Objective To study the anticoagulation effects of Supplemented Salvia Beverage (SSB) on myocardial ischemia in rats. Methods Rat myocardial ischemia model was induced by occluding the left anterior descending coronary artery,the effects of SSB (8.24,4.12,2.06 g/kg) on the activated partial thromboplastin time (APTT),prothrombin time (PT),thrombin Time (TT),platelet adherence rate,platelet aggregation,length of thrombus and the dry and wet weight of thrombus,as well as amount of Fibrinogen (Fg) were observed. 2.Delayed Protection of Ischemic Preconditioning against Myocardial Ischemia-reperfusion Injury in Rabbits

Objective To explore the delayed protection of ischemic preconditioning against myocardial ischemia-reperfusion injury in rabbits. Methods Twenty four healthy rabbits were randomly into sham, ischemia-reperfusion(I/R) and ischemic preconditioning(IP) groups, each group containing 8 rabbits. After 24h IP, rabbits in IP group received 60 min coronary artery occlusion followed by 120 min reperfusion. Myocardial morphological changes were observed by optics microscope and electron microscope, infarction size and the area at risk were determined by triphenyltetrazolium chloride(TTC) and Evan's blue dye, and concentrations of adenosine and calcitonin gene-related peptide(CGRP) in serum were assayed. Results There were a few of cardial fibers ruptured, focus hemorrhage and a mass of inflammatory cell infiltration, and the concentrations of adenosine and CGRP in serum also decreased in rabbits of I/R group. 3.Determination of Matrine in Matrine Ethosome Thermosensitive Gels by HPLC

Objective To establish an HPLC method for determination of matrine in matrine ethosome thermosensitive gels. Methods Matrine was detected in matrine ethosome thermosensitive gels by using a YMC-Pack NH2 column(4.6 mm× 150 mm,5 μm),with the mobile phase containing acetonitrile-ethanol(9: 1).The flow rate was 0.8 mL·min-1 ,the column temperature( 25 ± 2) ℃ and the UV detection wavelength 215 nm. Results The peak area correlated with matrine concentration within the range of 1.00-100.00 μg·mL-1 linearly,and the average correlation coefficient was 0.999 2.The average recoveries rate of low,medium and high concentration of matrine were 98.16%,98.15%,98.00%,respectively,and the RSD were 0.30%,0. 69%,0. 71%,respectively. Conclusion The method is simple,sensitive and accurate. With the advantages of specificity,high precision,good stability and repeatability,the HPLC method can be used for the determination of matrine in matrine ethosome thermosensitive gels.
4.Content Comparison of Three Ingredients in Notoginseng Radix et Rhizoma and Its Formula Granules

Objective:To compare the contents of notoginsenoside R1 , ginsenoside Rg1 and ginsenoside Rb1 between Notoginseng Radix et Rhizoma and its formula granules. Methods:An HPLC method was used with a SunFire C18 column (250mm × 4. 6 mm, 5μm),the flow rate was 1. 0 ml·min-1, the detection wavelength was set at 203 nm, and the column temperature was at 30 ℃. The mobile phase was acetonitrile( A)-water( B) with gradient elution. An HPLC was used to determine the contents of the three ingredi-ents between Notoginseng Radix et Rhizoma and its formula granules, and compare the differences. Results: The total content of the three ingredients in Notoginseng Radix et Rhizoma and its formula granules was 9. 214% and 8. 646%, respectively. The total content of the three ingredients was equivalent and the daily amount of the major components in the commercial formula granules was equivalent with that in the decoction of Notoginseng Radix et Rhizoma. Conclusion:The production process of the original formula granules is re-liable, and the quality of formula granules of Notoginseng Radix et Rhizoma is stable.
5.Comparison of Contents of Forsythoside A and Forsythin in Fructus Forsythia and Its Dispensing Granule

Objective To compare the contents of forsythoside A and forsythin in Fructus Forsythia and its dispensing granule. Methods HPLC-gradient elution method was used with SunFire C18 column (4.6 mm×250 mm, 5μm), mobile phase A of acetonitrile and B of acetic acid, flow rate of 1.0 mL/min, detection wavelength of 277 nm, and column temperature at 30 ℃. HPLC was used to determine the contents of forsythoside A and forsythin in Fructus Forsythia and its dispensing granule, and compare the difference between the two contents. Results The content of forsythoside A in dispensing granule was less than that of raw material of Fructus Forsythia, and the concentration of the major components in the commercial Lianqiao Granule were not equivalent to that in the decoction of Fructus Forsythia. The content of forsythin in dispensing granule was equivalent with that of raw material of Fructus Forsythia. Conclusion The original formula granule production process needs to be improved, and the standardized criteria for the quality control and reasonable quality standard of granule should be established.
6.Effect of wogonin on spatial memory and its possible mechanisms in chronic cerebral ischemia in rats

