1.Efficacy of Emergency Endoscopic Hemostasis Combined with Somatostatin in Treatment of Upper Gastrointestinal Hemorrhage

Objective:To discuss the efficacy of emergency endoscopic hemostasis combined with somatostatin in treatment of upper gastrointestinal hemorrhage.Methods:100 patients with upper gastrointestinal hemorrhage were selected and divided into two groups randomly.The control group (48 cases) was given conventional hemostatic measures.The observation group (52 cases) was given emergency endoscopic hemostasis combined with somatostatin.The efficacy of emergency endoscopic hemostasis combined with somatostatin in treatment of upper gastrointestinal hemorrhage was evaluated by efficacy,the curative success rate and the improvement situation of clinical symptoms.Results:The effective rate was 88.5 % in the observation group,and the effective rate was 70.8 % in the control group,and the effective rate of observation group was higher than that of the control group (P<0.05).The success rate for different lesion size was higher in the observation group compared with control group (P<0.05).According to the success rate,with the increased size of lesion,the hemostasis rate was decreased.The hospitalization,negative fecal occult and haematemesis disappeared time of observation group was shorter than that of the control group (P<0.05).The postoperative bleeding rate of observation group was lower than that of the control group (P<0.05).Conclusions:The emergency endoscopic hemostasis combined with somatostatin has a good therapeutic effect on upper gastrointestinal hemorrhage.It can improve the clinical symptoms and shorten the hospitalization time,but its effect is limited on large lesion of upper gastrointestinal hemorrhage.
2.Clinical research of functional reconstruction with BTA for treatment on equinovarus foot of spasitic cerebral palsy

Objective To observe the clinical therapial value of functional reconstruction with Botulinum Toxin A (BTA) on spasitic cerebral palsy. Methods Thirty-two patients were treated by Achilles tendon lengthening and anterior transfer of posterior tibial tendon.According to the spasticity of triceps surae muscle,all cases were arranged by BTA injection 2 months later after operation.Results From Jan.2000 to Jan.2009,thirty-two cases with equinovarus foot of spasticitical cerebral palsy were collected,the muscle strength of ankle dorsal extensor increased from 0-2 grades to 4-5 grades,there was significant difference between preoperational muscle strength and postoperational one.There was also significant improvement to adjust yarus degrees of ankle joint.the musclar tension of triceps muscle of calf decreased from Ⅱ-Ⅳ grades to Ⅰ-Ⅱ grades. Conclusion Anterior transfer of posterior tibial tendon corresponding with Botulinum Toxin A injection not only release muscle spasticity but also improve dorsal extending strength of ankle joint.The clinical effect of these methods was reliable on cerebral palsy.
3.Reason for dormancy of Cuscuta chinensis seed and solving method.

OBJECTIVETo study the reason for the deep dormancy of the aged Cuscuta chinensis seed and find the solving method.
METHODThe separated and combined treatments were applied in the orthogonal designed experiments.
RESULTThe aged seed had well water-absorbency; the water and ethanol extracts of the seeds showed an inhibition effect on germination capacity of the seeds.
CONCLUSIONThe main reason for the deep dormancy of aged C. chinensis seed is the inhibitors existed in seed. There are two methods to solve the problem. The seeds is immersed in 98% of H2SO4 for 2 min followed by 500 mg x L(-1) of GA3 treatment for 60 min, or in 100 mg x L(-1) of NaOH for 20 min followed by 500 mg x L(-1) of GA3 treatment for 120 min.

4.Effects of sevoflurane post treatment on cerebral ischemia-reperfusion injury in diabetic rats and non-diabetic rats

