Objective To investigate effects of CO_2 pneumoperitoneum on cytokines and peritoneal macrophages in a rat model with implanted liver tumor.Methods A total of 32 Wistar rats with implanted liver tumor were randomly divided into 4 groups((n=8)): Control Group(anesthesia only),Laparotomy Group,Gasless Group(gasless laparoscopy),and Pneumoperitoneum Group(laparoscopy under CO_2 pneumoperitoneum).Serum samples were collected at the 2nd and 24th hours after the procedure respectively for the detection of levels of interleukin-1?(IL-1?) and interleukin-6(IL-6).Samples of peritoneal macrophages were collected and incubated for the detection of levels of tumor necrosis factor-?(TNF-?),a product of macrophages.Results At the 2nd and 24th hours after surgery,levels of serum IL-6 in the Laparotomy Group(57.92?2.06 pg/ml and 35.49?1.15 pg/ml) were significantly greater than those in the Pneumoperitoneum Group(14.64?0.34 pg/ml and 15.39?0.86 pg/ml),the Gasless Group(24.75?1.53 pg/ml and 17.10?0.97 pg/ml),and the Control Group(17.75?1.60 pg/ml and 14.55?0.25 pg/ml)(P
2.Effects of TSA on promoter methylation and expression of E-cadherin gene in hepatocellular carcinoma cell lines

Objective To study the effects of histone deacertylase inhibitor (TSA) on promoter methylation and expression of E-cadherin gene in a hepatocellular carcinoma cell line SMMC7721. Methods Hepatocellular carcinoma cell line SMMC-7721 was treated with TSA (300 nm/L), MTT method was used to investigate the growth inhibition ratio, TUNNL was conducted to measure the apoptosis ratio, methylation-specific PCR (MSP) was employed to detect changes in the CpG island methylation of E-cad promoter region, Western blot technique was used to detect the expression of E-cad gene and DNMT3b before and after TSA treatment, respectively. Results TSA decreases the SMMC-7721 cell viability and induces apoptosis, the growth inhibition ratio was 21.85% compared with control group. The apoptosis ratio of control group was (4.69±0.56)% ,the apoptosis ratio of TSA treatment group was (14.94±0.91)%. The apoptosis ratio of TSA treatment group was significantly higher than that of control group(P = 0.000). Before treated with TSA, the CpG island of E-cad promoter region was methylated, and the expression of E-cad was negative. TSA treatment induces demethylation of the CpG island in E-cad promoter region, causes the re-expression of E-cad. TSA reduces the expression of DNMT3b. Conclusions TSA decreases the SMMC-7721 cell viability and induces apoptosis, reverses the methylation status of E-cad promoter region, and resumes E-cad gene expression. TSA may induce demethylation through down-regulating the expression of DNMT3b.
3.Application effect of bandage corneal contact lens in ptosis during surgical treatment

Objective To study the application effect of bandage corneal contact lens in ptosis during surgical treatment.Methods A total of 62 patients(73 eyes) with primary congenital ptosis in the First People's Hospital of Xiantao from March 2013 to March 2016 were randomly divided into group A,group B and group C.All of the three groups were treated by shortening levator palpebrae superioris muscle.The A group[20 cases(24 eyes)] began to wear bandage corneal contact lens during the operation.The B group [22 cases (25 eyes)] began to wear bandage corneal contact lens at the end of surgery.And the C group [20 cases (24 eyes)] took conventional methods for covering operative eyes.Postoperative follow-up lasted for 3 months,the postoperative FL staining scores and ocular pain severity were compared among the three groups.Results Comparison of postoperative FL staining scores:there was no statistically significant difference between B group and C group at postoperative 1 day(q =0.537,P > 0.05),but the differences between B group and A group were statistically significant (q =10.876,11.298,all P < 0.05),the differences among the three groups were statistically significant at the 7th day after surgery (q =8.682,4.048,4.722,all P < 0.05).There was no statistically significant difference between B group and A group at postoperative 2 weeks (q =1.635,P > 0.05),but the differences between group A/B and group C were statistically significant (q =8.747,7.201,all P < 0.05).There were no statistically significant differences among the three groups at postoperative 3 months (q =1.607,0.186,1.438,all P > 0.05).Comparison of postoperative ocular pain severity:there were no statistically significant differences among the three groups at postoperative 1 day (q =0.986,0.586,0.410,all P > 0.05).At postoperative 7 days,there was no statistically significant difference between A group and B group (q =0.948,P > 0.05),but the differences between A/B group and C group were statistically significant (q =4.743,3.843,all P < 0.05).There were no statistically significant differences among the three groups at postoperative 1 month (q =3.181,0.599,2.654,all P > 0.05).Postoperative upper eyelid radian and double eyelid plasty in all of 62 cases(73 eyes) was satisfied and the width of palpebral fissure when look straight was normal and locating above superior margin of pupil,but there were 16 eyes with slightly hypophasis which were all naturally closed good after 1 to 2 months without surgical incision infection and corneal infection.Conclusion It is effective to protect the cornea since intraoperative application of bandage corneal contact lens,and it with high safety to wear corneal contact lens shortly after surgery,which can promote corneal epithelial wound healing,reducing incidence of exposure keratitis,at the same time,it can obviously relieve ocular pain in patients.
4.Comparative clinical study of percutaneous and peratrial device closure of secundum atrial septal defects under single transesophageal echocardiographic guidance

