1.STUDY OF WHOLE-MOUNT CYTOSKELETON OF HUMAN GLIOBLASTOMA BT_(325) CELLS BY CARBONPLATINUM COATING AND IMMUNOELECTRON MICROSCOPY

Cells of BT_(325), a human glioblastoma cell line, were treated with 0.5% Triton X-100 and immunostained with a monoclonal antibody to vimentin and antiserum to glial fibrillary acidic protein (GFAP), and followed by fluorescence labeled secondary antibody. After observation with fluorescence microscope the cells were dehydrated in increasing grades of ethanol and then critical point-dried. The carbon-platinum coating replicas of cytoskeleton were observed by transmission electron microscopy. The morphology of whole cytoskeleton as well as the arrangement of microtubules, microfilaments and intermediate filaments were clearly observed by this technique. It was observed that the diameter of intermediate filaments highly increased in the cells stained with the monoclonal antibody to vimentin. In the samples stained with anti-GFAP intermediate filaments were decorated in some cells but undecorated in other cells. The results indicates that all BT_(325) cells express vimentin, but only a fraction of the cells express GFAP. It was also observed that if the cells were fixed with formaldehyde before immunostaing with the monoclonal antibody to vimentin, the diameter of the intermediate filaments only slightly increased. However, the diameter of the intermediate filaments highly increaed in the cells stained firstly with the monoclonal antibody to vimentin and followed by formaldehyde fixation. This result is in accord with that of our previous immunofluorescence study. It indicates that formaldehyde fixation may "mask" certain epitopes on vimentin molecules.
2.Reform research on biomedical engineering experiment teaching

Aiming at the problems and deficiencies of traditional experiment teaching of biomedical engineering, this paper explores a series of reforms to the original teaching mode and states clearly the direction of experimental teaching reform and the specific content, outstanding interdisciplinary subject. In the teaching reform, in order to cultivate students' basic skills and the ability to analyze and to solve the problem, in order to improve the overall quality of students and to cultivate high-level innovative talents as the ultimate goal, this article discusses the main content of the reform of the biomedical engineering from the teaching methods, teaching material construction, teaching contents, lab management, diversified evaluation methods. The experimental teaching reform fully arouses the enthusiasm of the students' learning, and enhances the students' ability of practice and innovation, and improves the intersection between different disciplines, and has obtained the good teaching effect.
3.Application of continuous subglottic irrigation and aspiration in patients with artificial airway

Objective To explore the effect of continuous subglottic irrigation and aspiration in patients with artificial airway. Methods One hundred and twenty-eight patients with mechanical ventilation were randomly divided into observation group and control group with 64 cases in each group.The observation group was treated by continuous subglottic irrigation and aspiration,and the control group received traditional nursing.The two groups were observed and compared in terms of incidence of aspiration and ventilator-associated pneumonia.Result The incidences of ventilator associated pneumonia(VAP)and aspiration in the observation group were significantly lower than that in the control group(P<0.05).Conclusion Continuous subglottic irrigation and aspiration can effectively prevent aspiration and VAP among patients with artificial airway.
4.Influence of military exercise on some biochemical indexes in soldiers

Objective To investigate the influence of military exercise on some biochemical indexes in soldiers.Methods The level of serum soluble tumor necrosis factor receptor-1(sTNFR-1)and soluble intercellular adhesion molecule-1(sICAM-1)were determined with ELISA in peripheral blood of 133 servicemen who participated in the excercise before and after the military exercise.At the same time,the levels of serum gastrin and tetosterone were determined by radioimmunassay(RIA),the level of superoxide dismutase(SOD)and malonyl dialdehyde(MDA)were determined by biochemical methods.The correlation between the changes in these contents and psychological stress was studied.Results The levels of serum sTNFR-1,sICAM-1,testosterone and MDA of 133 servicemen increased after the military exercise compared with that of before the exercise(P0.05).Conclusion As a strong stress,military exercise can inhibit the immunological function and produce changes in emotional state,gastroenterological hormones,sex hormones etc.It is necessary to carry out mental and physiological training,so that the ability of servicemen to withstand the military stress can be improved.

6.Urodynamic effects of electroacupuncture on neurogenic bladder in patients with spinal cord injury

ObjectiveTo investigate the effects of electroacupuncture in Guanyuan(Ren4) and Zhongji(Ren3) on the functions of neurogenic bladder in patients with spinal cord injury(SCI) with the standard urodynamic measure.Methods15 patients with neurogenic bladder after SCI were included into the study.During urodynamic measure,they accepted electroacupuncture in Guanyuan(Ren4) and Zhongji(Ren3).The changes of the bladder storage volume(VH2O),bladder pressure(Pves),detrusor pressure(Pdet) and bladder compliance(BC) before and after electroacupuncture were recorded.ResultsThe patients' VH2O increased from(124.00±77.37) ml to(144.47±85.87) ml(P<0.01),Pves decreased from(65.33±23.90) cmH2O to(55.27±17.52) cmH2O(P<0.05),Pdet decreased from(52.87±18.68) cmH2O to(44.93±15.82) cmH2O(P<0.05)and BC increased from(2.35±2.05) ml/cmH2O to(3.48±3.04) ml/cmH2O(P<0.01).ConclusionElectroacupuncture is effective on neurogenic bladder recovery in patients with SCI.
7.Effect of daily life activity training on self-care abilities of children with cerebral palsy

