1.Effects of using distal protection device on neuroendocrine in patients with acute myocardial infarction

Objective To study changes of neuroendocrine in patients with ST segment elevation acute myocardial infarction (STEMI) after using distal protection device (GuardWire PlusTM). Method Seventy patients with STEMI received percutaneous coronary intervention (PCI) in Municipal Hospital Qingdao, during September 2004 to December 2006. They were randomdy (random numbs) enrolled in this prospective and control study. All the patients were divided into 2 groups: the distal protection device group (GW) and the non-distal protection device group (NGW).The inclusion criteria were:onset within 6 hours, chest pain more than 30 minutes without response to nitroglycerin, two or more adjacent ST segnents elevated over 0.2 mv,the proximal or middle diameter of infarction artery over 3 mm, and the increased plasma creatine kinase. The exclusion criteria were fluctuation in hemodynamics, severe heart failure, arteriopathy of left main coronary artery, mechanical complications of acute myocardial infarction and multi-vessel disease scheduled for coronary artery bypass. The plasma levels of endothelin(ET) , plasma renin activity (PRA),aldosterone (ALD),angiotensin Ⅱ (Ang Ⅱ), norepinephrine (NE) and epinephrine (E) were measured on the day of operation and on the 1st,2nd,3rd and 5th day after operation, respectively. The t-test was used to compare those neuroendocrine elements between two groups. Results There were no differences in plasma levels of all the neuroendocrine elements between two groups before operation. Compared with the NGW group, the levels of neuroendocrine elements in the plasma rapidly decreased in the GW group at 1 d after the operation ( P < 0.05). Conclusions In patients with ST segment elevation acute myocardial infarction, the distal protection device can decrease the changes in neuroendocrine.
2.The feasibility of direct applying MRI to treatment planning system of radiotherapy for intra-cranial tumor

Objective Objective To study the feasibility of direct applying MRI to TPS by measuring the geometric distortion of MRI. Methods Phantom 1 was made by marking radical arrangement of fat pipelines every 15? in foam phantom and Phantom 2 by net arrangement at every 2.7?cm. Coronal and transverse scan parallel with the radial and net fat pipeline in the phantom was performed at sequence of cranial FSE T2WI by Marconi 1.5T superconduct MRI machine. Geometric distortion in 5 different fields of view (FOV) was calculated by measuring the absolute borderline error of direct scan and coronal digital reconstructed radiograph (DRR). Results When FOV was equal or smaller than 11.0?cm?11.0?cm, 22.0?cm?22.0?cm and 27.5?cm?27.5?cm, the geometric distortion was 0.0, 1.1 and 1.4?mm of direct scanning MRI and 1.0,1.4 and 2.0?mm of coronal DRR. Conclusion When the magnetic field of MR is high and the tissue shows uniform density, MRI can be applied on TPS directly.
3.Determination of Astragloside IV in Extraction of Radix Astragali by HPLC-ELSD

Objective: To develop a method to determine Astragaloside IV in spray-dry of Radix Astragali by HPLC-ELSD. Methods: ELSD was used to determine directly Astragaloside IV in spray-dry of Radix Astragali on Elite-OSD column, using CH 3CN-H 2O(36∶64) as a mobile phase. The flow rate was 1.0 ml/min. The temperature of drift tube for ELSD was 105 ?C and the flow rate of N 2 was 2.84 ml/min. Results: The recovery of the added sample was 91.68% and RSD 1.64%. Soluble amylum was helpful for spray-dry but it can influence was determination of Astragaloside IV. Conclusion: The method is accurate and can be used in the determination of Astragaloside IV in spray-dry of Radix Astragali. Better adjuvant should be found.
4.Immunological characteristics of Mycobacterium tuberculosis antigen Rv2628.

