1.In vitro Proliferation and Cytotoxicity of PBMC Induced by Autologous Tumor Specific Antigen Costimulated with IL-2

To observe its proliferation and cytolytic activity in vitro induced by autologous soluble tumor antigen and IL-2, PBMC costimulated with TSA extracted from gastric tumor by biochemical procedure and IL-2 was cultured. The proliferation and cytotoxicity were evaluated by MTT method and the phenotype was analysed by APAAP method. TSA alone did not induce PBMC proliferation while TSA (10?g/ml) with IL-2 presented syngenic stimulating effect which was time and concentration dependent. The cytolytic unit of PBMC induced with IL-2/TSA was 86.3% vs 48.6% with IL-2 alone. The phenotypic measurement demonstrated that TSA enhanced CD25 and CD8 expression significantly. The observation offered experimental data for further research work about tumor activated killer(TAK) cells. The results indicated that TAK cells would be a new type of immunoactive cells in adoptive immunotherapy against tumor.
2.The clinicopathological significance of transforming growth factor-β1, transforming growth factor-β1 receptor Ⅱ ,Smad4 and Smad7

Objective To investigate the clinicopathological significance of transforming growth factor-β1 (TGF-β1), transforming growth factor-β1 receptor Ⅱ (TGF-βRⅡ ), Smad4 and Smad7 protein expressions in gastric carcinomas. Methods 109 cases of gastric carcinomas,28 of high grade dysplasia,20 of low grade dysplasia,30 of chronic atrophic gastritis,29 of intestinal metaplasia, and 21 of normal gastric mucosa were selected for the detection of the expression level of TGF-β1,TGF-βRⅡ , Smad4 and Smad7 proteins by immunohistochemistry. Results The expression of TGF-β1,TGF-βRⅡ and Smad7 proteins in gastric carcinomas and dysplasia (high and low grade) were significantly higher than those in chronic atrophic gastritis, intestinal metaplasia and normal gastric mucosa (all P <0. 001 ) ,whereas the expression of Smad4 was significantly increased in high and low grade dysplasia (cytoplas-mic IS :4.89±2.38,5.80±1.54, respectively; nuclear IS: 3.89± 1.52,3.80±1.33, respectively), and decreased in gastric carcinomas (cytoplasmic IS:2.41±2.27,nuclear IS:2.02±2. 14) (P <0.001 ,P <0.001 ,respective-ly). Meanwhile,the expressions of TGF-β1,TGF-βRⅡ and Smad7 were higher in advanced gastric cancer (4.36± 2.66,3.05±1.93,4.84±3.06, respectively) than those in early gastric cancer ( 2.93±1.85,2. 17±1.87, 4. 14±2.46, respectively) ( P < 0.01, P < 0.05, P < 0. 05, respectively ) ; however the expression of Smad4 protein was much higher in early gastric cancer (P < 0. 001 ). The expression of Smad4 protein was significantly correlated with tumor size( P < 0.05 ), lymph node metastasis ( P < 0. 001 ), T stage ( P < 0. 001 ) and chnical stage ( P < 0.001) ,and the expressions of TGF-βRⅡ and Smad4 proteins were correlated with patients' 5-year survival rate (P<0.05 ,P < 0.01 ). TGF-β1 showed the positive correlation with TGF-βRⅡ and Smad7 (r1= 0.45 ,P <0. 05; r2 = 0.49, P <0. 05), but negative correlation with Smad4 ( r = - 0.21,P < 0.05 ). Conclusion TGF-β1, TGF-βR Ⅱ, Smad4 and Smad7 protein may play an important role in the carcinogenesis of gastric carcinoma,and smad4 is the key in TGF-β pathway. Also, the detection of these proteins expression can be a useful marker for predicting the gas-tile cancer invasion,metastasis and patients' prognosis.
3.Dynamic hip screw combined with reduction fixation of lesser trochanter prevents hip coxa vara after intertrochanteric fractures:a meta-analysis