Objective This study is to evaluate the advantages between percutaneous and peratrial device closure of secundum atrial septal defects(ASD) under single transesophageal echocardiographic (TEE) guidance.Methods From December 2010 to December 2012,53 patients with the ASD of≤25 mm in diameter underwent percutaneous device closure under simple TEE guidance(the percutaneous group).The device was implanted through the femoral vascular access.Fifty patients with similar age and similar-sized ASD to the percutaneous group,were selected from 350 consecutive patients who underwent peratrial device closure of ASD and assigned to the peratrial group.The ASDs were occluded through a right minithoracotomy approach.The success rate,intracardiac manipulation time,procedural time,postoperative stay and the follow-up results were recorded.Results When the maximum diameter of ASD was < 20 mm,the success rate of both groups was 100%.When the ASD diameter was 20 mm but 25 mm,the success rate was 84% in the percutaneous group and 100% in the peratrial group.Three patients failed in the percutaneous group with the ASD diameter of 20 mm and the aortic rim of 3 mm.They were successfully converted to peratrial device closure.The average intrcardiac manipulation time was(20±7) minutes in the percutaneous group and (5 ± 6) minutes in the peratrial group(P < 0.05).The average procedure time was(24 ± 7) minutes in the percutaneous group and (39 ± 6) minutes in the peratrial group(P < 0.05).The postoperative hospital stay was (3.0 ± 0.8) days in the percutaneous group and(4.7 ± 1.5) days in the peratrial group(P < 0.05).Conclusion The percutaneous device closure of ASD under simple TEE guidance is feasible,safe,and efficacious in patients with the ASD diameter of ≤25 mm.It has the advantages of less trauma,less procedural time,shorter hospital stay and better cosmetic results.However,when the ASD diameter was 20 mm and the aortic rim was 3 mm,the peratrial approach may be a better choice.
5.Biological variation in 32 clinical laboratory routine tests

Objective To set quality goals of conventional biochemical tests through the research of biological variation of the 32 routine items in Chinese population to provide the basis for Chinese clinical and laboratory standards.Methods According to the experimental designs and computing methods from foreign counterparts,the results of biological variation,individual indexes and quality goals were calculated through the serum detection of 22 subjects from clinical laboratory of Zhongshan Hospital in Shanghai (male 12,female 10,ages varying from 20 to 40 years old,median age 30) in short-term (five blood draws within one day at 8:00,10:00,12:00,14:00 and 16:00) and long-term (one blood draw at 8:00 in 6 weeks consecutively) and serum controls (mixed from healthy people).Results (1) Based on the results of shortterm and long-term biological variation in 32 routine itens,the individual indexes and quality goals were obtained.(2)The influence of diet on the biological variation of part of the test items could be observed,especially free fatty acid (the mean value of post-meal was less than pre-meal about 30%),and then followed by high-sensitivity C-reactive protein (the mean value of post-meal was lower than pre-meal about 20%) and triglyceride (the mean value of post-meal was higher than pre-meal about 10%).(3)There were some differences between the quality goals we accessed and the the indicators from Europe and CLIA.Conclusions (1)The results of apolipoprotein E and free fatty acid in this study made up for the inadequate of the European biology database.(2) Only a small part of the 32 routine items were affected by dietary factors.(3) Most quality goals obtained from this study generally consisted with Europe biology quality goals,but a few items existed different.(4)It's more practical and effective to use the results of biological variatiou than CLIA standards for setting up quality goals.
6.Safety and efficacy of transcatheter arterial angiography and embolization for endoscopic refractory gastrointestinal bleeding

Objective To evaluate the diagnostic and therapeutic value and safety of transcatheter arterial angiography and embolization in patients with endoscopic refractory gastrointestinal bleeding.Methods Thirty-one cases of endoscopic refractory gastrointestinal bleeding were performed DSA and treated with transcatheter arterial angiography and embolization.The safty and efficacy was evaluated.Results Angiographic positive rate of bleeding was 80.65% (25/31);28 cases was treated with embolization.The success rate of first embolization was 75.00% (21/28),and the total success rate was 82.14 % (23/28) by the second embolization.Seven patients received surgical resection after interventional therapy,including 2 cases of jejunal stromal tumors and 5 cases of gastric malignant tumors.Four cases of gastric cancer patients underwent rebleeding within 30 days after interventional therapy,of which 2 died of heart or lung function failure due to basic diseases.Except for 1 patient of anastomotic bleeding after gastrointestinal anastomosis occurred anastomotic fistula after embolization,who recovery with the support treatment,no other cases occurred serious gastrointestinal ischemic necrosis.Conclusion Interventional diagnosis and treatment for gastrointestinal bleeding hemostasis is effective and safety,and also can achieve good results especially for malignant gastric tumor hemorrhage,which can be used for endoscopic refractory gastrointestinal bleeding patients.
7.Simultaneous Detection of 25 Kinds of Veterinary Antibiotics in Soil, Manure and Water Samples Using Liquid Chromatography_Tandem Mass Spectrometry

