Objective To precious localize DNase Ⅰ hypersensive sites exactly in the promoter region of CD133 of cell line SW480 by inverse-PCR.Methods The colonel cancer cell SW480 nuclei were suspended in digested buffer,treated with DNase Ⅰ at the concentration of 10 U/ml for 10 min.The inversePCR was performed as follows.DNA treated by DNase Ⅰ was purified,fragmented with restricted enzyme EcoRI and Xmal Ⅰ.Then the ends were blunted,ligated by T4 ligase.PCR was performed,and production was sequenced.The restricted enzymes cut sites were near DNase Ⅰ cleavage sites.Results 9 DNase Ⅰ cut sites were identified in CD133 promoter region.The DNaseI hypersensitive sites all distributed in a region -300 bp--700 bp up to transcription start site.Conclusion The DNase Ⅰ cleavage sites could identified preciously by application of inverse-PCR.These sites locate in a region of-300 bp--700 bp up to transcription start site.
2.3-dimentional CT reconstruction in the diagnosis of upper urinary tract obstructive diseases (report of 37 cases)

Objective To study the diagnostic value of 3 dimentional CT reconstruction in upper urinary tract obstructive diseases. Methods 37 patients with upper urinary tract obstructive diseases underwent thin slice helical CT scan and 3 dimentional reconstruction of the urinary tract as well as 3 dimentional CT angiography when needed. Results All the cases have been definitely diagnosed including ureteral carcinoma in 8, bladder carcinoma in 2, ureteral calculi in 9, benign ureteral stricture in 7, congenital malformation in 7 and pelvic tumor in 4.All the patients except those with pelvic tumors underwent surgical intervention and the diagnosis were confirmed.The operative findings were in concord with the helical CT results. Conclusions 3 dimentional CT reconstruction of the urinary tract can provide distinct urinary tract images and helps determining the site and the cause of the obstruction. Compared with other methods, the procedure has the advantages of high resolution, rapid imaging and low invasiveness. It might become the most valuable means of detecting upper urinary tract diseases.
3.Let-7 and miR-24 in UVB-induced apoptosis

Objective To study the function of let-7 and miR-24 in UVB-induced apoptosis.Methods After NIH3T3 cells were irradiated with UVB.Hoechest33342/PI staining was used to study the cell apoptosis and RT-PCR was used to uralyzc the expression level of let-7 and miR-24.In addition,potential target genes of these miRNAs in PicTar were classified into different function categories through GOstat software.Results Compared to the control,the NIH3T3 cells exposure to UVB appeared typical apoptotic and necrotic ceils by fluorescence microscope.The exprossion level of let-7 and miR-24 in NIH3T3 cells after UVB irradiation was higher than that of the control.Among the target genes,casp3,bc1212,map3kl and cdk5 were also involved in UVB-induced apoptosis mechanism.Conclusion Let-7 and miR-24 play a role in UVB-induced apoptosis.
5.Analysis of the infectious complications following nonmyeloablative allogeneic peripheral blood stem cell transplantation

Objective To investigate the infectious complications after nonmyeloablative allogeneic peripheral blood stem cell transplantation (NASCT). Methods Ten patients with leukemia received NASCT from HLA-identical or 1-3 antigen mismatched sibling donors with conditioning regimens of Flu/Cy+BU+ALG. Five patients were acute leukemia (AL) and all of them is in complete remission (CR1). Cyclosporine in combination with methotrexate was administered for GVHD prophylaxis. Results WBC recovered to more than 0.5 ?10 9/L during postoperative 10 day to 38 day and platelet recovered to more than 20?10 9/L during postoperative 8 day to 16 day. Bacterial infections occurred in 3 patients (30 %) and cytomegalovirus (CMV) infections in 2 patients (20 %). Varicella zoster virus (VZV) infections occurred in one patient (10 %). No fungal infections were documented. No patients died as a result of infection . Conclusion NASCT is a safe and effective therapeutic method for leukemia. It reduces acute GVHD,regimen-related toxicity and early neutropenia associated with traditional allo-HSCT.

Objective To investigate the effects of paired associative stimulation (PAS) on the recovery of upper limb motor function in stroke patients,and to analyze the relationship between the change of motor cortex excitability in the contralesional hemisphere and the recovery of motor function in the affected upper limb.Methods Thirty hemiplegic stroke patients were divided randomly into a treatment group and a control group.Both groups were given routine rehabilitation therapy,but the treatment group also received PAS consisting of transcranial magnetic stimulation (TMS) of the intact motor cortex and electrical stimulation (ES) of the median nerve at the wrist of the intact arm with an interval of 10 ms between the TMS and ES (PAS10).The PAS10 was delivered at a frequency of 0.05 Hz and an intensity of 120% the resting motor threshold (RMT),once daily for 30 minutes,five times a week for 4 weeks.Corticospinal excitability was measured using motor evoked potentials (MEP) and the RMT.The FuglMeyer upper limb assessment (FMA),Brunnstrom staging and the modified Barthel index (MBI) were also applied before and at the end of the 4 weeks of treatment.Correlation was sought between any changes in MEP amplitude,the RMT of the contralesional hemisphere and changes in the FMA results.Results Before the intervention there were no significant differences between the two groups in terms of any of the assessments.After 4 weeks of treatment,all the assessments had changed significantly compared to those before the treatment,but there were still no significant differences between the two groups in terms of any the assessments.After 4 weeks of treatment,the differences in MEP amplitude from the contra-lesional hemisphere and the differences in FMA scores were positively and significantly correlated with a correlation coefficient of r =0.431.The lesioned hemisphere was also positively correlated with the differences in FMA scores with a significant correlation coefficient of r =0.608.Conclusion PAS10 can facilitate the recovery of upper limb motor function.The change in motor cortex excitability of the contra-lesional hemisphere significantly correlates with functional recovery in the upper limb.
8.Effects of paired associative stimulation and repetitive transcranial magnetic stimulation on the excitability of the motor cortex

