Current reseach of molecular targeted therapies in squamous cell carcinoma of head and neck(SCCHN) is particularly active.As epidermal growth factor receptor(EGFR) signaling pathway and angiogenesis play a key role in the growth of SCCHN,EGFR with its downstream effectors and molecular factors implicated in the angiogenesis process,such as vascular endothelial growth factor and its receptors,represent the main targets of new therapeutic agents now.
2.Changes of IL-10 mRNA expression and activator protein-1 activity in lung of rats with SIRS-acute lung injury induced by lipopolysaccharides

AIM: To establish the model of systemic inflammatory response syndrome (SIRS)/acute lung injury (ALI) by LPS administration in rats and to measure the content of interleukin-10 massage RNA(IL-10 mRNA) and activator protein-1 (AP-1) activity in order to investigate the anti-inflammatory mechanism.METHODS: Wistar rats were injured with increased dose of LPS to set up the SIRS/ALI model. Reverse transcriptase-polymerase chain reaction(RT-PCR) and electrophoretic mobility shift assays (EMSAs) were used to measure IL-10 mRNA content and the AP-1 binding activity in rat lung, respectively.RESULTS: ①LPS could be applied to simulate SIRS-ALI in rats.②Acute respiratory distress syndrome (ARDS) could be induced under condition of LPS≥6 mg/kg, which was similar to that of the excessive expression of SIRS. ③ LPS may cause the increase content of IL-10 mRNA and AP-1 activity in lung in rat. ④Content of IL-10 mRNA and AP-1 activity were increased significantly under LPS≥6 mg/kg. CONCLUSIONS: ①LPS≥6 mg/kg might cause the SIRS/acute lung injury in rats.②The rats with excessive SIRS/acute lung injury had an obvious increase of transcription of IL-10 gene and upregulated AP-1. ③ Intensified anti-inflammatory mechanism plays a pathological role in SIRS/acute lung injury.

Objective To explore how to improve the immunogenicity of short epitope peptides of triggering melanoma MART 1 specific CD8 +T cell responses Methods Therapeutic peptides based on the immunodominant MART 127 35, HIV Tat49 57CCP sequence and a tetanus toxoid universal Th epitope were designed and synthesized The immunological functions were studied in PBMCs from HLA A2 + melanoma patients Results The results demonstrated that the peptides could trigger vigorous MART 1 specific CD8 + CTL activities in vitro The function of peptide containing MART 127 35 and tetanus universal Th epitope was more vigorous than that of MART 127 35 peptide, and the immunogenicty of the peptides with HIV Tat49 57CCP sequence, MART 127 35 and tetanus universal Th epitope was the most vigorous Conclusion Linkage of HIV Tat49 57CCP sequence and a tetanus universal Th epitope could dramatically improve the immunogenictiy of the MART 127 35 epitope peptide
5.RhoA gene expression and clinicopathology parameters in HCC patients

Objective To investigate the expression of RhoA gene in human hepatocellular carcinoma (HCC) and to discuss its significance during hepatocellular carcinogenesis. Methods Intratumor RhoA expression level was determined and compared with that in adjacent nontumorous hepatic tissues using quantitative real time reverse transcription polymerase chain reaction and Western blot in 64 HCC patients. Results The mRNA levels of RhoA were significantly higher in tumor tissues than that in the unaffected portions (t =3.445 ,P =0.0006). The expression of RhoA mRNA in the primary lesion was higher in patients with venous invasion (t = 2.667, P = 0.009), microscopic satellite lesions (t=2.172,P =0.038), and advanced pTNM stage (stage Ⅱ/Ⅳ; t=2.551,P=0.013) than in those without. There was a significant difference between high RhoA protein levels in the tumor tissues and noncancerous liver tissues in HCC patients (t = 3.532, P = 0.0002), and there was a significant association between high tumor RhoA protein levels and the presence of venous invasion (t = 2.087, P = 0.042), microscopic satellite lesions (t = 2.254, P = 0.031), and advanced pTNM stage (t = 2.812, P = 0.007). Conclusions There is a significant correlation between RhoA expression, tumor stage, and intrahepatic metastasis. The expression of RhoA could be used as a good tumor marker for invasive and advanced carcinoma as well as a prognosis predictor.
6.Preparation and Certification of Reference Materials for 3-Amino-2-oxazolidinone Residue in Eel Muscle Lyophilisates

