1.Effects of Anticancer Chinese Medicines Screened Out by Data Mining on Proliferation and Migration of Triple-negative Breast Cancer Cells

Objective To screen out some Chinese medicines with obvious inhibitory actions on triple-negative breast cancer based on data mining. Methods Literatures of breast cancer effectively treated by Chinese medicine issued in recent 15 years were collected, and then the frequency analysis and cluster analysis were used to screen the frequently-used herbs. According to the above results, the serum containing the obtained drugs were prepared respectively. And then the proliferation, migration and invasion abilities of breast cancer cells MDB-MB-231 treated with the serum containing various Chinese medicines for 12, 24 h were observed by methyl thiazolyl tetrazolium(MTT) assay and transwell cell invasion and migration experiments, respectively. Results The frequently-used herbs were screened out as Rhizoma Chuanxiong, Herba Taraxaci, Herba Hedyotis Diffusae, Radix Trichosanthis, Radix Achyranthis Bidentatae. The proliferation of MDB-MB-231 cells was significantly inhibited by serum seperately containing Rhizoma Chuanxiong, Herba Taraxaci, Herba Hedyotis Diffusae, Radix Trichosanthis, and Radix Achyranthis Bidentatae to different degrees, and the migration abilities of MDB-MB-231 cells were obviously inhibited by serum containing Herba Taraxaci, Herba Hedyotis Diffusae, Radix Achyranthis Bidentatae, respectively. And the invasion abilities of MDB-MB-231 cells were obviously inhibited by serum containing Herba Taraxaci, Rhizoma Chuanxiong, Herba Hedyotis Diffusae, respectively. Conclusion Chinese medicines such as Rhizoma Chuanxiong, Herba Taraxaci, Herba Hedyotis Diffusae, Radix richosanthis, Radix Achyranthis Bidentatae obtained by data mining technology have obviously inhibitory effects on triple-negative breast cancer.
2.Evaluation on performance of Sysmex CS2000i Automatic Blood Coagulation Analysis System

Objective To systematically evaluate the performance of the Sysmex CS2000i Automatic Blood Coagulation Analysis System .Methods According to the standards of the Clinical and Laboratory Standards Institute (CLSI) ,the precision ,accuracy ,lin‐earity ,biological reference interval and carry‐over rates of the Sysmex CS2000i Automatic Blood Coagulation Analysis System were detected ,meanwhile the detections of 5 clinical specimens were compared between this system and the Sysmex CA 1500 Automatic Blood Coagulation Analysis System .Results The intra‐assay precision coefficient of variation (CV) ,inter‐day precisions CV carry‐over rate ,accuracy and linearity of plasma prothrombin time ,activated partial thromboplastin time ,fibrinogen and D‐dimmer detec‐ted by the Sysmex CS2000i Automatic Blood Coagulation Analysis System were all consistent with the quality target requirements of our laboratory .Conclusion The Sysmex CS2000i Automatic Blood Coagulation Analysis System has better performance in vari‐ous aspects ,the detection results could be used for the clinical diagnosis and treatment of related diseases .
3.Vascularized distal radius graft of 1, 2 intercompartmental supraretinacular artery for scaphoid nonunion

Objective To study the techniques and outcomes of using a retrograde pedicled vascularized radius bone flap based on the 1st,2nd intercompartmental supraretinacular artery (1,2ICSRA) for scaphoid nonunion through dorsal and palmar approaches.Methods Between March 2013 and April 2015,16 patients with scaphoid nonunion were treated by surgery through palmar and dorsal approaches.There were 14 males and 2 females with an average age of 29.1 years (range,19-51 years).The location of lesion was 6 cases on the left and 10 cases on the right.5 cases were treated with plaster fixation.11 cases weren't treated after wrists injured.The preoperative time was 6-26 months,with an average of 16.5 months.The operation was completed within one incision.The bone flaps based on 1,2ICSRA were taken through dorsal approach.The broken ends of the scaphoid fracture were cleaned,and the deformity was corrected with bone grafting and internal fixation through palmar approach.Time of fracture union and wrist pain resolution was evaluated.Wrist motion and grip strength were measured and compared.The result of the latest follow-up was used as the criterion for efficacy evaluation.The wrist pain was evaluated using visual analogue scale (VAS) and the postoperative wrist function based on the modified Mayo wrist score was recorded.Results Post-operative follow up ranged from 6-19 months,with an average of 11 months.Bone union was achieved in all the cases,so the healing rate was 100%.7 cases were healed in 11 weeks.5 cases were healed in 13 weeks.3 cases were healed in 15 weeks.1 case was healed in 17 weeks.The average healing time was 12.5 weeks.The humpback deformity of Scaphoid and DISI of 7 cases were corrected.The bending of wrist could reach 60.2°±3.2°,and the elongation could reach 51.3°± 3.5°.The radial deviation of wrist could reach 13.6°±1.42°,and ulnar deviation could reach 24.4°±1.8°.The range of grip strength was 29-64 kg,with an average of 45 kg.The 16 patients returned to normal work.The pain after wrist movement of 14 cases was completely disappeared,and the VAS was 0.Two cases felt slight pain when wrist was overworked,and the VAS were 0.9 and 1.2.The wrist joint function of patients recovered well.The modified Mayo score was 69-99,with an average of 90.75.The functional results were 12 excellent,2 good,and 2 fair,and the excellent and good rate was 87.5% (14/16).No complications such as infection,failure of screws,orthopaedic arthritis or scaphoid necrosis were found during the follow-up period.Conclusion The technique of 1,2 ICSRA pedicled bone graft and palmar bone grafting for nonunion of scaphoid can provide convenience for us to deal with the broken ends of the scaphoid fracture,correct the malformation and graft bone through dorsal and palmar approaches.This method can protect the blood supply of the scaphoid,and promote scaphoid union.
4.Analysis of anatomical structure and variation related disease of os-tiomeatal complex with three-dimensional reconstruction of MSCT