Objective To compare the effects of paired associative stimulation (PAS) and repetitive transcranial magnetic stimulation (rTMS) on motor cortex excitability.Methods The baseline corticospinal excitability of the left hemispheres of 10 healthy subjects was measured in terms of resting motor threshold (RMT) and other indicators of motor evoked potentials (MEP).On the following day they received PAS composed of trascranial magnetic stimulation (TMS) to the motor cortex of the left hemisphere and electric stimulation (ES) of the median nerve contralateral to the motor cortex,with an interval of 10ms between the TMS and ES (termed PAS10).The PAS10 was delivered at a frequency of 0.05 Hz and an intensity of 120% of the RMT,for a total of 90 pulses.The MEP amplitude,MEP latency and RMT were evaluated one minute after the stimulation.After the PAS intervention,an interval of one week was allowed to eliminate any effect of PAS on motor cortex excitability.Then rTMS was delivered to the subjects' left motor cortex at the same time of day at a frequency of 1 Hz and an intensity of 120% of the RMT,for a total of 1000 pulses.MEP amplitude,MEP latency and RMT were evaluated one minute after the stimulation.The two interventions were compared in terms of MEP amplitude,MEP latency and RMT.Results The average MEP amplitude,MEP latency and RMT at baseline were (2.93 ± 0.99) mV,(20.97 ± 1.67) ms,and (46.06 ±5.32) %,respectively.One minute after PAS10,the MEP amplitude,MEP latency and RMT were (1.14 ± 0.76) mV,(21.87 ± 1.09) ms and (52.06 ±4.20) %,respectively.One minute after rTMS,the MEP amplitude and latency and the RMT were (2.24 ± 0.79) mV,(20.88 ± 1.94) ms,and (49.00 ± 4.54) %,respectively.The differences in MEP amplitude,MEP latency and RMT pre-and post-intervention were (0.69 ± 0.10) mV,(0.09 ±0.05) ms and (3.94 ± 0.93) %,respectively for rTMS.For PAS10 they were (1.83 ± 0.14) mV,(0.90 ± 0.26)ms and (6.00 ± 1.13)%,respectively.The differences in MEP amplitude decrease and MEP latency lengthening between the two stimulation protocols were significant,but the difference in RMT elevation was not.Conclusion Both PAS10 and low frequency rTMS suppressed motor cortex excitability,but the suppressive effect of PAS10 is more significant.
9.Infection situations of fungi and trichomonas and changes of cell counts in patients with non-bacterial vaginosis

Objective To explore the infection situation of fungi and trichomonas and the changes of pus cell and epithelial cell counts in patients with non-bacterial vaginosis in Lanzhou .Methods The pus and epithelial cell counts of vaginal secretion samples from patients with vaginitis were detected .Fungi and trichomoniasis were checked under high power microscope ,and the samples were defined positive if fungal spores or pseudohyphae were found .Results In the 4 404 cases of patients with vaginitis ,the total rate of fungi and trichomoniasis infection was 28 .63% (1 261/4 404) .The infection rate for fungi was 25 .39% ,and for trichomonad was 3 .25% ,respectively .The dual infection rate of fungi and trichomonad was 0 .54% .Compared with ≤20 age group ,the counts of pus cells and epithelial cells in other age groups were significantly different(P<0 .05) .Conclusion Fungi were the primary in-fection etiology of non-bacterial vaginosis in Lanzhou .
10.Atorvastatin alleviates pulmonary fibrosis-induced by bleomycin in rats

Objective To investigate the therapeutic effects of atorvastatin on pulmonary fibrosis of rats induced by Bleomycin (BLM). Methods Fourty-two female Wistar rats were randomly divided into 7 groups: the control group(group C) , the model group which was furtherly divided into group 2-week (M2) , 4-week (M4) , 6-week (M6) , and atorvastatin-treatment group which was furtherly divided into group 2-week (A2) , 4-week (A4) , 6-week(A6). Group M and A were induced to pulmonary fibrosis by the method of BLM endotracheal injection, while group C was injected with saline. On 2nd day, group A were given orally atorvastatin by 10 mg/kg · d. Rats were seperately killed on 2nd, 4th and 6th week. After intratracheal injection of BLM, alveolitis and pulmonary fi-brosis were evaluated by pathology, hydroxyproline concentration and PaO_2. Results The lung coefficient of group M2, M4, A2 and A4 was significantly higher than that of group C. The degree of pulmonary fibrosis in group A4 and A6 was improved as compared with group M4, M6. Alveolitis and pulmonary fibrosis in group A were improved compared with those in group M. Hydroxyproline concentration in group A and M were significantly higher than that in group C. While A4 was lower than M4 (P <0. 01). There was a decrease in PaO_2 after exposure to BLM, espe-cially in group M2 (P <0. 01). Improvement in PaO_2 was documented in groups A, compared with M. Conclusion Atorvastatin has centtain efficacy in treatment of pulmonary fibrosis of rats. The effect was significant on 6th week after treatment, which suggest that the effect is correlated positively to the carly treatment.
