China Pharmacist 2015;(5):794-796
Objective:To discuss the influence and curative effect of bifidobacterium tetravaccine tablets on serum substance P ( SP) and neuropeptide Y ( NPY) levels in the patients with irritable bowel syndrome ( IBS) . Methods: Totally 84 cases of patients with IBS were selected and divided into the observation group and the control group at random. The patients in the two groups were both given the medical treatment, such as diet adjustment, malntenance of water and electrolyte acid-base balance, administration of gastro-intestinal antispasmodic and antidiarrheal agents. The patients in the observation group were additionally given 0. 15g bifidobacterium tetravaccine tablets per time, three times a day for 8 weeks. The changes of serum SP and NPY levels in the two groups before and after the treatment were observed, and the clinical curative effect and adverse drug reaction ( ADR) were compared. Results:After the 8-week medical treatment, the serum SP level in the two groups was obviously declined than that before the treatment, while the NPY lev-el was obviously ralsed than that before the treatment (P<0. 05 or P<0. 01), and the declining or increase in the observation group was much higher than that in the control group (P <0.05). Meanwhile, the total clinical efficiency in the observation group was 95. 24%, which was much higher than that in the control group (78. 57%, P<0. 05). After comparing the occurrence of ADR be-tween the two groups, no statistical differences were appeared (P>0. 05). Conclusion:Bifidobacterium tetravaccine tablets show reli-able curative effect on IBS with high security, and the curative effect has close relationship with inhibiting the release of SP, enhancing the release of NPY, adjusting the excitement of braln gut axis and inhibiting the imbalance of braln gut petide in the patients with IBS.

Chinese Journal of Medical Ultrasound (Electronic Edition) 2014;(9):749-754
Objective To summarize the sonographic features and differential diagnosis points of mass-type cornual pregnancy. Methods The sonographic ifndings of 23 pathological proven mass-type cornual pregnancy cases enrolled in PUMCH from 2011 January to 2013 January were retrospectively analyzed. Results All pathological proven mass-type cornual pregnancy were located at one corner of the uterus presenting as a heterogenous outward mass. Well-deifned margins were found in 20 cases, and interstitial-line signs were found in 15 cases. The surrounding muscle thickness is 0.1-0.3 cm. Typical hyperechoic villi were found on sonography in cases with bloodβ-hCG>20 000 IU/L. On Doppler, the lesion showed abundant peripheral vascularity with low resistance in 22 cases, 9 lesions also showed abundant internal vascularity. Among 23 mass-type cornual pregnancy cases, 7 cases were misdiagnosed as gestational trophoblastic neoplasia (GTN) due to the similar sonographic characteristics including mixed-echo and abundant vascularity with low resistance. Sixteen cases were diagnosed by ultrasound preoperatively, with featured sonographic signs including mass located in the endometrial extension line;clear margin;peripheral vascularity;or detection of interstitial-line sign and typical villus. Conclusions Mass-type cornual pregnancy may be correctly diagnosed according to the location, boundary of the mass and the distribution of blood flow combining with clinical manifestation and bloodβ-hCG level. Transvaginal sonography could play an important role in diagnosis of cornual pregnancy.

Objective To investigate the expression of miR-125a-5p in diffuse large B-cell lymphoma and the relationship among the levels of NF-κB/p65,Bcl2,Caspase3 and Ki-67 protein in clinicopathological features, and the potential prognostic value. Methods A total of 84 cases of DLBCL were enrolled in this study. At the same time,10 cases of reactive hyperplasia tissues were used as the control group. The expression of miR-125a-5p was examined by SYBR Green real-time reverse transcriptase polymerase chain reaction(real-time RT-PCR). The expression of NF-κB/p65,Bcl2,Caspase3,Ki-67 was detected by the immunohistochemical staining assay. The clinical pathological data was collected and some patients were followed up. The clinical data was used for analysis of the prognosis of patients. Results MiR-125a-5p was shown up-regulated in the DLBCL samples ,which was (4.99 ± 8.22)times of that in the reactive hyperplasia samples(P=0.002). The up-regulation of miR-125a-5p in DLBCL was correlated with non-GCB subtype(P=0.027). The up-regulation of miR-125a-5p in DLBCL was sig-nificantly correlated with the expression of NF-κB/p65,Bcl2 and Caspase3(P=0.031,0.030,0.022). There was a significant correlation between non-GCB subtype and NF-κB/p65 level in DLBCL(P=0.002). Kaplan-Mei-er survival analysis showed that the high expression of miR-125a-5p was associated with the prognosis of DLBCL pa-tients(P=0.038). Multivariate Cox regression analysis showed that the high expression of miR-125a-5p may be an independent risk factor for the prognosis of DLBCL(P=0.013). Conclusions MiR-125a-5p may play an im-portant role in the diagnosis of DLBCL. The up-regulation of miR-125a-5p in DLBCL may affect the activation of NFκB/p65,especially in the non-GCB subtype. The up-regulation of miR-125a-5p in DLBCL may influence the de-velopment of tumor via apoptosis pathway. The expression of miR-125a-5p is associated with the poor prognosis of patients, which can be regarded as an independent risk factor for the prognosis of DLBCL.

