1.Suicide Prevention, Suicide Attitude and Related Factors among Undergraduates in Changsha

Objective:To understand suicide prevention knowledge, suicide attitude and related factors among undergraduates in Changsha.Methods:Using multistage stratified cluster sampling, a total of 1664 undergraduates from six universities in Changsha were tested with Suicide Awareness Questionnaire, Suicide Attitude Questionnaire (QSA), Adolescent Self-Rating Life Events Check List (ASLEC), Social Support Questionnaire (SSQ) and Symptom Checklist 90 (SCL-90). Results:The mean score of suicide awareness among undergraduates was 10.8?0.9 and the correct response rate was 54.5%. The result of suicide awareness score showed, female undergraduates had a lower score than males (9.2?0.8, 12.3?0.8;t=2.33, P=0.020), medical undergraduates had a higher score than ones of humanities and social science or engineering (13.9?0.8, 10.0?1.0, 9.7?0.7;F=10.47, P=0.000), undergraduates from one-child families had a higher score than ones from non-one-child families (11.9?0.7, 9.1?0.9;t=3.25, P=0.034), and college students from nuclear family had a higher score than ones from extended families or one-parent families (14.0?0.9, 10.3?0.8, 9.4?0.7;F=7.49, P=0.000). The major factors related to suicide awareness were gender, specialty and family type (?=0.087, 0.342 and 0.084, respectively). And the major factors related to attitude towards attributes of suicide behaviors were specialty, gender, suicide ideation and mental health status (?=0.688, -0.055, 0.810 and 0.731, respectively). Conclusions:College students in Changsha have severe misunderstands about suicide prevention. It is necessary to pay more attention to the education of suicide knowledge and to cultivate the correct suicide attitude among college students.
2.Investigation of Osmotic Pressure of Citicoline Sodium Injection

Objective:To analyze the osmotic pressure of citicoline sodium injection from different manufacturers to provide experi-mental basis for improving the national standard of the product. Methods:The cryoscopic method was used to detect the osmotic pres-sure of 479 batches of citicoline sodium injection from 45 pharmaceutical factories. Results:The osmotic pressure of citicoline sodium injection was within the range of 359-692 mOsmol · kg-1 , that of citicoline sodium for injection was within the range of 401-1408 mOsmol·kg-1 , and that of citicoline sodium chloride injection was within the range of 278-299 mOsmol·kg-1 . Conclusion:The os-motic pressure of citicoline sodium injection and citicoline sodium for injection from different manufacturers is obviously different. It is necessary to detect osmotic pressure in the quality standard in order to evaluate the quality of preparation process.
3.Treatment of fractures of thoracolumbar vertebral body with paraplegia by anterior route decompression and internal fixation

Objective To study the clinical outcome of thoracolumbar fractures treated by ante rior route decompression,bone-graft-fusion and internal fixation.Methods 51cases of thoracolumbar fractures from Jun1994to Mar 2002were treated by anterior route decompression,bone-graft-fusion and internal fixation,of w hich32cases by Kaneda system and 19cases by Z-Plate system.Their clinical data were analyzed.Results The patients were followed up from five months to eight years(average of 3.5years).45cases had a good anatomic reduction and maintain thoracic-lu mbar lordosis.48cases gained one to three Frankel grades of neurologic r e-covery as a consequence of operation.Conclusion Kaneda internal fixation has the characteristics of low cost,neurological decompression and reliable fixation;and Z-Plate systems has the advantages of simple manipul ation,intrinsic stability,less complications and good biocompatibility.
4.The Effect of Dehydration after a Sauna and Intermittent Hypoxia Stimuli on Heat Stress Protein 70 in Human Leucocytes and Aerobic Capacity during Incremental Exercise