BACKGROUND:More and more elderly patients have hip fractures because of diverse reasons, and most of the fractures are unstable fractures combined with the displacement of the lesser trochanter. At present, there are stil many reports about using dynamic hip screw to repair unstable intertrochanteric fractures, and the effect is obvious. However, there are stil great controversies regarding whether lesser trochanter fixation is performed during the repair process. OBJECTIVE:To evaluate the effect of using dynamic hip screw combined with the lesser trochanter fixation to repair intertrochanteric fractures and the occurrence of hip coxa vara after repair through a meta-analysis. METHODS:The PubMed, EMCC, CBM, CNKI, VIP and Wanfang databases were retrieved by computer for randomized controled trials on whether lesser trochanter reduction fixation was performed during the process of dynamic hip screw internal fixation for repair of intertrochanteric fractures. The quality of the included studies was evaluated according to the pre-designated inclusion and exclusion criteria. The available data were extracted and analyzed using the RevMan5.2 software. RESULTS AND CONCLUSION:A total of 10 randomized controled trials involving 604 patients were included. Among them, 298 cases were assigned to the dynamic hip screw combined with lesser trochanter fixation group, and 306 cases to the lesser trochanter unfixed group. The meta-analysis results indicated that hip coxa vara, other postoperative complications, the excelent and good rate of efficacy in the dynamic hip screw combined with lesser trochanter fixation group were al superior to those in the lesser trochanter unfixed group (alP < 0.000 1), and for hip coxa vara, the parameters were odds ratio=0.17, 95% confidence interval (0.07, 0.41),Z=3.99,P< 0.000 1. The operation time and the amount of blood loss in the lesser trochanter unfixed group were superior to those in the dynamic hip screw combined with lesser trochanter fixation group (alP < 0.000 1). These results suggest that the lesser trochanter unfixed therapy has the advantages of relatively simple operation, shorter operative time and less blood loss during the process of dynamic hip screw internal fixation for repair of unstable intertrochanteric fractures. However, the excelent and good rates of efficacy, hip coxa vara and other postoperative complications were superior in patients with intertrochanteric fracture above the Tronzo-Evans III type who received dynamic hip screw combined with lesser trochanter fixation to those who received lesser trochanter unfixed therapy. The results indicate that dynamic hip screw combined with lesser trochanter fixation for repair of intertrochanteric fracture is beneficial to prevent hip coxa vara and other complications.
4.AN IMMUNOCYTOCHEMICAL STUDY OF THE DISTRIBUTION OF T AND B LYMPHOCYTES AND THEIR SUBSETS IN HUMAN SPLEEN

A series of monoclonal antibodies have been used to study the distribution of T and B lymphocytes and their subsets in human spleen (5 from normal human and 2 from patients with portal hypertension). The results indicate that the T cells are mostly located in the periarterial lymphatic sheath, and in which a few B lymphocytes can be seen. The B cells are concentrated in lymphoid follicles, but also contain some T lymphocytes such as Leu 1, Leu 3a and Leu 4 positive T cells, these cells are necessary for forming of germinal center. Whereas the marginal zone, is composed of a mixture of T and B cells as well as the T_(ac) positive cells. The red pulp is composed of a mixture of T and B ceils, but the T and B cells are distributed randomly. In this report, the LN-2 monoclonal antibody is used first to study the B lymphocytes in human spleen. So far it is a unique antibody to react with nuclear membrane of B lymphocytes, the activity of LN-2 antigen do not influenced by B-5 fixation and paraffin embedding. From our data, there is no difference in staining feature and charateristic distribution between the normal human spleen and spleen of portal hypertensive patients. Although the periarterial lymphatic sheath in cases with portal hypertension seems to be narrower than the normal.
5.Investigation of Drinking Water Quality in Some Rural Areas in Fujian

Objective To know the drinking water quality in the rural areas in Fujian Province in order to provide the scientific basis for water improvement in the rural areas.Methods Fourteen counties were selected in Fujian,2004,10 monitoring sites in each county based on the population proportion of different drinking water types.Sixteen indicators were determined for every water sample.Results In the 14 investigated counties,38.71% of total population drinking water supplied with surface water,and 61.29% with ground water source.Drinking water from centralized water supply system was supplied to 58.24% of total population.The qualified rate of water samples was 26.39% for total 288 water samples,35.38% for surface water samples and 18.99% for ground water samples respectively.The hygienic state of surface water was much better(P
6.THE CORDYCEPS SINESIS INFLUENCE ON THE IgG AND IgM ANTIBODY-FORMING CELLS IN MOUSE SPLEEN

A sensitive immunocytochemical technique has been used to study the effect of Cordyceps Sinesis on the IgM and IgG antibody-forming cells in mouse spleen. The results indicate that after the mice have been treated for 7 days with Cordyceps Sinesis, their splenic nodules were increasing, the germinal centers became prominent and the marginal zones were thickening. The immunocytochemical results disclosed that a huge amount of IgM and IgG lymphocytes appeared in the splenic nodules, especially in the marginal zones, which were thickening due to the proliferation of the IgM-lymphocytes. In the red pulp, a large amount of IgM- and IgG-plasma cells were peripherally located in the terminal arteries or sinuses, indicating that it may be convenient for the plasma cells to release the antibodies into blood circulation. The results suggest that the Cordyceps Sinesis could stimulate the IgM-and IgG-lymphocytts to proliferate and promote the forming of antibodies.
7.Balloon dilation treatment for tuberculous airway stenosis