A methodology for preparing and certifying the reference material of 3-amino-2-oxazolidinone(AOZ) in eel muscle lyophilisates was presented. Furazolidone was accessed to eel by dipping fish in pond with furazolidone solution at a dosage of ca 0.16 mg/L. With the metabolism of furazolidone in eel, the muscles contain a certain concentration of AOZ as furazolidone metabolite was obtained. Lyophilization of the muscles was performed in one batch and 400 bags of samples were obtained by the procedure of homogenation, cryodesiccation and irradiation. The homogeneity and stability of the sample was examined. The value of the chemical constituent of the sample was certified through the collaborative analysis program participated by 11 laboratories using isotope dilution liquid chromatography-tandem mass spectrometry, and the uncertainty assessment was performed. The reference materials have been approved as certified reference materials by AQSIQ, China (State General Administration of the People′s Republic of China for Quality Supervision and Inspection and Quarantine) in 2009 after one year of trial period. The serial numbers is GBW(E)100180.
7.Application of laparoscopic distal pancreatectomy for treatment of insulinoma

Objective To evaluate the safety and efficacy of laparoscopic distal pancreatectomy in treatment of insulinoma.Methods Clinical data of 8 cases of insulinoma treated by laparoscopic distal pancreatectomy from Apr.2015 to Apr.2017 were retrospectively reviewed.Results Locations of the insulinoma in distal pancreas were all identified preoperatively by enhanced CT,MRI or somatostatin receptor scintigraphy (SRS).Laparoscopic distal pancreatectomy was applied to 8 cases,including combined splenectomy to 1 case.The operation time,bleeding volume,and postoperative hospital stay was (159±44) min,(125±119) ml and (5.5±1.4) days,respectively.Grade B fistula happened to one patient after surgery.The level of postoperative blood glucoses was normal in all cases.Conclusion Laparoscopic distal panreatectomy is safe,effective,and less invasive in treating insulinoma,with quick recovery and high efficacy in spleen preservation.
8.Application of Monse′s solution in bleeding in endoscopic sclerotherapy for esophageal varices

ObjectiveTo investigate the effects of Monsel′s solution on bleeding in endoscopic sclerotherapy for esophageal varices. MethodsOne hundred and sixteen patients with cirrhosis and esophageal variceal bleeding who were admitted to our hospital from February 2013 to February 2015 were enrolled as subjects. All patients received endoscopic sclerotherapy. Forty-six patients had hemorrhage after removal of needles, and they were treated with topical spraying of 15% Monsel′s solution. The instant and long-term hemostatic effects were evaluated. ResultsAfter spraying of 15% Monsel′s solution, the immediate formation of coagulum and no bleeding were found in the bleeding spots of 46 patients undergoing bleeding in endoscopic sclerotherapy. At one week after surgery, the blood pressure, pulse, fecal occult blood, and routine blood indices were evaluated, and no signs of active bleeding were found. ConclusionThe response rate of topical spraying of Monsel′s solution is 100%. Moreover, with a convenient preparation, few adverse reactions, and low economic burden on patients, the simple method is the best choice in the treatment of bleeding in sclerotherapy.
9.Diagnosis and treatment of thyroglossal duct cyst in infants

Objective To explore the imageology features and curative effect of operation for thyroglossal duct cyst in infants.Methods CT and B-mode ultrasound features of 23 infant cases diagnosed as thyroglossal duct cyst were retrospectively analysed.The patients underwent classic Sistrunk operation.And ampliate Sistrunk operation was performed for those with postoperative recurrence.Results CT showed a cystic mass in the anterior triangle of neck,relevant to the hyoid bone and thyroid.B-mode ultrasound showed hypoechoic and heterogeneous image companied with posterior echo enhancement.None cyst had a solid component.Twenty-three cases got satisfied curative effect after repeat operation.There were no complications such as trachyphonia,bucking and dyspnea.Following up more than 2 years,no recurrence case occurred.Conclusions CT and B-mode ultrasound scan can help to make accurate diagnosis of the thyroglossal duct cyst,based on the special location and the typical manifestations.Thyroglossal cyst had high recurrence rate after operation.Sistrunk operation is the basic procedure for the complete removal of the thyroglossal cyst in infants.

Objective This study was to evaluate the usefulness of the model of end-stage liver disease (MELD) score in comparison with the Child-Pugh classification to predict postoperative mortality and short-term survival in liver transplant patients.