Objective To investigate the value of multislice spiral CT (MSCT) with three-dimensional reconstruction technique in the diagnosis of anatomical structure and variation related disease of ostiomeatal complex. Methods A to-tal of 200 cases of chronic sinusitis patients with normal or variant ostiomeatal complex were retrospectively analyzed with thin layer reconstruction CT images. Results Among these 200 cases, deviation of nasal septum was the majority, accounting for 62.0%. Excessive gasification of ethmoid bulla and middle turbinate gasification were 37.0% and 36.5%respectively . No significant difference was observed between them . The haller cells and uncinate deviation bend accounted for 33.0% and 22.0% separately. Curved middle turbinate accounted for 13.0%. The agger nasi was the least, accounting for 12.5%. Conclusion Three-dimensional reconstruction of MSCT can show the anatomical variations of sinus-nasal as well as the extent of chronic sinusitis clearly, which has important clinical significance on treatment.
5.The application of multiple end-to-side anastomosis in free anterolateral thigh perforator flap transplantation

Objective To explore the clinical application effect of multiple end-to-side anastomosis in free anterolateral thigh perforator flap transplantation.Methods From January,2013 to October,2017,29 cases were applied the technology of multiple end-to-side anastomosis to the same recipient vessel in anterolateral thigh perforator flap transplantation for wound repair.Fifteen cases treated with multiple end-to-side anastomosis on flap arteries,and 2-3 arteries were anastomosed,with the average of 2.13.Nine cases were treated with venous multiple end-to-side anastomosis,and 2-4 veins were anastomosed,with the average of 3.11.Five cases treated with arterial and venous multiple end-to-side anastomosis,and 4-6 vessels were anastomosed,with the average of 5.20.Distribution of wounds:10 cases with hands and wrists wounded,15 cases with forearms and elbow joints wounded,and 4 cases with lower legs and feet wounded.The soft tissue defect size was 5 cm×13 cm-11 cm×27 cm,and the flap area was 6 cm× 15 cm-12 cm×29 cm.Postoperative followed-up was performed every 3-6 months to review flap survival.Results All flaps of 29 cases survived.Venous congestion occurred in 2 flaps within 48 h after the operation,among which,1 was overcomed after released the dressing and sutures,and the other underwent surgical exploration.The venous end-to-side anastomotic stomas were unobstructed,and hematoma was formed.After the hematoma compression was removed,circulation was recovered and the flap survived.With followed-up for 6 months to 2.5 years,both donor site and recipient site of the flaps healed well and the injured distal limbs had no hemodynamic disorder.Conclusion The application of multiple end-to-side anastomosis to the same recipient vessel for free transplantation of anterolateral thigh perforator flaps is safe and reliable.
6.The role of endothelial biomarkers in predicting damp-heat syndrome in diabetic kidney disease