The purpose of this study was to compare Heat Stress Protein70 (HSP70) in human leucocytes, VO2max and Blood Lactate (BLa) during incremental exercise under a thermoneutral condition without taking a sauna with those during the same workload and under same thermoneutral condition after a sauna and after intermittent hypoxia stimuli. Ten unacclimated men performed an incremental test to exhaustion on a treadmill under a thermoneutral condition without taking a sauna (N25℃, relative humidity 65%) and during the same workload and under same thermoneutral condition after a sauna (D25℃, relative humidity 65%), and after intermittent hypoxia stimuli (LN25℃, relative humidity 65%) . 5.Effects of several Chinese herbal medicines on SMMC-7721 liver cancer cells by FTIR spectroscopy

Objective:To study the effects of several herbal medicines on SMMC-7721 liver cancer cells with Fourier transform infrared spectrometer(FTIR). Methods: FTIR was employed to determine the infrared spectra(IRs) of SMMC-7721 liver cancer cells cultivated for 20 h with the extracts of Spica prunellae, Herba houttuyniae, Radix bupleuri and Herba artemisiae scopariae. Cluster analysis of IRs was also performed. Results: IR spectral parameters such as band shape, intensity and frequency of the blank, control and herbal-extract-treated cells were compared. There existed obvious blue shift of ? s(PO 2 -), ? as (PO 2 -) bands, red shift of ? as (CH 3), ?(CH 2) bands on the herbal-extract-treated cells IRs. The decreasing ratio of ? as (CH 3) to ? s(CH 2) peak intensity and the increasing ratio of ? s(PO 2 -) to ?(N-H) peak area indicated the destructive effect of herbal extracts on the membrane structure of SMMU-7721 cells and inhibitory effect on the DNA replication respectively. Cluster analysis successfully discriminated the herbal-extract-treated cells from the blank cells and the liver-oriented medicines from the non-liver-oriented medicine. Conclusion: FTIR provides another fast and effective approach to analyze the changes of cells treated with Chinese herbal medicines, which may help to illuminate the functional mechanism of Chinese herbal medicines.
6.Expression of TRAIL and TRAILR in human hepatocellular carcinoma and its therapeutic significance

Objective To evaluate the expression of tumor necrosis factor related apoptosis-inducing ligand (TRAIL) and its receptor(TRAILR) in human hepatocellular carcinoma (HCC), and its therapeutic potential for human HCC. Methods Expressions of TRAIL and TRAILR were determined in and adjacent to carcinoma tissues, and normal liver tissues. The cytotoxic effects of TRAIL in combination with chemotherapeutic agents on HCC cell lines were detected. Results Expression of TRAIL in tumor adjacent tissue was higher than in cancer tissue. 7.Intraoperative radiotherapy using low-energy X rays:dosimetric characteristics and potential limitations in clinical application

Objective To test intraoperative radiotherapy with mobile photon beam using the INTRABEAM system ( Germany) , and to analyze the dosimetric characteristics of low?energy photon beam using X?ray source and spherical applicators and explore its potential limitations in clinical application. Methods A special water phantom, a parallel?plate ionization chamber, and an electrometer were used to measure the depth dose rates and isotropy of dose distribution in x/y plane of X?ray source and different spherical applicators in the INTRABEAM system. Those data were then compared with the system data. Results For the X?ray source, the deviation of observed depth dose rate and isotropy in the x/y plane from the system data were-2.16%± 1. 36% and-1.9%~ 2. 1%, respectively. For applicators with different diameters, the deviation of observed depth dose rate, transfer coefficient, and isotropy in x/y plane from the system data were-10.0%~2. 3%,-8.9%~4. 2%, and-1.6%~2. 6%, respectively. Surface dose rate and dose gradient became larger with the decrease in the diameter of the spherical applicator. The measurement of depth dose rate and isotropy of X?ray source and spherical applicators showed good repeatability. The influencing factors for measurement accuracy included the positioning error of ionization chamber, energy response, noise current, and correction factor f ’ ( R ) . Conclusions This study reveals the dosimetric characteristics of the INTRABEAM system, verifies the accuracy of the system data, and obtains the data for clinical application and routine quality assurance. However, large dose gradient and small therapeutic range may limit its wide clinical application.
8.Effects of esculentoside A on production of IL-1 and TNF by rabbit synovial cells