Antigen Rv2628 of Mycobacterium tuberculosis is associated with latent tuberculosis infection. In this study, Rv2628 was prokaryotic expressed and purified, its immunological characteristics was evaluated with macrophage cell line RAW264.7 and BALB/c mice. The results show that Rv2628 was mainly expressed in form of inclusion body confirmed by SDS-PAGE, and could react with rabbit anti-H37Rv polyclonal antibody detected by Western blotting assay, indicating that the protein had an effective immunoreactivity. The interactions between Rv2628 and macrophage cell line RAW264.7 confirmed that it could effectively induce cells to produce pro-inflammatory cytokines, the relative expression level of IL-6 mRNA was higher than the control group in 1-12 h. BALB/c mice were subcutaneously immunized with Rv2628 protein, the production of IFN-gamma and IL-4 in the spleen cells was determined by Sandwich ELISA, in the Rv2628 immunized group, the level of IFN-gamma was significantly higher than that of IL-4 (P < 0.000 1). It indicated the protein induced Th1-tendency immune responses. At the same time, Rv2628(11-30) peptide used as coating antigen, the murine serum antibody titer detected by indirect-ELISA was 1:1 600, which demonstrated that Rv2628 could also induce humoral immune responses. In summary, Rv2628 could induce specific pro-inflammatory cytokines, affectively induce strongly Th1-tendency immune response and humoral response, it could be a potential target for developing subunit vaccine against TB. In addition, it laid foundation for probing the cross-talk between M. tb and host.

5.Content Determination of Baicalin in Chang'an Capsule by RP-HPLC

OBJECTIVE:To establish the method for determination of baicalin in Chang’an capsule by RP-HPLC. METHODS:HPLC was performed on C 18 column with methanol-0.07%phosphoric acid solution(44∶56)as mobile phase at a flow rate of lml/min and the temperature of column kept at room temperature.The UV detection wavelength was280nm.RESULTS:The linear range of baicalin was sample size0.1054?g~1.0540?g(r=0.9996),the average recovery was97.44%(RSD=2.92%,n=5).CONCLUSION:The method was simple,accurate and fast,and can be used for the determination of baicalin.
6.Study on neuroendocrine change in patients with ST segment elevation in acute myocardial infarction using thrombectomy

Objective To study neuroendocrine change and clinical value of percutaneous thrombectomy system(GuardWire PlusTM)in treatment of patients with ST-elevation acute myocardial infarction(STEMI). Methods 72 patients with STEMI underwent percutaneous coronary intervention(PCI)were divided into A group(38 patients)with direct stent placement after thrombectomy and B group(34 patients) with primary PCI. The plasma levels of ET, PRA, ALD, AngⅡ, NE, E were measured on the day of operation and the first, second, third and fifth days after PCI. Left ventricular ejection fraction(LVEF) was measured by echocardiography at one week and three months after PCI. Results The stents were successfully implanted in two groups. All the neuroendocrine factors have no difference between the two groups before operation. The first and second day after PCI, the levels of ET, PRA, ALD, AngⅡand E were significantly lower in A gronp than those in B group(P 0.05). Conclusions Deteriorated neuroendocrine changes are significantly improved with thrombectomy, providing potential benefits on heart function.
7.Development of an interferon-gamma ELISPOT for bovine tuberculosis.

We established an ELISPOT for bovine interferon-gamma (BoIFN-γ), and applied it in the diagnosis of bovine tuberculosis (bTB). Monoclonal antibodies that can bind with native BoIFN-γ were screened as the coating antibody and detecting antibody. After optimization of detecting conditions including coating antibody concentration, cell number, and detecting antibody concentration, the ELISPOT assay was established. Peripheral mononuclear cells (PBMCs) isolated from 30 cows were co-cultured with PPD, and detected with the ELISPOT assay. The optimal conditions of ELISPOT assay were 2.5 μg/mL coating antibody 2G5, 2.5 x 10(5) cells/well, and 1 μg/mL detecting antibody Bio-5E11. In these 30 cows tested both with the ELISPOT assay and the BOVIGAM kit, 11 cows were proved to be positive in ELISOPT assay with the sensitivity of 78.6%, and 12 cows were proved to be negative in ELISOPT assay with the specificity of 75%. The ELISPOT assay for BoIFN-γ could be used to detect bTB efficiently and it might be an alternative method for the diagnosis of bTB.

8.Depyrogenation in key manufacturing processes of Reduning injection.