A liquid chromatography_tandem mass spectrometric method was established for the determination of 25 kinds of veterinary antibiotics in manure, soil and water by ultrasonic_solid phase extraction. Purification efficiency of SAX cartridges for soil and manure samples extract and the procedure that manure samples defatted with hexane was also verified. The results showed that the average recoveries of the 25 target antibiotics were 50. 0%-121. 9%, the relative standard deviations (RSD) were 1. 1%-14. 71% (n=9), and the limits of detection (LOD) ranged from 0. 0002-0. 0560 μg/kg for soil and manure and ranged from 0. 002-0. 28 ng/L for water; After adding SAX cartridges, the matrix effects of manure samples reduced to 55%-120%, and soil samples to 75%-160%; after defatting by hexane extraction, the matrix effects of manure sample reduced to 55%-120%. This method has been employed to detect the veterinary antibiotics in environmental samples of livestock farm.
8.Evaluation of value of indirect method in establishing reference intervals of CA72-4

Objective To establish the reference interval for CA 72-4 in indirect method.Methods All results for CA72-4 that were stored in our laboratory information system of Zhongshan hospital between Jan.2010 and Dec.2012 were included in this study.Outliers were identified and omitted using Stem-and-Leaf&Box Plots in SPSS statistical software.The treated data was divided into several groups according to gender and age.Nonparametric rank sum test was used to observe the difference between male and female participants and Spearman correlation analysis was used to examine the correlation between CA 72-4 and age.Nonparametric reference intervals for CA 72-4 were estimated statistically in two gender sub-groups.Results After 139 cases excluded, there were 1 548 cases of male (Median 1.7, 0.4 to 18.9) and 773 cases of female ( Median 1.8, 0.2 to 18.9 ).There was a significant difference in serum CA 72-4 between male and female participants.No significant difference was found in serum CA 72-4 among age sub-groups.Indirect reference values for CA 72-4 of male and female were respectively 0 to 8.9 U/ml and 0 to 11.6 U/ml.Conclusion Indirect method to establish biological reference interval is a relatively simple and less expensive method under the high rapid development of the hospital information network .It can be used in the periodical review and establishing the reference intervals where the direct method can not be used.
9.Study on physiological and germination characteristics of Tulipa edulis seed.

OBJECTIVECurrent study was conducted to investigate the seed physiological characteristics of Tulipa edulis and improve germination rate.
METHODAnatomical characteristics was observed. Seed water absorption curve was tested by soaking method. Dynamic of embryo development and germination rate as well as germination index under different conditions were recorded. And the biological test of cabbage seed was used for detecting the germination inhibitors.
RESULTThe embryo rate of newly matured seeds was about 10%, and there was no obstacle of water absorption on testa of T. edulis. The optimum method for embryo development was exposure to 300 mg x L(-1) gibberellin solution for 24 hours, and stratification at 25 degrees C for 70 days followed by stratification at 5 degrees C for 40 days. The germintion rate and germination index of dormancy-broken seeds under the dark environment at 10 degrees C and 15 degrees C were significantly higher than those under other conditions. Additionally, there were some germination inhibitory substances in dry seeds.
CONCLUSIONThe seed of T. edulis can be classified as having complex morphophysiological dormancy, and the morphological embryo dormancy played a leading role. Warm and cold stratification resulted in a fast dormancy breaking effect, and a high germination rate more than 90% could be obtained under the optimum conditions.

10.Effects of light intensity on growth and photosynthetic characteristics of Tulipa edulis.

OBJECTIVEPresent study was conducted to explore the growth and photosynthetic characteristics of Tulipa edulis under different light conditions (23%, 45%, 63%, 78%, 100% of full sunlight) and to determine the optimum light intensity for growth of T. edulis.
METHODThe leaf area and biomass indicators as well as reproductive characteristics were measured. The photosynthetic basic parameters and light response curve were determined by a LI-6400XT portable photosynthesis system, and the light response curve characteristic parameters was determined. Additionally, chlorophyll fluorescence parameters were determined by assorted fluorescence leaf chamber of LI-6400XT.
RESULTThe lowest biomass yield was observed in the 23% and 100% of full sunlight treatments while the highest value was found under the 78% of full sunlight conditions. With the reduction of light availability, the success rate of sexual reproduction, light saturation point (LSP) and light compensation point (LCP) reduced, while apparent quantum yield (AQY) increased. 23% and 45% of full sunlight treatments led to lower photosynthesis rate (Pn) and higher apparent quantum yield (AQY) in comparison with other treatents. The highest photosynthesis rate was observed in the 78% and 100% of full sunlight treatments. In addition, 78% of full sunlight treatments led to highest Fv/Fm, Fv'/Fm', PhiPS II, ETR, and qP.
CONCLUSIONT. edulis was able to adapt in a wide range of light intensity, and 78% of full sunlinght was the most suitable light condition for growth of T. edulis.
