1.Relevance between Gastrointestinal Microbiota and Helicobacter pylori Eradication Therapy

Gastrointestinal microbiota is a huge and complex system that has multiple influential factors. As an exogenous pathogen,Helicobacter pylori(Hp)may interact with gastrointestinal microbiota. It has been revealed that Hp infection led to an increase in Lactobacillus acidophilus in intestinal tract and a decrease in Actinobacteria,Bacteroidetes and Firmacutes in stomach. Proton pump inhibitors and antibiotics,the major components of eradication regimens for Hp infection,may also have an impact on gastrointestinal microbiota. On the other hand,gastrointestinal microbiota interferes with Hp’s attachment into gastric mucosa. Probiotics combined with eradication therapy could benefit the eradication rate of Hp infection and reduce adverse events.

3.Serum progesterone levels in early pregnant women and its relation to luteal maintenance therapy for pregnancy protection

Objective To investigate serum levels of β-human chorionic gonadotrophin (β-HCG) and progesterone (P) in early pregnant women,and their relation to luteal maintenance therapy for early pregnancy protection.Methods One hundred and thirty five infertility women treated in Department of Gynecology and Obstetrics of Beijing Chaoyang Hospital from January 2007 to December 2011.Total 150pregnancy cycles,including 84 with intrauterine insemination (IUI) and 66 with natural conception,were divided into two groups:normal intrauterine pregnancy group (group A,n =80) and early pregnancy loss group (group B,n =70).Medical history,ultrasonic findings,serum female hormone,P and β-HCG levels at early pregnancy stage were analyzed.Results The age of group A and group B was (30.0 ± 3.9) years and(30.7 ± 4.9) years,respectively (P > 0.05).The follicle-stimulating hormone/luteinizing hormone in group A and group B was 1.57 ±0.96 and 1.56 ± 1.08 ; the estradiol levels in two groups were (152 ±66) pmol/L and (147 ± 69) pmol/L,respectively (both P > 0.05).There were no differences in dominant follicles and endometrial thickness between groups A and B (P > 0.05).Ovulation promotion and luteal support treatments were adopted in both groups:50% (40/80) of cycles in group A received ovulation promotion,73% (58/80) of cycles received luteal support,while 44% (31/70) and 76% (53/70) received in groups B,respectively.The levels of serum progesterone in group A during 14-21 d,22-27 d and ≥ 28 d after ovulation were higher than those in group B at each time points (P < 0.0l).There were no significant differences in serum progesterone levels between women with luteal support treatment and those without luteal support treatment in both groups (P > 0.05).Conclusion Dynamic monitoring of serum progesterone level in early pregnancy may be used as an auxiliary index for prediction of pregnancy outcome,but it may not be an indicator for luteal maintenance therapy.

5.Expression and clinical significance of transcription factor Sox2 in human prostate cancer

Objective To study the expression of Sox2 in human prostate cancer and pericarcinomatous tissue,and the relation between its expression and the pathological type and clinical stage of prostate cancer.Methods Immunohistochemisty,Western blot and reverse transcription-polymerase chain reaction (RT-PCR)were used to detect the expression of Sox2 in 86 cases of prostate cancer tissues and 40 cases of pericarcinomatous tissues.The relation between Sox2 expression and the pathological type and clinical stage of prostate cancer was analyzed.Results The expression of Sox2 in prostate cancer tissue (71.9%) was significantly higher than that in pericarcinomatous tissue (7.5%),and the difference was statistically significant (x2 =53.98,P < 0.05).Sox2 expression in prostate cancer tissues was significantly related to Gleason score (x2 =19.49,P < 0.05).The positive rate of Sox2 in lymphatic metastasis group and negative lymphatic metastasis group was 92.3% and 73.3% respectively,and the difference was statistically significant (x2 =3.95,P < 0.05).While there was no significant difference among clinical stages (x2 =0.16,P > 0.05).Moreover,Western blot and RT-PCR revealed that the expression of Sox2 in prostate cancer tissue was significantly higher than that in pericarcinomatous tissue (t =-2.93,P < 0.05 ; t =3.29,P < 0.05).Conclusion The high expression of Sox2 is closely related to the Gleason score and lymphatic metastasis of prostate cancer,but not to the clinical stage.The overexpression of Sox2 may play an important role in formation,differentiation and metastasis of prostate cancer.

