1.Effect of lamivudine on clinical relevant indicators of patients with liver cirrhosis after splenectomy

Objective To explore the effect of lamivudine on clinical relevant indicators of patients with liver cirrhosis after splenectomy.Methods 30 cases of patients with liver cirrhosis after splenectomy from July 2002 to July 2012 were selected in Ningxia Shizuishan First People's Hospital, and randomly divided into control group and observation group, each had 15 cases.Control group underwent splenectomy only, the observation group was given oral lamivudine, 100 mg/times, one times/d, taking more than one years after splenectomy.The changes of HBV-DNA ration, liver function, prothrombin time and other indexes of the two groups before and after treatment were observed in the treatment process, and the complications after treatment were also observed.Results The hepatitis B virus gene ( HBV-DNA ) quantitative examination results of patients in observation group with negative rate was 33.33%, the control group had no change; total bilirubin(T-BIL), alanine aminotransferase(ALT) in observation group decreased significantly than control group(P <0.05), albumin(ALB) increased significantly(P <0.05), prothrombin time(PT) also decreased significantly ( P<0.05).There were two cases of complications such as ascites, gastrointestinal bleeding in the observation group, which eight cases in control group, the difference was significant.Conclusion The combination of lamivudine treatment on patients with cirrhosis after splenectomy improves the life quality of patients, safely and effectively promoting the rehabilitation of patients.
2.Effects of Shengji Huayu Recipe and its decomposed formulas on synthesis of collagen types I and III in granulation tissue of rats in early wound healing

OBJECTIVE: To study the effects of Shengji Huayu Recipe (a traditional Chinese medicine compound recipe for resolving stagnation and promoting granulation) and its decomposed formulas (Huayu Recipe for resolving stagnation and Shengji Recipe for promoting granulation) on the synthesis of collagen types I and III in granulation tissue of rats in early wound healing. METHODS: Twenty-four male Sprague-Dawley (SD) rats with full-thickness skin lesion were randomized into 4 groups: Shengji Huayu Recipe-treated group, Shengji Recipe-treated group, Huayu Recipe-treated group and untreated group. Collagen types I and III in granulation tissue of the rats were tested with immunohistochemical methods and image analysis. RESULTS: On the third day of wound healing, collagen I of the rats in both Shengji Huayu Recipe-treated group and Shengji Recipe-treated group was higher than that in the untreated group, and collagen I of the rats in Huayu Recipe-treated group was lower than that in the untreated group (P<0.05). Collagen III of the rats in the three treated groups were lower than that in the untreated group (P<0.05). On the seventh day of wound healing, Collagen I of the rats in both Shengji Huayu Recipe-treated group and Shengji Recipe-treated group was higher than that in the untreated group (P<0.05), and collagen III of the rats in both Shengji Recipe-treated group and Huayu Recipe-treated group was higher than that in the untreated group (P<0.05). CONCLUSION: Resolving stagnation and promoting granulation therapy can promote the wound healing in rats.
3.Regulation of Transforming Growth Factor -beta 1/Smad3 Signaling Pathway of Asthmatic Rat Airway Remodeling Model by Back Three-acupoint Electroacupuncture

Objective To observe the effect of electroacupuncture on back three-acupoints ( namely bilateral Dazhu, Fengmen, Feishu) on the protein and mRNA expression of transforming growth factor-beta 1 (TGF-β1) and Smad3 in asthmatic rat airway remodeling model, and to explore its therapeutic efficacy and molecular mechanism. Methods Rat asthma model was established by inhalation of ovalbumin. After sensitization for 6 weeks, rats were killed. And then the airway morphological parameters of rats were measured by image analysis. The protein and mRNA expression of TGF-β1 and Smad3 in lung tissues were detected by immunohistochemistry and real-time quantitative polymerase chain reaction (qRT-PCR) respectively. Results Compared with the blank group, the standardized values of muscle cross-sectional area including airway smooth muscle area (WAm) /perimeter of the basement membrane (Pbm), and bronchial inner wall area (WAi)/Pbm were increased in the model group. The protein and mRNA expression levels of TGF-β1 and Smad3 were also increased in the model group. In electroacupuncture group, the above observation indexes were decreased, and the differences were statistically significant ( P<0.05 compared with the model group). Conclusion The experimental results indicated that back three-acupoint electroacupuncture has an effect on remodeling airway, and one of the mechanisms is probably associated with the regulation of TGF-β1/Smad3 signaling pathway.
4.Effects of resolving stagnation and promoting granulation therapy on expressions of Bax and Bcl-2 in granulation tissue of diabetic rats during wound healing