Objective:To explore the role of endothelial biomarkers in predicting damp-heat syndrome in diabetic kidney disease (DKD).Methods:A total of 183 patients with DKD were divided into 3 groups:the early DKD group,established DKD group,and advanced DKD group.All patients were classified according to traditional Chinese medicine (TCM) syndrome type,and clinical indexes were collected for statistical analysis.Results:A total of 183 DKD patients were included in this study.Fibroblast growth factor 23 (FGF23),chitinase-3-like protein 1 (CHI3L1),endocan,tumor necrosis factor receptor 1 (TNFR1),secretory leukocyte protease inhibitor (SLPI),and vascular endothelial growth factor A (VEGF-A) were increased in advanced DKD.FGF23,CHI3L1,endocan,SLPI,and TNFR1 showed a negative correlation with estimated glomerular filtration rate (eGFR),while they had a positive correlation with 24 h urine protein.After adjusting for age,gender,diabetes duration,body mass index (BMI),hemoglobin,glucose,uric acid,24 h urine protein,cholesterol,triglyceride,low-density lipoprotein,and hemoglobin A1c (HbA1c),the multiple regression analysis showed that FGF23,endocan,TNFR1,and SLPI significantly correlated with eGFR.Conclusions:FGF23,endocan,TNFR1,and SLPI are elevated in advanced DKD compared with early stage,and they may take part in the pathogenesis and progression of DKD.Our study provides useful bio-markers for predicting the appearance of damp-heat syndrome,including FGF23,endocan,TNFR1,and SLPI.
7.Correlation study between cerebral microbleeds and white matter lesions in patients with ischemic cerebrovascular disease

Objective To explore distribution characteristics and risk factors of cerebral microbleeds (CMBs),and the correlation between CMBs and white matter lesions (WML) in patients with ischemic cerebrovascular disease(ICVD).Methods 180 patients with ICVD in neurology department of Hebei General Hospital from February 2015 to January 2017 were recruited.Those patients were underwent brain magnetic resonance imaging (MRI),and magnetic susceptibility weighted imaging (SWI).Recorded the baseline data and risk factors of high blood pressure,diabetes,hyperlipidemia,and high homocysteine were recorded.Patients with CMBs were counted and graded to understand the characteristics of CMBs distribution.Logisitic regression analysis was used to analyze the influencing factors.ICVD patients were divided into CMBs group and non CMBs group.CMBs group was further divided into 4 groups according to the severity,which was divided into level 1-3.The correlation between CMBs influencing factors and classification was further studied.Then patients with ICVD were divided into WML group and non WML group.WML group scored each region with age-related white matter changes rating scale (ARWMCrs).The correlation between WML and CMBs classification was further studied.Results (1) The overall prevalence of CMBs in patients with ICVD was 61.7% (111/180).The most common location of CMBs in patients with ICVD was the cortical and subcortical regions (80/111,72.1%),followed by the basal ganglia and thalamus regions (61/111,55.0%),and the infratentorial regions(38/111,34.2%).The difference between them were significant (x2 =32.061,P=0.000).In cortical and subcortical regions of CMBs,temporal lobe was the most common (61.3%).(2) Age(B=0.046,Or=1.047,95%CI =1.017~ 1.077,P=0.002) and the high homocysteine (B =1.458,Or=4.299,95% CI =2.114 ~ 8.744,P<0.001) were the risk factors for CMBs.(3) Four classification of CMBs was positively correlated with and WML total score (r=0.393,P=0.393).Conclusion The temporal lobe was the most common region for CMBs in patients with ICVD.Age and high homocysteine were risk factors for CMBs.With the increase of WML total score,severity of CMBs was also increased.
8.Simultaneous Determination of Blood Concentrations of Five Antiretrovirals in Human Plasma by Ultra High Performance Liquid Chromatography-tandem Mass Spectormetry Method

Objective To develop an ultra-performance liquid chromatography-mass spectrometry(UPLC-MS/MS)method for the simultaneous quantification of dolutegravir,raltegravir,efavirenz,lamivudine and tenofovir in human plasma and to apply it to the therapeutic monitoring.Methods Dolutegravir-D5,raltegravir-D4,efavirenz-D5,lamivudine-13 C-15 N2 and tenofovir-D7 were used as internal standard,respectively.All samples were extracted using the protein precipitation method with acetonitrile and then diluted for analysis.Chromatographic separation was performed on Shim-pack XR-ODS Ⅲ(2.0 mmx50 mm,1.6 μm)column.Mobile phases A and B consisted of 0.1%formic acid in water and acetonitrile respectively.A programmed mobile phase gradient was used at a flow rate of 0.3 mL·min-1 and column temperature of 40 ℃.The tandem mass spectrometer was equipped with an electrospray ionization(ESI)source operating in multiple reaction monitoring(MRM)modes.After methodological validation,it can be used for therapeutic drug monitoring in HIV patients.Results There was good linearity in the validated concentration ranges of 62.5-3 000 ng·mL-1 for dolutegravir,10-500 ng·mL-1 for raltegravir,125-6 000 ng·mL-1for efavirenz,10-500 ng·mL-1 for lamivudine and 10-500 ng·mL-1 for tenofovir with the linear correlation coeffificients of determination(R2)of all higher than 0.998.The accuracy of both intra-day and inter-day studies ranged from 94.0%-109.3%,and the relative standard deviations were less than 7%.The IS-normalized matrix factor and extraction recoveries of all analytes were 95.7%-106.0%and 98.7%-104.5%at all concentrations.All analytes were stable in plasma at a certain storage environment.The trough blood concentrations of dolutegravir,efavirenz,lamivudine and tenofovir were 107.7-2 366.0,740.0-3 410.0,38.5-1 229.3,31.6-224.4ng·mL-1 in HIV patients,respectively.Conclusion The method is highly aceurate,easy to perform,low-cost,and suitable for therapeutic drug monitoring of dolutegravir,raltegravir,efavirenz,lamivudine and tenofovir in HIV patients.
9.Biological characteristics and whole-genome sequencing analysis of a strain of sheep-derived Streptococcus equi