ObjectiveTo investigate the inhibitory effects and related toxicity of all-trans-retinoic acid(ATRA) combined with arsenic trioxide(As2O3) on transplanted human bladder neoplasms in nude mice.MethodsThe subcutaneously transplanted tumor models of human bladder neoplasms in nude mice were established and then it was randomly divided into four teams: Saline group, ATRA group, As2 O3 group and the combination of ATRA and As2 O3 group.Each group composed of 10 nude mice and received intratumor injection in the following 14 d.The tumor growth inhibitory rate was calculated.The pathological changes of tumor,cardiac, liver, and kidney were observed after H.E.staining.The expression of MVD (Marked with CD43) and VEGF were detected by SP of immunohistochemistry.Blood routine examination and hepatic and nephritic functional examination was carried out to evaluate the side-effects.Results The tumor growth was significantly suppressed in ATRA group(inhibitory rate was 41.82%) and As2O3 group (inhibitory rate was 43.77%) compared with saline group.When ATRA combined with As2O3 ,inhibitory effects were improved(inhibitory rate was 68.55%).The difference of growth inhibitory rate between each group was obviously significant(x2 =26.81, P <0.01).There was less expression of MVD (Marked with CD43) and VEGF in ATRA group and As2 O3 group and the combination group compared with saline group (ODVEGF = 19.23 + 2.32,20.72 + 2.01,17.16 ± 1.59,27.33 ± 2.17, respectively; ODMVD = 44.77 + 8.25,43.39 + 7.41, 30.56 + 7.71,141.12 + 8.38, respectively), and the suppressed effect in the combination group was most significant (t = 3.16,3.08,3.37, P < 0.01).WBC inhibition was seen in ATRA group and As2O3 group and the combination group, but no significantly difference among them(P >0.05).Hepatic and nephritic toxicity were not found.ConclusionsCombination of ATRA and .As2O3 had obvious synergistic effects on transplantéd tumor of human bladder neoplasm in nude mice, and moderate white blood cell inhibition and no hepatic or nephritic toxicity.

Lung adenocarcinoma is usually thought to follow a linear multistep progression schema,that is precancerous lesions progress to adenocarcinoma in situ,and progress to micro-invasive adenocarcinoma,eventually some lung adenocarcinoma happen diffusion.However,recent findings reveal that this linear progression schema might not occur in all lung adenocarcinoma subtypes,and some subtypes show a kind of non-linear progression schema.The study of these two kinds of schemas can help for early detection of lung adenocarcinoma.

10.Development and validation of a GC-FID method for quantitative analysis of oleic acid and related fatty acids☆

Oleic acid is a common pharmaceutical excipient that has been widely used in various dosage forms. Gas chromatography (GC) has often been used as the quantitation method for fatty acids normally requiring a derivatization step. The aim of this study was to develop a simple, robust, and derivatization-free GC method that is suitable for routine analysis of all the major components in oleic acid USP-NF (United States Pharmacopeia-National Formulary) material. A gas chromatography-flame ionization detection (GC-FID) method was developed for direct quantitative analysis of oleic acid and related fatty acids in oleic acid USP-NF material. Fifteen fatty acids were separated using a DB-FFAP (nitroterephthalic acid modified polyethylene glycol) capillary GC column (30 m × 0.32 mm i.d.) with a total run time of 20 min. The method was validated in terms of specificity, linearity, precision, accuracy, sensitivity, and robustness. The method can be routinely used for the purpose of oleic acid USP-NF material analysis.