OBJECTIVE: To study the effects of resolving stagnation and promoting granulation therapy on expressions of Bax and Bcl-2 in granulation tissue of diabetic rats during wound healing. METHODS: Seventy-two male SD diabetic rats with full-thickness skin lesion were randomly divided into 3 groups: SJHYR 1-treated group, SJHYR 2-treated group and normal saline (NS) control group. SJHYR 1 was prepared with Shengji Recipe (SJR, a compound traditional Chinese herbal medicine for promoting granulation) and Huayu Recipe (HYR, a compound traditional Chinese herbal medicine for resolving stagnation) at a ratio of 1:2, while SJHYR 2 was prepared with SJR and HYR at a ratio of 1:1. Immunohistochemical method was used to assess Bax and Bcl-2 protein levels in granulation tissue. RESULTS: SJHYR 1 could accelerate wound healing as compared with SJHYR 2 and NS (P<0.05). On the third day in experiment, Bax and Bcl-2 proteins were not found in any groups, but on the seventh and eleventh day in experiment, Bax and Bcl-2 proteins in SJHYR 1-treated group were much higher than those in the other two groups (P<0.05). CONCLUSION: SJR and HYR in different ratios may all have a role in regulating Bax and Bcl-2 expression in granulation tissue of diabetic rats during wound healing.
5.Interferon-induced protein 44 is correlated with clinical features of patients with systemic lupus erythematosus

Objective To investigate the expression of interferon-induced protein 44 (IFI44) gene in the leukocytes of the peripheral blood samples from patients with systemic lupus erythematosus (SLE), and to evaluate the relationship between the expression level and disease activity. Methods Mononuclear cells in the peripheral blood samples from 100 SLE patients were compared with those of 40 disease controls and 40 healthy donors (HD) and the expression of the IFI44 was evaluated by quantitative real-time PCR.Comparisons between groups were performed with ANOVA, and the correlation analysis between the level of expression was higher in SLE patients than disease controls and healthy donors (26.8±5.3, 7.4±2.7, 5.2±2.0,respectively) (P=0.0012, P=0.005), but no difference was found between disease controls and healthy donors. Mild disease activity and the SLE patients with stable disease (63.1±22.4, 28.0±7.2, 9.2±1.8, respectively)and 24 hours urine protein level (r=0.42, P=0.000). Conclusion IFI44 is demonstrated to be highly expressed in SLE patients. The level of IFI44 may be a promising candidate biomarker for identifying SLE activity.
6.Inhibiting Effect of Ultra-micro Powder Qiweibaizhusan on Bacteria in Vitro

Objective To observe the bacteriostatic effect of single herbs, traditional complex prescription and ultra-micro powder of Qiweibaizhusan. Methods The inhibiting zone and MIC of single herb and compound of Qiweibaizhusan on Escherichia coli, Staphylococcus aureus, Eubacterium aerofaciens, Pseudomonas aeruginosa, Salmonella sp, Saccharomyces cerevisiaes and Candida glabrata were measured by filter paper method. Results The growth of the tested bacteria and yeast except Staphylococcus aureus and Salmonella sp were inhibited by ginseng. The antibacterial effect of licorice was the best, and only Pseudomonas aeruginosa’s growth was not inhibited by licorice. The growth of Staphylococcus aureus and Eubacterium aerofaciens were inhibited by Agastache rugosa. The growth of Escherichia coli, Staphylococcus aureus, Eubacterium aerofaciens and Pseudomonas aeruginosa were inhibited by Poria cocos. Only Eubacterium aerofaciens’s growth was inhibited by Radix aucklandiae and fried Atractylodes macrocephala. The growth of all the bacteria and yeast were not inhibited by Radix puerariae. The growth of Staphylococcus aureus, Eubacterium aerofaciens and Salmonella sp were inhibited by the traditional decoction and ultra-micro powder of complex prescription of Qiweibaizhusan, and all the MIC of ultra-micro powder were smaller than the traditional decoction. Conclusion The main antibacterial component of Qiweibaizhusan was ginseng and licorice. The inhibiting effect of ultra-micro powder on bacteria was better than traditional decoction of Qiweibaizhusan in vitro.
7.Progress on identification and management of Parkinson′s disease-related fatigue

Fatigue is one of the most common and disabling symptoms of Parkinson′s disease (PD), which has an important impact on the quality of life. Due to the lack of a strict and unified definition and classification of PD-related fatigue, as well as a clear quantitative standard, clinical studies on fatigue in PD have drawn different conclusions, making fatigue research challenging. Normative studies in this area have been complemented by the publication of the 2016 expert consensus on diagnostic criteria for PD associated fatigue. In this paper, the incidence, influencing factors, clinical evaluation measures and treatment of PD-related fatigue were reviewed to provide the latest information for clinical identification and management of PD-related fatigue patients.
8.Carrier detection and prenatal diagnosis of hemophilia Alpha.

