1.Investigation of flurbiprofen axetil on analgesic efficacy in nerve-injured rats

Objective To investigate the influence of different doses of flurbiprofen axetil on neurological behavior in CCI rats.Methods All SD rats were randomly assigned to 3 groups:control group(C group),operation but without CCI,n=5;CCI group,without injection,n=5;flurbiprofen axetil (FA) group,different doses of flurbiprofen axetil were intraperitoneally administered every 12 hours for 7 days beginning on postoperative 6 hours in CCI rats (FA1 group:2mg/kg,FA2 group:4mg/kg,FA3 group:8mg/kg,FA4 group:16mg/kg),5 rats in each subgroup.The thermal withdrawal latency(TWL) and mechanical withdrawal threshold(MWT) were tested on preoperative 1,3,5 and 7 days and postoperative 1,3,5 and 7 days.Results Firstly,compared with C group[TWL:(12.396±1.144)s;MWT:(12.801±0.494)g],the TWL and MWT of CCI and FA groups[TWL:(5.645±1.144)s,(5.901±0.914)s,(6.300±0.835)s,(7.631±0.597)s,(7.894±0.526)s;MWT:(5.14±0.71)g,(5.28±0.60)g,(6.74±0.50)g,(6.74±0.25)g,(6.70±0.42)g]were significantly declined after surgery (tTWL=26.973,31.047,31.176,30.373,30.772;tMWT=44.192,48.132,51.136,54.326,46.937,all P<0.05),and the TWL and MWT of FA groups significantly improved with the doses from 2mg/kg to 16mg/kg of flurbiprofen axetil[TWL:the minimum (5.030±0.240)s,the maximum (8.300±0.430)s;MWT:the minimum (4.56±0.14)g,the maximum (7.11±0.20)g].Secondly,there were no statistically significant differences in TWL and MWT between CCI group and FA1 group(all P>0.05).However,the TWL and MWT of FA2 group,FA3 group and FA4 group[TWL:(6.300±0.835)s,(7.631±0.597)s,(7.894±0.526)s;MWT:(6.74±0.50)g,(6.74±0.25)g,(6.70±0.42)g] increased significantly compared with CCI group(tTWL=-3.768,-11.569,-13.867,tMWT=-3.898,-15.233,-14.801,all P<0.05).Thirdly,there were no statistically significant differences in TWL and MWT between FA3 group and FA4 group (all P>0.05).Conclusion Over 4mg/kg of flurbiprofen axetil can significantly reduce the thermal hyperalgesia and mechanical allodynia in CCI rats,however,there is a ceiling effect of flurbiprofen axetil,and the dosage of 8mg/kg can provide a maximum analgesic effect.
2.The effect of lornoxicam on the expression of growth associated protein-43 and nerve growth factor in the dorsal root ganglion of rats during the development of neuropathic pain

Objective To investigate the effect of lornoxicam on the.neurological behavior change and the expressions of growth associated protein-43 (GAP-43) and nerve growth factor (NGF) in dorsal root ganglion (DRG) on the peripheral nerve chronic constriction injury (CCI) model of rats.Methods Fifty Wistar rats were randomly divided into four groups:normal control group (n =5),CCI model group (CCI group),normal saline control group (NS group),and lomoxicam therapy group (L group) ; CCI,NS,and L groups were subdivided into 3 groups according to the different postoperative interval:3,7 and 14 days (n =5 each subgroup),respectively.The right sciatic nerve of rat was to be chronic constriction injure.Group L was given the rat 1.3 mg/kg of lomoxicam every 12 hours; then,the mechanical withdrawal threshold (MWT) and thermal withdrawal latency (TWL) of the rats of CCI,NS and L groups were measured at the different postoperative interval (3,7 and 14 days).After that,the DRG to the injured sciatic nerves were harvested.Immunohistochemistry was used to examine the expressions of GAP-43 and NGF.Results Comparing with the normal group,the latencies of MWT and TWL at CCI,NS and L groups were significantly declined at the third,seventh and fourteenth day after surgery (P < 0.05),and the expressions of GAP-43 and NGF in DRG of CCI,NS and L groups were significantly increased after surgery (P < 0.05).Comparing with the CCI group,the MWT and TWL of group L were significantly increased at the same time subgroup (P < 0.05).The expressions of GAP-43 and NGF in DRG of group L were significantly declined at the same time subgroup (P < 0.05).Conclusions Lornoxicam could relieve the symptoms of heat and mechanical hyperalgesia after sciatic nerve chronic constriction injury.It proved that lornoxicam was effective in the therapy of neuropathic pain.

