1.BIOCHANIN A PROTECTS DOPAMINERGIC NEURONS BY INHIBITING MICROGLIA ACTIVATION

Objective:To explore the protective effect of biochanin A(Bioch A) on lipopolysaccharide(LPS) -induced dopaminergic neurodegeneration. Method:An in vitro model of LPS-induced dopaminergic neurodegeneration was used to investigate the protective effect of Bioch A on dopaminergic neurons against LPS-induced neurotoxicity by measuring [3H] DA uptake and counting TH-immunoreactive cells. Microglia were visualized by staining for the CR3 complement receptor with monoclonal antibody OX-42. The production of nitric oxide(NO) was determined by measuring the accumulated levels of nitrite in the culture supernatant with the Griess reagent,and release of tumor necrosis factor-?(TNF-?) was measured by immunosorbent assay. The production of superoxide was determined by measuring the superoxide dismutase(SOD) -inhibitable reduction of cytochrome C. Results:BiochA dose-dependently attenuated LPS-induced decrease in dopamine(DA) uptake and the number of dopaminergic neurons in rat mesencephalic neuron-glia cultures. BiochA also significantly inhibited LPS-induced activation of microglia and production of TNF-?,NO and superoxide in rat mesencephalic neuron-glia cultures and microglia-enriched cultures. Conclusion:Biochanin A may protect dopaminergic neurons from LPS-induced injury and its effectiveness in inhibiting microglia activation may underlie one of the mechanisms.
2.X-ray and CT findings of costal eosinophilic granuloma

Objective To study the X-ray and CT features of costal eosinophilic granuloma for a better understanding.Methods Eight patients with costal eosinophilic granuloma proved by surgery or biopsy were analyzed retrospectively.All patients had X-ray plain film, 6 patients had CT examination,including a case of enhanced CT scan.Results All 8 lesions were solitary.Six lesions were in the anterior rib and 2 in the posterior rib.On X-ray, all case showed single cavity and oval lesion with clear boundary.On CT images, 5 lesions demonstrated expansile destruction of bone with cortical bone thinning, and 3 were osteolystic destruction with soft tissue mass around.On the patient with enhanced CT scan, the lesions showed a moderate and uniform enhancement.Conclusion The X-ray and CT findings of costal eosinophilic granuloma are characteristic.
3.Evaluation of endovascular radiofrequency ablation combined with TACE in treatment of portal vein tumor thrombus of primary hepatocellular carcinoma

Objective To cvaluate the efficacy and safety of HabibTM VesOpen intravascular radiofrequency catheter percutaneous radiofrequency ablation combined with TACE in the treatment of portal vein tumor thrombus of primary hepatocellular carcinoma.Methods Thirty-nine patients with primary hepatocellular carcinoma complicated with portal vein thrombosis were treated with HabibTM VesOpen intracavitary catheter ablation combined with TACE.The postoperative success rate,complication,blood index,α-fetoprotein (AFP),portal vein patency and tumor thrombus imaging changes were assessed.Results All of 39 patients underwent surgery successfully without occurrence of perforation,infection,hepatapostema,intra-abdominal hemorrhage nor other complications associated with surgery.Blood routine examinations (WBC,RBC,PLT) after 2 weeks and 4 weeks of surgery had no statistical difference compared with those of preoperation.Liver function indexes including ALT,AST,ALB had statistical difference before and after the surgery except for TBIL.AFP had statistical difference before and after surgery.Doppler ultrasonography after 4 weeks showed blood flow in original blocked portal vein.Enhanced CT or MR examinations after 8 weeks on the abdomen,suggested that tumors were shrinked or disappeared.Conclusion Radiofrequency catheter ablation with intracavitary radiofrequency catheter of HabibTM VesOpen is an effective method in the treatment of portal vein tumor thrombus in primary hepatocellular carcinoma.
4.Preliminary application of multi-slice CT direct venography in lower extremity vein