OBJECTIVETo establish an effective laboratory examination system for carrier detection and prenatal diagnosis of haemophilia Alpha(HA) with a variety of molecular biological methods which are simple,rapid and easy to use.
METHODSDetection of inversion involving intron 22 in the FVIII gene was completed by long distance polymerase chain reaction (PCR) and linkage analysis was performed by using several genetic polymorphisms including an intragenic Bcl I RFLP, 2 STRs and an extragenic St14 VNTR.
RESULTSIntron 22 inversion was observed in 10 out of the 21 (47.6%) pedigrees examined. Prenatal diagnosis was completed in 3 pedigrees. A further combination of the four intragenic and extragenic polymorphic loci gave an informative rate of 94.7%.
CONCLUSIONSFemale relatives in HA families with inversion can be detected with direct diagnostic procedure. The application of long distance PCR makes the detection much more simple and rapid. For families without inversions,it is easier and more cost-effective to undertake linkage analysis of genetic polymorphism based on PCR.

9.Risk factors for hypoparathyroidism after thyroid papillary carcinoma

Objective To determine the clinicopathological risk factors and reliable biochemical predictors of the development of hypoparathyroidism after total thyroidectomy plus central compartment node dissection in papillary thyroid carcinoma (PTC).Methods A study was performed on 93 patients who underwent a total thyroidectomy with central compartment node dissection due to PTC.The rate of hypoparathyroidism was calculated.We evaluated the correlations between hypoparathyroidism and clinicopathological factors by chi-square test and logistic regression model for multivariate analysis.The prediction value of PTH and serum calcium level was assessed by a 2×2 contingency table and ROC curve analysis.Results Postoperative hypoparathyroidism was found in 46 patients (49.5%) and 2 with permanent hypoparathyroidism.Univariate analysis showed that tumor size (P=0.034),perithyroidal extension (P=0.003),bilateral cancer(P=0.045)and bilateral central neck dissection (P=0.028)were risk factors for postoperative hypoparathyroidism in patients with PTC.Multivariate analysis showed that perithyroidal extension (P=0.003) and bilateral central neck dissection(P=0.044)were independent risk factors for postoperative hypoparathyroidism in patients with PTC.ROC curve analysis showed that PTH level in the first after operation played significant roles in predicting hypoparathyroidism(AUC 0.875).Conclusions Hypoparathyroidism is the most common complication after total/near-total thyroidectomy.Perithyroidal extension and bilateral central neck dissection are the important risk factors of hypoparathyroidism.The level of PTH is a reliable and early predictive indicator of postoperative hypoparathyroidism.
10.Study on extraction technology of Fuyuan Xingnao granule

Objective To study the extraction technology and optimize the extraction process of Fuyuan Xingnao gran-ule.Methods The preliminary extraction process was designed by routine extraction method combined with the results of pharmacological activity experiment.The extraction process was optimized with orthogonal test.Results The extract was ob-tained by ethanol reflux extraction,water decoction extraction or ethanol precipitation followed by filtration and concentration. The optimal process for ethanol reflux extraction was extracting twice with 50% ethanol.The ratio of liquor to dry material was 8∶1 for the first time and 6∶1 for the second time.The extraction time was 120 minutes for the first time and 80 minutes for the second time.The optimal process for water decoction extraction was extracting twice with 120 minutes and 80 minutes each time.The ratio of liquor to dry material was 10∶1 for the first time and 8∶1 for the second time.The extraction temper-ature was100 ℃.The best ethanol precipitation process was 16 hour extraction with 80% ethanol.The ratio of ethanol to dry material was 2∶1.Conclusion The quality of Fuyuan Xingnao granule extract with this optimized extraction process was in line with requirements of Pharmacopoeiaof the People's Republic of China 2015,4 Volumes(0104 granule).
