3.Influences of simulated microgravity on human endothelial cells and platelets

The change of gravity causes a series of physiological dysfunctions,such as redistribution of blood,destruction of skeleton,reduction of hemopoietic tissue and so on.Since the success of mission to the moon and construction of space station,the study on change of human physiological functions under microgravity has been underway.With the machine of microgravity developing maturely,associated researches have made big steps forward.However,intensive study on the changes of human hemostasis and thrombosis under the microgravity are still needed,especially in the area of high incidences of bleeding and thrombotic diseases.The result will help to prevent hemostasis and thrombosis on astronaut and promote the development of our national aerospace industry.
4.Rehabilitative therapy for the elderly chronic somatic disease complicated with depression

BACKGROUND: There is a complicated relation between somatic disease and mental disorder. Somatic disease may induce functional disorder of brain, and generate psychiatric symptoms due to the change of psychological and social environment. The proportion of depression in elder people based on the chronic somatic disease is much higher, and thus the fatality of them increases.OBJECTIVE: To investigate the influence of single antidepressant, and therapeutic alliance of psychological and drug treatment on the rehabilitation of patients with elderly chronic somatic disease complicated with depression. DESIGN: Randomized and controlled grouping experiment.SETTING: Department of Senile Disease of Tongji Hospital Affiliated to Huazhong University of Science and Technology.PARTICIPANTS: Totally 102 patients were hospitalized in the Department of Senile Disease of Tongji Hospital Affiliated to Huazhong University of Science and Technology from March 2000 to January 2003, and all the patients had sleep disorder to a different extent, and suffered from symptoms of heart and alimentary tract which could not be explained as organic diseases, and chronic pain or systemic symptom. Their score of Zung scale was > 40 points with average of (48.83±4.15)points. The patients were randomly divided into the drug group (n=46) and the combined treatment group (n=56). Each patient had more than two somatic symptoms in the two groups and more than 81.0% of them had three somatic symptoms. METHODS: According to the different primary diseases in patients of the two groups, regular treatment was given routinely. Oral thymoleptics fluoxetine of 20 mg/d 5-hydroxyltryptamine reuptake restrainer with 20 mg/d was administered, the dose of which could be added to 40 mg/d according to the pathogenetic conditions. At the beginning of the administration of oral antidepressant drugs, the patients in the combined treatment group were The focal point of depression and fear was identified, and the comfort and edge related to the disease was supplied, and the pathogenetic condition and the prognosis were told to the patients objectively. The family and society were encouraged to learn the rehabilitative skill and improve their possible, and the family and society were encouraged to give the patients adequate psychological support and life support. The patient and the family were assisted to establish favorable communication. The feeling of loneliness and isolation was reduced, and the sense of safety was increased. The depression status and the quality of life of the patients in the two groups were evaluated before the treatment and at 2 and 8 weeks after treatment with the Zung scale and the Spitzer quality of life index score (QLI), including 5 aspects: Activity, daily life, health, support and perspective. The highest score was 10 points. The lowest was 0, and the higher the score, the better the quality of life is.MAIN OUTCOME MEASURES: The results of the Zung scale and the QLI score evaluated before the treatment and at 2 and 8 weeks after the treatment.RESULTS: The treatment of the patients in the two groups was all finZung scale in the drug group and the combined treatment group at 2 weeks after the treatment indicated that there was significant difference in comparison with those before the treatment (41.12±2.60,48.83±4.15,t =10.67,P < 0.05); (40.88±3.43,49.03±4.21 ,t=11.24,P < 0.05). There was not significant difference in group comparison. The total points of Zung scale at 8 weeks were significantly decreased (36.11 ±2.87 ), (30.71±3.62),and there was significant difference in comparison with that before the treatment. The combined treatment group was better than the drug group in difference of QLI score at 2 weeks between the drug group and combined treatment group before and after the treatment (5.47 ± 1.33,5.32 ± 1.04,5.55±1.18,5.27±1.35). The QLI score of drug group and combined treat ment group at 8 weeks after treatment began to rise 7.21±0.13 and 8.83 ±0.32 ,respectively, and there was significant difference in comparison withthose before the treatment, and there was also significant difference in group comparison.CONCLUSION: The positive and effective combined therapy of anti-depression significantly improved the somatic symptoms of patients, no complications appeared, and most patients presented optimistic attitude to the prognosis of the primary disease. To identify the depression of elder people in earlier period is very important to promote rehabilitation of the primary disease.
5.Progression of triple-negative breast cancer

Studies about epidemiology,molecular characteristics,chemotherapy and targeted-therapy are ongoing,In particular,preliminary results of stage II or III clinical trials indicate new chemotherapy or combination targeted therapy may have a good efficacy.
6.Effect of microgravity on biological characteristics of vascular endothelial cell

