1.Effects of Dibutyl Phthalate on Rat Sperm Production and Quality

Objective:To evaluate the effects of dibutyl phthalate on rat sperm production and quality. Methods:Totally 150 male rats were randomly divided into the low dose group (50 mg·kg-1), the middle dose group (200 mg·kg-1), the high dose group (1 000 mg·kg-1 ) , the blank control group and the solvent control group ( peanut oil as the control) with 30 ones in each. After continu-ous administration for every 30 days, 10 rats from each group were anatomized, the weight of testes and epididymides were determined, and one side of epididymis was used to carry out the sperm analysis including counting, survival rate and morphology. Results:After intragastric administration for 90 days, the sperm count and survival rate, the weight of testis and epididymis and organ coefficient in the middle dose group and high dose group were decreased significantly(P<0. 05 or 0. 01). Conclusion:The long-term administration of dibutyl phthalate at high dose exhibits notable toxicity on rat reproductive function.
2.Next generation of antibody therapy for cancer.

Monoclonal antibodies (mAbs) have become a major class of therapeutic agents providing effective alternatives to treating various human diseases. To date, 15 mAbs have been approved by regulatory agencies in the world for clinical use in oncology indications. The selectivity and specificity, the unique pharmacokinetics, and the ability to engage and activate the host immune system differentiate these biologics from traditional small molecule anticancer drugs. mAb-based regimens have brought clinical benefits, including improvements in overall survival, to patients with a variety of cancers. Many challenges still remain, however, to fully realize the potential of these new medicines. With our further understanding of cancer biology, mechanism of antibody action, and advancement of antibody engineering technologies, many novel antibody formats or antibody-derived molecules are emerging as promising new generation therapeutics. Carefully designed and engineered, they retain the advantage of specificity and selectivity of original antibodies, but in the meantime acquire additional special features such as improved pharmacokinetics, increased selectivity, and enhanced anticancer efficacy. Promising clinical results are being generated with these newly improved antibody-based therapeutics.

3.Therapeutic effects of KH902 eye drops on corneal alkali burns by confocal microscopy

Objective Corneal alkali burn is a major cause of corneal neovascularization ( CNV) .Confocal microscopy was used to observe the therapeutic effects of KH902 eye drops for the treatment of corneal alkali burns. Methods 24 adult rabbits were randomly divided into two groups( n=12) , alkali burn model being established:experimental group by KH902 eye drops and control group by saline solution 3 times a day.Confocal microscopy was given at 3, 7, 14, 28 d post-operatively (po). Results Corneal epithelium deletion or vacuolar necrosis was found at 3 d po, along with stromal edema.There was no obvious inflammatory cell or no immunocyte infiltration.No difference was found in the corneal structures between two groups.CNV appeared in the peripheral stroma in both groups at 7d po.Inflammatory cell infiltration was more severe in the limbus and peripheral sections in control group than in control group.At 14 d po, inflammatory cells declined gradually and CNV took shape.At 28 d po, scarring and decreased inflammatory cells were found in both groups, while the experimental group had fe-wer and smaller blood vessels than the control group.As to the area of CNV, it was respectively (35.42 ±6.40) mm2, (60.23 ±5.35) mm2, (60.23 ±5.35)mm2 at 7, 14, 28d po in control group, statis-tical difference being found among different time points(P<0.05).While in experiment group, the area of CNV was respectively (22.11 ±5.45)mm2, (31.62 ±7.19)mm2, (28.54 ±3.01) mm2at 7, 14, 28 d po without statistical difference.(P>0.05).The CNV area in experiment group is significantly less than that in the control group at 14 d and 30 d po(P<0.05).The inflammatory cell density in control group was respectively (74.21 ±9.33)mm2, (1883.39 ±43.11)mm2, (2532.10 ±98.00)mm2, (723.05 ± 23.34)mm2at 3,7, 14, 28 d po, and (58.0 ±10.22)mm2, (656.90 ±33.01)mm2, (432.32 ±60.11)mm2, (122.11 ±30.37) mm2respectively at 3,7, 14, 28d po in experiment group, significant difference was found at different time points in both groups(P<0.05), among which significant decreased inflammatory cells was found in experiment group compared with control group (P<0.05). Conclusion KH902 eye drops can be used for the inhibition of CNV and inflammatory cell infiltration after alkali burn.

5.Best Operation Mode of AP-960 Full-automated ELISA System

Objective To compare the operation modes of AP-960 full automated ELISA system.Methods Two models were tested: beginning with adding specimens(model 1) and beginning with adding enzymes(model 2).The following factors were compared: the consumption of suckers,usage of time,result of test and the usual alarm during the test.Results The consumptions of suckers were 564 and 84 respectively.The needing time of the former mode was 125 minutes and that of the latter mode was 90 minutes.Conclusion The mode beginning with adding enzymes(model 2) is better for the current status of our clinical laboratory.

Purpose:To Explore the requirements and accuracy for applying Microwave-ELISA Two-Steps-Test to the factory physical examination.Methods:Do the Microwave-ELISA Two-Steps-Test for 1,128 factory physical examination specimens that need the HBsAg and HBeAg inspection;comparing the result to the conventional ELISA test;and retest the specimens with the HBsAg-positive results.Results:There are 119 HBsAg-positive and 32 HBeAg-positive results for the ELISA test;there are 122 HBsAg-positive and 34 HBeAg-positive results for the microwave test;all the HBsAg-positive results are proved to be positive by using the colloid gold HbsAg rapid testing paper.Conclusion:Microwave-ELISA Two-Steps-Test is timesaving,simple and high sensitive for testing Hepatitis-B signs,which can reduce the hook phenomenon and can be used to do the large number of HBsAg and HBeAg checks in factory physical examinations.It can hasten the result report and greatly improve the efficiency.