Objective:By analyzing clinical data, discuss the clinical efficacy of balloon dilatation for the treatment of tuberculous scarred airway stenosis and factors affecting efficacy. Methods:Selected 13 cases airway stenosis caused by tracheobronchial tuberculosis.Airway stenosis was confirmed and measured by bronchoscopy and CT before balloon dilation. Balloon dilatation with forming expansion was conducted once a week. We evaluated the shortness of breath scores, measuring FEV1, FVC and the diameter of airway before and after dilation. The long-term outcome and lung function improvement were evaluated. Results:Thirteen cases were treated by balloon dilation with fiberoptic bronchoscopy. The airway diameter before and after the treatment, shortness of breath score, FEV1, FVC have improved significantly, the difference was significant(t=15.596, t=-27.657, t=-14.604, t=-41.766;P<0.05). Conclusion:Balloon dilation treatment for tuberculous scarred airway stenosis can achieve better results.
8.Transcatheter hepatic arterial chemoembolization in treatment of GIST with metastatic liver cancer

Objective To observe the clinical curative effect of transcatheter hepatic arterial chemoembolization (TACE) in treatment of gastrointestinal stromal tumors(GIST) with metastatic liver cancer. Methods Four GIST patients with 19 lesions of liver metastasis were treated with 2 ~ 4 times of TACE. The diameters of the metastatic tumor were from 2 ~ 5 cm. FuDR, CDDP, EPI-ADM, together with lipiodol ultrafluid and gelatin sponge to from a mirture was prepared for arterial embolization. Results DSA showed abundant tumor vasculature and tumor stain on hepatoarterial angiography. The size of tumor showed no decrease after TACE and lipiodol was cleared away in a short period. No patient achieved CR or PR until two of them received imatinib. Conclusions TACE is not effective in treating hepatic metastasis from GIST shown in limited cases. 9.Clinicopathological and prognostic significance of NQO1 protein expression in patients with colorectal carcinoma

Purpose To investigate the prognostic significance of NQO1 protein expression in colorectal carcinoma ( CRC) patients. Methods 192 cases of primary CRC, 28 of colonic dysplasia, and 44 of adjacent non-tumor tissues were selected for immunohisto-chemical staining of NQO1 protein. Correlation between NQO1 overexpression and clinicopathologic characteristics, and the survival rates were calculated by the statistical methods. Results The strongly positive rate of NQO1 protein in CRC was significantly higher than that in gastric dysplasia and adjacent non-tumor tissues (P<0. 01, respectively). NQO1 high-expression rate was positively cor-related with differentiation, serosal invasion, lymph node metastasis, and clinical stage (P <0. 05, respectively). Survival curve showed that the disease-free survival and 5-year survival rates of the patients with high NQO1 expression were obviously shorter than those of patients with low NQO1 expression (P<0. 001, respectively). Further analysis showed that, high expression of NQO1 predic-ted the lower disease-free survival and 5-year survival rates in late-stage patients (P<0. 01, respectively). Importantly, NQO1 was an independent risk factor for the prognosis of CRC using Cox proportional hazards regression model ( HR: 1. 398,95%CI: 1. 011 ~1. 934, P=0. 043). Conclusions Detection of NQO1 protein expression in CRC has an important clinical significance, and it is ex-pected to become a new biomarker for CRC.
10.Effects of chronic intrauterine hypoxia on intimal-media thickening and strain rate of abdominal aorta of male offspring rabbits during adult age

Objective To investigate the the effects of chronic intrauterine hypoxia on intimal-media thickening(IMT) and strain rate(SR) in abdominal aorta of male offspring rabbits during adult age.Methods Sixten New-Zealand rabbits were assigned randomly to 2 groups: chronic intrauterine hypoxia group (CIH, 12% O_2, n = 8) and normal oxygen group (NO,21%O_2, n = 8).After delivery,2 male offspring rabbits per litter were selected and breast-fed for 3 months, randomly divided into high-fat diet and normal diet groups.Finally, there were 4 groups in this experiment:chronic intrauterine hypoxia with high fat diet (CIH + HFD, n = 8) ,non-chronic intrauterine hypoxia with high fat diet (NCIH + HFD, n = 8),chronic intrauterine hypoxia with normal diet (CIH + ND, n = 8) and normal control (NC, n = 8).At sixth months of age, Serum levels of total cholesterol(TC) and triglyceride(TG) were assayed.SR in the abdominal aorta of male offspring rabbits was evaluated by ultrasonography.Then, abdominal aorta was taken out and observed by electron microscope and IMT was measured.Results CIH increased the levels of TC and TG (P < 0.01), thickened the IMT (P<0.05) and decreased the SR(P<0.05) of abdominal aorta of male offspring rabbits during adult age.There were relevant pathological changes in different groups.All these above-mentioned profiles were aggravated significantly after feeding high fat diet (P<0.05 or P<0.01).Conclusions CIH increased the IMT and decreased the SR of abdominal aorta of male offspring rabbits during adult age.