ObjectiveTo investigate the effect of daily living activity training on the self-care ability of children with cerebral palsy.Methods Sixty children with cerebral palsy were equally divided into control group and observation group. The former group were managed with rehabilitative therapy and the latter were given ADL training consecutively for 6 months apart from rehabilitative therapy.The two groups were compared in terms of score on ADL after 6 months treatment.Results There was no difference in the score on activities of daily living between the two groups before intervention (P>0.05),but the score after treatment in the obsertation group was significantly higher than that of the control group (P<0.01).Conclusion The ADL training is effective for improving the self-care ability of children with cerebral palsy and promoting the rehabilitation of children.
8.Influence of health management on compliance and complication in postpartum reexamination

Objective To explore the effect of health management on compliance and complication in postpartum reexamination.Methods According to the admission time, 2,217 parturients were assigned into control group and another 2,216 into experiment group.The latter was managed with health management including establishment of health records, telephone follow-up and follow-up by visits and the former received routine nursing care.The two groups were compared in respect of maternal postpartum reexamination and health status.Result The compliance rate in the experimental group was higher than that in the control group and the incidence of complications was lower than that in the control group(P<0.05).Conclusion The implementation of health management can improve postpartum compliance in reexamination and reduce postpartum complications.
9.Effects of celecoxib combined with arsenic trioxide on bcr-abl protein and signal transduction in CML primary cells

Objective To explore the effects of celecoxib combined with arsenic trioxide (As2O3) on the mRNA expression,protein expression and protein tyrosine kinase (PTK) activity of bcr-abl fusion gene and its downstream signal transduction in chronic myeloid leukemia (CML) primary cells.Methods The cells were incubated with celecoxib (40 μmol/L) or As2O3 (2 μmol/L) alone and celecoxib (40 μmol/L) combined with As2O3 (2 μmol/L) for 36 hours.The changes of mRNA expression,p210 expression and PTK activity of bcr-abl fusion gene in each group were examined respectively by real time quantitative reverse transcriptase polymerase chain reaction (RQ-RT-PCR),Western blot and PTK activity analysis.The important proteins STAT1,STAT5 and their phosphorylatic proteins p-STAT1 and p-STAT5 and inhibitor of DNA binding 1 (ID1)a common downstream target of oncogenic tyrosine kinase were also analyzed by Western blot.Results The mRNA expressions in control group,celecoxib group,As203 group and celecoxib combined with As2O3 group were (5.97±0.53) %,(5.74±0.46) %,(5.94±0.57) % and (3.06±0.41) % respectively,and the statistical difference was found only between celecoxib combined with As2O3 group and control group (t =28.35,P =0.00).The similar statistic difference was only observed between the two groups from PTK activity tests (t =4.38,P =0.04).Western blot also showed that p210,STAT1,STAT5,p-STAT1,p-STAT5 and ID1 were more extraordinaryly downregulated by celecoxib combined with As2O3 than others treatments.Conclusion Celecoxib combined with As2O3 can downregulate mRNA,p210 expression,PTK activity of bcr-abl fusion gene and inhibit STAT and ID1 signal transduction pathways in a synergistic way.
10.Effects of celecoxib on bcr-abl fusion protein in chronic myeloid leukemia primary cells

Objective To explore the effects of celecoxib, a cyclooxygenase-2 (COX-2) specific inhibitor, on the mRNA expression protein expression and protein tyrosine kinase (PTK) activity of bcr-abl fusion gene in chronic myeloid leukemia (CML) primary cells. Methods The primary cells were incubated with various concentration of celecoxib (0, 10, 20, 40, 80, 160 μmol/L) for 36 hours, then the changes of mRNA expression, p210 expression and PTK activity of bcr-abl fusion gene in each group were examined respectively by real time quantitative reverse transcriptase polymerase chain reaction (RQ-RT-PCR), Western blotting and PTK activity analysis. Results The mRNA expression was downregulated with high concentration of celecoxib (80-160 μmol/L), and p210 expression was decreased gradually after increasing celecoxib. The PTK activity was inhibited in a concentration-independent way, and the statistic difference was observed only above 40μmol/L concentration of celecoxib. Conclusion Celecoxib can downregulate mRNA,and the protein expression of bcr-abl fusion gene; and inhibit the PTK activity by defferent extent.
