1.Expression of Inducible Nitric Oxide Synthase and NF-κ Bp65 in Patients with Psoriasis Vulgaris and Their Clinical Significance

Objective To investigate the internal relationship between the expression of inducible nitric oxide synthase (iNOS) and the activation of NF-κ Bp65 in the skin of patients with psoriasis vulgaris(PV) and their roles in the pathogenesis of PV.Methods The expression and distribution of iNOS and NF-κ Bp65 were studied by in situ hybridization and immunohistochemistry in 20 patients with PV and 12 healthy controls.The severity of PV was assessed by Psoriasis and Severity Index(PASI) score.Results ① The expression of both iNOS mRNA and protein was significantly higher in lesional epidermis than those in non-lesional epidermis and epidermis of the controls (P<0.01).Weak exprssion of iNOS mRNA was found in basal cell layer in a small number of healthy controls, however, strong expression was observed in entire basal cell layer and focal parts of prickle cell layer in all PV epidermis.② There was significantly positive correlation between iNOS and NF-κ Bp65 protein expression in lesional epidermis (P< 0.01).③ There was significantly positive correlation between the level of expression of iNOS mRNA in lesional epidermis and PASI score of patients (P< 0.01).Conclusions The expression of iNOS and NF-κ Bp65 is markedly increased in lesional epidermis of patient with PV.The activation of NF-κ B probably takes part in the pathogenesis of PV.High-level expression of iNOS may be due to the activation of NF-κ B.

3.Detection of Nitric Oxide, Glutathione and Superoxide Dismutase in Ovarian Neoplasm

Objective: Our aim was to study the relationship between nitric oxide and ovarian neoplasm. The concentrationg of nitrite /nitrate (NO-2/NO-3),glutathione(GSH), and the activity of superoxide dismutase(SOD) were detected in ovarian tissues.Methods:A modified copper-coated cadium reduction method was used to detect NO-2/NO-3. Dithio-bis-nitrobenzoic acid (DTNB) method was used to detect the GSH concentration. The activity of SOD was measured by an ultramicroanalytic and rapid assay. All the data were obtained from 21 patients with ovarian neoplasm(10 with benign ovarian neoplasms,and 11 with ovarian carcinomas). This paper is concerned with the analysis of matched case-control studies. The carcinoma tissues and normal ovarian tissues were obtained from the same patient. We performed Paired-Samples t test to determine whether the NO-2/NO-3,GSH and the activity of SOD in carcinomas and normal ovarian tissues have significantly difference. Results:No significant difference for the concentration of NO-2/NO-3,GSH and activity of SOD was shown in the normal ovarian tissues and neoplasm tissues of benign neoplasm . The concentrations of GSH and activity of SOD in normal ovarian tissues were significantly higher than ovarian carcinomas(P<0.05). No significant difference of concentration of NO-2/NO-3 was shown in normal ovary and ovarian carcinomas. In the normal tissues of all patients, NO-2/NO-3 was positively correlated with GSH and SOD. Glutathione was positively correlated with SOD. In the ovarian neoplasms, there was no significant correlation between NO-2/NO-3 and GSH, NO-2/NO-3 and SOD,GSH and SOD.Conclusion: Nitric oxide and the other matters which attend the course of its metastasis may closely related to ovary neoplasms.
4.Inhibition of transdifferentiation of human renal tubular epithelial cells by valsartan

Objective To investigate the possible role of valsartan(Val) in transdifferentiation of human renal tubular epithelial cell line(HKC ). Methods HKC cells were divided into four groups: (1) serum-free (negative control); (2) MCP-1 + AAI-treated (positive control); (3) Val-treated alone; (4) Val-inhibition(treated with MCP-1 + AAI + Val). Then the expression of vimentin, ?-smooth muscle actin (?-SMA ) of HKC cells were assessed by indirect enzyme immunohistochemistry (IEI), and the percentage of ?-SMA(+ ) HKC cells was assessed by flow cytometry. Results No difference in expressions of vimentin and ?-SMA by IEI and the percentage of ?-SMA(+ ) HKC cells by cytometry were found between serum-free control HKC cells and Val treated ones. The expressions of vimentin and ?-SMA in positive controls were markedly stronger than negative controls; while these expressions in Val + MCP-1 + AAI-treated HKC cells were less strong than those in positive controls. Objective To investigate behavioral disposition of nurses in emergency room and analyze influential factors.Methods A total of 200 nurses from the emergency room participated in the survey using demographic questionnaire and the nursing behavior questionnaire.Results The total average score of the nursing behavior was(3.13±0.54),and the influential factors included the level of hospital,the time span for the job and the continued education on the infectious diseases(all P<0.01).Conclusions The nursing behaviors was at the medium level.The nursing administrators should provide the nurses with personal protection apparatus,strengthen education on infectious diseases and use incentive mechanism so as to improve the ER nurses' nursing behavior.