Objective To investigate the effects of sevoflurane post treatment on cerebral ischemia-reperfusion injury in diabetic rats and non-diabetic rats.Methods Thirty-two male SD rats,aged 3.0-3.5 months,weighing 280-320 g,were divided into four groups by random number method (n =8):non diabetic ischemia reperfusion group (group NDC);diabetic ischemia reperfusion group (group DC2);non diabetic ischemia reperfusion group with the sevoflurane post-treatment (group NDS);diabetic ischemia reperfusion group with the sevoflurane post-treatment (group DS).The middle cerebral artery occlusion method and streptozotocin were used to establish the ischemiareperfusion injury model and diabetic model.2 h after ischemia and 24 h after reperfusion,the rats nerve defect scale was detected,the cerebral infarction volume was evaluated by TTC method,and Western blot method was used to detect angiogenin-1 (Ang 1) expression.Results Between the four groups of rats,nerve deficit score and infarct volume were significantly higher,Ang-1 protein relative expression was significantly lower in group DC than those in group NDC (P<0.05);neural deficit score and infarct volume were significantly lower,Ang-1 protein relative expression was significantly higher in group NDS than those in group NDC (P<0.05);nerve deficit score and infarct volume were significantly lower,Ang-1 protein relative expression was significantly higher in group DS than those in group DC (P<0.05).Conclusion In the rats after cerebral ischemia reperfusion,diabetes could aggravate the nerve defect,increase the volume of cerebral infarction,reduce the expression of Ang-1;sevoflurane could reduce nerve defects,reduce infarct volume,increase the expression of Ang-1.The expression of Ang-1 and degree of injury after cerebral ischemia reperfusion in rats have relationship.
5.Establishment and application of LAMP-Taqman detection method of porcine pseudorabies virus

A LAMP-Taqman rapid detection system for porcine pseudorabies virus(PRV)was de-veloped based on LAMP and quantitative PCR.LAMP primers were designed for PRV conserved sequences,and the loop primer modified by the fluorescent quenching group was used as the Taq-man probe.The composition optimization,specificity,sensitivity and repeatability of the LAMP-Taqman system were tested using positive samples and recombinant plasmid as templates.Thirty-eight samples of pork swabs were tested with the commercial LAMP detection kit in parallel to verify the actual detection effect of the LAMP-Taqman detection system.The results showed that the optimal final concentration of each component was as follows:PRV-FIP/BIP 0.8 μmol/L,Bst DNA polymerase 0.7 U/μL,Taq DNA polymerase 0.24 U/μL,dNTPs 1.6 mmol/L,MgSO47.2 mmol/L.This system had good specificity and did not cross-react with other virus samples.The linear correlation coefficient of gradient samples was 0.995,the coefficient of variation of repeatable tests was less than 3.000%,and the minimum detection limit could reach 2.81 ×102 copies/μL.The test results of the actual swab samples were consistent with the commercial isothermal fluores-cence detection reagents.In conclusion,the PRV detection system established by LAMP-Taqman method in this study is specific,sensitive,stable and accurate,and is a reliable technical method suitable for the accurate detection of porcine PRV.
6.Antibody persistence 3 to 5 years after vaccination with measles, mumps and rubella combined attenuated live vaccine in children

Objective:To assess the antibody persistence 3-5 years following vaccination of measles and rubella combined live-attenuated vaccine (MR) at 8 months of age and measles, mumps and rubella combined attenuated live vaccine (MMR) at 18 months of age.Methods:In 2016, 18-month-old children who were vaccinated with one dose of MR vaccine at the age of 8 months were recruited in Hebei Province as group 1; 4-, 5- and 6-year-old children who were vaccinated with one dose of MR vaccine at the age of 8 months and one dose of MMR vaccine at 18 months of age were recruited in Shanxi, Inner Mongolia and Beijing as group 2, group 3 and group 4, respectively. Serum samples were collected to detect IgG antibodies against measles, mumps and rubella by ELISA. Geometric mean concentrations (GMCs) of measles, mumps, and rubella antibodies were compared among groups by analysis of variance or non-parametric test. Seropositive rates were compared among groups by Chi-square test or Fisher′s exact test. Results:A total of 650 children were included in this study. Seropositive rates of measles, mumps and rubella antibodies 30 d after vaccination of 150 18-month-old children with one dose of MMR vaccine were 100%, 91.33% and 100%, respectively, and the GMCs were 1 846.87 mIU/ml, 299.91 IU/ml and 111.33 IU/ml, respectively. Seropositive rates of measles, mumps and rubella antibodies 3-5 years after vaccination one dose of MR vaccine at 8 months of age and one dose of MMR vaccine at 18 months of age were above 94%, 79% and 71%, respectively, and the GMCs were above 830 mIU/ml, 240 IU/ml and 31 IU/ml. No significant difference in the seropositive rates of the three antibodies was observed among groups 2, 3 and 4 ( P>0.05). There was no significant difference in the GMCs of measles or mumps antibodies among the three groups ( P>0.05), but the differences in the GMCs of rubella antibodies were statistically significant ( P=0.034). Conclusions:Measles, mumps and rubella antibodies persisted for 3-5 years without significant decrease after vaccination one dose of MR vaccine at 8 months of age and one dose of MMR vaccine at 18 months of age.