OBJECTIVETo investigate the effect of removing bacterial endotoxin in the key processes of Reduning injection.
METHODThe content of bacterial endotoxins was detected by kenitic-turbidimetry and the removal efficacy was studied before and after using 0.8% of activated carbon and ultrafiltration with molecular weight cut-off of 10 x 10(3).
RESULTThe adsorption rate of bacterial endotoxins was 78.7% by using activated carbon, while the removal efficacy of bacterial endotoxins was 99.6% with ultrafiltration membrane at cut-off molecular weight 10 x 10(3).
CONCLUSIONThe key technology can effectively guarantee the safety of Reduning injection.

9.NF-kappaB/I-kappaB pathway during ischemia reperfusion injury of rat liver.

OBJECTIVETo determine the role of the NF-kappaB/I-kappaB pathway during ischemia reperfusion (I/R) injury to rat liver.
METHODSA group of rats underwent partial hepatic ischemia and reperfusion. The left and median lobe of the liver was subjected to ischemia for 90 minutes followed by reperfusion for previously specified periods. NF-kappaB activity was analyzed by electrophoretic mobility shift assay (EMSA). The protein level of I-kappaB was assessed using Western blot analysis. A semiquantitative reverse transcriptase polymerase chain reaction was used to analyze TNF-alpha and ICAM-1 mRNA levels.
RESULTSDuring liver I/R injury, NF-kappaB activation was induced in a time-dependent fashion. NF-kappaB was activated within 1 hour and 2 hours after the initiation of reperfusion and decreased afer 4 hours. The I-kappaB protein level was decreased in the cytoplasm after 2 hours, and the messenger RNA expression of TNF-alpha and ICAM-1 were increased simultaneously.
CONCLUSIONSThe data suggest that I-kappaB protein was degraded during hepatic I/R injury, NF kappaB was released and bound to special sequence in the promoters of budget genes, which regulated the expression of TNF-alpha and ICAM-1 mRNA. This provides evidence that the NF-kappaB/I-kappaB pathway plays an important modulating role in the expression of proinflammatory genes relevant to the development of ischemia reperfusion injury.

10.Application effect of timing theory in educational intervention for mothers of premature infants

Objective:To explore the effect of educational intervention based on timing theory on mothers of premature infants.Methods:Using the convenience sampling method, 80 mothers of premature infants hospitalized in the Neonatology Department of Affiliated Hospital of Youjiang Medical University for Nationalities from May 2019 to October 2020 were included in this study. According to the time of admission, they were divided into the control group (42 cases) and the observation group (38 cases). The mothers in the control group were given routine educational guidance during the hospitalization of premature infants, while the mothers in the observation group were given comprehensive educational intervention based on timing theory on the basis of the control group. The mothers′ caring ability during the transition period, breastfeeding self-efficacy and breastfeeding rate at a month after discharge, and mother′s coping ability at 3 months after discharge were compared between the two groups.Results:There were 35 cases in each group completed the study. The observation group scored (100.86 ± 6.22) on the maternal care ability of premature infants in transition period, and the control group scored (89.51 ± 4.17), the difference between the two groups was statistically significant ( t=-8.97, P<0.05). The breastfeeding self-efficacy score of the observation group a month after discharge from hospital was (47.83 ± 2.54) points, which was higher than (41.20 ± 1.97) points of the control group, and the difference was statistically significant ( t=-12.21, P<0.05). The breastfeeding rate a month after discharge in the observation group was 62.9% (22/35), which was higher than 37.1% (13/35) in the control group, and the difference was statistically significant ( χ2=4.63, P<0.05). The maternal coping ability scores of the premature infants in the observation group and control group 3 months after discharge were (119.29 ± 6.03) and (113.66 ± 6.59) points respectively, and the difference between the two groups was statistically significant ( t=-3.73, P<0.05). Conclusions:The educational intervention based on timing theory can help mothers of premature infants master the nursing knowledge and skills, strengthen the mother′s transitional care ability and post-discharge coping ability, improve the breastfeeding self-efficacy and breastfeeding rate of premature infants, promote and development the growth of premature infants, worthy of clinical application.
