1.Anti- Tumor Effect of Vaccination with Irradiation Tumor Cells Transduced with Mouse Interferon-Gamma Gene

The whole length mouse interferon-gamma (mIFN-?) cDNA was obtained using RT-PCR and was introduced into mouse hepatoma cell line H22 by retroviral vector. We examined the tumorigenicity of mIFN-? gene modified H22 in experimental animals. The results indicated that the gene modified tumor cells had less tumorigenicity than the parental tumor cells and the tumor vaccine had therapeatic effects on tumor-bearing animals.
2.Study on the inhibitory effects and related toxicity of all-trans-retinoic acid combined with arsenic trioxide on transplanted tumor of human bladder neoplasms in nude mice

ObjectiveTo investigate the inhibitory effects and related toxicity of all-trans-retinoic acid(ATRA) combined with arsenic trioxide(As2O3) on transplanted human bladder neoplasms in nude mice.MethodsThe subcutaneously transplanted tumor models of human bladder neoplasms in nude mice were established and then it was randomly divided into four teams: Saline group, ATRA group, As2 O3 group and the combination of ATRA and As2 O3 group.Each group composed of 10 nude mice and received intratumor injection in the following 14 d.The tumor growth inhibitory rate was calculated.The pathological changes of tumor,cardiac, liver, and kidney were observed after H.E.staining.The expression of MVD (Marked with CD43) and VEGF were detected by SP of immunohistochemistry.Blood routine examination and hepatic and nephritic functional examination was carried out to evaluate the side-effects.Results The tumor growth was significantly suppressed in ATRA group(inhibitory rate was 41.82%) and As2O3 group (inhibitory rate was 43.77%) compared with saline group.When ATRA combined with As2O3 ,inhibitory effects were improved(inhibitory rate was 68.55%).The difference of growth inhibitory rate between each group was obviously significant(x2 =26.81, P <0.01).There was less expression of MVD (Marked with CD43) and VEGF in ATRA group and As2 O3 group and the combination group compared with saline group (ODVEGF = 19.23 + 2.32,20.72 + 2.01,17.16 ± 1.59,27.33 ± 2.17, respectively; ODMVD = 44.77 + 8.25,43.39 + 7.41, 30.56 + 7.71,141.12 + 8.38, respectively), and the suppressed effect in the combination group was most significant (t = 3.16,3.08,3.37, P < 0.01).WBC inhibition was seen in ATRA group and As2O3 group and the combination group, but no significantly difference among them(P >0.05).Hepatic and nephritic toxicity were not found.ConclusionsCombination of ATRA and .As2O3 had obvious synergistic effects on transplantéd tumor of human bladder neoplasm in nude mice, and moderate white blood cell inhibition and no hepatic or nephritic toxicity.

4.Fluid balance and myocardial function relate to mortality of patients with septic shock

Objective To evaluate the correlation between fluid balance and myocardial function and the mortality in patients with septic shock.Methods Ninety two septic shock patients enrolled between January 2013 and January 2015 were divided into survival group and non-survival group based on 28-day mortality.Fluid balance,haemodynamic parameters and B-type natriuretic peptide (BNP) were collected on days1,3,7 after admission.Results Forty four patients survived the crisis while 48 died.The 7 day average fluid balance volume in survival group was lower than those in non-survival group [(55 ± 9)ml/kg vs.(187 ± 14)ml/kg,t =54.260,P <0.01].The initial left ventricle ejection fraction (LVEF) in survival group was lower than that in non-survival group [(42% ±4%) vs.(54% ±6%),t =10.100,P <0.01].Serum BNP level was significantly lower than those in non-survival group [(267 ± 54) ng/L vs.(681 ± 22) nig/L,t =48.580,P <0.01].Multivariate logistic regression analysis showed that fluid positive balance,LVEF and acute physiology and chronic health evaluation Ⅱ (APACHEⅡ) were the independent predictors for 28 day mortality (P < 0.05).Conclusions Fluid balance and myocardial function can serve as risk predictors for septic shock patients.
5.Predictive value of serum macrophage migration inhibitory factor for myocardial depression in severe sepsis patients

