1.Flexible dilatation combined with topical betamethasone cream in treatment of child phimosis

Two hundred and twenty five children aged 4-12 years with phimosis were treated by flexible dilatation combined with topical betamethasone cream from January 2003 to January 2015.Two hundred and eight cases (92.44%) were completely cured and penile edema and urination were returned to normal 7 d after a single operation;while,17 children received 2-3 additional flexible expansion.During follow-up of 2-24 months,221 children achieved the cure standards with an overall cure rate of 98.22%.Flexible dilatation combined with topical betamethasone cream is effective modality for phimosis in children,with less surgical trauma and low complication rate.
2.Relationship between apolipoprotein E ε4,ε2 alleles and intrauterine growth

Objective To investigate the relationship between apolipoprotein E (ApoE) ε4,ε2 alleles and intrauterine growth.Methods ApoE genotypes of 1418 people born in Peking Union Medical College Hospital were analyzed,allele frequencies were calculated and their parameters at birth were collected.To compare ApoE ε4,ε2 alleles with parameters at birth through single factor analysis and Logistic regression analysis.Results The alleLic frequencies of e2,83 and 84 were 8.11%,83.39% and 8.50% in the group.The results of single factor analysis showed that there was significant difference between the distribution of ponderal index (PI) in the ApoE ε2 allele group(χ2=4.87 ,P=0.027).While there was no significant difference between the distribution of head circumference at birth,placental weight and gestational age in the ApoE ε2 allele group.ApoE ε2 allele showed negative correlation with small PI in the logistic regression analysis(χ2=5.077 ,P=0.024),after adjusted for gender,age,head circumference at birth,placental weight,gestational age,parity and maternal age at delivery.No association between ApoE ε4 allele and parameters at birth was found.Conclusions ApoE ε2 allele may have protective effect on PI.No association was found between ApoE ε4 allele and intrauterine growth.
3.Proliferation activating effects of Mtb-Ag on γδT cells and its effects on expression of CD69 molecules inγδT cells

Objective To activate and amplify γδT cells with low molecular peptide antigen of Mycobacterium tuberculosis low molecular peptide antigen (Mtb-Ag),and to investigate the expression of CD69 molecules on γδT cellular surface.Methods Healthy human peripheral blood mononuclear cells (PBMC) were obtained and separated,then positive cells were isolated by immuno-magnetic beads selection,and the proportion of γδT cells in the PBMCs was detected by fluorescent monoclonal TCR γδT-PE staining and flow cytometry.Expression of CD69 molecules in γδT cells was detected by γδ-PE/CD69 FITC double staining.Results The proportion of γδT cells was (4.9±1.85)% in freshly obtained PBMC,(69.2±6.57)% after 10 d of Mtb-Ag activation,and (99.3±8.92)% after immuno-magnetic beads selection.The expression of CD69 molecules in γδT cells reached the peak (75.2%) at 24 h after initial Mtb-Ag stimulation,and reached the peak (72.0%) at 6 h after second stimulation.Conclusions MtbAg can specifically stimulate the proliferation of γδT cells in the PBMC.Both its initial and the second stimulation can specifically activate γδT cells.
4.Rhytidectomy by multi-method joint application of W incisions in temporal hair line

Objective To introduce an operative method of rhytidectomy with W-incision and fewer complications for facial rejuvenation.Methods A total of 84 patients were treated with rhytidectomy by multi-methods and followed up from January 2007 to December 2012.The rhytidectomy was done along the temporal hairline edge through the combined procedures,including sharp and blunt separation,liposuction and filling with autologous fat,tighting and fixing the fascial flap of superficial musculoaponeurotic system,as well as the suspension of the zygomatic cheek fiber fat pad.Results Follow-up for 3 moths to 2 years after surgery in 84 patients showed that the wound was healing with good concealment and without complications such as facial nerve injury or local uneven.All the cases achieved good results in facelift with high satisfaction rate after the combined operations.Conclusions This combined rhytidectomy is safe,effective,simple and easy.And it is a good and ideal approach of facial rejuvenation worthy of clinical application.
5.The application of delayed skin grafting combined traction in severe joint cicatricial contracture.

OBJECTIVETo investigate the effect of delayed skin grafting combined traction in severe joint cicatricial contracture.
METHODSAt the first stage, the joint cicatricial contracture was released completely with protection of vessels, nerves and tendons. The wound was covered with allogenetic skin or biomaterials. After skin traction for 7-14 days, the joint could reach the extension position. Then the skin graft was performed on the wound. 25 cases were treated from Mar. 2000 to May. 2013.
RESULTSPrimary healing was achieved at the second stage in all the cases. The skin graft had a satisfactory color and elasticity. Joint function was normal. All the patients were followed up for 3 months to 11 years with no hypertrophic scar and contraction relapse, except for one case who didn' t have enough active exercise on shoulder joint.
CONCLUSIONDelayed skin grafting combined traction can effectively increase the skin graft survival rate and improve the joint function recovery.