Objective To investigate the effect of wogonin on ethology and its possible mechanisms in chronic cerebral ischemia in rats.Methods Rats were randomly divided into a sham operation group,a wogonin intervention group,and a phosphate buffered solution (PBS) control group.A rat model of chronic cerebral ischemia was induced by the two-vessel occlusion method.Six weeks after modeling,the rats in the wogonin intervention group and the PBS control group were intragastric administrated with wogonin (50 μmol/L,10 ml/kg,once a day) and PBS with equal volume for 14 days.Morris water maze test was used to evaluate the spatial learning and memory function.Laser confocal three-dimensional vascular imaging was used to detect the vascular proliferation of ischemic brain tissue.5-Bromo-2-deoxyuridine (BrdU)immunochemical staining was used to detect the cell proliferation in ischemic brain tissue.Transmission electron microscope was used to observe the morphological changes of neural cells in cerebral ischernic region.Results The Morris water maze (n =8) showed that the trains of escape latency from the second to the fifth day in the wogonin intervention group were 43.45 ± 8.64 s,37.12 ± 1.31 s,34.75 ± 5.36 s,and 24.36 ± 5.43 s,respectively.They were significantly shorter than 51.69 ± 5.32 s,43.65 ± 9.21 s,50.19 ± 10.31 s,and 53.65 ± 7.15 s in the PBS control group (all P < 0.05).The first quadrant swimming time of the wogonin intervention group was significantly longer than that of the PBS control group (26.16 ±3.29 s vs.14.38 ±2.16 s; P<0.01).Laser confocal three-dimensional vascular imaging (n=4) showed that the capillary inner diameter in cerebral ischemia region of the wogonin intervention group was reduced significantly compared to the PBS control group (3.02 ±0.21 μm vs.3.35 ±0.18 μm; P <0.05),vascular density was increased significantly (205.80 ± 12.70/0.002 mm3vs.158.42 ± 10.92/0.002 mm3; P<0.01),and total microvascular area was increased significantly (83 389 ± 4 026 μm2/0.002 mm3 vs.73 349 ±3 986 μm2/0.002 mm3; P<0.01).Immunohistochemical staining (n =6) showed that the number of BrdU positive cells in the ischemic brain tissue of the wogonin intervention group was increased significantly compared to the PBS control group (24.62 ±3.25/HPF vs.9.87 ±2.89/HPF; P<0.01).The observation of transmission electron microscope showed that the inflammatory edema in the intercellular spaces of the wogonin intervention group was significantly reduced compare to the PBS control group.Conclusions Wogonin can significantly improve the spatial learning and memory ability of chronic cerebral ischemia in rats,and its possible mechanisms may include the promotion of proliferation and angiogenesis in ischemic region and angiogenesis,and reduce inflammatory response.
7.Study on HPLC Fingerprints of Forsytiae Suspensa Fructus from Shiyan District

Objective:To establish the HPLC fingerprints of chemical constituents in Forsytiae suspensa Fructus from Shiyan dis-trict to provide scientific evidence for the quality control of Forsytiae suspensa Fructus. Methods: HPLC was performed on a Waters SunFire C18 (250 mm × 4. 6 mm, 5 μm) column and a Diamonsil C18 guard column, and the mobile phase consisted of acetonitrile-0. 4% acetic acid solution with gradient elution. The flow rate was 1 ml·min-1 , the detection wavelength was 277 nm, and the col-umn temperature was 30℃. Results:There were 14 common peaks in the HPLC fingerprints of chemical constituents in 12 batches of Forsytiae suspensa Fructus from Shiyan. The 14 characteristic peaks of Forsytiae suspensa Fructus from different habitats were under clustering analysis, and the similarity showed little difference. The contents of forsythoside A and forsythin exhibited significant differ-ence in the samples. Conclusion:The HPLC fingerprint method is simple, rapid and feasible in the quality control of Forsytiae suspen-sa Fructus.

According to ZHANG Zhong-jing, the main reason for the stroke was the exogenous wind, and the most important pathogenesis was the deficiency of qi and exogenous pathogenic factor. Three methods are stressed to cure the stroke: the equal treatment focused on the liver and the spleen, the use of tranquilizing medicine to stop the wind, and the use of strengthening body resistance to get rid of the exogenous pathogenic factors. While YE Tian-shi considered that the main reason for stroke was the deficiency of liver-yin and kidney-yin. The chief pathogenesis in stroke was the yang transforms to the interior wind. There are three therapeutic features in the treatment: the equal treatment focused on the liver and the spleen, the synchronous treatment of the liver and stomach, and select the medicine according to the four seasons climate. ZHANG Zhong-jing's theory of exogenous wind and YE Tian-shi's theory of yang transforms to the interior wind not only reflect the different angles in treatment for stroke, but also reflect the development of pathogenesis of stroke in the history.
10.Effects of Jiawei Danshen Decoction on Endothelial Cell Function and Ultrastructure in Atherosclerosis Model Mice

Objective To investigate the effects of Jiawei Danshen Decoction on endothelial cell ultrastructure, serum NO and ET in apolipoprotein E-knockout atherosclerosis (AS) mice;To discuss its mechanism of anti-atherosclerosis.Methods Totally 24 7-8 weeks old ApoE -/- mice were fed with a high fat diet for 12 weeks until the a mature atherosclerotic plaque was formed. They were randomly divided into four groups:model group, control group,Jiawei Danshen Decoction of low-dose group and high-dose group, 6 mice in each group. Another 6 normal C57BL/6J mice with the same strain were set as the blank group, and fed with general diet. After medicine intervention by gavage for 8 weeks, blood was extracted from orbital venous to measure serum NO and ET;aortic endothelial cell ultrastructure changes were observed by transmission electron microscope.Results Compared with the blank group, the level of NO obviously decreased and ET significantly increased in the model group (P<0.01). Compared with model group, medicine intervention increased NO and reduce ET level (P<0.05). Electron microscopy results showed that the model group showed fatty streaks stage performance;endothelial damage in each administration group was improved compared with model group.Conclusion Jiawei Danshen Decoction can improve the endothelial cell ultrastructure and functions and protect endothelial cells, with a purpose to delay the AS process.
