1.Studies on Cytotoxicity of Nitric Oxide to Schistosomula of Schistosoma japonicum

Objective To study the in vitro larvicidal activity of nitric oxide (NO) to the juvenile Schistosoma japoni-cum. Methods Macrophages were induced by LPS or LPS + IFN-? to produce NO, schistosomula obtained mechanically from cercariae were added to the medium with activated macrophages, the larvicidal activity was observed within 48 h . In order to further confirm the effect of NO, an inhibitor of iNOS,L-NNA (N?-nitro-L-arginine), was used to inhibit the production of NO, larvicidal activity was measured by the same methods and the difference of dead larvae ratio was compared between the inhibited and uninhibited groups. Results LPS and LPS + IFN-? can induce macrophages effectively, with the NO production of (109.96?3.70)?mol/L and (113.50?7.38) ?mol/L respectively, accordingly the larvicidal effect reached to 91.07% ?2.92% and 96.86%?2.36% respectively. This activity can be inhibited by L-NNA. NO production and dead larvae ratio were reduced significantly in the inhibited group than in the uninhibited one. Conclusion NO produced by activated macrophages can kill schistosomula of Schistosoma japonicum.
2.An analyses of 104 cases spontaneous rupture of hepatocellular carcinoma

Objective To investigate the clinical management of Spontaneous rupture of hepatocellular carcinoma(SRHCC) . Methods This was a retrospective review of the clinical data of patients with SRHCC who underwent treated in the affiliate hospi‐tal of Luzhou medical college from January 2001 to December 2014 .Results Among 104 patients ,small hepatocellular carcinoma (<5 cm) were found in 11 cases ,and large hepatocellular carcinoma(>5 cm) in 93 cases .Thirty‐one cases which underwent surgi‐cal treatment ,were cured;44 underwent transcatheter arterial embolization (TAE) ,5 cases died of liver function failure;29 cases were treated conservatively ,11 cases died of huge bleeding ,18 cases gave up discharged .Twenty‐two small and medium‐sized SRH‐CC cases underwent hepatectomy survived 1to‐10 years ;8 huge sized SRHCC cases survived 5to‐13 months;one case who under‐went partial filling pressure hemostasis and hepatic artery ligation ,but died of tumor rupture again after 34 days .Sixteen cases un‐derwent TAE were followed up ,14 cases survived 3to‐10 moths ,the survival time of two cases were 3 years and 5 years ,respective‐ly .Conservative treatment group has not been followed up .Conclusion The tumours should be surgical resection as soon as possi‐ble in those whose lesions confined to the liver and may be removed ,systemic condition is good;TAE should be used for other pa‐tients .
3.Neural Cell Apoptosis in Postnatal Developing Spiral Ganglion of Rat

Objective To study cell apoptosis and differentiation in postnatal developing spiral ganglion(SG)of rat.Methods By means of transmission electron microscope(TEM),cell apoptosis and the ultrastructure of rat SG neurons were investigated during postnatal days 1,3,5,7,10,14,and 30.Results In P5 and P7,neural cell apoptosis was observed in rat SG neuron.The typical morphological alterations of apoptotic cells included a crescent-shaped band of condensed chromation round the nuclear periphery,apoptotic bodies,and cell shrinkage.The first appeared type 2 neurons with densely packed neurofilaments in the cytoplasm could be recognized with confidence at postnatal day 7.The type 2 neurons at this time showed obvious signs of cell shrinkage.Cell apoptosis disappeared subsequently.In subsequent developing period,the morphological characteristics of type 1 and type 2 neurons became more remarkable and gradually achieved their mature structure.Conclusion The neural cell apoptosis was an important event in the postnatal development of rat SGCS.Cell apoptosis,occurred prior to appearance of the type 2 neurons,may be related to remodeling of the outer hair cell innervation and type 2 neuron differentiation.
4.Exploration on the experiment teaching reform of microbiology examination based on MES mode

According to the course character and training objective, we analyzed the necessity of the experiment teaching reform of microbiology examination . The modules of employable Skills (MES) was applied to the experiment teaching of microbiology examination tentatively. On the basis of the needs of microbiology laboratory jobs, the contents of experimental course were integrated and optimized to skills module , comprehensive training module and application module , which include fourteen study units such as basic techniques of identification of bacteria,the separation and identifi-cation of pathogenic bacteria, microbiology examination of clinical specimen etc. Intensive teaching and multiplex teaching methods were applied to each module according to the module's characteristic, teaching objectives and cognitive rules of students. This teaching reform has achieved initial results.
5.The change in clinical presentation of patients with hepatobiliary stones: an analysis of 2 359 patients treated in a single hospital

