1.THE EFFECT OF KERATIN 13 GENE ON LARYNGEAL CARCINOGENESIS

Objective To investigate the role of Keratin 13 gene in laryngeal carcinogenesis. Methods To detect the deletion frequency of keratin 13 sequence indirectly by LOH analysis at DNA level using 5 STR primers within and near keratin 13 gene in 100 cases. Results LOH was found in all of the microsatellite loci, and the LOH frequencies were 30.48%, 26.02%, 21.62%, 37.66% and 21.51% at D17S1964E, D17S2092, D17S791, D17S1665, and D17S808 positions respectively. The frequencies of LOH were not related to the type of laryngeal carcinoma. Conclusion Keratin 13 gene might play an important role in the laryngeal carcinogenesis,and further research is necessary to confirm it.
3.Effect of dexmedetomidine and midazolam on hemodynamics and sedation in patients with nasal intubation

Objective To study the effect of dexmedetomidine and midazolam on hemodynamics and sedation in patients with nasal intubation.Methods Forty patients whose ASA grade Ⅰ-Ⅱ and anticipated difficult airway were randomly divided into dexmedetomidine group(group D,20 cases)and midazolam group(group M,20 cases)according to the admission number.In group D,dexmedetomidine 1 μ g/kg were constant speed pumped in 10 minutes.In group M,midazolam 0.03 mg/kg were intravenous injected.Then nasal intubation were carried.Systolic blood pressure(SBP),diastolic blood pressure(DBP),mean arterial blood pressure(MAP),heart rate(HR),pulse oxygen saturation(SpO2),Ramsay sedation score,rate-pressure product(RPP),tip perfusion index(TPI)were recorded and compared before anesthesia (T0),fiberoptic bronchoscope pass by later nostril(T1),to spy on epiglottis(T2),intubation succeed(T3),after intubation 1 minute(T4)and after intubation 3 minutes(T5).Airway score and postoperative visit were evaluated.Results SBP,DBP,MAP,HR,RPP in group M were significantly higher at T1-T3 than those at T0 (P<0.05),and were significantly higher than those in group D at the same time(P< 0.05).There was no significant difference in group D(P > 0.05).Ramsay sedation score and TPI at T1-T3 in group M were significantly lower than those at To(P <0.05).Ramsay sedation score and TPI at T1-T5 in group D were significantly higher than those at T0(P < 0.05),and were significantly higher than those in group M at the same time(P < 0.05).The rate of airway score 1 score and intubation satisfaction in group D were significantly higher than those in group M[100%(20/20)vs.30%(6/20),90%(18/20)vs.50%(10/20)](P< 0.05).The rate of throat ache in group D was significantly lower than that in group M[5%(1/20)vs.35%(7/20)](P <0.05).Conclusions For difficult airway patients with nasal intubation during dexmedetomidine infusion,hemodynamics is stable and sedation is satisfied.
4.The synergistic effect and its mechanism of PLLA and PCCL on antisenility

Aim To study the synergistic effect and its mechanism of polysaccharide from Ligustrum Lucidum Ait(PLLA)and polysaccharide from Cuscuta Chinensis Lam (PCCL)on antisenility. Methods To observe the effect on immune organs and antioxidation systems of mice with senile model caused by D-galatose (D-gal) and meanwhile administered with PLLA∶PCCL 50 ∶50 mg or 50 mg ∶200 mg or 200 mg ∶50 mg or 200 mg ∶200 mg?kg -1?d -1 designed by orthogonal method. Results Thymus index and spleen index differently rose,MDA differently dropped,activities of SOD and GSH-PX differently ascended in liver and kidneys,LF differently descended in brain of mouse with senile model administered with 50 mg∶50 mg or 50 mg ∶200 mg or 200 mg ∶50 mg or 200 mg ∶200 mg?kg -1?d -1 PLLA ∶PCCL.The statistical analysis indicated that the combination of PLLA with PCCL yielded of notable or extremely notable reciprocal effect on the experimental index of antisenility. Conclusion The combination of PLLA with PCCL can yield notable synergistic effect on antisenility,the optimal combination of both drugs is PLLA ∶PCCL 50 mg ∶200 mg, its mechanism is probably connected with immunity inhacement,eliminati on of oxyen free radicals and active oxyen and antilipoperoxidation.
5.Effect of alcoholic extract from Fructus Polygoni Orientalis on lipid peroxidation in rat tissues

