1. Effects of geldanamycin on cell proliferation and apoptosis of human ovarian cancer cell line SKOV3 and their mechanism

Objective: To investigate the cytotoxic effects of geldanamycin on human ovarian cancer SKOV3 cells, and to explore the possible mechanisms of apoptosis and cell proliferation inhibition of SKOV3 cells induced by geldanamycin. Methods: MTT method was performed to evaluate the inhibitory effects of geldanamycin alone or in combination with cisplatin on proliferation of SKOV3 cells. Cell cycle and apoptosis rate of SKOV3 cells treated with geldanamycin were detected by flow cytometry. The expressions of Akt and Raf-1 proteins were examined by immunocytochemistry and flow cytometry. Results: The proliferation of SKOV3 cells was inhibited by different concentrations of geldanamycin in a time- and dose-dependent manners (P<0.05). Different concentrations of geldanamycin strengthened the inhibitory effect of cisplatin on proliferation of SKOV3 cells, and this inhibitory effect was strengthened with the increased concentration of geldanamycin within a certain concentration range. The apoptosis rate of SKOV3 cells was raised up gradually after treatment with 0-2000 nmol/L geldanamycin for 48 h, and the number of cells in G2/M phase was increased significantly (P<0.05). The results of immunocytochemistry and flow cytometry showed that the expressions of Akt and Raf-1 proteins in SKOV3 cells induced by geldanamycin were significantly down-regulated in a dose-dependent manner (P<0.05). Conclusion: Geldanamycin can inhibit the cell proliferation and induce the apoptosis of human ovarian cancer SKOV3 cells in vitro , which may be associated with downregulation of the expressions of Akt and Raf-1 proteins. 2. Inhibitory effects of manumycin combined with cisplatin on the proliferation of human ovarian cancer cell line 3AO in vitro and in vivo

Objective: To investigate the inhibition effects of manumycin combined with cisplatin (DDP) on the proliferation of human ovarian cancer cell line 3AO and the growth of transplantation tumors in nude mice, and to analyze its possible mechanisms. Methods: MTT colorimetric assay was performed to evaluate the cytotoxic effect of manumycin combined with DDP on 3AO cells. The model of nude mice bearing ovarian transplantation tumor was established. The expressions of survivin, NF-κB, vascular endothelial growth factor (VEGF) and caspase-3 proteins were detected by immunohistochemical method. The cell cycle distribution and apoptosis rate were examined by flow cytometry. Results: The proliferation of 3AO cells was inhibited by manumycin in a time- and dose-dependent manner (P <0.05). The manumycin of 10, 20, 40 or 60 μmol/L could strengthen the cytotoxic effect of DDP on 3AO cells. The tumor size in nude mice of each drug-treated group was significantly smaller than that of the control (untreated) group (P <0.05). Compared with the DDP alone and the manumycin alone group, the expressions of survivin, NF-κB and VEGF proteins were significantly down-regulated while the expression of caspase-3 protein was significantly up-regulated (P <0.05) in the combination (manumycin plus DDP) group (P <0.05). The apoptosis rates were increased gradually in the control group, manumycin group, DDP group and the combination group. In each drug-treated group, the number of cells in G0/ G 1 phase was decreased significantly, while which in G2/M phase was increased significantly (P <0.05), as compared with the control group. The number of cells in S phase did not show any changes in each group. Conclusion: The combination of manumycin and DDP shows an enhanced ability to inhibit the proliferation of human ovarian cancer cells 3AO and increase their apoptosis. 3.Minimally Invasive Percutaneous Nephrolithotomy for the Treatment of Impacted Upper Ureteral Calculi: A Report of 49 Cases

Objective To investigate the efficacy of minimally invasive percutaneous nephrolithotomy(MPCNL) for impacted upper ureteric calculi.Methods Retro-urethral catheterization was conducted in the diseased ureter under cyctoscope.After a channel from the skin surface into the middle calyx was established under the guidance of X-ray or B ultrasonography,pneumatic lithotripsy was conducted to break up stones under ureteroscopy.Results The success rate of pneumatic lithotripsy was 100%.The rate of postoperative macroscopic hematuria was 55.1%(27/49).The symptom lasted one to three days in 26 cases,and 1 case had apparent hematuria with blood clot flowing out from nephrostomy tube,which lasted 8 days.10.2%(5/49) of cases was complicated with postoperative pyrexia.The stone-free rate in one week and one month postoperatively was 93.9%(46/49) 98.0%(48/49),respectively.Follow-up observations in 49 cases for 1-12 months(mean,5 months) showed no major complications.Conclusions MPCNL for impacted upper ureteral calculi has the advantages of simple performance,less complications and satisfactory efficacy.
4.Dynamic analysis of anti-Cov antibody and SARS coronavirus in the plasma of 25 SARS patients