Objective To discuss the injective method, contrast agent concentration and delay time of MSCT direct venography in lower extremity vein, in order to make optimal venographic schedule. Methods Thirty-two patients with lower extremity vein diseases underwent MSCT direct venography in using different concentration and injective rate of contrast agent, and the images were analyzed. The displaying ability of MSCT direct venography in lower extremity vein segment was compared with that of X-ray venography in 22 patients. Results Blood flow correlation constructed defect was the main factor impacting image quality of CT venography in lower extremity. The image scores between the groups of injective rate 1.5 ml/s and the group of 2.5 ml/s were statistically different (χ~2=5.495, P=0.019), whereas no difference of image score was found between the group of concentration of 25% and 15% (χ~2=3.333, P=0.068). The displaying ability of CT and X-ray venography in anterior tibial vein, posterior tibial vein, popliteal vein and femoral vein was not statistically different (χ~2=2.095, P=0.148). For iliac vein, the displaying ability of CT outstripped that of X-ray venography (χ~2=8.324, P=0.004). Conclusion With the schedule of delay time 50 s, injective rate 2.5 ml/s and concentration 45 mgI/ml (nonionic 300 mgI/ml and diluted 15%), MSCT direct venography in lower extremity can acquire satisfactory image, and its displaying ability of iliac vein is better than that of X-ray venography.
5.Progress in Pre-mRNA Alternative Splicing in Gliomas

Gliomas constitute the most common type of primary brain tumor. Alternative pre-mRNA splicing may play roles in the carcinogenesis, cell growth and invasion in various types of gliomas. Factors regulating the alternative spicing in gliomas include cis-regulatory element (for example, ESE, ISS and ESS) and trans-regulatory factors (such as SRp55, SC35, SF2/ASF and PTB). Recent progresses demonstrate that many genes associated with gliomas, including those encoding tumor suppressors or promoters, enzymes, receptors and ion channels are subject to regulation by alternative pre-mRNA splicing. Therefore, studying the alternative pre-mRNA splicing in gliomas will be of benefit to understanding the molecular basis of gliomas and identifying new targets potentially useful for early diagnose as well as therapy of gliomas.
6.The Expression of SRp38 Gene in Mouse Retina and It's Regulation of Alternative Splicing of GluR-B Minigene

SR proteins play important roles in regulating alternative pre-mRNA splicing. As a newly discovered neural and reproductive tissue specific SR protein, SRp38 regulates the alternative splicing of several genes important for neural function, such as GluR-B, Trk-C and NCAML1. It also acts as a splicing inhibitor during mitosis or stress response in order to prevent wrong splicing. The expression of SRp38 in mouse retina was investigated by Western blot and immunohistochemistry (IHC) analyses. The result shows that the expression of SRp38 proteins in mouse retina is region-specific, with extensive distribution in the outer and inner plexiform layers, inner nuclear layer and ganglion cell layer, but no expression in outer nuclear layer. Double staining of isolated retina cells with anti-SRp38 and anti-Trk-C antibodies showed that SRp38 is localized in the dendrites, somata and axon terminals of rod-bipolar cells. By transient co-transmission of over-expressed SRp38 plasmid and RT-PCR analyses, the further results showed that overexpressed SRp38 could promote the splicing of the Flip isoform of GluR-B minegene in R28 cells. The result suggests that SRp38 may play important roles in the retinal function, possibly via regulating the neural-specific alternative splicing of genes as GluR-B.
7.A Simultaneous Determination of Five Components in Radix et Rhizoma Salviae Miltiorrhizae by RP-HPLC

Objective To develop a method for simultaneous determination of three hydrophilic components and two lipophilic components in Radix et Rhizoma Salviae Miltiorrhizae. Methods The RP-HPLC method was performed by using a Welchrom C18 column(250 mm×4.6 mm,5 μm)with a mobile phase of acetonitrile (A)-0.1%phosphoric acid(B). The gradient elution program was as follows:0-15 min,10%→12%A;-35 min,12%→20%A;-45 min,20%→60%A;-65 min,60%→65%A;-80 min,65%→80%A;-90 min,10%A. The flow rate was kept at 1.0 mL?min-1 . The detection wavelength was set at 280 nm. The column temperature was 30 ℃ . Results A good linearity was obtained over 0.059 5-2.380 0 μg for tanshinol, 0.346 0-13. 840 0 μg for rosmarinic acid, 0. 656 0 - 26. 240 0 μg for salviamolic acid B, 0. 420 0 - 16.800 0 μg for cryptotanshinone and 0.414 0- 16.560 0 μg for tanshinoneIIA, respectively ( r = 0.999 9). The average recovery rates were between 98.69%-100.91% with RSD less than 1.2%(n = 6). Conclusion The method is rapid, accurate, credible and repeatable, and can provide basis for the quality control of Radix et Rhizoma Salviae Miltiorrhiza.
8.Application of point massage combined with physiotherapy in the treatment of insomnia for the community elderly

