Objective To explore the induced effects of colchicine in different concentration and times on Foeniculum vulgare polyploidy. Methods Seed germination rate and mutation rate, morphology of radicle induced mutants, chromosome number, tissue structure, and content and component of essential oil, which induced by colchicine in different concentiation and times were investigaled using the method of soaking seeds. Results The results showed that 0.13% of colchicine concentration and 24 h inducing time had the best induction on F.vulgare polyploidy.Compared to the control, the radicle of mutant was thicker, the chromosome number increased clearly, and the cell number in radicle manifolded distinctly.Among the four main components of essential oil,the contents of Dill apiol in anamorphosis radicle was a little lower than that of the control, but the contents of limonene,(E)-anethole,and camphene in anamorphosis radicle were all remarkably higher than that of the control after induced by colchicine.ConclusionThe content of main essential oil components in fennel mutant induced by colchicine is remarkably enhanced. The study provides the theoretical basis for the breeding of F.vulgare new varieties of high oil content.

Objective To characterize the viability and transgene expression of articular chon-drocytes cultured in 3-Dimensional scaffolds provided by four types of carriers. Methods Articular chondrocytes from rabbit knees were cultured and infected with adenovirus that could express green fluo-rescence protein (AdGFP) and GL3 luciferase (AdGL3-Luc).The viability and gene expression were determined with fluorescence microscopy and luciferase assays in four types of scaffolds;type I collagen sponge, fibrin glue, hyaluronan and open-cell polylactic acid (OPLA).Cartilage matrix production was assessed by Alcian blue staining. Results Articular chondrocytes of rabbits were effectively infected by AdGFP and exhibited sustained GFP expression. All the tested scaffolds supported the survival and gene expression of the infected chondrocytes. However, the highest transgene expression was observed in the OPLA carrier (P<0.01).Alcian blue-positive matrix materials were readily detected in OPLA cultures four weeks later. Conclusion OPLA supports the highest transgene expression and is the most conduc-tive scaffold for matrix production, suggesting that OPLA may be a suitable scaffold for cell-based gene therapy of articular cartilage repair.

Variation in pH value of oral cavity circumstance causes decayed tooth. In this paper, corrosions of Ti-Fe-Mo-Mn-Nb-Zr dental alloys in various pH lactic acids were studied by immersion test. In the case of pH=4, which is an acidity slightly larger than that causes decayed tooth, Ti-Fe-Mo-Mn-Nb-Zr alloys are entirely corrosion-resistant, and the corrosion type is pitting. With the increase in acidity, the corrosion-resistance of Ti-Fe-Mo-Mn-Nb-Zr alloys deteriorates, and the corrosion type changes from pitting to intergranular corrosion. Fe ion is the most dissolved substance in impregnation, which means Fe goes against the corrosion-resistance of Ti alloys. Both results of thermodynamics calculation and X-ray photoelectron spectroscopy have shown that Mn2O3, Nb2O5 and TiO2 form oxide film on the surface of Ti-Fe-Mo-Mn-Nb-Zr alloys.

Objective To establish a monkey model of muscle atrophy by fixing the right lower limb and set up e-valuation criteria for that model. Methods Twelve healthy,6-7 year old male cynomolgus monkeys(body weight 6-7 kg)were used in this study. All animals were normal and had no malformed lower limb bones. Right lower limbs of the an-imals were fixed with fiberglass bandage(from knee joint to anklebone)for 15 weeks. Body weight, crus perimeter and crus volume were recorded every two weeks. MRI examinations of the gastrocnemius muscle were conducted at weeks 11,13 and 15. Muscle biopsy was taken for pathological examination at week 15. Results There were no obvious changes of body weight during the experiment. At 15 weeks after modeling, the right crus perimeter and crus volume of the experimental an-imals were significantly decreased(P <0.05),and some biochemical indexes such as serum creatinine, gloucose and al-kaline phosphatase were significantly decreased(P <0.05), while cholesterol was significantly increased(P <0.05).MRI showed atrophy of the right gastrocnemius muscle. Muscle biopsy also showed muscle atrophy and muscle fibers be-came thinner in the right gastrocnemius, and the cross-section area of the muscle fibers was significantly decreased. Con?clusions It is easy to operate and establish a muscle atrophy model in Cynomolgus monkeys by fixing the right lower limb. This model is suitable for objective and easy evaluation of muscle atrophy by measurement of crus perimeter and crus volume of the limb, blood biochemical parameters,MRI examination, and histopathological examination in combination.