In order to study the biological characteristics and whole-genome sequence of Streptococ-cus equi,a sheep-derived Streptococcus equi preserved in our laboratory was subjected to recovery,biochemical experiments,drug susceptibility tests and animal experiments,and followed by whole genome sequencing and annotation of the gene functions of the genome by using the biogenetic da-tabase.The biochemical identification results showed that this strain could ferment sugars,but the results of nitrate,catalase,V-P test,and M-R test were negative;the drug susceptibility test results showed that this strain was highly sensitive to most antibiotics and was resistant to kanaphylaxis.It was resistant to amikacin and amikacin;animal experiments showed that the lethality rate of this strain to mice was 100%,and the median lethal dose of this strain was measured to be 4.86X 106 CFU/kg.According to the pathological section results of mice,it showed that the lungs,liver,kidneys,and spleen all had varying degrees of lesions.The whole genome sequencing results showed that the total genome length of this strain is 2 272 497 bp,the G+C content was 41.1%,and it was predicted to contain 2 124 CDS regions.The RNA prediction results showed that the number of rRNA and tRNA was 15,and the number of tRNA was 57.There were four prophages and gene islands,and there were 362 pathogenicity-related genes in the VFDB database without CRISPR sequences.This study analyzed the complete genome of Streptococcus equi derived from sheep,and also provided a theoretical basis for the treatment,prevention and control of the disease.
10.Prediction of osteoporotic vertebral compression fracture based on comprehensive index of lumbar vertebral bone strength

BACKGROUND:Osteoporotic vertebral compression fracture is a common fracture secondary to osteoporosis.At present,there is no effective prediction index and method for osteoporotic vertebral compression fracture. OBJECTIVE:To investigate the predictive effect of the comprehensive index of lumbar vertebral body bone strength on osteoporotic vertebral compression fracture. METHODS:233 patients with osteoporosis were divided into a fracture group and a non-fracture group according to whether a vertebral fracture occurred.The demography,body mass index,vertebral bone mineral density and other details were collected.Lateral X-ray films of the lumbar spine were photographed.The vertebral body width,vertebral body length,sacral slope,pelvic tilt,pelvic incidence,lumbar compressive strength index and the lumbar impact strength index were measured,calculated,and analyzed by univariate and multivariate,and the receiver operating characteristic curve was analyzed.The survival analysis was conducted according to the cut-off value. RESULTS AND CONCLUSION:(1)All patients were followed up for 2-4 years,with an average of 3.1 years.During the follow-up period,99 cases(38 cases of L1 vertebral body,61 cases of L2 vertebral body)had fractures(fracture group),and 134 cases(52 cases of L1 vertebral body,82 cases of L2 vertebral body)had no fractures(non-fracture group).Univariate analysis showed that there was no significant difference in age,sex,height,body mass,body mass index and fracture segment between the two groups(P>0.05).(2)Lumbar compressive strength index and lumbar impact strength index in the fracture group were lower than those in the non-fracture group(P<0.05).Pelvic incidence and pelvic tilt in the fracture group were higher than those in the non-fracture group(P<0.05).(3)Multivariate analysis showed that lumbar compressive strength index,lumbar impact strength index and pelvic tilt were risk factors for osteoporotic vertebral compression fractures(P<0.05).(4)Receiver operating characteristic curve analysis showed that the cutoff values of vertebral bone mineral density,lumbar compressive strength index,lumbar impact strength index,pelvic tilt and pelvic incidence were 0.913 5 g/cm2,1.932,0.903,21.5° and 55°,respectively;areas under the curve were 0.630,0.800,0.911,0.633 and 0.568,respectively.(5)According to the survival analysis(with osteoporotic vertebral compression fracture as the end point),the average survival time of the patients with lumbar impact strength index≥0.903 was significantly longer than that of the patients with lumbar impact strength index<0.903(P<0.05).(6)These findings conclude that the comprehensive index of lumbar vertebral body bone strength is more accurate than the bone mineral density of the vertebral body and spine-pelvis sagittal parameters in predicting osteoporotic vertebral compression fractures,which is helpful for early prevention and treatment of osteoporotic vertebral compression fractures.