Objective To evaluate the predictive value of serum macrophage migration inhibitory factor (MIF) for myocardial depression in severe sepsis patients.Methods Taken prospective study method,42 cases of severe sepsis patients were enrolled from December 2011 to June 2013.The patients were monitored by PICCO system after informing consent into ICU day,and given the corresponding treatment.According to the hemodynamic parameters were divided into two groups,myocardial depression group:cardiac index < 3 L/(min· m2) and global end diastolic volume index > 800 ml/m2,the remaining were in non myocardial depression group,and the patients were divided into survival group and death group according to 28-d mortality.Hemodynamic parameters and serum MIF,B-type natriuretic peptide (BNP),cardiac troponin Ⅰ (cTnI) level at the 1st,3rd,Sth day after admission.Results Fortty-two severe sepsis patients,non myocardial depression in 22 cases (non myocardial depression group),myocardial suppression in 20 cases (myocardial depression group).At 28 d,20 patients of survival (survival group),22 patients of death (death group).The serum MIF in death group was higher than that in survival group at the 1st,3rd,5th day (P < 0.01) ; BNP and cTnI at the 1st day in two groups had no statistical significance (P >0.05),BNP and cTnI at the 3rd,5th day in death group was higher than that in survival group(P < 0.01).The serum MIF at each time point in myocardial depression group was higher than that in non myocardial depression group [survival:8.70(3.53,16.80) μ g/L vs.1.20(0.80,1.77) μ g/L,2.30(1.33,8.40) μ g/L vs.0.60 (0.60,0.99) μg/L,0.50 (0.31,2.50) μg/Lvs.0.16 (0.15,0.20) μg/L;death:11.43(8.10,17.16) μ g/L vs.2.30(1.96,3.69) μ g/L,9.70(6.55,14.65) μ g/L vs.1.90(1.88,5.27) μ g/L,7.50(5.15,14.20)μ g/L vs.2.40(0.80,8.46) μ g/L] (P < 0.05).The serum MIF of death patients in two groups was higher than that of survival patients (P < 0.05).MIF at the 5th day had biggest prediction value,AUC was 0.952,when the cut-off point of 0.65 μg/L,the sensitivity was 100% (22/22) and specificity was 85% (17/20).Multivariable Logistic regression analysis results showed that only APACHE Ⅱ scores was the independent risk factor for 28-d mortality (P < 0.01),while MIF couldn' t independent forecast 28-d mortality (P > 0.05).Conclusions Elevation of serum MIF prompts severe sepsis patient with poor outcome,and dynamic changes of MIF is helpful to evaluate the prognosis and severity of severe sepsis patients with myocardial depression,but MIF is not an independent risk factor for predicting 28-d mortality.
6.Minimally invasive treatment for hypertensive intracerebral hemorrhage through a straight incision and keyhole craniotomy

Objective To evaluate the effect of surgical treatment of hypertensive intracerebral hemorrhage (HICH) through a straight incision and keyhole minimally invasive craniotomy. Methods According to the location of the hematoma revealed by preoperative CT scans, a straight skin incision was made 4~5 cm in length, and then a keyhole craniotomy 2 cm in diameter was performed. The underlying cortex was incised the hematoma was exposed and removed under microscope. Results The hematomas were thoroughly cleared in 17 cases. The clearance rate was 90% in 18 cases and 80% in 4 cases. Re-hemorrhage occurred in 2 cases after operation. A total of 35 cases was followed for 0.5~3 years (mean, 2.1 years). The quality of life was assessed by activity of daily living (ADL) classification, which revealed 9 cases of grade 1, 12 cases of grade 2, 9 cases of grade 3, 4 cases of grade 4, and 1 case of grade 5 at the 6th postoperative month. The mortality of this series was 10.3% (4/39). Conclusions Straight incision keyhole minimally invasive craniotomy is a rapid, effective, and safe technique for the removal of hypertensive cerebral hemorrhage. The method herein provides an effective decompression of hematoma, with low recurrence rate and good prognosis compared with conventional surgery.
7.Transcriptional activation function of intracellular domain of EPOR and G-CSFR protein in yeast: a comparative study

Objective To investigate the feasibility of searching for proteins which interact with intracellular domain of hematopoietic growth factor receptors using yeast two-hybrid system. Methods RT-PCR method was performed to amplify the genes of intracellular domains of G-CSF receptor and EPO receptor in NFS-60 and BET-2 cells of mice. The genes were cloned into yeast expression plasmid pGBKT7 vector,and then transformed into yeast AH109. The yeast proteins were isolated and analyzed with Western blotting. Transcriptional activation was analyzed by the ?-galactosidase colony-lift filter assay. Results The intracellular domains of G-CSF receptor and EPO receptor genes were successfully cloned into pGBKT7 vector. The results of Western blotting assay showed that both proteins were expressed in the yeast cells. The ?-galactosidase colony-lift filter assay demonstrated that G-CSF receptor alone had no activity of transcriptional activation,while the EPO receptor alone could activate transcription. Conclusion The findings suggested that intracellular domain of G-CSF receptor could be used as a bait to find interacting proteins using yeast two-hybrid system,while that of the EPO receptor could not. Therefore the system could not be applied to all hematopoietic factor receptor.
8.The predictive value of serum presepsin (sCD14-ST) for myocardial depression in septic patients

