1.Isolation,characterization and application of umbilical cord mesenchymal stem cells in the treatment of liver diseases

Human umbilical cord (UC) has been attracting increasing research effort in recent years.Since UC is a postnatal organ discarded after birth,the collection of its cells is non-invasive and raises no ethical con-cerns.Human umbilical cord provides large amount of mesenchymal stem cells(MSCs) which hold unlimited ca-pability of proliferation and great potential of differentiation and. MSCs have stable biological properties and low-er immunogenicity and can function well after amplifications.Studies have demonstrated that MSCs have the po-tential of inhibiting inflammatory response, generation of fibrosis, apoptosis and stimulating regeneration of hepa-tocytes.Therefore, UCMSCs could be an ideal cell source for the therapy of liver diseases.
2.Application of visible simulation surgery in the typeⅡB odontoid fractures

Objective Through the exploration of the reality technology to find the treatment of type Ⅱ odontoid fractures in anterior fixation of odontoid fractures with hollow screw .Methods Based on 64 row spiral CT continuous 2-dimensional images of akull ,atlas and axis were reconstructed by the Mimics software ,Mimics software reconstruction atlas ,the atlantoaxial 3D visualization structure in Mimics, simulated surgery was carried out in the MedCAD .Results The structure of three-dimensional reconstruction image was clear ,structure of hollow screw into pin-point was accurate in virtual simulation surgery ,the screw path is correct .Conclusion Mimics Software has great val-ues in the diagnosis of odontoid fractures ,preoperative risk evaluation ,place the hollow screw and clinical teaching ,which provides an objec-tive basis for choice of treatment program .
3.Knowledge,attitude and related factors associate with HIV/AIDS among female prisoners in Shaanxi province

Objective To investigate the knowledge,attitude and related factors associated with HIV/AIDS among female prisoners in Shaanxi province.Method A cross-sectional survey with questionnaire was conducted in 837 female prisoners.Results Of the studied female prisoners 94% had heard of HIV/AIDS.The rate of correct answer on basic knowledge about AIDS was 19.8%-87.3%.The rate of correct answer on the transmission route was 11.6%-87.6%.The rate of correct answer on non-transmission route was 23.4%-48.6%.The rate of correct answer on prevention was 36.7%-57.5%.Of them 46.6%-55.7% were afraid of AIDS,and 26.6%-90.0% had prdjudice and stigma to HIV/AIDS and PLWHA(people living with HIV/AIDS).The factors affecting HIV/AIDS related knowledge included marriage status,career,education background and number of family members.The factors affecting attitude included age and education background.Conclusion The studied female prisoners have wide but superficial knowledge about HIV/AIDS and have expressed over fear for HIV/AIDS.They also cannot tolerate AIDS patients.
4.Preparation of GM-CSF Loaded Chitosan Nanoparticles

OBJECTIVE: To study the feasibility of using biodegradable chitosan nanoparticles as new type carriers for controlled GM-CSF delivery system. METHODS: GM-CSF and BSA loaded chitosan nanoparticles were prepared by ion cross linking method using tripolyphosphate as cross-linking agent with its morphology observed by transmission electron microscope. The encapsulating efficiencies of GM-CSF and BSA were determined respectively by fluorescence scanning and ultraviolet spectrophotometer. The in vitro drug release properties of chitosan nanoparticles were studied as well. RESULTS: Morphologically, the nanoparticles were spherical with mean particle size of 201nm. The encapsulating efficiencies of GM-CSF and BSA were 62.1% and 58.5%, respectively, and their in vitro accumulative drug release rates on the 3rd day were 69% and 82% respectively. CONCLUSION: The ion cross linking method is fit for the preparation of GM-CSF loaded chitosan nanoparticles.

7.Effects of Environmental Lead Exposure on Immune System in Preschool Children

Objective To explore the effects of environmental lead exposure on immune system in preschool children. Methods The blood lead levels of 217 preschool children were determined by graphite furnance atomic absorption spectrophotometry. The distribution of T_lymphocyte subsets: CD 3+,CD 3+CD 4+,CD 3+CD 8+, B cells (CD 3- CD 19+), NK cells (CD 3-CD 16+CD 56-) were analyzed by flow cytometer, the levels of serum immunoglobulin G and immunoglobulin M were determined by scattering turbidimetry, the levels of serum immunoglobulin E were examined with ELISA. Results The mean level of blood lead of 217 preschool children was (0.46?0.27)?mol/L(range:0.11~2.71 ?mol/L). The blood lead levels of 63 preschool children were ≥0.48 ?mol/L. 38 preschool children among 63 preschool children with blood lead level of ≥0.48 ?mol/L were selected as the high_blood_lead group, 35 preschool children with blood lead levels of 0.05). Condusion The blood lead levels of ≥0.48 ?mol/L presented adverse effects on the T_lymphocyte subsets.

The in vitro modulation effect of insulin on DNA and IL-2 production of spleen lymphocytes in diabetic mice were studied. The results suggest that the DNA and IL-2 production of the lymphoc-ytes are significantly inhibited in the mice. When suspend the lymphocytes to the culture medium containing insulin, the DNA and IL -2 production of the lymphocytes are remarkably increased. Therefore, insulin is an important immunoregulation hormone.
9.Isolation and purification of breast cancer stem cells by suspension culture combined with chemotherapeutic agents

Background and purpose:The scarcity in quantity and continuous differentiation ability severely compromise the isolation and purification of cancer stem cells(CSCs).The existing sorting methods can only enrich CSCs as a heterogeneous population.However,more primitive CSCs closer to the source still have not been identified.Based on the CSC hypothesis,we hereby explored the possibility of isolation,culture,purification and identification of murine breast cancer stem cells in suspension culture combined with anticancer regimens. Methods :TM40D murine breast cancer cells were cultured in serum-free medium.The expression of CD44 + CD24-was measured by flow cytometry.Cells from the passage of TM40D cells with the highest expression of CD44 + CD24-were treated in combination with anticancer agents pacilitaxel and epirubicin at different peak plasma concentrations(PPC) for 24 hours,and then maintained under suspension culture.The rate of apoptosis was examined by flow cytometry with Annexin-V FITC/PI double staining method.Selected cells in different amounts were injected subcutaneously into BALB/C mice to observe tumor formation. Results :Cells of passage 10 in suspension culture had the highest percentage of CD44 + CD24-(about 77 percent).As few as 100 cells in 0.35 PPC could generate tumors in BALB/C mice. Conclusions :TM40D cell line contains breast cancer stem cells.This study suggested that suspension culture combined with anticancer regimens is a feasible approach for screening tumor stem cells.
10.Large-sample Analysis of the Normal Reference Value of Hematological and Serum Biochemical Parameters in Healthy Beagle Dogs

Objective To establish the normal reference value range of hematological and serum biochemical parameters in Beagle dogs for our own research center and to supply references for establishing the national standard.Methods CV Beagle dogs aged 4~6 months were selected as the investigative objects.Of the Beagle dogs,263 were male with body weight of(8.09?0.78)kg,263 were female with body weight of(6.84?0.87)kg.Blood was collected from cephalic vein at the endgame of the experiment,then both hematological and serum biochemical parameters were measured.The acquired data were analyzed by the "Independent-Samples T Test" process of SPSS V 13.0.Regarded the males and the females as two independent samples,the difference between sexes was also compared.Results The mean normal reference values and the standard deviation of hematological and serum biochemical parameters in both male and female Beagle dogs were obtained by analysis.Conclusions Comparison of sexes showed that there exists obviously difference between male and female Beagle dogs referring to 3 serum biochemical parameters of T-Bil,GLU and ALB.