Objective To study the activities of the alcoholic extract from Fructus Polygoni Orientalis (EFPO) against antioxidation. Methods The peroxidation in homogenate of rat heart, liver and kidney was induced by ?OH generation system Fe2++ascorbic acid. Malondialdehyde (MDA) was determined by TBA colorimetric method. Superoxide (O-2) from zymosan-stimulated neutrophils of rat was measured by NBT reduction method. H2O2-caused hemolysis of erythrocytes was measured by spectrometry. 6.Standardized application of clinical laboratory module of mobile medical unit during field operations

Objective To solve the problems of clinical laboratory module in mobile medical unit such as miscellaneous and mixed equipment,disordered packages,and weak mobility and responsiveness.Methods The concept of modularization was used for the secondary optimization of clinical laboratory equipment and consumables.The specialized vehicle for clinical laboratory was designed with considerations on the advantages of clinical laboratory shelter in equipment and medical vehicle in mobility,so as the values of clinical laboratory module were enhanced in mobile medical unit.Results Modular and standardized management of clinical laboratory equipment was realized to solve the existing problems of equipment.Conclusion The equipment supportability and mobility are enhanced in mobile medical unit,so that the requirements can be fulfilled for medical support missions at peace time and wartime.
7.Research progress on immunomodulatory effects and mechanisms of bacterial flagellin

Flagellum is a slender and wavy protein-attached filament on the surface of certain bacterial cells. It not only plays an important role in the movement and pathogenic ability of bacteria, but also participates in a variety of host immune regulation. Flagellin is a structural protein that forms the main part of flagellar filaments and can be recognized by TLR5 and other receptors in the host cell to induce the body′s immune response. At present, flagellin is widely used in the research of new immune adjuvants due to its immune activation, and its inflammation inhibitory effect also has good prospects against immune pathological damage. In this review, we summarized and analyzed the recent progress on the basic structure and function of flagellin, the host recognition mechanism, and its role in regulating the host immune system.
8.Screening, identification and culture optimization of a newly isolated aromatic nitrilase-producing bacterium--Pseudomonas putida CGMCC3830.

Microbial nitrilases have attracted increasing attention in nitrile hydrolysis for carboxylic acid production in recent years. A bacterium with nitrilase activity was isolated and identified as Pseudomonas putida CGMCC3830 based on its morphology, physiological and biochemical characteristics, as well as 16S rRNA gene sequence. The nitrilase production was optimized by varying culture conditions using the one-factor-at-a-time method and response surface methodology. Glycerol 13.54 g/L, tryptone 11.59 g/L, yeast extract 5.21 g/L, KH2PO4 1 g/L, NaCl 1 g/L, urea 1 g/L, initial pH 6.0 and culture temperature 30 degrees C were proved to be the optimal culture conditions. It resulted in the maximal nitrilase production of 36.12 U/mL from 2.02 U/mL. Investigations on substrate specificity demonstrate P. putida nitrilase preferentially hydrolyze aromatic nitriles. When applied in nicotinic acid synthesis, 2 mg/mL P. putida cells completely hydrolyzed 20.8 g/L 3-cyanopyridine into nicotinic acid in 90 min. The results indicated P. putida CGMCC3830 displayed potential for industrial production of nicotinic acid.