Objective To review the clinical presentation of patients with hepatobiliary stones (HS).Method 2 359 patients with HS were divided into group A and B according to the presentation of these patients before or after 2002.Their clinical data were retrospectively analyzed.Results The age,the percentage of patients with a case history > 10 years,the admission rate for relapse,the intrahepatic to extrahepatic stone ratio,the number of patients complicated with liver cirrhosis/portal hypertension,the elective operation rate,the ratio of biliary drainage operation,or the ratio of biliary drainage combined with hepatic resection in group B were 54.02 ± 13.54 years,68.99%,53.07%,73.18%,13.41%,80.80%,83.81%,44.74%,respectively.The corresponding figures for group A were 48.65 ± 14.47 years,46.25%,32.0%,62.02%,4.63%,63.92%,41.45%,19.05%,all P <0.01.However,the rates of biliary ascariasis,acute cholangitis of severe type (ACST),hepatic abscess,bleeding or perforation of the biliary tract,non-operative mortality,emergency operation rate and stone residual rate in group B were 6.56%,6.15%,0.84%,0,0,1.71%,5.18%,18.70%,respectively.All these were significantly lower than those in group A (12.11%,33.72%,1.95%,0.37%,0.67%,25.62%,respectively,all P < 0.01).Conclusions The popularization of medical insurance and the increase in hospital admission rate,but not the actual increase in HS,led to the increase in hospitalization of patients.There was a tendency of less patients presenting with severe disease due to delay in treatment.Routine choledochoscopic stone extraction intraoperatively or postoperatively and the increased liver resection rate had decreased the residual stone rate.There should be a strict restriction on the use of choledochojejunostomy.
6.Shear bond strength of recycled metal brackets

BACKGROUND: Bracket dropping is often seen in clinic. Can these shed brackets be used again; what should be done to treat these brackets; is the bond strength of these recycled brackets different from those new ones, these questions are often concerned by orthodontists in their clinic and are the purpose of our study as well.OBJECTIVE: To evaluate the shear bond strength and compare two base-cure methods of recycled brackets. DESIGN: Grouping control study. SITTING: Department of Orthodontics, the Affiliated Hospital of Medical College, Qingdao University; Department of Materials, Dental School, Beijing University. MATERIALS: The teeth were collected from Oral Surgery, the Affiliated Hospital of Medical College, Qingdao University. The age was from 11 to 19 years. The gender was half male and half female. Intact enamel of the crown was required, but decayed, fracture, tetracycline pigmentation teeth or fluorosis teeth were excluded. Patients provided the confirmed consent for this experiment.METHODS: The experiment was carried out in Stomatology College of Peaking University in December 2005. Thirty ex vivo teeth were selected randomly from those prepared enamel surfaces, used as sample for rebounding. The edgewise brackets of swallow-tailed brackets were bonded on each tooth and rebonded after 24 hours. The teeth were cleaned and divided randomly into 3 groups. There were 10 teeth in each group. Group 1 was bonded with new brackets; group 2 with recycled brackets whose adhesive remnant were burned and got rid of; group 3 with recycled brackets whose adhesive remnant in the groove of the base were stored. Bracket was provided by Standard edgewise, Orsu Company, Hangzhou, China. After 24 hours, the shear bond strength was tested with material testing machine (Autogragh, Shimadzu, Japan). Adhesive remnant index (ARI) was added up. MAIN OUTCOME MEASURES: Shear strength of old metal bracket after rebonding and adhesive remnant.RESULTS: ① Shear bond strength: The shear bond strengths were (10.094±3.158 9) in group 1, (10.266±2.406 0) in group 2 and (8.898±1.365 9) in group 3. There were no statistically significant differences among the three groups (P > 0.05). ② ARI scores: ARI scores were 51.7%, 58.3% and 35.0% in the three groups, respectively. There were statistically significant differences between group 2 and 3. CONCLUSION: In vitro study indicates that the recycled metal brackets can reach good bond strength and that maybe there is chemical bond between new and old adhesive.
7.Effect of orthodontic extraction treatment and non-extraction treatment on esthetical smile

