2.Effects of Different Extraction Methods on the Content of Cholalic Acid from Artificial Bezoar
China Pharmacy 2001;0(09):-
OBJECTIVE:To study the effects of different extraction methods on the content of cholalic acid from Artificial Bezoar. METHODS: Artificial Bezoar from 3 different habitats was used to extract cholalic acid by hydrolization vs. hydrolization-free method, and the content of cholalic acid was determined by HPLC-ELSD(high performance liquid chromatography with evaporative light scattering detection). RESULTS: The content of cholalic acid in the hydrolyzed sample was higher than that in the non-hydrolyzed sample of the same batch. CONCLUSION: Through hydrolization, the binding cholalic acid in the artificial bezoar was transformed into free cholalic acid so that the content of cholalic acid was increased. Therefore, it is more reasonable to control the quality of Artificial Bezoar by taking the contents of total cholalic acid and free cholalic acid as indexes.

Objective: To study the effects of clarificant chitosan on contents of zinc, manganese, calcium and heavy metal lead in the water extraction liquid of chinese herbal medicines. Methods: The contents of these elements were determined by atomic absorption spectrophotometry. Results: The results indicated that the chitosan clarification could raise the transfer rate of some inorganic elements such as zinc, manganese and calcium as compared with water extraction and alcohol precipitation method,And it could also reduce the amount of heavy metal lead. Conclusion: chitosan as a clarificant is promising in the field of refining water extraction liquid of chinese herbal medicines.

Object To study the degree of deacetylation of chitosan and its residual amount left over in the clarified products when chitosan was used as a clarificant. Methods The free amino group in the deacetylated chitosan was determined by linear titration. Results The degree of deacetylation of chitosan was found to be about 80%. The residual amounts left over in the clarified products were lower than 10 -4 . Conclusion The results suggested that it is feasible to use chitosan as a clarificant for refining water extract of TCM.

OBJECTIVE:To study the implementation of scoring diseases payment under the control of total budget in some ar-eas of China, and provide reference for the further improvement of the payment. METHODS:Based on the operation process of scoring disease payment in the control of toal budget, the specific practices of Huaian,Nanchang,Zhongshan and Taiwan area, were compared. RESULTS:It showed differences in the control and allocation of total budget, the establishment of scoring diseas-es,the setup of adjustment coefficient, the development of balance methods, the complementary mechanism and the monitor exami-nation setting. CONCLUSIONS:Whether to classify the diseases according to diagnosis and treatment methods, how to setup ad-justment coefficient appropriately, how to reimburse the patients in stages and how to strengthen supervision are worthy of further thinking and exploring.

Thiols, which are components of many proteins and simple molecules, play an important role in the cellular antioxidant defense system. The quantitative determination of thiols is important in biochemistry and clinical chemistry. Fluorescent probes, which have its apparent advantages in sensitivity and, most importantly, in imaging thiols in vivo, even in single living cells, appear to be particularly attractive. In this review, we classify the fluorescent probes based on their different reaction mechanisms with thiols and summarize the recent progresses of thiols fluorescent probes with fifty-one

Objective To investigate the protective effects of recombinant human erythropoietin (rHuEPO) on acute lung injury (ALI) induced by lipopolysaccharide (LPS).Methods Fourty-five rats were randomly (random number) assigned to three groups, namely control group, model group, and rHuEPO group.ALI was induced by intravenous injection of LPS (6 mg/kg).The rHuEPO (5000 U/kg) was injected intravenously into rats 60 min before LPS challenge. The general status of rats was observed.Twelve hours after modeling, the rats were sacrificed and the tissue samples including lung tissue and blood were collected.PaO2,PaCO2,pH,the lung wet/dry weight ratio, plasma cytokines [interleukin (IL) IL-6 and tumor necrosis factor-alpha (TNF-α)],and inducible nitric oxide synthase (iNOS) were detected.Cytokines were assayed with ELISA method.Pathological changes of lung tissues were observed under light microscope and transmission electron microscopy. Results (1) Compared with the control group,PaO2,pH in the model group and in the rHuEPO group were significantly lower (P < 0.05),and PaCO2 were significantly higher (P < 0.05).Compared with the model group,PaO2,pH in the rHuEPO group were significantly higher (P < 0.05),and PaCO2 were significantly lower (P < 0.05).(2) Compared with the control group, the W/D weight ratio of lung tissues in the model group and the rHuEPO group was significantly higher (P < 0.05).Compared with the model group, the W/D weight ratio of lung tissues in the rHuEPO group significantly lower (P < 0.05).(3) The levels of TNF-o,IL-6 and iNOS in serum of rats in the control group were lower than those in the model group and the rHuEPO group (P <0.01).The serum levels of TNF-α,IL-6 and iNOS of rats in the rHuEPO group were significantly lower compared with the model group (P < 0.01).(4) The light microscopy and the transmission electron microscopy showed the model group had histopathologic changes with acute diffuse lung injury manifested by intra-alveolar hemorrhage, exudate, inflammatory cells infiltration,Ⅰ type and Ⅱ type epithelial cell necrosis and detachment, and the pathological changes of lung tissue in the rHuEPO group were not as serious as those in the LPS group, showing only a little inflammatory cells infiltration of focal alveoli. Conclusions Recombinant human erythropoietin can inhibit the genesis of TNF-α,IL-6 and iNOS in serum, modifying the inflammatory response and providing protective effects against acute lung injury induced by sepsis.

Chinese Journal of Experimental Ophthalmology 2014;32(4):340-344
Background Poly(adenosine diphosphate-ribose) polymerase 1 (PARP-1) plays an important role in the pathogenesis of diabetic retinopathy (DR),and Notch1 signal pathway is one of the important signal transduction pathways in the organism which may antagonize retinal vascular diseases. However,if Notch1 signaling pathway is involved in pathogenesis of DR has not been confirmed yet. Objective This study was to investigate the expressions of Notch1,Dll4,PARP-1,Akt,nuclear factor-κB (NF-κB) and caspase-3 in the retina of diabetic mouse model and retinal vascular endothelial cells (RVECs) under the high glucose. Methods The expressions of Notch1,Dll4,PARP-1,Akt,NF-κB and caspase-3 in the retina of diabetic mouse models were investigated using immunochemistry and Western blot method after the diabetic mouse models were established. And these proteins were detected in retinal RVECs under the high glucose by Western blot. Results The expressions of Notch1,Dll4 and phosphorylated Akt (p-Akt) in retinas reduced significantly and simultaneously companied with increases of PARP-1 and caspase-3 in diabetic mice compared with normal mice (all at P<0.05).However,no obvious change was found in the expression of NF-κB (P>0.05).Expressions of Notch1 and p-Akt in RVECs increased with the increase of glucose concentration, but expressions of cleaved PARP-1 and caspase-3 decreased, especially in the 30 mmol/L group, showing significant differences in comparison with the normal control group (all at P<0.05).But no altering of NF-κB expression was seen in the mice with diabetes mellitus. Conclusions The expressions of cleaved-PARP-1 and caspase-3 in the retinas is up-regulated,but the expressions of Notch1 and p-Akt are down-regulated in diabetic mice.