Objective To evaluate the predictive value of serum presepsin (sCD14-ST) for septic myocardial depression (SMD) in patients with severe sepsis or septic shock.Methods This was a prospective cohort study.A total of 84 patients with severe sepsis or septic shock were monitored by pulse indicator continuous cardiac output (PiCCO) system and divided into myocardial depression group (cardiac function index [CFI]<4.1 L/min, n=37) and non-myocardial depression group (CFI≥4.1 L/min, n=47) according to cardiac function index (CFI).Left ventricular ejection fraction (LVEF) was measured by doppler echocardiography at admission.The patients were divided into survival group (n=40) and non-survival group (n=44) based on 28-day mortality.Serum sCD14-ST,B-type natriuretic peptide(BNP), tumor necrosis factor-α(TNF-α), cardiac troponin Ⅰ (cTnⅠ) levels and hemodynamic parameters were observed dynamically at day 1,3,5 after admission.Quantitative data with normal distribution were analyzed using t-test and abnormal distribution data were analyzed using Mann-Whitney U test.Categorical data were analyzed using χ2 test.Results The serum levels of cardiac index (CI), global ejection fraction (GEF) and left ventricular contractility index (dPmax) in myocardial depression group were all significantly lower than those in non-depression group at day 1 after admission (all P<0.01).The serum levels of sCD14-ST, TNF-α and BNP in myocardial depression group were all significantly higher than those in non-depression group (all P<0.01).Serum sCD14-ST levels in 84 septic patients were positively correlated with both BNP and TNF-α (r=0.204, P<0.05 and r=0.516, P<0.01, respectively).The area under the receiver operating characteristics curve (AUC) of sCD14-ST was 0.782 for myocardial depression in patients with sepsis, with a sensitivity of 89.2% and a specificity of 78.0% at the cut-off point of 988 mg/mL.The predictive value of sCD14-ST was inferior to BNP and TNF-α (both P<0.05).The combination of sCD14-ST, TNF-α and BNP could provide better prediction value in septic myocardial depression.Logistic regression analysis showed that serum sCD14-ST was not the in dependent predictor for septic myocardial depression (P>0.05).There were 24 cases died in myocardial depression group.The mortality of myocardial depression group was significantly higher than that of non-depression group (64.9% vs 42.6%, χ2=4.132, P =0.042).The serum levels of sCD14-ST at day 1 and day 3 in myocardial depression group were significantly higher than those in non-myocardial depression group (both P<0.01).sCD14-ST levels in both groups showed downtrend.The serum level of sCD14-ST in non-survival group was significantly higher than that in survival group (P<0.01).Conclusions Myocardial depression is common in patients with severe sepsis and septic shock.High serum level of sCD14-ST is correlated with myocardial depression to some extent, but not an independent predictor.The combination of sCD4-ST, BNP and TNF-α can improve the predictive value for myocardial depression.
9.Endoscopic-Assisted Correction of Facial Contour Deformities

Objective To evaluate the efficacy of endoscopic techniques for corrections of facial contour deformities.MethodsA total of 58 patients(48 women and 10 men)with square face(38),prominent zygoma(12),or crooked noses(8)were treated by endoscopic-assisted technique in our hospital.Results The operation time ranged from 40 to 86 min(mean,65 min),and intraoperative blood loss was 60-120 ml(mean,78 ml).The patients were discharged from the hospital in 3 to 7 days(mean 4)after the surgery,and was followed up for 3 to 24 months.During the period,no one had unexpected fracture,injuries to the mental and facial nerves and vessels,or infection.All the patients were satisfied with the surgical outcomes.ConclusionsEndoscopic-assisted correction of facial contour deformities is feasible,safe,and effective.
10.The Expression of TGF-?_1 And Its Significance in Human Gastric Cancer

Objective To study the role TGF-? 1 expression in the gastric cancer and to evaluate its clinical and pathological significance.Methods The expression of TGF-? 1 was studied by using S-P immunohistochemical technique in 20 cases of normal gastric tissue and 46 cases of gastric carcinoma.Results in the gastric cancer patients,the positive rate of TGF-? 1 expression was 100%,but there was 50% positive rate in the normal gastric tissues.The TGF-? 1 expression in group with lymph node metastasis was higher than that without lymph node metastasis.Conclusion TGF-? 1 plays an important role in the progress of gastric cancer.