Objective To observe dynamic responsive regularity of specific IgM and IgG antibodies in SARS patients. Methods 145 specimens of plasma from 25 cases of clinically diagnosed SARS patients were examined in the study. ELISA was employed to detect IgM and IgG antibodies against SARS coronaviral antigens. Nested RT-PCR was used to qualitatively determine SARS coronavirus (SARS-CoV). Results 49.0% (71/145) and 54.5% (79/145) of the samples were positive for IgM and IgG antibodies, respectively. Both antibodies were found to be detected in 84.0% of the patients. The antibodies were found to be detectable from the 2nd to 4th week after the onset of disease in most patients, and there was a fendeney of sising in positive rate until the 5th week after the onset. Thereafter, the detectable rate of IgM antibody began to decline, while that of IgG antibody remained to rise. Positive rate for serum SARS-CoV was 15.8% (18/114) for all samples or 40.0% (10/25) of patients. Most virus-positive samples were those which were collected within 4 weeks after disease onset. Conclusions Anti-SARS-CoV IgM/IgG antibody and detection of virus in plasma could serve as practical diagnostic indicators for SARS. In most cases, when the serum was pasitive for antibody the serum virus positive rate would soon declined. However, concomitant existence of antibody and viral sequence in plasma was observed in a few patients.

6. The feasibility of comprehensive staging surgery via laparoscopic operation in patients with ovarian cancer and its clinical significance

Objective: To explore the feasibility and safety of comprehensive staging surgery via laparoscopic operation and its clinical significance. Methods: A retrospective analysis of medical records was conducted in 35 inpatients diagnosed of ovarian cancer (stage I, n = 29; stage II, n = 4; stage III, n = 2) in Department of Gynecology of Fourth Hospital of Hebei Medical University between January 2010 and January 2013. Of the 35 patients, 15 underwent comprehensive staging operation via laparoscopic operation and 20 via open operation. The general conditions during and after operation and the postoperative survival were observed and compared between the two groups. Results: All operations completed successfully in 15 patients in the laparoscopic operation group, and no perioperative complications were observed. The intraoperative blood loss and the rate of postoperative analgesia in the laparoscopic operation group were both less than those in the open operation group (P < 0.05), but the differences in duration of operation, number of abdominal and pelvic lymph nodes resected, recovery time of gastrointestinal function, rate of postoperative complications, rate of postoperative recurrence, rate of increase in pathological stage, hospitalization expenses, duration of hospital stay after operation and the rate of chemotherapy after operation were not statistically significant between the two groups (P > 0.05). There was also no significant difference in the cumulative survival rate between the two groups (P > 0.05). Conclusion: Comprehensive staging operation for ovarian cancer especially for early-stage cancer is feasible and safe. It has the merits of smaller wound, less blood loss during the operation and more rapid recovery. Copyright© 2014 by TUMOR.
7.Candida parapsilosis-c aused candidiasis characterized by verrucous nodules: two case reports

Objective To report two cases of Candida parapsilosis-caused candidiasis characterized by verrucous nodules and masses,and to assess their clinical features,diagnosis and treatment.Methods A medical history including medication history and therapeutic response was carefully collected from two male patients.Physical examination was carried out with a focus on skin lesions.Diagnosis was made according to medical history as well as physical,mycological and histopathological examination findings.Antifungal agents were given at a high enough dose in time.After lesions improved,the doses of antifungal agents were tapered gradually,and drugs were withdrawn until patients completely healed.Compound preparations containing both antifungal agents and glucocorticoids were also topically applied in the early period of treatment.Results Both patients were diagnosed as Candida parapsilosis-caused candidiasis characterized by verrucous nodules and masses,and cured after 100-day supporting treatment and antifungal treatment with oral itraconazole,etc.Conclusions Candida parapsilosis-caused candidiasis should be managed with combination therapy mainly including antifungal agents.To achieve a satisfying efficacy,antifungal agents should be given early at a high enough dose for a long enough period.
8.Nursing cooperation for patients with severe aortic stenosis receiving transcatheter aortic valve replacement using balloon-expandable aortic valve stent

Objective To discuss the nursing cooperation for patients with severe aortic stenosis who are receiving transcatheter aortic valve replacement using balloon-expandable aortic valve stent. Methods Percutaneous interventional transcatheter aortic valve replacement was employed in 9 patients with severe aortic stenosis. Active preoperative preparation, sufficient psychological care, close observation of vital signs and effective surgery cooperation during the operation were carried out. The clinical results were analyzed. Results Successful operation was accomplished in all the nine patients. After the surgery, all patients were in good condition, and no nursing-related complications occurred. Conclusion Correct understanding of the cooperation with the procedure of transcatheter aortic valve replacement using balloon-expandable aortic valve stent, excellent preoperative psychological care and active preoperative preparation, etc. are the most important things to ensure a successful surgery.