The influences of Ce on the microstructure and mechanics performances of Ti-Fe-Mo-Mn-Nb-Zr alloys were studied and presented in this paper. The microstructure of no-Ce Ti-Fe-Mo-Mn-Nb-Zr alloy was thick, long and dendritic, and the microstructure of alloy was fined by Ce. With the increase in Ce wt%, the microstructure became equiaxial gradually. Hardness decreased with the increase in Ce wt%. The compression yield strength became greater with the increasing of Ce wt%, but after the Ce wt% increased to a certian degree, the compression yield strength became smaller with the further increasing of Ce wt%. This phenomenon was explained in the light of bond energy and X-ray diffraction experiment. X-ray diffraction experiment showed that the alloys were beta-Ti.

Chinese Journal of Biotechnology 2011;27(12):1773-1779
To illustrate the complex fermentation process of submerged culture of Antrodia camphorata ATCC 200183, we observed the morphology change of this filamentous fungus. Then we used two optimization models namely response surface methodology (RSM) and artificial neural network (ANN) to model the fermentation process of Antrodia camphorata. By genetic algorithm (GA), we optimized the inoculum size and medium components for Antrodia camphorata production. The results show that fitness and prediction accuracy of ANN model was higher when compared to those of RSM model. Using GA, we optimized the input space of ANN model, and obtained maximum biomass of 6.2 g/L at the GA-optimized concentrations of spore (1.76x 10(5) /mL) and medium components (glucose, 29.1 g/L; peptone, 9.3 g/L; and soybean flour, 2.8 g/L). The biomass obtained using the ANN-GA designed medium was (6.1+/-0.2) g/L which was in good agreement with the predicted value. The same optimization process may be used to improve the production of mycelia and bioactive metabolites from potent medicinal fungi by changing the fermentation parameters.

Chinese Journal of Medical Genetics 2002;19(2):120-123
OBJECTIVETo investigate the role of transglutaminase 3 (TGM3) gene in laryngeal carcinogenesis.
METHODSThe authors detected the deletion indirectly through loss of heterozygosity (LOH) analysis at DNA level using 4 STR primers within and near TGM3 gene in 72 cases, and detected the differential expression of TGM3 gene in 8 cases of paired normal and cancerous tissue of laryngeal carcinoma by Northern blot.
RESULTSLOH was found existing in all of the microsatellite loci, and the LOH frequencies were 25.76%, 20.00%, 38.10% and 18.75% at D20S17, D20S607, D20S99 and D20S841 respectively; LOH concerning at least one polymorphism locus accounted for 61.11%. No correlation of clinical stage, lymph node metastasis and differentiation with the LOH of TGM3 gene was observed, P>0.05. TGM3 gene expressed significantly higher in normal tissues than in paired cancerous tissues.
CONCLUSIONTGM3 gene might play an important role in laryngeal carcinogenesis and further researches will be needed to clarify the possible mechanisms.

Objective To evaluate the therapeutic effects and safety of acitretin for severe inherited keratodermas in children and adolescents. Methods Acitretin was given to 23 children and adolescents with either lamellar ichthyosis, bulbous ichthyosiform erythroderma, pityriasis rubra pillars, progressive sym- metrical erythrokeratoderma, keratitis ichthyosis deafness syndrome, generalized porokeratosis, inflammatory liner verrucous epidermal nevus, ichthyosis hystrix and non-bullous ichthyosiform erythroderma. The thera- peutic dosage was 0.67-1.07 mg/(kg?d),and maintenance dosage 0.08-0.94 mg/(kg?d).The effects on the patients' growth and development of the drug were evaluated based on the changes of body weight and height in the children. The total follow-up period was 6-35 months in an interval of 1-3 months. Results The considerable overall improvement was achieved after 1-6 months' treatment, with an overall clinical cure rate of 82.6%. Only one case responded poorly to the therapy. The excellent responses were observed in patients with bulbous ichthyosiform erythroderma, lamellar ichthyosis, and pityriasis rubra pillars, etc, and the much poor responses in ichthyosis hystrix. The most frequent adverse reaction was mild to moderate dry lips (65.2%),the next were pruritus(39.1%),skin fragility(34.8%),and dry mouth(30.4%).The less frequent adverse reactions were alopecia(13%),anorexia(8.7%),headache (4.3%) and hypoacusis (4.3%).No effects on the growth and development were found in those children during the follow up period. Conclusions The considerable overall improvement is achieved with the acitretin therapy for children and adolescents with inherited keratodermas, with only mild to moderate adverse reactions and no effects on the growth and development in the children.
