Objective To study the effect of rising [Ca 2+ ]i on the expression of Caspase 3, Bax and Bcl 2 in human breast cancer MCF 7 cell lines. Methods After the MCF 7 cells treated with different dose of thapsigargin(TG), intracellular calcium concentrations in MCF 7 cells were measured by calcium sensitive fluorescence indicator Fura 2/AM technique; and immunohistochemical method was used to detected the expressions of Caspase 3, Bax and Bcl 2. Results Bax was over expressed and Bcl 2 was down expressed in a [Ca 2+ ]i concentration dependent way;nevertheless, the expression of Caspase 3 was negative in all the TG treated cells. Conclusions The increase of intracellular calcium concentration may have an inductive effect of apoptosis on MCF 7 cells, which is independent to Caspase 3.

Objective To study the apoptotic induction effect of Thapsigargin on estrogen receptor positive human breast cancer cell lines MCF 7. Methods Cells were treated with Thapsigargin and 5 FU in vitro . The rate of cell apoptosis and distribution of cell cycle were detected on flow cytometry. The cell viability was measured by MTT assay and ultrastructural changes in apoptotic cells were confirmed by transmission electron microscopy.Results Thapsigargin could increase the rates both of cell apoptosis and growth supression of MCF 7 cells induced by 5 FU and alter the distribution of cell cycle. Under electron microscope, apoptotic bodies in MCF 7 cells considerably increased.Conclusion Thapsigargin apparently enhances the effect of apoptotic induction of 5 FU on MCF 7 cells, it is worthy of being further studied.

Objective To study the correlation between the polymorphism of GSTM1 and GSTT1 genes and the susceptibility to the development of COPD. Methods Polymerase chain reaction (PCR) was used to detect the GSTM1 and GSTT1 genotypes in 91 cases of the patients with COPD (COPD group) and 44 cases of the subjects without COPD (control group), and their susceptibility to the development of COPD were analyzed according to their gender, age and smoking history. AIM: To compare the quality of Liuwei Dihuang Pill among the pharmaceutical companies by determining the content and the dissolution of paeonol of Liuwei Dihuang Pill. METHODS: A method of determining paeonol and the dissolution from Liuwei Dihuang Pill was establishod. HPLC was used with methanol-water (70 ∶30) as a mobile phase and detection wavelength was at 274 nm. The flow rate was 1.0 mLmin, injection volume was 20 ?L. The percentage of dissolution was determined in the 30% methanol- 0.1 mol1 HCL solution. RESULTS: A good linearity was over the weight range of 1.4 ?g-140 ?g, the average recovery was 97.38% , the RSD for reproducibility was 1.44% (n=6). The parameter of Tianjin’s production was T_ 50 = 156.77 , T_d= 256.55 ,M= 0.734 and the parameter of Beijing’s production was T_ 50 = 110.66 , T_d= 197.19 , M= 0.583 . CONCLUSION: The method is simple, and can be used to determine the content and the percentage of dissolution of paeonol in Liuwei Dihuang Pill. The experiment indicates that the percentage of dissolution has no obvious distinction in products from different manufactures; but has obvious distinction in the batches of same manufacture.

Objective Study on the intracellular calcium and growth suppression of human breast cancer cells exposed by high intensity ultrasound (HIU). Methods Exposed human breast cancer cells MDA-MB-231 and MCF-7 in vitro with HIU (50 W/cm2). Examined the intracellular calcium from exposed cells with Fura-2 fluorescence probe. The cell viabilities were measured by MTT assay. The rate of cell apoptosis and distribution of cell cycle were detected on flow cytometry. Results The lever of intracellular calcium went up in pace with exposed time of HIU, MCF-7 cells were (572±20.1), (670±18.9), (815± 16.3) nmol/L (F = 663.65, P＜0.001), MDA-MB-231 cell was (582±16.3), (687±19.7), (843± 14.8) nmol/L (F = 863.06, P＜0.001), and the distribution of cell cycle waved to G0-G1, the ratios of G0-G1 in MCF-7 and MDA-MB-231 were (60.5±5.5)%, (66.3±7.0)%, (74.5±8.2)% (F=8.17, P = 0.002) and (58.5± 6.3) %, (66.1±6.3)%, (71.2±7.9) % (F=7.51, F= 0.003). Apoptotic rate upgraded gradually, the apoptotic rates of MCF-7 and MDA-MB-231 were (7.3±1.7)%, (13.2±3.5) %, (19.3±3.7)% (F= 18.73, P＜0.001) and (6.3±1.8)%, (11.4±2.31)%, (16.4±3.3)% (F = 19.26, P＜0.001). Under MTT assay, the rate of cell growth suppression increased significantly, the rates of cell growth suppression in MCF-7 and MDA-MB-231 were (9.2±2.2) %, (24.3±3.9)%, (48.6±5.5)%(F=117.16, P ＜0.001) and (9.0±1.7)%, (22.3±3.5) %, (416± 6.4)% (F =71.25, P＜0.001). Conclusion HIU enhanced the intracellular calcium of human breast cancer cells within given time and promoted the distribution of cell cycle to G0-G1. The rate of cell apoptosis and the cell's death rate increased evidently.

