Objective To evaluate the serum Krebs von den Lungen (KL)-6 for the diagnosis of interstitial lung disease (ILD) associated with connective tissue diseases (CTD) and its lung-CT subtypes. Methods Seventy-five CTD patients were employed for this study,44 CTD with ILD and 31 ILD without ILD.The 44 CTD patients with ILD were further divided into different subgroups based on lung-CT imaging and clinical indexes. The enzyme-linked immune sorbent assay (ELISA) was used to measure the serum KL-6 level. For those data that was abnormally distributed, the differences between groups was compared with independent samples nonparametric tests. Results The level of serum KL-6 in the CTD with ILD was significantly higher than that without ILD [(1 118±877) U/ml vs (253±144) U/ml] (Z=-6.047,P<0.01).By using a criterion of 500 U/ml,our data suggested that the serum KL-6 level was useful for the ILD-CTD diagnosis;the sensitivity, specificity, positive and negative predictive values were 72.7%,87.1%,88.9% and 69.2%,respectively.The serum KL-6 level, however, showed no statistical differences between ILD subtypes,i.e.,usual interstitial pneumonia (UIP),nonspecific interstitial pneumonia (NSIP) and indeterminate [(1 104±843) U/ml,(1 242±1 039) U/ml,(815±400) U/ml,respectively] (x2=0.35,P=0.84).Our data further showed that the KL-6 level was significantly higher in CTD-ILD patients with intensive lung lesions than those with limited lung lesions [(1 910±918) U/ml vs (459±268) U/ml] (Z=-4.364,P<0.01).In addition, the KL-6 level was significantly higher in active ILDs than in inactive ILDs[(1 478±917) U/ml vs (598±475) U/ml] (Z=-3.915,P<0.01).Conclusion The serum KL-6 is a valuable biomarker for CTD-ILD diagnosis and even for the assessment of the extent and activity of lung damage.

Objective To investigate clinical results of shoulder hemiarthroplasty in treating comminuted proximal humeral fractures. Methods Shoulder hemiarthroplasty was performed from May 1997 to June 2004 in 28 cases with comminuted proximal humeral fractures who were followed up for mean 2.1 years. American Shoulder & Elbow Surgeon (ASES) score was adopted for evaluating pain after operation, range of active movement and function of daily work. Results Mean ASES score was 87.9(67-96 scores), with mean forward flexion of 132?, external rotation of 40?, mean VAS score of 2.6 and internal rotation at T_ 10 level. Strength and function of 23 cases reached 80% of the normal side six months postoperatively and 90% of the normal side one year postoperatively. Of all cases, 82% were satisfied with the treatment. Conclusion Only reconstruction of normal humeral length, tuberosity fixation and persevering rehabilitation can contribute to a successful result of hemiarthroplasty for the comminuted proximal humeral fractures.

Objective To study the effect of TBTCl on the immunity of mice. Methods The mice were randomly divided into 5 groups, namely groups of negative control, low dose (0.8 mg/kg), moderate dose (4 mg/kg), high dose (20 mg/kg) and positive control. Gavage was employed to administrate TBTCl to mice for two consecutive weeks, the number of T-lymphocyte in blood and invert ability of T-lymphocytes in spleen, hemolysin in serum, clearance exponent and viscera quotiety were determined to study the effect of TBTCl on immunity of rats. Results Compared with the control, no significantly increased T-lymphocytes were seen in low dose group, in moderate dose group the number of T-lmphocyte and the production of hemolysin increased, no obvious changes in the clearance exponent and viscera quotiety were seen. In high dose group, a significantly decreased T-lymphocytes, the clearance exponent and viscera quotiety were observed. Conclusion Low level TBTCl exposure may stimulate the immune system of mice, high exposed to TBTCl may intensively inhibit the immune system.

The authors retrospectively analysed 67(63 in cervical cord)cases of traumatic SCIWOAR with positive MR abnormalities. It was deduced that the cases of SCIWOAR might be related with (1)those traumatic forces resulting in SCI being not so heavy and severe, (2)the cases orignally with cervical spondylopathy,(3)the limitation of the conventional X-ray examination for SCI,and (4)the cases of children relativly rare in our country and hospital. The ALL hematomas are the pathognomonic signs in the case of the acute cervical SCI within one week. The analysis demonstrated that MR investigation of the traumatic spine could give the clinical activities the correct diagnosis of different degree of SCI and dependable reference.

Objective To investigate the effects of inhibition of STAT5 gene expression by RNA interference technology on apoptosis of human hepatocellular carcinoma cell line SMMC-7721. Methods Three siRNA eukaryotic expression vectors against STAT5 were constructed and transfected with lipofectamineTM 2000 into SMMC-7721 cells. The changes in STAT5 expression were detected by semi-quantitative RT-PCR and Western blot. Cell apoptosis was assayed by flow cytometry (FCM). Results The sequence-specific siRNA could effectively and specifically inhibit STAT5 gene expression at both mRNA and protein levels. The inhibition rates of STAT5 mRNA expression were 70.43%, 43.02%, and 45.07%, respectively. The inhibition rates of STAT5 protein expression were 67.45%, 37.36%, and 41.86%, respectively. At 48 h after transfection, apoptosis rate was 25.61%. Conclusion siRNA against STAT5 can inhibit STAT5 gene expression in SMMC-7721 cells effectively and specifically and induce apoptosis of SMMC-7721 cells. siRNA targeting STAT5 has a great potential value in gene therapy of hepatocellular carcinoma.

Objective To observe the inhibitive effect of electro-acupuncture (EA)at Zusanli points (ST36)on inflammatory mediators of postoperative intra-abdominal adhesions and study the relationship between EA and cholinergic anti-inflammatory pathway. Methods Forty-eight male Wistar rats were divided into 6 groups (each =8):Group A (control),Group B(abdominal adhesions model),Group C (abdominal adhesions plus EA),Group D(sham acu-point control),Group E (abdominal adhesions plus α-bungarotoxin )and Group F (abdominal adhesions plus EA after α-bungarotoxin).Animal models of abdominal adhesion were produced by Chiang’s path.Bilateral Zusanli points (ST36) and shame acupoints were electro-acupunctured at a constant voltage for 1 hour while rats were awake. The ɑ-BGT(1 μg/kg)was injected into the abdominal cavity after surgery. All the rats were sacrificed on the 3rd day,and the levels of inflammatory mediators (TNF-ɑ,NO and NOS)in tissues were evaluated. Results Three days after surgery, the damaged cecum of abdominal adhesion groups developed obvious edema that did not adhere with other tissues.Compared with sham control, the abdominal adhesion resulted in significant elevation of inflammatory mediators (TNF-ɑ,NO and NOS).EA at Zusanli points obviously lowered the elevated levels of inflammatory mediators (P <0.01 and P <0.05).EA at Zusanli points following the injection of ɑ-BGT showed less anti-inflammatory effect(P <0.01).Conclusion EA at Zusanli points significantly lowers the elevated levels of inflammatory mediators after abdominal adhesion challenge. The activation of cholinergic anti-inflammatory pathway might be one of the mechanisms by which Zusanli points exert anti-inflammatory effects.
