1.Establishment and optimization of the amplified fragment length polymorphism reaction system of Carthamus tinctorins L.genome

Objective:To study the factors affecting amplified fragment length polymorphism(AFLP) of Carthamus tinctorins L.genome,and to develop and optimize the AFLP reaction system,laying a foundation for Marker-assisted breeding of Carthamus tinctorins L..Methods: Cetyl-trimethyl-ammonium-bromide(CTAB) method was used to extract the genomic DNA of Carthamus tinctorins L.,whose concentration and purity were determined by UV spectrophotometry.The genomic DNA was digested by restricted enzyme and ligated with one-step method or two-step method so as to find out which one is more suitable.Then the products were prepared with different dilution multiples(5,10,15,20,25 and 30) for pre-amplification and with different concentrations of gradients(10,25,50,75,100,150 and 200)for selected amplification.Then they were subjected to PAGE electrophoresis and silver-staining after denaturing at 95℃ for 8 min.Results: The AFLP reaction system for Carthamus tinctorins L.genome included the following steps: the genomic DNA was precipitated with alcohol by CTAB method to avoid contamination by any inhibitors;the DNA was completely digested by MseⅠ and EcoRⅠ at 37℃ for 3 hours.Two-step method was superior to one-step method in this AFLP system.The digestion time was 3 hours at 37℃ and ligated at 16℃ over night,with Buffer 2(NEB Company) as the specific buffer.Furthermore,the optimal dilution was 25 folds for pre-amplification and 75 folds for selected amplification. In this reaction system,the strips in PAGE electrophoresis were clear and less decomposed.Conclusion: The AFLP reaction system in this experiment is suitable for silver-stained AFLP of Carthamus tinctorins L.analysis.

3.Analysis of 137 Adverse Drug Reaction Cases Induced by Levofloxacin Injection in Our Hospital

OBJECTIVE: To analyze the characteristics of adverse drug reaction (ADR) induced by Levofloxacin (LVFX) injection and to promote rational use of drugs in the clinic. METHODS: 137 LVFX injection-induced ADR cases reported in our hospital during 2006~2009 were collected and analyzed statistically with Excel in respect of age, gender, medication and clinical manifestation, etc. RESULTS: ADR cases occurred nearly in all ages of people, and 32.85% were appeared in people over 60 years. Main clinical manifestations were lesion of skin and its appedants (62.04%) and gastrointestinal injury (16.06%). Most of them were mild and medium ADR. CONCLUSION: Great importance should be attached to the rational use of LVFX injection and ADR monitoring to guarantee the safety of patients.
4.Study on the efficacy and mechanism of probiotic assisted glucocorticoid on the treatment of the patients with eczema

Objective To study the efficacy and mechanism of probiotic assisted glucocorticoid on the treatment of the patients with eczema. Methods 92 cases with eczema from February 2015 to March 2017 were divided into two groups according to the treatment methods, the conventional group and the combined group, 46 cases in each group. The conventional group were given glucocorticoid, and the combined treatment group were received probiotics combined with glucocorticoid. The levels of serum inflammatory factors, the treatment effects of eczema and the incidence of adverse reactions were compared between the two groups before and after treatment. Results The treatment effect of eczema in combined group was higher than that in routine group (P<0.05). No severe adverse reactions occurred in the two groups. There was no significant difference in serum inflammatory factors between the two groups before treatment. The level of serum inflammatory factors in combined group was significantly better than that in tconventional group (P<0.05). Conclusion Probiotic assisted glucocorticoid is effective in the treatment of eczema. It can lower the level of inflammation and improve the condition of the body. It has no obvious adverse reaction which has the characteristics of safety and effectiveness.
5.Effect of miRNA-146a on life cycle of hepatitis B virus in vitro