6.The value of rubella IgG avidity test in the diagnosis of recent rubella infection

Objective To evaluate the value of rubella IgG avidity test in the diagnosis of recent rubella infection.Methods We detected the rubella IgG avidity of three groups of serum samples:48 acute infection samples from BIOMEX seroconversion panels RP-001,RP-014 and RP-011,38 rubella IgM positive samples,978 past infection samples (IgG positive IgM negative).Results All the acute infection samples from BIOMEX seroconversion panels RP-001 (50 days after infection),RP-014 (42 days after infection),RP-011 (71 days after infection) show IgG low avidity.Of the 38 rubella IgM positive samples,24 samples show IgG low avidity,9 samples show IgG high avidity,6 samples show grey zone.Of the 978 past infection samples,963 samples show IgG high avidity,9 samples show IgG low avidity,6 samples show grey zone.Conclusions Rubella IgG avidity test can be used to rapidly distinguish between past and acute infection.Of the rubella IgM positive samples,about 25% show IgG high avidity,indicate past infection,so it is important to detect IgG avidity for this population.
7.A novel splicing mutation in COL1A1 gene caused type I osteogenesis imperfecta in a Chinese family.

OBJECTIVETo study a family affected with osteogenesis imperfecta for potential mutations in COL1A1 gene.
METHODSClinical data of an affected family was collected. Potential mutation of the COL1A1 gene was screened using polymerase chain reaction and direct sequencing. Suspected mutation was detected in 20 unaffected relatives and 200 unrelated healthy controls.
RESULTSAnalysis of RNA splicing has revealed a c.3208G/A mutation, which created a new splice sites and led to a frameshift mutation. The same mutation was not detected in the unaffected relatives or the 200 healthy controls.
CONCLUSIONMutations of the COL1A1 gene are one of the major causes of osteogenesis imperfecta in Chinese population. Our finding has enriched the mutation spectrum of type I collagen genes.

8.Morphology and pathogenesis of 47, XYY/47, XY, +mar identified in patients with super male syndrome.

OBJECTIVETo explore the source and morphology of supernumerary markers from patients with 47,XYY/47,XY, +mar and supermale syndrome.
METHODSConventional GTG banded karyotyping and dual-color fluorescence in situ hybridization (FISH) were performed on 21 such patients.
RESULTSAmong these cases, 18 had their small supernumerary marker derived from the Y chromosome. Three were derived from autosomal chromosomes. Those derived from Y chromosome were small fragments with centromeres, while those derived from autosomes were in the ring form.
CONCLUSIONIn children with supermale syndrome and 47,XYY/47,XY,+ mar, the supernumerary marker chromosomes primarily derive from sex chromosomes. These small chromosomes mainly have the forms of small segments with centromeres or rings. For such children, molecular cytogenetic analysis can facilitate genetic counseling and prenatal diagnosis.

9.Application of eight-probe fluorescence in situ hybridization and R-banding karyotype analysis for the diagnosis of acute lymphoblastic leukemia.

OBJECTIVETo assess the value of eight-probe fluorescence in situ hybridization (FISH) and R-banding karyotype analysis for the diagnosis of acute lymphoblastic leukemia (ALL).
METHODSWith the eight-probe FISH (using probes for MYC, P16, E2A, CHIC2/D10Z1/D17Z1, TEL/AMLl, MLL, BCR/ABL1, and IGH) and R-banding karyotype analysis, 237 cases of ALL were analyzed.
RESULTSCytogenetic changes were detected in 135 (56.96%) of all cases, which have involved MYC, P16, E2A, CHIC2/D10Z1/D17Z1, TEL/AMLl, MLL, BCR/ABL1, and IGH polyploidies. R-banding karyotype analysis has only detected abnormalities in 48 of such cases, in addition with 14 abnormalities missed by the FISH probes, which have given a total positive rate of 26.16%. The detection rate of the two methods has differed significantly(P<0.05).
CONCLUSIONCompared with the R-banding karyotype analysis, the eight-probe FISH is more accurate and efficient. Diagnosis of cytogenetic abnormalities for children with ALL using the combined method can provide a basis for evaluation of prognosis as well as personalized therapy.

10.Effect of Acupuncture on Cx43 Protein Expression in the Hippocampal Region in Rats with Acute Cerebral Infarction

Objective To investigate the effect of acupuncture on gap junction protein Cx43 expression in the hippocampal region in ischemia/reperfusion rats.Methods Wistar rats were randomized into normal, model, non-point acupuncture and acupuncture groups. A rat model of focal cerebral ischemia/reperfusion was made by modified middle cerebral artery thread occlusion. The model, non-point acupuncture and acupuncture groups were separately divide into four subgroups: 30, 60, 180 and 360 min ischemia/reperfusion, 10 rats each. The middle and lateral lines of vertex were given electroacupuncture in the acupuncture group of rats. A subcostal fixed point 10 mm above the iliac crest on the affect side was selected as an acupuncture point in the non-point acupuncture group. The model group was not treated. Cx43 protein expression was determined by an immunohistochemical method.Results There was a statistically significant difference in the behavior disorder (Bederson’s) score between the acupuncture group and the non-point acupuncture or model group after different times of cerebral ischemia/ reperfusion (P<0.05). There was a statistically significant difference in hippocampal Cx43 content between model, acupuncture or non-point acupuncture group and the normal group and between the acupuncture group and the model or non-point acupuncture group after different times of cerebral ischemia/reperfusion (P<0.05).Conclusions Acupuncture can inhibit the overexpression of Cx43, intervene in cerebral ischemia-reperfusion injury and produce a neuroprotective effect in ischemic brain injury.
