Mongolian gerbil is a multifunctional laboratory anima1 resource with Chinese characteristics.It has many advantages for some research fields, and play an important role.The more biological characteristics of Mongolian ger-bils discovered with in-depth research will promote diverse application of this resoure.In this paper, we reviewed the devel-opment of application of Mongolian gerbils in taxonomy, parasitology, microbiology, cerebral ischemia, lipid metabolism, neurological diseases, and cancer researches.
2.Determination of Vitexin in Cajanus cajan(L.) Millsp. by HPLC

Objective To develop a quantitative method to determine vitexin in Cajanus cajan (L.) Millsp. Methods The chromatographic conditions were as follows: column C18(250 mm? 4.6 mm, 5 ? m) , mobile phase being MeOH ∶ 1 % HAC (25 ∶ 75) for 20 mins and then being MeOH ∶ 1 % HAC (30 ∶ 70) after 20 mins, flow rate at 1.0 mL/min, and wavelength at 339 nm. Results The good linearity of this method was in the range of 0.104 4~ 0.522 ? g (r=0.999 4), and the average recovery of vitexin was 100.11 % ( RSD =0.63 % ).Conclusion The method is simple and sensitive with good stability, it can be suitable for the quality control of vitexin in Cajanus cajan (L.) Millsp..

Objective To observe the effect of Changji’an Prescription (CP) on depressing the mast cells in the irritable bowel syndrome(IBS)rats. Methods The postnatal rats aged 8~21 days were given continous inflammation stimulation inside the colon to replicate the animal model of the IBS. The number of mast cells and their degranulation were observed by modified Toluidine Blue staining method under light microscope and electron microscope. Objective To observe the effect of Changji'an Prescription(CP) on T lymphocytes count and 5-hydroxytryptamine(5-HT) secretion in rats with diarrhea predominant irritable bowel syndrome(IBS).Methods Forty-six neonatal SD rats were randomized into normal group,model group,low-,middle-and high-dose CP groups(0.94,1.88 and 3.76g?kg-1?d-1 respectively),and Bupi Yifei Pills group(6.76g?kg-1?d-1).Continuous internal rectal stimulation with acetate was used to induce the rat model of intestinal hypersensitive IBS.Flow cytometer was used for the quantitative analysis of peripheral levels of T lymphocytes subsets CD+3,CD+4 and CD+8,and immunohistochemical method was used to observe 5-HT-positive cells number and the immunostaining intensity in the bowels of rats.Results In the model group,the defecation volume and the intestinal sensitivity were increased,CD+4 level and CD+4/CD+8 decreased,CD+8 level increased,5-HT-positive cells number and the immunostaining intensity elevated(P

6.The optimization scheme of Mongolian gerbils superovulation induced by PMSG and hCG

Objective To optimize the scheme of Mongolian gerbil superovulation.Methods Based on the analysis of the animal-week-old, the dose and time interval of hormone, we got the best animal-week-old, dose and time interval of hormone for Mongolian gerbil superovulation.Results The 6 week old female Mongolian gerbils which injected of 10 IU PMSG and followed by 10 IU hCG in 70 hours later could get the best superovulation.We collected eggs at 16 hour after mate with male gerbils.The ovum pick-up rate reached 80%, the number of oocytes were 32.6 ±3.0, the number of the fertilized egg developed to 2-cell are 24.8 ±5.4.Conclusions This study summarizes the optimization scheme of Mongolian gerbil superovulation induced by PMSG and hCG and it supported the foundation for Mongolian gerbils embryo biotechnology.

8.A new monacolin analogue from Xuezhikang capsule.

Xuezhikang capsule (ethanol extract of red yeast rice) which produced by Beijing WBL Peking University Biotech Co., Ltd., is a traditional Chinese medication with 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase inhibiting activity. Clinical trials indicated that Xuezhikang with lipid-lowering action could reduce the risk of cardiac events and total mortality of Chinese coronary heart disease patients. To exactly explain the clinical features of Xuezhikang, we undertook a complete study of the chemical constituents of Xuezhikang. This study resulted in the isolation of a new monacolin analogue, named alpha, beta-dehydromonacolin L (1), along with two known compounds: monacolin L (2) and 3-(2, 6-dimethyl-1, 2, 4a, 5, 6, 7, 8, 8a-octahydronaphthalen-1-yl)propanoic acid (3). The chemical structures were determined by extensive 1D and 2D NMR and MS spectroscopic analysis.
10.Study of the effects of integrin-linked kinase on proliferation and differentiation of fibroblast in hypertrophic scar.

OBJECTIVETo study the role of integrin-linked kinase (ILK) on the proliferation and differentiation of human fibroblast in hypertrophic scar and its effect on the scar formation.
METHODSThe human scar fibroblasts were isolated and cultured in vitro. The cells were divided into 4 groups. (1) control group: only contains DMEM; (2) jetPRIME group: DMEM with 200 microl jetPRIME buffer and 4 microl jetPRIME; (3) ILK siRNA group: DMEM and ILK siRNA; (4) ILK cDNA group: DMEM and ILK cDNA. The cell proliferation was detected by XTT assay and the mRNA and protein expressions of ILK and alpha-SMA were detected by Real-time qPCR and Western blot.
RESULTS(1) XTT results showed that the cellular proliferation level after 48 h in four groups were 0.820 +/- 0.065, 0.873 +/- 0.041, 0.554 +/- 0.013 and 1.296 +/- 0.094, respectively. The cellular proliferation curve showed that the cellular proliferation level was very flat in ILK siRNA group while the cellular proliferation level gradually increased from 12 h. 48 h after transfection, the cellular proliferation level in ILK siRNA group was significant lower than those in other groups (P value were 0.021, 0.034, 0), while the cellular proliferation level in ILK cDNA group was the highest among all 4 groups (P value were 0.017, 0.009, 0). (2) The Real-time qPCR showed that the expressions of ILK mRNA and alpha-SMA mRNA were 0.693 +/- 0.412 and 0.422 +/- 0.037 in control group, were 0.621 +/- 0.183 and 0.388 +/- 0.005 in jetPRIME group, were 0.052 +/- 0.019 and 0.073 +/- 0.023 in ILK siRNA group, were 240.193 +/- 35.170 and 138.056 +/- 24.060 in ILK cDNA group. The expressions of ILK mRNA and alpha-SMA mRNA in ILK siRNA group were significantly lower than those in other three groups (P < 0.05). And the expressions of ILK mRNA and alpha-SMA mRNA in ILK cDNA group were significantly higher than those in other three groups (P < 0.05). (3) The Western blot also showed that the expression of ILK and alpha-SMA proteins were decreased in ILK siRNA group and increased in ILK cDNA group.
CONCLUSIONILK may promote the proliferation and differentiation of human scar fibroblast. It may play an important role in scar formation and contracture.