Objective To regulate nutrition support nursing, facilitate the development of nursing specialty, cultivate nutrition support nurse. Methods Establish nursing specialist team on nutrition support. 30 nurses received theory training, skill training, clinical practice, and completion reporting. Conduct nutrition knowledge management survey. Establish standardized wards of nutrition support nursing and conduct special inspection. Results Nursing specialist team on nutrition support was established, and 29 nutrition support nurses were cultivated. Before training, nurses' knowledge of nutrition and nutrition nursing management score were (3.46 ± 0.47), (0.70 ± 0.24) points, and the score were (4.63±0.14), (0.90±0.27) points after training, results was statistically significant (t=12.848, 2.981, P<0.01). Before and after special inspection of enteral nutrition, enteral nutrition pump and special infusion tube cases were 106,60 and 106, 60, results were statistically significant (χ2=4.747, 4.471, P<0.05). Before and after special inspection of parenteral nutrition, correct infusion and speed cases were 90, 92 and 61, 54, results were statistically significant (χ2=4.642, 7.644, P<0.05 or 0.01). Conclusions The development of nursing specialty of nutrition support was promoted, nurses′ comprehensive ability and self-identity was improved, management quality of nutrition support was.

Objective To observe the clinical efficacy of acupuncture plus medicine and function training in treating post-stroke shoulder-hand syndrome. Method Sixty patients with post-stroke shoulder-hand syndrome were randomized into a treatment group of 30 cases and a control group of 30 cases. The treatment group was intervened by acupuncture plus medicine and functional training, while the control group was only by functional training. Before and after intervention, the hand swelling degree, Visual Analogue Scale (VAS), shoulder joint motion scale, Fugl-Meyer Assessment (FMA) scale were used to measure the swelling and pain of hand, motion of shoulder, and motor function of upper limb. Result After intervention, the swelling and pain of hand, motion range of shoulder and motor function of upper limb were significantly improved in both groups (P<0.01), and the improvements in the treatment group were more significant than that in the control group (P<0.05). Conclusion Acupuncture plus medicine and function training can enhance the efficacy in treating post-stroke shoulder-hand syndrome, superior to pure function training.

Objective To explore the status of seroepidemiology on hepatitis A,B and C in primary and middle school students in Shufu county,Xinjiang Uygur Autonomous Region of China (Xinjiang) and to evaluate the effect of related immunization.Methods Students in four towns and villages were selected by cluster random sampling method.HAV-IgG,HBsAg,HBsAb and HCV-IgG were detected in Feb to May,2015.Results The overall HAV-IgG positive rate was 99.75％,among 4 830 primary and middle school students.The positive rates were seen 99.92％ in boys and 99.57％ in girls,with difference statistically significant (x2=5.798,P=0.016).The overall HBsAg positive rate appeared as 3.02％,with 3.55％ for boys and 2.47％ for girls,with difference statistically significant (x 2=4.782,P=0.029).The difference between age specific HBsAg positive rates also showed statistically significant (x2=71.990,P=0.000).HBsAg positive rate in the students in rural area (3.28％) was higher than that in the students in urban area (1.61％,x2=6.019,P=0.014).HBsAb positive rate was 38.84％,and the differences between the age specific HBsAb positive rates appeared statistically significant (x2=837.699,P=0.000).HBsAg positive rate in students from the urban area (42.36％) was higher than those from the rural area (38.20％,x2=4.598,P=0.032).2 815 students,accounting for 58.28％ of the total students,showed negative on both HBsAg and HBsAb.The overall HCV-IgG positive rate was 0.19％,and all appeared in students fiom the rural areas,with ethnicity solely as Uygur.Conclusions The effect of hepatitis A vaccine was satisfactory in primary and middle school students in Shufu county but quiet a number of the students missed the vaccination.The infection rate of hepatitis C was low.Publicity and health education on hepatitis immunization and control should be revved up.Programs regarding primary and supplementary immunization on hepatitis,should be carried out timely for children of school age.