Under the condition of microgravity, human body will experience a series of cardiovascular deconditioning. The vascular endothelial cells are the most important component of the vessel wall, so the changes of biological characteristics of vascular endothelial cells are directly related to cardiovascular deconditioning. Therefore, it is important to prevent and treat the cardiovascular deconditioning in astronauts by studying the effect of microgravity on biological characteristics of vascular endothelial cells.
7.Recombinant Lactobacilli for Ethanol Production

Recombinant plasmids pHY-PA, pBBR-PA were constructed in which genes pdc and adhB were placed under the control of tac promoter, respectively, and had successfully expressed in Escherichia coli. Then these recombinant plasmids were electroporated into Lactobacillus strains for ethanol production. Preliminary ethanol fermentation using these Lactobacillus strains and their recombinants was carried out using 42℃ as fermentation temperature. The results indicated that introducing pdc and adhB, ethanologenic pathway was successfully constructed in L.plantanum CICIM B0080. 0.4% (V/V) ethanol was detected at the end of fermentation with 6.7% glucose, and that is 67-fold as the wild-type B0080. Two-fold of ethanol production was detected in L.amylovorus B0112 (pHY-PA) and L.acidophilus B0068 (pBBR-PA). Introducing both pdc and adhB, and meanwhile knock-outing the lactate dehydrogrnase gene may better convert carbon flux to ethanologenic direction.
8.Effect of visual function on posterior capsule opacification before and after Nd:YAG laser posterior capsulotomy

Objective To explore the influence of stereoscopic vision on the development of posterior capsule opacification (PCO) after intraocular lens implantation. The efficacy of Nd: YAG laser was studied. Design Case-controlled study. Participants 57 consecutive patients with POC(59 eyes with phacoemulsification and IOL implantation). Methods A prospective observational study was conducted comprising 57 patients (59 eyes) who had postoperative PCO. Neodymium: YAG (Nd: YAG) laser posterior capsulatomy was performed on all patients following a standardizes procedure. Preoperative and postoperative evaluation included LogMAR visual acuity, best-corrected visual acuity (BCVA), contrast sensitivity and stereoscopic vision. Main Outcome Measures LogMAR visual acuity, BCVA, contrast sensitivity and stereoscopic vision. Results The average LogMAR eyesight median of sick eye before laser treatment was (0.22?0.31) and the contrast sensitive degree of each frequency area was relatively lower than the opposite eye; After the treatment, there was evident improvement of the sick eye, and the LogMAR eyesight median of that was 0.00; The ratio of reaching the central spatial eyesight and the average sharpness degree both improve much than ahead of treatment. Those whose spatial eyesight sharpness don't get normal after the low best remedy eyesight. Irregular dipodic and previous eye disease. Conclusions LogMAR visual acuity, BCVA, contrast sensitivity and stereoscopic vision were damaged markedly with PCO. Our results indicate that Nd: YAG capsulotomy may affect the reconstruction of stereoscopic vision. Nd:YAG laser posterior capsulotomy is a valuable and clinically relevant method for clinical treatment of PCO.
9.SUBACUTE ORAL TOXICITY STUDIES OF BCDMH EFFERVESCENT TABLETS FOR HOS-PITAL SEWAGE

Objective To study subacute oral toxicity of a disinfectant for hospital sewage which contained 1-bromo -3 -chloro -5, 5 -dimethylhydantoin (BCDMH) in order to know about its toxicological safety .Methods Animal test and biochemical examination methods were adopted to evaluate its subacute toxicity .Results During the subacute toxicity test, none of the rats died.At the end of the experiment , urine routine and hematological exami -nation were in the normal ranges .There were no difference compared with the control (p >0.05).But serum alanine aminotransferase in both sexes as well as creatinine in male SD rats of the high dose group were increased obviously compared with the control (p <0.05).Mild steatosis and spotty necrosis of liver cells were found in the high dose group during the pathological examination .Conclusion The BCDMH effervescent tablets'no -observed -adverse -effect -level (NOAEL) is 84 mg? kg-1 for male SD rats, and 98 mg? kg-1 for female SD rats.
10.Comparison of Blinding Sample Size Adjustment with Unblinding One in Clinical Trials Through Simulations

Objective To determine the appropriate methods to sample size adjustment for adaptive design through simulations by comparing sample size adjustment of blinding internal pilot design with that of unwinding one in clinical trials. Methods Compare type I error and the power of blinding and unblinding internal pilot design through Monte Carlo simulation. Results Either type I error rates or the powers are not substantially distinct in two lands of settings. Conclusion Blinding sample size adjustment is more preferable.