Objective To determine the effect of microRNA-146a (miR-146a) on the life cycle of hepatitis B virus (HBV) and investigate the underlying mechanisms.Methods The miRNA expression profiles were compared by miRNA array between HepG2 and HepG2.2.15 cells.Then miR-146a was chosen as objective,and its expression level was further confirmed by RT-PCR.After miR-146a mimic and inhibitor were transfected into HepG2.2.15 cells respectively,the quantification of HBV replication was determined by RT-PCR,and the levels of HBsAg and HBeAg in the supernatant were measured by ELISA,and the expression of HS3ST3B1 at mRNA and protein levels were tested by RT-PCR and Western blotting.Dualluciferase reporter assay was used to detect the interaction between miR-146a and potential target HS3ST3B1.Results The expression levels of totally 72 miRNAs were changed in HepG2.2.15 cells,with 27 upregulated and 45 down-regulated.RT-PCR showed the expression level of miR-146a was significantly higher in HepG2.2.15 cells than HepG2 cells (1.55-± 0.13 vs 1.00 ± 0.01,P < 0.05).Transfection of miR-146a mimic into HepG2.2.15 cells resulted in significantly increased HBV replication and levels of HBsAg and HBeAg (P < 0.05),while the transfection of its inhibited caused opposite results (P < 0.05).Bioinformatic analysis showed that HS3ST3B1 was a potential target of miR-146a.The reporter luciferase reporter system indicated that the reported fluorescence intensity of HS3ST3B1 wild type vector was significantly lower than that of the control group (P < 0.05),but showed no significant difference between HS3ST3B1 mutant vector and control group (P >0.05).The mRNA level of HS3ST3B1 was not significantly changed in HepG2.2.15 cells transfected with miR-146a mimic (P > 0.05),but its protein level was significantly decreased (P < 0.05).Conclusions miR-146a affects the life cycle of HBV,which may be through suppressing the translation of HBV inhibitory factor HS3ST3B1 3'UTR.
6.Effects of Clarificant Chitosan on Contents of Zinc,Manganese,Calcium and Heavy Metal Lead in Water extraction Liquid of Chinese Herbal Medicines

Objective: To study the effects of clarificant chitosan on contents of zinc,manganese,calcium and heavy metal lead in the water extraction liquid of chinese herbal medicines. Methods: The contents of these elements were determined by atomic absorption spectrophotometry.Results: The results indicated that the chitosan clarification could raise the transfer rate of some inorganic elements such as zinc,manganese and calcium as compared with water extraction and alcohol precipitation method,And it could also reduce the amount of heavy metal lead.Conclusion: chitosan as a clarificant is promising in the field of refining water extraction liquid of chinese herbal medicines.
7.Studies on degree of deacetylation of chitosan and its residual amount left over in clarified products when used as a clarificant

Object To study the degree of deacetylation of chitosan and its residual amount left over in the clarified products when chitosan was used as a clarificant. Methods The free amino group in the deacetylated chitosan was determined by linear titration. Results The degree of deacetylation of chitosan was found to be about 80%. The residual amounts left over in the clarified products were lower than 10 -4 . Conclusion The results suggested that it is feasible to use chitosan as a clarificant for refining water extract of TCM.
8.Establishment of evaluation index system for training effect of rural oriented clinical students based on Delphi method

Objective:To establish an evaluation index system for training effect of order-oriented clinical medical students in Xinjiang by employers, so as to lay a foundation for training effect evaluation.Methods:On the basis of questionnaires and interviews with the training requirements of employers, Delphi method was used to determine the index system and weight.Results:The training requirements of employing units for the ability of free and directional medical students in autonomous regions were in turn competent for work, knowledge and practical ability, and good moral quality. The cultivation requirements for students' personal qualities of free and directional medical students were good interpersonal communication and moral quality. After three rounds of Delphi expert consultation, the evaluation index system was established, and three dimensions (knowledge, professional ability, professional ethics literacy), nine secondary indicators and 33 tertiary indicators were screened out, and the weights were determined.Conclusion:The evaluation index of training effect of directional clinical medical students established by Delphi expert consultation method needs further validation. Through the follow-up empirical application in directional clinical students, the evaluation index system can be constantly improved, which can also provide reference for the establishment of evaluation index system for other positions.
10.CAD fabrication of the mandible All-on-4 implant photoelastic model

Objective:To explore the method of the establishment of edentulous mandible All-on-4 implant photoelastic model with various distally tilted implants by computer-aided design(CAD).Methods:The edentulous mandible specimen was scanned by CT. Three-dimensional models of the mandible consisting of implant location were reconstructed and edited with Mimics and Geomagic Stu-dio software.Each of the four models had four implant sockets in the interforaminal area.In the first model,the bilateral distal im-plants were placed vertically.In the other models,the posterior implants were respectively inclined by 1 5°,30°and 45°distally.The four models were manufactured by RP and then the All-on-4 photoelastic models were made by traditional ways.Results:The ob-tained ethoxyline resin models featured bright surface,homogeneous structure,faintly yellowish,high optical sensibility,precision im-plants location and with no natural stress.The model offered a good experimental basis for the stress measurement.Conclusion:The method of fabricating ethoxyline resin models by CAD is reproducible,which simplifies the operating process.