AIM:To investigate the inhibitory effect of dihydro-?-erythroidine on nicotine induced-current in cultured superior cervical ganglion of neonatal rats.METHODS:Pneumatic pressure administration of drug and whole-cell recording techniques were performed to compare induced-current amplitude.RESULTS:Dihydro-?-erythroidine competitively antagonized nicotinic effect,and EC50 was about 0.015mmol/L.CONCLUSION:Dihydro-?-erythroidine is a competitive antagonist of nicotinic receptors in sympathetic neurons.
4.The effect of necrostatin-1 on expression of liver monocyte chemotactic protein-1 in septic rats

Objective To investigate the effect of necrostatin-1 (Nec-1) on the expression of liver monocyte chemotactic protein-1 (MCP-1) in septic rats and its mechanism. Methods Forty-eight male Sprague-Dawley (SD) rats were randomly divided into sham group, model group, and Nec-1 group by randomized digital number method, with 16 rats in each group. The model of sepsis was reproduced by cecal ligation and puncture (CLP). Rats in sham group received anesthesia, and flipping the cecum followed by closure of the abdomen without ligation of the cecum. Rats in Nec-1 group were given 1 mg/kg Nec-1 [25 mg Nec-1 solution dissolved in 2.5 mL of dimethyl sulfoxide (DMSO)] through caudal vein 30 minutes before operation, while the rats in model group were given 0.1 mL/kg of DMSO only. Blood from abdominal aorta and liver tissue in each group were collected at 0 hour and 8 hours after operation. Serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were determined with automatic biochemistry analyzer. The pathological changes in liver were observed under light microscope using hematoxylin-eosin (HE) staining. The serum levels of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) were determined by enzyme linked immunosorbent assay (ELISA). The MCP-1 mRNA expression in the liver was determined by reverse transcription-polymerase chain reaction (RT-PCR). Results There was no significant differences in the levels of serum ALT, AST, TNF-α, IL-6 and expressions of liver MCP-1 mRNA at 0 hour among three groups, and the liver cellular structure was normal. At 8 hours, compared with sham group, the expressions of serum ALT, AST, TNF-α, IL-6 and liver MCP-1 mRNA were significantly increased in model group and Nec-1 group [ALT (U/L): 172.35±21.88, 129.67±18.20 vs. 60.04±11.74, AST (U/L): 511.03±34.92, 363.51±25.25 vs. 254.83±31.04, TNF-α(ng/L): 603.96±24.18, 483.87±26.60 vs. 265.74±15.14, IL-6 (ng/L): 975.62±65.37, 712.09±45.47 vs. 310.42±13.88, MCP-1 mRNA (2-ΔΔCt): 7.09±0.18, 5.51±0.45 vs. 0.99±0.06, all P < 0.05]. Levels of the above parameters in Nec-1 group at 8 hours were significantly decreased compared with those of model group (all P < 0.05). Under light microscopy, it was noted that the structure of hepatic lobules was destroyed, with exacerbation of immunocyte infiltration at 8 hours in model group. At 8 hours, it was found that Nec-1 alleviated the pathological damage in Nec-1 group. Conclusion Nec-1 can protect the liver of rats with sepsis, lower the expression of serum TNF-α and serum IL-6 and liver MCP-1 mRNA, and obviously reduce the damage of inflammation.
5.Effect of IKVAV peptide nanofiber on proliferation, adhesion and differentiation into neurocytes of bone marrow stromal cells.

This study examined the effect of IKVAV peptide nanofiber on proliferation, adhesion and differentiation into neurocytes of bone marrow stromal cells (BMSCs). IKVAV Peptide-amphiphile was synthesized and purified. Then, hydrogen chloride was added to the diluted aqueous solutions of PA to induce spontaneous formation of nanofiber in vitro. The resultant samples was observed under transmission electron microscope. BMSCs were cultured with IKVAV peptide nanofiber. The effect of IKVAV nanofiber on the proliferation, adhesion and induction differentiation of BMSCs was observed by inverted microscopy, calcein-AM/PI staining, cell counting and immunofluorescence staining. The results demonstrated that IKVAV peptide-amphiphile could self-assemble to form nanofiber gel. BMSCs cultured in combination with IKVAV peptide nanofiber gel grew well and the percentage of live cells was over 90%. IKVAV peptide nanofiber gel exerted no influence on the proliferation of BMSCs and could promote the adhesion of BMSCs and raise the ratio of neurons when BMSCs were induced to differentiate into neurocytes. It is concluded that BMSCs could proliferate and adhere well and yield more neurons during when induced to differente into neurocytes on IKVAV peptide nanofiber gel.
6.Regulation of Recipe Xiaoban No.1 on expression of P53 in dystrophy of vulva