Objective To observe secretive ability of three C-C chemokines, macrophage inflammatory protein-1?(MIP-1?),MIP-1? and regulated-upon activation.normal T-cell expressed and secretory factor(RANTES)by lympocytes and moncytes of cord blood from chinese necnates and peripheral blood from chinese adults.Method Using Ficoll density-gradient centrifugation and gelatin-coated flask,the Iympocytes and monocytes were isolated from cord blood samples of seventeen Chinese neonates and peripheral blood samples of twenty Chinese adults.Purified lymphocytes and monocytes were incubated with PMA and LPS,respectively.Supernants were tested by ELISA for concentrations of MIP-1?,MIP-1? and RANTES.Results Concentrations of MIP-1?, MIP-1? and RANTES in cord blood lymphocyte and monocyte were (3920?730)pg/ml, (4910?590)pg/ml,(1470?410)pg/ml,(3240?980)pg/ml,(1960?1300)pg/ml and (240?120)pg/ml,respectively,and in adults peripheral blood were (6560?840)pg/ml,(5810?1150)pg/ml,(2250?570)pg/ml,(3010?1350) pg/ml,(2280?870)pg/ml,(690?430)pg/ml,respectively.Cord blood lymphocyte and monocyte showed diminished ability to secrete RANTES than adult peripheral blood.Conclusions The diminished RANTES secretion of cord blood lymphocyte and monocyte may be releted to the pathogenesis of Chinese neonatal HIV infection,and indicated some immunotherapies are feasible for preventing neonate from HIV mother-to-child transmission.
7.The role of anti-annexin A2 antibodies in antiphospholipid syndrome

Objective This study has explored the role of antibody against annexin A2 in patients with antiphospholipid syndrome (APS) and systemic lupus erythematosus (SLE). Methods Using purified recombinant annexin A2, IgG anti-annexin A2 antibody was measured by ELISA in 101 APS patients, 41 SLE patients with thrombosis, 124 SLE patients without thrombosis and 120 healthy controls. Results The positive rate of IgG anti-annexin A2 antibody in APS patients and SLE patients with thrombosis was 21.8%, 26.8%, respectively, they were all significantly higher than in SLE patients without thrombosis (6.5%). IgG anti-annexin A2 antibody was associated with thrombosis and/or pregnancy morbidity (P<0.01). Conclusion Anti-annexin A2 antibody is associated with thrombosis and/or pregnancy mnrbidity. It suggests that anti-annexin A2 antibody may be helpful in identifying in some potential AIRS.

10.Recognition of Specific Antigens by Specific IgG and IgE During Anaphylactic Shock Induced by Echinococcus granulosus in Sheep

Objective To determine the specific recognition of Echinococcus granulosus (E.g.) cyst fluid crude antigen (EgCF) and antigen B (EgB) by serum specific IgG and IgE using Western blotting during anaphylactic shock induced by E. g. in sheep, and to investigate the importance of defined characteristics and molecular weight of the specific antigens. Methods EgCF was obtained through local slaughterhouses in Urumqi from the cysts of infected sheep liver. Western blotting was used to analyze total specific IgG and IgE antibodies in serum from sheep infected with E.g. using either crude antigen of E. g. and EgB, and to understand specific recognition of hydatid cyst antigens by serum total IgG and specific IgE. Results SDS-PAGE and Western blotting showed that there were differences between EgB and EgCF in electrophoresis pattern. EgB was recognized by IgG from sera of infected sheep in a series of bands with molecular weight ranging from 31, 43 to 66.2 kDa. No binding of IgG against EgCF was observed in any serum from the infected sheep during anaphylactic shock. In contrast, specific IgE antibodies in E. g. infected sheep obviously recognized the single 43 kDa subunit of EgCF, but no binding of specific IgE against EgB was observed in sera of the infected sheep. Conclusion EgCF is consisted of antigenic components in which there is a specific antigen against IgE with a molecular weight of over 43 kDa. This component may lead to an anaphylactic shock induced by E. g. . EgB is a specific antigen against the total IgG but not to the specific IgE.