Objective:To explore the mediating effect of empathy between personality traits and death attitude in nursing students.Methods:From April to May 2022, a total of 237 undergraduate nursing students were surveyed by general information questionnaire, big five inventory, Jefferson scale of empathy for nursing students, and death attitude profile-revised. Common method bias test, correlation analysis and descriptive analysis were conducted by SPSS 26.0 software.PROCESS macro program was used to test the mediating effect.Results:Agreeableness(32.78±4.92), empathy(110.03±16.83)were positively correlated with positive death attitude(57.95±12.35)( r=0.274, 0.571, both P<0.01), neuroticism(23.00±4.78)was positively correlated with negative death attitude(31.81±10.04)( r=0.199, P<0.01), empathy was negatively correlated with negative death attitude ( r=-0.226, P<0.01). Empathy partially mediated the relationship between neuroticism and negative death attitude, the mediating effect accounted for 16.08%(0.032/0.199) of the total effect, and empathy played a completely mediating role between the agreeableness and positive death attitude. Conclusion:The death attitude of undergraduate nursing students is mainly natural acceptance. Personality traits can directly affect death attitude and also indirectly affect death attitude through empathy.

ObjectiveTo investigate the effects of Asari Radix et Rhizoma-Zingiberis Rhizoma herb pair （XGHP） on lung and liver lipid metabolism in rats with chronic obstructive pulmonary disease （COPD）. MethodForty SD male rats were divided into a normal group （10 rats） and a model group （30 rats）. The method of cigarette smoke + tracheal injection of lipopolysaccharide（LPS） + cold stimulation was used to replicate COPD model with the syndrome of cold phlegm obstruction in lung. A COPD group， a XGHP group （5.4 g·kg^-1·d^-1）， and an aminophylline group （0.5 g·kg^-1·d^-1） were established after successfully inducing the model， with 10 rats in each group. After treatment， the serum triglyceride （TG）， total cholesterol （TC）， high-density lipoprotein cholesterol （HDL-C）， and low-density lipoprotein cholesterol （LDL-C） levels of rats in each group were measured. Gas chromatography-mass spectrometer （GC-MS） was used to detect the differential metabolites in the lung and liver tissues of rats in each group， and the relevant targets of the differential metabolites were predicted by network pharmacology. Molecular docking was used to verify the binding ability of key components in XGHP to the relevant targets in network pharmacology. The mRNA and protein expression levels of peroxisome proliferator-activated receptor α （PPARα） and fatty acid binding protein 4 （FABP4） in lung and liver tissues of rats in each group were detected by real-time polymerase chain reaction （PCR） and Western blot. ResultXGHP significantly increased the levels of TG， TC， and LDL-C in serum （P<0.05）， and decreased the level of HDL-C （P<0.05） in rats with COPD. GC-MS results showed that there were 8 lung differential metabolites and 17 liver differential metabolites in the COPD group and XGHP group. Network pharmacology predicted 59 common targets for the two differential metabolites， mainly enriched in the PPAR signaling pathway. Molecular docking results showed that the main components in XGHP were well combined with both PPARα and FABP4. Real-time PCR showed that XGHP effectively up-regulated the expression levels of PPARα and FABP4 mRNA （P<0.05）， and Western blot showed that XGHP effectively up-regulated the expression levels of PPARα and FABP4 proteins （P<0.05） in lung and liver tissues of rats with COPD. ConclusionXGHP effectively improves the blood lipid levels of rats with COPD， which may be related to the increase of the expression levels of PPARα and FABP4 mRNA and proteins in the PPAR signaling pathway， thus regulating lung and liver lipid metabolism.
