1.Effects of islet B cells of rats on the differentiation of T-helper lymphocytes

Objective To investigate the effects of islet B cells of rat on the differentiation of T-helper lymphocytes and its potential mechanism.Methods Forty male SD rats were equally divided into five groups.Group-A:render diabetes by injection of streptozotocin (STZ);Group-B:OVA sensitized and then OVA challenged to render asthma;Group-C:render diabetes and then OVA sensitized,and challenged;Group-D:render diabetes,and then insulin subcutaneous injection,OVA sensitized,and challenged;Group-E:nondiabetes,nonsensitized,only OVA challenged.Bronchoalveolar lavage (BAL) was performed in rats from each group 24h after challenged and BAL fluid was used to detect IFN-? and IL-4.Blood glucose was estimated,total and differential leukocyte counts were carried out in blood samples and C-peptide concentration was tested in serum.Results The C-peptide concentration in serum was decreased significantly,while INF-? expression was higher,and IL-4 expression was markedly lower in BAL fluid in both group-A and group-C when compared with that of controls,respectively.Compared with group-C,IL-4 expression was increased and INF-? were significantly lower,respectively in BAL fluid of group-D after insulin subcutaneous injection.Conclusion Islet B cells of rat might modulate the differentiation of T-helper lymphocyte.
2.Proteomic identification of malignant transformation-related proteins in esophageal squamous cell carcinoma

Objective:To identify differentially expressed proteins related with malignant transformation of esophageal squamous cell carcinoma (ESCC) using proteomic analysis. Methods:Two-dimensional gel electrophoresis (2-DE) and matrix-assisted laser desorption/ionization timE-of flight mass spectrometry (MALDI-TOF-MS) in combination with protein database searching were used to determine and identify differentially expressed proteins in esophageal cancer cell lines (EC1, EC18, and EC109) and immortal cell line (NECA-E6E7-hTERT). Western blotting and immunocytochemistry were used to verify the differential expression of annexin 2 in esophageal cancer cell lines and immortal cell line (NECA-E6E7-hTERT). Real-time fluorogentic quantitative PCR(RFQ-PCR) was performed to analyze the expression level of annexin A2 mRNA.Results: A total of 15 differentially expressed proteins were identified with more than 5 folds difference. Among them three proteins were down-regulated and 12 proteins were up-regulated. Western blotting and immunocytochemical analysis verified the down-regulation of annexin A2 protein in ESCC cell lines. However, differential expression pattern of annexin A2 mRNA was not consistant with its protein expression in ESCC cell lines and immortal cell line (NECA-E6E7-hTERT). Conclusion:The findings provide important clues for identifying the candidate biomarkers for high-risk population screening and early diagnosis of ESCC. Post-translative regulation/modification contributes to the down-regulation of annexin A2 protein.
3. An analysis of the new "three tubes" method in the treatment of spontaneous esophageal rupture

To investigate the clinical efficacy, feasibility and safety of new "three tubes" method in the treatment of spontaneous esophageal rupture. A total of 22 patients with spontaneous esophageal rupture were retrospectively analyzed. Through the new "three tubes" method of treatment, patients achieved leak cured with reduced hospital stay, less medical expenses and early resumption of oral diet. The new "three tubes" method for spontaneous esophageal rupture has the advantages of easy handling, minimal invasion, few complication and exact curative effect.
4.Endoscopic trans-fistula drainage for gastroesophageal anastomotic fistula with para-fistula abscess after esophagectomy.

Objective: To evaluate the efficiency and safety of endoscopic trans-fistula drainage (ETFD) for gastroesophageal anastomotic fistula with para-fistula abscess after esophagectomy. Methods: Among 456 esophageal cancer patients receiving esophagectomy between February 2012 and February 2017 in Sir Run Run Shaw Hospital, 15 cases were diagnosed as gastroesophageal anastomotic fistula with para-fistula abscess after surgery. Seven cases received ETFD treatment (ETFD group), and 8 cases received conventional treatment (control group). Recovery of inflammatory markers and fistula, length of hospital stay after esophagectomy and total medical expenses were compared between ETFD group and control group. Results: All patients recovered in ETFD group. Time of white cell count returning to normal and decline of C-reactive protein, time of fistula healing and length of hospital stay after esophagectomy in ETFD group were significantly shorter than those of control group (all P<0.05). And medical expenses in ETFD group was also lower (P<0.05). Conclusion: ETFD is effective and safe for gastroesophageal anastomotic fistula with para-fistula abscess after esophagectomy.