Objective:To explore the regulation of Recipe Xiaoban No.1 on the expression of p53 in dystrophy of vulva.Methods:Expression of p53 was determined by immunohistochemical technique in skin lesion of prior treatment and posttreatment from 30 cases with chronic vulvar dystrophy,and normal skin from 23 health adults were served as controls.Results: Expression of p53 in various of chronic dystrophy of vulva before treatment was obviously higher than control group(P0.05).Conclusion:Recipe Xiaoban No.1 can obviously improve the clinical symptoms and decrease the expression rate of P53.
7.Epidemiological characteristics and associated factors of delayed voiding among middle school students

Objective:
To analyze the epidemiological characteristics and associated factors of delayed voiding among middle school students, and to provide the basis for health education.
Methods:
A total of 1 260 middle school students were selected from Sanmen country in Taizhou by using multi-stage stratified sampling method. A cross-sectional survey was conducted to investigate the demographic characteristics, epidemiological characteristics and associated factors of delayed voiding by questionnaire.
Results:
The proportion of frequent delayed voiding among middle school students was 14.37%(181). The proportion among senior high school students was 15.51%(90), and among junior high school students was 13.67%(91),no significant differences were found by grade(χ2=0.57, P=0.45). The multivariate Logistic regression analysis showed that residential school, homework per day >2 hours, games and reduce the frequency of drinking ≥7 times/week were risk factors for delayed voiding frequently in junior high school students(OR=3.71, 1.70, 3.02,6.52,P<0.05). Higher than 2 hours of daily homework, a supportive or indifferent attitude to delayed voiding, staying in bed after waking up, drinking <0.5 L/d or ≥2.0 L/d, and reduce the frequency of drinking ≥7 times/week were risk factors for delayed voiding behavior in high school students(OR=2.23, 1.81, 2.22, 3.48, 2.69, 3.00, P<0.05).
Conclusion
The prevalence of frequent delayed voiding was high among middle school students. Attitude, environment and the quality of drinking water were the primary factors of frequent delayed voiding.
8.Management of main hepatic vein injury in hepatectomy for hepatic neoplasm of segment Ⅷ

Objective To summarize the experience in management of main hepatic vein injury due to hepatectomy for hepatic neoplasm of segment Ⅷ. Methods Clinical data of 64 patients suffering from main hepatic vein injury due to hepatectomy of hepatic neoplasm of segment Ⅷ in our hospital from October 1996 to October 2008 were retrospectively analyzed. Results Both the main trunks of the middle and right hepatic vein were injured in 34 patients, single right hepatic vein in 13 and middle hepatic vein in 17. In these patients with hepatic vein injury, the main trunk of the hepatic vein was repaired in 39, vessels ligated in 12 and direct liver wound surfaces sutured in 12. The hepatectomy and hemostasis were successfully performed during operation in all patients. After operation, 3 patients had active bleeding and 2 patients were reoperated on to sew up the bleeding points by wadding with the gelatin sponge and discharged after rehabilitation. One patient gave up treatment and was discharged automatically. Conclusion Main hepatic vein injury in hepatectomy of hepatic neoplasm of segment Ⅷ can be managed effectively by hepatic vein repair, hepatic vein ligation and suture of the liver section that can be chosen to control the bleeding of hepatic vein injury according to the actual conditions.

10.The free flap from the superficial palmar branch of radial artery innervated by the lateral cutaneous nerve of the forearm repair the defects of finger pulp

Objective To explore the outcomes of free flap from the radial artery superficial palmar branch (RASP) innervated by the lateral cutaneous nerve of the forearm for repairing the defects of finger pulp. Methods From April, 2013 to February, 2015, 20 fingers in 20 cases were treated with free flap from RASP innervated by the lateral cutaneous nerve of the forearm. The area of flap was from 1.8 cm×2.0 cm to 2.0 cm×4.0 cm. Postoperative fol-low up was done termly. Results All 20 flaps survived. All cases were followed-up for 7-20 months ( averag, 12 months). The flaps appeared well with good texture. The two-point discrimination was from 7 mm to 12 mm, and grad-ed with S3 and S3+sensation. There was little scar noted at the donor site in the wrist. There was no influence of hand and wrist function. Conclusion The method of using free flap from SASP innervated by the lateral cutaneous nerve of the forearm to repair the defects of finger pulp has good clinical effect.