Objective To investigate the effect of orthodontic tooth extraction and non-extraction on dental arch width and esthetical smile. Methods 100 patients treated without extraction and 100 patients treated by 4 first-premolars extraction were selected. The study models of the patients were measured before and after the treatment and compared statistically. Measurements were made in the maxillary and mandibular canine regions from the most labial aspect of the buccal axial surfaces of the canine roots. Results Before treatment, maxillary and mandibular arch widths were the same between both groups (P>0.05). In non-extraction group, mandibular arch width of posttreatment was 0.88 mm larger than that of pretreatment (P<0.001), and maxillary arch width of posttreatment was 0.84 mm large (P<0.001). In extraction group, mandibular arch width of posttreatment was 1.64 mm larger than that of pretreatment (P<0.001), and maxillary arch width of posttreatment was 1.50 mm large (P<0.001). After treatment, the width of mandibular arch in the extraction group was 0.59 mm larger than that in the non-extraction group (P>0.05), while the width of maxillary arch in the extraction group was 0.10 mm less (P>0.05).Conclusion Both extraction treatment and non-extraction treatment do not result in narrower dental arch, but wider. The view that orthodontic extraction results in narrower arch widths and unaesthetic smiling is untenable.
8.EFFECT OF NITRIC OXIDE ON LIVER FIBROSIS OF MICE INFECTED WITH SCHISTOSOMA JAPONICUM

Objective To investigate the effect of nitric oxide on the liver fibrosis of mice infected with Schistosoma japonicum during the early stage of schistosomiasis. Methods NMRI mice were treated with AMG-an iNOS inhibitor from day 23 post infection(p.i) to the sacrificed and the livers were collected at 38 days, 45 days p.i respectively. The expression and distribution of collagen typeⅠ, Ⅲ (ColⅠand ColⅢ) in liver tissues were investigated with the Picric acid-Sirius red staining techniques and differentiated with the polarization microscopy combining with picture analysis system. Hydroxyproline concentration in liver homogenate was measured by the biochemical methods. Results At 38 day p.i, the Picric acid-Sirius red staining showed that the hyperplasia of Col Ⅰ and ColⅢ in the livers of AMG treated mice increased significantly compared with the control livers, but there was no significant difference of hydroxyproline content between the two groups. At 45 days p.i, only the hyperplasia of Col Ⅰ in AMG treated group increased significantly compared with the control livers, and moreover, content of hydroxyproline of the inhibited mice was significantly higher than that of the control mice (P
9.Treatment of massive variceal bleeding secondary to localized pancreatitis-associated portal hypertension

Objective To study treatment of massive variceal bleeding secondary to localized pancreatitis-associated portal hypertension (MVBPAPH).Methods A retrospective study on the clinical data of patients with MVBPAPH was carried out.Of 24 patients with MVBPAPH,20 had pancreatic pseudocysts.12 were operated after failure of treatment with endovascular intervention for variceal bleeding (including 10 patients with splenectomy and gastric fundus-body peripheral vessels amputation and 2 patients with pancreatic pseudocystogastrostomy).8 patients underwent partial splenic embolization and left gastroepiploic artery embolization.4 patients directly underwent splenectomy and gastric fundus-body peripheral vessels amputation for variceal bleeding.Results Left pleural effusion developed in 5 patients who underwent arterial embolization.Left pleural effusion and lung infection occurred in 2 patients who underwent operation.All patients recovered well and were discharged home.During the follow-up period of 2 to 72 months,no rebleeding occurred in these patients (including 2 patients had passed little interval melena).Gastroscopy re-examination showed that variceal veins were not found in 18 patients.Variceal veins which were detected in the remaining 6 patients were obviously less severe.Conclusion Individualized treatment should be given to patients with MVBPAPH and according to the specific type of pancreatitis and the onset time of any accompanying pseudocyst.
10.Detection of Pathogens and Antimicrobial Resistance of Surgical Incision Infection

OBJECTIVE To investigate the resistance status of pathogens from surgical incision infection.METHODS The secretion of infected wounds was cultured to detect pathogens by routine methods from Jan 2001 to Dec 2006.The identifications and antimicrobial-susceptible tests of pathogens causing incision infection were determined by Full Automated Analyzer.All data were analyzed retrospectively.RESULTS A total of 246 pathogen strains were cultured from the secretion of surgical incision infection in 6 years,of which Gram-negative bacilli,Gram-positive cocci and fungi accounted for 57.7%,34.1% and 8.2%,respectively.The first place of isolates was Escherichia coli,followed by Staphylococcus aureus,Pseudomonas aeruginosa,Klebsiella pneumoniae,Acinetobacter baumannii,etc.74.5% of S.aureus isolates and 83.3% of S.epidermidis isolates were resistant to oxacillin.45.1% of E.coli of the isolates and 35.0% of isolates of K.pneumoniae were extended-spectrum ?-lactamases producing.Almost all of the detected resistant strains showed serious multiple resistance.Vancomycin and imipenem still had better activity for antimicrobial multiple resistant bacteria.CONCLUSIONS Surgical incision infection is serious.Infection surveillance should be taken to control surgical infection.Pathogens infected surgical incision are multi-resistant to antibiotics.
