in 3T3-L1 adipocyte by reducing oxidative stress, and the increase of leptin and adiponectin were most obvious, which indicated that schisanhenol could play a role in the treatment of diabetes by Chinese herb wuweizi.
3.Association of SUMO4 Met55Val variation with increased insulin resistance in newly diagnosed type 2 diabetes in a Chinese population.

SUMO4 Met55Val variation was shown to be related to type 2 diabetes susceptibility and the vascular complications in Asian people. To further examine the related mechanisms, this study was designed to evaluate the association of SUMO4 Met55Val polymorphism with insulin resistance and β cell function in newly diagnosed type 2 diabetic patients in a Chinese population. Four hundred and twenty seven newly diagnosed type 2 diabetic patients were selected for SUMO4 Met55Val polymorphism genotype analysis. All subjects underwent a 75-g oral glucose tolerance test (OGTT) to estimate the insulin sensitivity and β cell function. Anthropometrics and a metabolic profile were used for phenotyping analysis. The results showed that the SUMO4 Met55Val polymorphism was associated with higher insulin resistance (P<0.001) and lower insulin sensitivity (P<0.001). Patients with GG genotype had higher levels of plasma glucose, insulin and C peptide. Insulin sensitivity index (ISI) was closely correlated with body mass index (BMI) in patients with GG genotype in comparison to the counterparts with AG or AA genotype (r= -0.504 vs. r= -0.430 vs. r= -0.340). Multiple regression linear analysis showed that SUMO4 Met55Val polymorphism was an independent determinant for insulin sensitivity (P=0.001), which, along with triglyceride, BMI and sex, could account for 20.1% of the variation in ISI. The result remained the same after adjusting for BMI and sex. No association was found between SUMO4 Met55Val polymorphism and β cell function (all P>0.05). It was concluded that SUMO4 Met55Val variant was associated with increased insulin resistance in Chinese patients with newly diagnosed type 2 diabetes.

5.Effects of LAMC2 on macrophage polarization and regulation of NF-κB and ERK1/2 signaling pathways

Objective:To explore the role and potential mechanism of middle layer adhesion γ2 (LAMC2) in the regulation of macrophage polarization in diabetic nephropathy (DKD).Methods:Using GEO database, LAMC2 was identified as the main research object. Mouse mononuclear macrophages RAW264.7 were cultured in vitro and divided into the following 6 groups: Control group (blank control group), HG group (high glucose group), Treated with 30 mmol/L glucose, si-NC group (treated with 30 mmol/L glucose and transfected with negative control), si-LAMC2 group -1# (treated with 30 mmol/L glucose and transfected with shRNA-LAMC2 construct 1), si-LAMC2 group -2# (treated with 30 mmol/L glucose and transfected with SHRnA-LAMC2 construct 1), and SI-LAMC2 group-2# (treated with 30 mmol/L glucose) Group 3# of si-LAMC2 was treated with mmol/L glucose and transfected with shRNA-LAMC2 construct 2, and Group 3# of Si-LAMC2 was treated with 30 mmol/L glucose and transfected with shRNA-LAMC2 construct 3. Real-time quantitative fluorescent polymerase chain reaction (qRT-PCR) and Western Blot assay were used to detect LAMC2 expression in macrophages, small interfering RNA (siRNA) technique was used to inhibit LAMC2 expression, cell proliferation was evaluated by EdU staining, and cell invasion ability was evaluated by Transwell assay. The expressions of inducible nitric oxide synthase (iNOS), interleukin (IL)-12, Tumor necrosis factor-α (TNF-α), Arg-1, CD163 and IL-10 were detected by qRT-PCR, the expressions of CD86 and CD206 were detected by cellular immunofluorescence staining, and the expressions of key proteins in nuclear factor-κB (NF-κB) and extracellular signal regulated kinase (ERK) signaling pathways were detected by western blot assay.Results:Gene analysis in GEO database showed that the expression of LAMC2 in DKD and M1 macrophages was significantly up-regulated (all P<0.01). Compared with the blank Control group, the mRNA and protein expression of LAMC2 in macrophages of HG group were higher (all P<0.01). EdU staining and transwell statistics showed that, the EdU positive rate in HG and si -NC groups was lower than that in the Control group, and the number of invasive cells was significantly higher than that in the Control group (all P<0.05). The EdU positive rate of the si-LAMC2 group was higher than that of the si-NC group, and the number of invaded cells was significantly lower than that of the si-NC group (all P<0.05). Compared with Control group, the mRNA expression levels of M1 macrophage markers iNOS, IL-12 and TNF-α were higher in HG group and si-NC group (all P<0.01). Compared with the si-NC group, the mRNA expression levels of the above three markers in the si-LAMC2 group were lower (all P<0.01). Compared with the Control group, the mRNA expression levels of M2 macrophage markers Arg-1, CD163 and IL-10 in HG and si-NC groups were significantly decreased (all P<0.01). Compared with the si-NC group, the mRNA expression levels of the above three markers in the si-LAMC2 group were higher (all P<0.01). Compared with the Control group, the fluorescence intensity of CD86 in HG and si-NC groups was significantly higher, and the fluorescence intensity of CD206 was lower. Compared with the si-NC group, the fluorescence intensity of CD86 in the si-LAMC2 group was significantly lower, and CD206 were higher. Compared with the Control group, the expression levels of p-ERK1/2 and p-NF-κBP65 in HG and si-NC groups were significantly higher (all P<0.01). Compared with the si-NC group, the expression levels of two proteins in the si-LAMC2 group were significantly lower (all P<0.01). Conclusions:LAMC2 may play important physiological and pathological functions in diabetic nephropathy by promoting the polarization of macrophage M1 and affecting NF-κB and ERK1/2 signaling pathways.
6.Fixation of Sanders type ⅡB calcaneal fractures with hollow screws: a finite element analysis