9.Spontaneous hypertension in cynomolgus monkeys and an analysis of its risk factors

Objective To establish reference values for blood pressure in cynomolgus monkeys in different ages.Methods The blood pressures and blood lipids indexes were detected in 521 cynomolgus monkeys using an American BECKMAN-CX4 automatic biochemical analyzer and a wrist electronic blood pressure monitor.Statistical tests were performed to analyze the data.Results Significant differences were found in blood pressure values of cynomolgus monkeys in different ages.Blood pressure values in the elderly group were higher than those of other groups.The morbidity of hypertension in the elderly group was higher than those of the other groups.Body mass index (BMI) in the hypertension group was higher than that of normal group in the same age.The incidence of hypertension in the elderly group with hyperlipemia was higher than that of other groups.Logistic regression analysis showed that age, BMI and hyperlipidemia in the hypertensive group were 1.435, 1.218, and 2.337 times higher than those of the normal group when predicting the risk of hypertension.Conclusions We have initially established reference values of blood pressure in cynomolgus monkeys in different ages.Age, BMI and hyperlipidemia are risk factors of spontaneous hypertension in cynomolgus monkeys, and the measurement of blood pressure may provide a basis for the screening of cynomolgus monkey model of spontaneous hypertensive and related research.
10.Effect comparison of laser peripheral iridoplasty at different sites of iris in pigment rabbit glaucoma

Background Laser peripheral iridoplasty (LPI) is widely used in the treatment of glaucoma by flattening the iris and widening angle of anterior chamber (AA).However,no evidence suggests the optimal site of LPI in iris.Objective This study was to compare the therapeutic effects of LPI at different sites of iris for glaucoma.Methods Glaucoma models were established in the right eyes of 40 healthy adult male pigment rabbits by intrachamber injection of 0.1 ml compound carbomer solution with 0.3% carbomer and 0.025% dexamethasone.The models were randomly divided into model control group,corneoscleral limbus group,one spot from corneoscleral limbus group and two spots from corneoscleral limbus group.LPI was performed at corresponding site of iris by 532 nm argon laser with the spot diameter 500 μm,energy 300 mW,exposure time 0.3 seconds and laser number 24 spots,and the rabbits in the model control group did not receive LPI.Intraocular pressure (IOP),coefficient of outflow facility (C value) were measured and calculated with Schi(o)tz tonometer before LPI and 2,4,7,14 and 30 days after LPI,and anterior chamber depth (ACD),AA,anterior chamber angle opening distance within 500 μm radius from scleral spur (AOD500) were measured with ultrasound biomicroscope (UBM).The eyeballs were extracted 30 days after LPI,and the chamber angle were observed under the optical microscope after hematoxylin and eosin staining.The use and care of the animals complied with the Guide for the Care and Use of Laboratory Animals of the National Institutes of Health.Results UBM showed that compared with the model control group,the anterior chamber angle was evidently widened in all the LPI groups,with the best effectiveness in the one spot from corneoscleral limbus group and the worst one in the two spots from corneoscleral limbus group.Compared with the model control group,the IOP was evidently reduced,and C values,AA and AOD500 were significantly increased in the corneoscleral limbus group,one spot from corneoscleral limbus group and two spots from corneoscleral limbus group after LPI,showing significant differences among the four groups (IOP:Fgroup =16.848,P < 0.01;C value:Fgroup =9.629,P < 0.01;AA:Fgroup =62.336,P<0.01;AOD500:Fgroup =77.779,P < 0.01).IOP was reduced and C value,AA and AOD500 were increased in 2,4,7,14 and 30 days after LPI as compared with before LPI,with significant differences over time (IOP:Ftime =3.041,P =0.011;C value:Ftime =4.311,P<0.01;AA:Ftime =14.627,P<0.01;AOD500:Ftime =20.378,P<0.01).Compared with the model control group,the ACD was significantly increased in the corneoscleral limbus group and one spot from corneoscleral limbus group,and that in the two spots from corneoscleral limbus group was significantly reduced,and the ACD was insignificantly increased over time after LPI (Fgroup =18.017,P<0.01;Ftime =0.022,P =1.000).Hematoxylin and eosin staining showed that the trabecular meshwork and adhesion of tissure were reopened and the anterior chamber angle was widened after LPI.Conclusions LPI can widen anterior chamber angle and lower the IOP.The best therapeutic outcome for glaucoma is displayed when LPI is performed at the iris site corresponding to one spot from the corneoscleral limbus.