Objective: To investigate the influence of esculentoside A(EsA) on production of IL-1 and TNF by rabbit synovial cells induced by LPS. Methods: levels of IL-1 and TNF in the supernatant of rabbit synovial cell were determined by examining proliferation of thymic cells and by bioassay L929 cells as target cells, respectively. Results: EsA in 5-40 μg/ml could significantly inhibit the production of IL-1 and TNF from rabbit synovial cells induced by LPS. Conclusion: EsA can inhibit the production of IL-1 and TNF from synovial cells. It suggests that EsA may play a role in improving the rheumatoid arthritis.
9.Differentiation from bone marrow stem cells into neuronal-like cells induced by fetal spinal cord tissue

BACKGROUND: It is demonstrated that bone marrow stem cells (BMSCs) can generate neurosphere structures, which is similar to cloning sphere of neuron-specific enolase (NSE), in a specially induced system in vitro; therefore, BMSCs draw more and more attention as seed cells to repair central nerve injury.OBJECTIVE: To investigate the differentiation from BMSCs into neuron-like calls induced by fetal spinal cord tissue.DESIGN: Observational study.SETTING: Department of Spine Surgery, Second People's Hospital of Shenzhen.MATERIALS: SD rats (16 pregnant days old and 2 months old) were provided by Animal Center, Tongji Medical College,Huazhong University of Science and Technology. Single antibody of NSE, multi-antibody of glial fibrillary acidic protein (GFAP) and single antibody of neurofilament (NF200) were provided by Wuhan Boster Company.METHODS: The experiment was carried out in Immune Opening Laboratory and Central Laboratory, Basic Medical Department, Tongji Medical College, Huazhong University of Science and Technology in September 2006. Bones of lower extremities of rats were collected to centrifuge BMSCs. Fetal spinal cord tissue homogenate was extracted from 16 pregnant days old rats to make inducing solution and induce differentiation of BMSCs. Otherwise, embryo muscle tissue was used to make muscle tissue homogenate as the same way so as to regard as the controls.MAIN OUTCOME MEASURES: BMSCs underwent morphological observation after induction; in addition, anti-NSE, NF200 and anti-GFAP were used to label neurons and astrocytes, respectively. Ten non-overlapping sights were randomly selected from positive reactive-induced cells after immunohistochemical staining under optic microscope to calculate ratio of positive cells of NSE and NF200 counting for total numbers of cells.RESULTS: ① Morphological changes of BMSCs after induction: During early induction, optic microscope indicated that soma of partial cells rebounded; whose apophysis was long and thin; apophysis of differentiated cells grew gradually and intercrossed each other. It was similar to nerve cells and some branches were similar to dendrite branches. However,morphological changes of cells in the control group were not obvious. ② Expression of relevant antigens differentiated from BMSCs into neuron-like cells at one week after induction: Most cells in spinal cord homogenate group expressed as positive NSE and NF200; a few of cells expressed as GFAP. While, positive staining of nerve cell antibody was not observed in the control group; meanwhile, positive reaction of nerve cell antigen was not observed in the control group,too. Immunohistochemistry examination demonstrated that positive rates of NSE and NF200 expressions were (68±1.7)%and (76.2±2.9)%, respectively.CONCLUSION: Fetal spinal cord tissue homogenate can induce differentiation from BMSCs into neuron-like cells.
10.Effect of esculentoside A on apoptosis of mouse thymocyte

Objective:To investigate the effect of esculentoside (EsA) on apoptosis of murine thymocyte. Methods:Using electronic microscope, DNA agarose electrophoresis and flow cytometry analysis,the effect of EsA on apoptosis of murine thymocyte was examined. Results:The result showed that apoptosis of activated thymocyte by ConA was markedly promoted by 2.5, 5, 10 ?g/ml EsA in murine thymocyte culture for 3 h, but the spontaneous apoptosis was not affected by EsA. Conclusion:The results suggest that EsA has the positive effect on apoptosis of activated murine thymocyte.