Objective:To investigate the biomechanical stabilities of different internal screw fixations for Sanders type ⅡB calcaneal fractures using finite element analysis.Methods:Based on the CT scan data of a male volunteer's calcaneus (26 years old, 173 cm tall, weighing 70 kg), a three-dimensional finite element model of the calcaneus was established using digital medical software such as Mimics 22.0, Geomagic 12.0, Solidworks 2019, and ANSYS Workbench 2022. A three-dimensional finite element model of Sanders type ⅡB calcaneal fracture was also established. The following 3 kinds of screws were used: ① longitudinal screws: from the calcaneal tuberosity to the anterior part; ② transverse screws: from the lateral wall of the calcaneus to the sustentaculum tali; ③ vertical screws: from the base of the calcaneus to the talar dome, perpendicular to the posterior articular surface. The scheme of screw configuration was as follows: the longitudinal screws were divided into the upper, middle, and lower groups based on their entry points from the upper, middle, and lower parts of the calcaneal tuberosity. Based on different configurations (double transverse screws + single vertical screw, single transverse screw + single vertical screw, double transverse screws without a vertical screw, and single transverse screw without a vertical screw) 4 subgroups were further subdivided, resulting in a total of 12 models (a, b, c, d, e, f, g, h, i, j, k, l). Forces of 420 N and 200 N were applied to the posterior and middle subtalar joints, respectively, to simulate the load on the calcaneus during single-leg standing in adults. In each model, total deformation displacement, relative displacement of the sustentaculum tali fracture gap, relative displacement of the anterior fracture gap, relative displacement of the articular surface fracture gap, peak stress at the fracture ends and peak screw stress were analyzed.Results:The total deformation displacement was the lowest in models i and j, while models k and l exhibited the highest deformation displacement. The maximum deformation values in the remaining models varied only slightly. The average relative displacement of the sustentaculum tali fracture gap was the lowest in the upper group (0.27 mm) and relatively higher in the lower group, with the greatest variation seen in models k and l. The average relative displacement of the anterior fracture gap was the smallest in the lower group (0.16 mm), with models i and k performing the best. The relative displacement of the articular surface fracture gap remained at a low level across all models. The upper group had the lowest average peak stress at the fracture ends (60.20 MPa), while the middle and lower groups had similar values (103.88 MPa and 99.76 MPa, respectively). The upper group had the lowest average screw stress peak value (222.77 MPa), whereas models k and l in the lower group had the highest stress peaks, reaching 331.48 MPa and 349.82 MPa, respectively.Conclusions:Model i (inferior longitudinal screw + double transverse screws + single vertical screw) can be recommended in clinic because it demonstrates balanced performance in displacement and stress and a notable advantage in reducing overall deformation displacement and anterior fracture gap displacement. As longitudinal, vertical, and transverse screws each play their distinct role in calcaneal fracture fixation, surgeons should select an appropriate screw configuration based on the patient's needs.
7.Preparation and Evaluation of Compound Nanxing Pain Relief Gel

Objective To prepare Compound Nanxing Pain Relief Gel(CNPRG)and evaluate its quality and sensitization.Methods CNPRG uses Carbopol 980 NF as the matrix;Appearance,viscosity,coating,centrifugal stability,cold stability,thermal stability as comprehensive indicators,single test and Box-Behnken effector method to optimize the prescription;The quality evaluation methods of appearance,pH,viscosity,stability,vapor phase identification of volatile components,and determination of diaconitine and eugenol content of CNPRG were preliminarily established;CNPRG sensitization was assessed by Active Cutaneous Anaphylaxis.Results The best prescription for CNPRG was Carbopol 980 NF 0.35 g,drug 1.02 g,glycerol 5.00 g,and pH 6.20.CNPRG ′s appearance likes jelly,is smooth,uniform and delicate;pH=6.20±0.03;viscosity 68.43±1.14 Pa·s;Centrifugation,high,low temperature stability,no stratification and precipitation;Identify camphor,(±)-Borneol and(±)-Isoborneol,Cinnamaldehyde,eugenol,phenol;Hypaconitine content 0.2983±0.0073 μg·g-1;Eugenol content 155.66±0.97 μg·g-1;CNPRG confirmed no sensitization.Conclusion CNPRG has good appearance,stable quality and no sensitization,and it can provide a choice for the development of new dosage forms of compound Nanxing pain relief cream to alleviate sensitization.