[Summary] A total of 128 individuals with type 2 diabetes underwent continuous glucose monitoring for 3 consecutive days. The dawn phenomenon was defined by three different parameters according to the previous research:(1)the absolute increase of glucose level from nocturnal nadir to prebreakfast value(?G) above 20 mg/dl;(2)?G above 10 mg/dl;( 3 ) insulin requirement increased at least 20%.The participants were secondarily separated by presence/absence of a dawn phenomenon based on the definitions above. The impact on blood glucose fluctuation of different groups was assessed according to the standard deviation of blood glucose( SDBG) , the area under curve above 10 mmol/L ( AUC ) , and the mean amplitude of glycemic excursions ( MAGE ) , etc.The frequencies of dawn phenomenon were 64.8%(?G≥20mg/dl), 85.2%(?G≥10 mg/dl), and 59.4%(rise in insulin requirement≥20%)respectively.The impacts on SDBG, AUC, MAGE, and MODD were without statistical difference(P>0.05) between the presence and absence of the dawn phenomenon patients when?G≥10 mg/dl.However, the differences reached statistical significance(P<0.05) when ?G≥20 mg/dl and the increase in insulin requirement≥20%. Besides, the incidence of dawn phenomenon was positively correlated with HOMA-IR, HbA1C , and free C-peptide.Dawn phenomenon is a very frequent event in type 2 diabetes and not only impacts the overall glycemic control but also exaggerates glucose fluctuation. To be clinically relevant, ?G≥20mg/dl should be taken as the quantitative criterion of the dawn phenomenon.

OBJECTIVETo study the effect of GABA transporter (GAT-1) on the analgesic action of oxysophoridine (OSR) in the central nervous system of mice.
METHODHot plate test was used to observe and analyze the effect of gamma aminobutyric acid and the inhibitor of GAT-1 (NO-711) on the analgesic action of oxysophoridine. Real time RT-PCR was used to investigate the influence of OSR on the expression of GAT-1 mRNA induced by formalin in spinal cord and brain of mice.
RESULTBoth GABA (2.0 mg x kg(-1), icv) and NO-711(0.125 mg x kg(-1), icv) enhanced the analgesic action of OSR (32.0 mg x kg(-1), iv) in the hot plate test, and the latencies was markedly increased (P < 0.05, P < 0.01). OSR (500.0 mg x kg(-1), iv) significantly inhibited the expression of GAT-1 mRNA induced by formalin (P < 0.05).
CONCLUSIONGAT-1 was involved in the analgesia effect of OSR and the down-regulation of GAT-1 mRNA enhanced the analgesic effect.

Chinese Journal of Lung Cancer 2005;8(4):270-273
BACKGROUNDIt has been known that different degrees of hypoxia can produce different effects on tumor. The study is to investigate the effect of hypoxia on the infiltration and migration of lung adenocarcinoma cells.
METHODSLung adenocarcinoma cells were exposed to normoxic (air, 5%O₂), hypoxic (1%O₂, 5%CO₂, 94%N₂) or anoxic (95%N₂, 5%CO₂) condition for 48 hours. The migration ability of the cells was assayed by wound healing methods. The infiltration ability was assayed by HABM-HEM model. The cells exposed to hypoxia were planted subcutaneously in nude mice, and the growth of cells and the rate of metastasis to lymph node or lung were observed. The levels of matrix metalloproteinase 2 (MMP-2) and tissue inhibitor of metalloproteinase II (TIMP-2) in culture media were assayed by ELISA.
RESULTSComparing with normoxia group, the infiltration, migration and metastasis of hypoxia group were increased significantly (P < 0.05) , as well as the level of MMP-2 (P < 0.01), and the TIMP-2 level was remarkably decreased (P < 0.05) . In anoxia group, the levels of MMP-2 and TIMP-2 were both significantly decreased (P < 0.01).
CONCLUSIONSModerate hypoxia can up-regulate the expression of MMP-2, down-regulate the expression of TIMP-2, and increase the infiltration and migration of lung cancer cells. But serious hypoxia can decrease the expression of MMP-2 and TIMP-2, and inhibit the proliferation, infiltration and migration of lung cancer cells.