Objective To observe the application effect of point massage combined with physiotherapy in the treatment of insomnia for community elderly.Methods 30 elderly patients with insomnia who accepted point massage combined with physiotherapy were selected as observation group,and 30 elderly patients treated in tertiary hospital with insomnia who took alprazolamum for treatment were selected as control group.The SRSS score of the two groups before and after treatment,as well as treatment efficacy were compared.Results The insomnia treatment efficacy of the observation group was higher than that of the control group(Z =2.45,P <0.05).The SRSS rating of the observation group before treatment was (30.76 ±7.1 9)points and after treatment was (1 5.43 ±3.95)points.The SRSS rating of the control group before treatment was (32.33 ±6.98)points,which after treatment was (21 .93 ±6.73)points.The SRSS scores of both two groups after treatment were decreased (t =1 0.23,5.87,all P <0.05),with obvious differ-ences.The SRSS scores of the observation group decreased more obviously and was lower than that of the control group (t =4.56,P <0.05).Conclusion The point massage combined with physiotherapy can effectively improve insomnia of the elderly,which is also with advantages of easy operation and no obvious side effects and high cooperation degree etc,and it is suitable to be promoted in community.
9.Experimental research on the characteristics of pancreatic cells regeneration in an animal model for isolated pancreatic trauma

Objective To study the relationships between tissue damage and the ability of the pancreatic cells to regenerate ,and analyze the alteration of the pancreatic cells regeneration .Methods Sixty rats were divided into two groups :impact group(the pan‐creas was injured by a BIM‐Ⅲ biotical impact machine ,40 rats) and control group(sham operated ,20 rats) .All rats were sacrificed at 6 h ,24 h ,72 h ,7 d after operation .The level of AMS ,LPS in the serum were detected by spectrophotometry ,pancreatic cells re‐generation were examined and analyzed by TUNEL staining and flow cytomertry ,and the Bcl‐2 and Bax expression were measured by Western blot .Results In the impact groups ,LPS was activated later than AMS ,and lasted persistently .The results from TUNEL stain ,flow cytometry and Western blot indicated that pancreatic trauma induces cell death and the compensatory prolifera‐tion of pancreatic cells .The characteristics of pancreatic cells regeneration in the animal model of isolated pancreatic trauma indicate that the proper remedial time is in the first 24h after the pancreatic trauma .Conclusion Detecting AMS and LPS at the same time can help us to determine the exocrine function of pancrease .
10.Clinical efficacy of fractional laser (AffirmTM) with combined apex pulse and multiplex technology on post-acne scars and other skin lesions

Objective To evaluate the clinical efficacy of factional multiplex laser (AffirmTM) with combined apex pulse (CAP) technology in treating depressive acne scars,post inflammatory erythema and enlarged pores and other lesions.By following up,the correlative factors were analyzed to guide the further use of the factional laser (AffirmTM) with CAP technology.Methods Patients who received the AffirmTM laser therapy from Sept.2007 to July 2008 were enrolled in this retrospective follow-up study.Pictures were taken before and after each treatment during the therapy.Then the pictures were evaluated by dermatologists to attain object variables.Other subject variables from patients were recorded during survey.The relation between the efficacy and age,sex and treatment frequencies were analyzed statistically.Results The effective rate was 25.34 % for post-acne scars,36.84 % for post-acne erythema.28.57 % for enlarged pores,and 14.29 % for rythids.Nonparametric test showed no statistical difference with gender or age.There was correlation between efficacy and treatment frequencies.No severe adverse effect was observed.Conclusions AffirmTM laser with CAP technology has a good effect on treating post-acne depressed scars and erythema,enlarged pores and fine rythids.