AIM To study the flavonoids from the leaves of Astragalus membranaceus (Fish.) Bge..METHODS The ethyl acetate and n-butyl alcohol fractions of 75% ethanol extract from A.membranaceus leaves were isolated and purified by silica,ODS and preparative HPLC column,then the structures of obtained compounds were identified by spectral data.RESULTS Thirteen compounds were isolated and identified as quercetin (1),quercetin-3-O-β-D-glucopyranoside (2),rhamnocitrin-3-O-β-D-glucopyranoside (3),rhamnocitrin-3-O-β-neohesperidoside (4),rhamnocitrin-3-O-3-D-glucopyranoside (1'''→2'')-β-D-apiofuranosyl (5),complanatuside (6),glycitein (7),4',7-dihydroxy-3-methoxy isoflavone (8),genistein (9),calycosin-7-O-β-D-glucopyranoside (10),genistin (11),glycitin (12),tiliroside (13).CONCLUSION Compounds 5,8,13 are isolated from genus Astragalus for the first time,and compound 2 is first isolated from this plant.
9.Effects of early enteral nutrition combined with probiotics on intestinal flora and immune function in patients with severe ischemic stroke

Objective To explore the effects of early enteral nutrition (EN) combined with probiotics on intestinal flora and immune function in patients with severe ischemic stroke. Methods Sixty-nine severe ischemic stroke patients were admitted and continuously enrolled in Taizhou First People's Hospital from June 2017 to June 2018, and they were randomly divided into an EN combined with probiotics group (35 cases) and a simple EN group (34 cases). Early EN support was given to both groups and probiotics (Live Combined Bifidobacterium, Lactobacillus and Enterococcus capsules) was added to the EN combined with probiotics group, 0.42 g each time, 3 times a day for 14 days. The changes of serum inflammatory markers [hypersensitive C-reactive protein (hs-CRP), procalcitonin (PCT), tumor necrosis factor-α (TNF-α), interleukin-10 (IL-10)], intestinal flora (Bifidobacterium, Lactobacillus, Clostridium, Enterobacter, Enterococcus, Bacteroides), intestinal mucosal barrier [endotoxin, D-lactic acid, diamine oxidase (DAO), intestinal fatty acid binding protein (I-FABP) ], and immune indexes [immunoglobulins (IgA, IgG, IgM), human leukocyte DR antigen (HLA-DR)] were observed in two groups of patients after treatment. Results With the prolongation of time, Bifidobacteria, Lactobacilli, HLA-DR and IgA, IgG, IgM after EN in both groups all decreased first and then had a tendency of increase, all reaching the lowest value on the EN 3rd day and then gradually elevated arriving at the peak value on the EN 14th day, and the levels in EN combined with probiotics group were significantly higher than those in the simple EN group [Bifidobacterium (×107 cfu/g): 8.31±1.49 vs. 7.49±1.32, Lactobacillus (×107 cfu/g): 8.04±1.45 vs. 7.19 ±1.37, HLA-DR: (67.22±9.11)% vs. (61.21±9.69)%, IgA (mg/L): 170.34±40.13 vs. 149.54±38.76, IgG (g/L):4.88±0.88 vs. 4.31±0.86, IgM (mg/L): 879.47±100.82 vs. 821.52±97.75, all P < 0.05]. With the prolongation of time, the Clostridium, Enterobacter, Enterococcus, Bacteroides, hs-CRP, PCT, TNF-α, endotoxin, D-lactic acid, DAO, I-FABP after En in both groups all increased first and then had a tendency of decrease, reaching the highest level on the EN 3rd day, then gradually decreased arriving at the valley value on the EN 14th day, and the levels in the EN combined with probiotics group were significantly lower than those in the simple EN group [Clostridium (×107 cfu/g): 5.23±0.87 vs. 5.79±0.91, Enterobacter (×107 cfu/g): 7.45±1.21 vs. 8.62±1.32, Enterococcus (×107 cfu/g): 7.32±1.05 vs. 8.12±1.23, Bacteroides (×107 cfu/g): 9.16±1.35 vs. 9.87±1.42, hs-CRP (mg/L): 18.45±12.98 vs. 25.47±15.55, PCT (ng/L): 3.24±1.21 vs. 4.18±1.32, TNF-α (ng/L): 9.43±8.69 vs. 13.59±9.45, IL-10 (μg/L): 39.45±10.72 vs. 48.52±11.42, endotoxin (U/L): 6.74±2.12 vs. 9.21±3.28, D-lactic acid (mg/L): 98.74±20.74 vs. 114.78±19.89, DAO (mg/L): 21.45±8.49 vs. 29.47±9.41, I-FABP (ng/L): 1.4±0.2 vs. 1.5±0.2, all P < 0.05]. Conclusion Early EN combined with probiotics can effectively regulate the intestinal flora and intestinal mucosal barrier function, reduce the level of inflammatory response and enhance the body immunity in patients with severe ischemic stroke.
